酶法转化液中L-瓜氨酸的分光光度法测定   总被引:8,自引:0,他引:8  
根据L-瓜氨酸在强酸性溶液中与二乙酰一肟的专一性显色反应及反应复合物在490nm处吸光度与L-瓜氨酸浓度呈线性关系的特点,通过考察酶转化液中转化底物L-精氨酸、副产物L-鸟氨酸、增效剂、磷酸盐缓冲溶液、细胞自溶释放物以及显色剂用量、显色反应时间等条件对L-瓜氨酸测定的影响,建立了一种准确、简便、灵敏的L-精氨酸酶法转化液中L-瓜氨酸的光度分析方法。该方法的相关系数r=0.999,样品回收率达99.58%。  相似文献

A short and convenient synthesis of 3'- and 5'-O-levulinyl-2'-deoxynucleosides has been developed from the corresponding 3',5'-di-O-levulinyl derivatives by regioselective enzymatic hydrolysis, avoiding several tedious chemical protection/deprotection steps. Thus, Candida antartica lipase B (CAL-B) was found to selectively hydrolyze the 5'-levulinate esters, furnishing 3'-O-levulinyl-2'-deoxynucleosides 3 in >80% isolated yields. On the other hand, immobilized Pseudomonas cepacia lipase (PSL-C) and Candida antarctica lipase A (CAL-A) exhibit the opposite selectivity toward the hydrolysis at the 3'-position, affording 5'-O-levulinyl derivatives 4 in >70% yields. A similar hydrolysis procedure was successfully extended to the synthesis of 3'- and 5'-O-levulinyl-protected 2'-O-alkylribonucleosides 7 and 8. This work demonstrates for the first time application of commercial CAL-B and PSL-C toward regioselective hydrolysis of levulinyl esters with excellent selectivity and yields. It is noteworthy that protected cytidine and adenosine base derivatives were not adequate substrates for the enzymatic hydrolysis with CAL-B, whereas PSL-C was able to accommodate protected bases during selective hydrolysis. In addition, we report an improved synthesis of dilevulinyl esters using a polymer-bound carbodiimide as a replacement for dicyclohexylcarbodiimide (DCC), thus considerably simplifying the workup for esterification reactions.  相似文献

The suspending behaviors of multiple-wall carbon nanotubes (MWNTs), including pristine MWNTs (p-MWNTs) and acid-mixture-treated MWNTs (MWNTCOOH), stabilized by cationic single-chain surfactant, dodecyltrimethylammonium bromide (DTAB), and cationic gemini surfactant hexyl-alpha,beta-bis(dodecyldimethylammonium bromide) (C 12C 6C 12Br 2) were studied systematically. The surfactant structure influences the suspendability of MWNTs dramatically as well as the surfactant adsorption behavior on the nanotubes. Although both the surfactants can disperse the MWNTs effectively, they actually show different stabilizing ability. DTAB is not capable of stabilizing these two MWNTs below critical micelle concentration (CMC). However, C 12C 6C 12Br 2 can suspend both the nanotubes effectively even well below its CMC. Moreover, the adsorption of these two surfactants reaches equilibrium at twice the CMC with the original MWNT concentration of 2 mg/mL, 2 mM for C 12C 6C 12Br 2, and 30 mM for DTAB. After the adsorption equilibrium, the maximum amounts of the two suspended MWNTs in C 12C 6C 12Br 2 solution are about twice as much as those in DTAB solution. The strong hydrophobic interaction among the C 12C 6C 12Br 2 molecules and between the C 12C 6C 12Br 2 molecules and the nanotubes as well as the high charge capacity of C 12C 6C 12Br 2 lead to its much stronger adsorption ability on the MWNTs and result in its superior stabilizing ability for the MWNTs in aqueous phase. The gemini surfactant provides a possibility to effectively stabilize the MWNTs in aqueous solutions even at very low surfactant concentration well below its CMC.  相似文献

The nature and properties of the low-lying singlet and triplet valence excited states of 2,2':5',2'-terthiophene (terthiophene) and 2,2':5',2':5',2'-quaterthiophene (tetrathiophene) are discussed on the basis of high-level ab initio computations. The spectroscopic features determined experimentally for short alpha-oligothiophenes are rationalised on theoretical grounds. Special attention is devoted to the nonradiative decay process through intersystem crossing (ISC) from the singlet to the triplet manifold, which is known to be relatively less efficient in tetrathiophene. Along the geometry relaxation of the S1 state of terthiophene, the S1 and T2 states become degenerate, which leads to a favourable situation for the occurrence of ISC. The parallel process is expected to be less favoured in tetrathiophene because of the less efficient spin-orbit coupling and the increase of the S1-T2 energy gap.  相似文献

Lithium sodium aluminium fluoride was obtained as a white powder by melting a stoichiometric mixture of AlF3, NaF and LiF at 1223 K, and then cooling to 923 K and sintering at this temperature for 4 h. The ab initio crystal structure determination was carried out using X‐ray powder diffraction techniques. The monoclinic structure of LiNa2AlF6 can be related to cubic elpasolite. The Li and Al atoms lie on inversion centres. The main octahedral AlF6 structural elements are not deformed, but are rotated slightly with respect to the unit‐cell axes. The Li atoms have octahedral coordinations, whereas the Na atoms have cubo‐octahedral coordinations. The Na coordination polyhedron is distorted in comparison with that of elpasolite.  相似文献

The title compound, cadmium bismuth dioxide chloride, CdBiO2Cl, was obtained as a white powder by reaction of solid BiOCl with CdO at 973 K. Ab initio crystal structure determination was carried out using X-ray powder diffraction techniques, including direct methods for atom location and Rietveld fitting for the final refinement. Being monoclinic, the crystal structure can be related to tetragonal Sillen layered phases. The main structural elements present are CdBiO2+ pleated metal–oxy­gen layers alternating with Cl layers along the c axis, whereas along the b axis, all atoms are on mirror planes. The formation of a strong Cd—Cl bond draws the layers together, causing layer deformation and a monoclinic distortion in the layer arrangement.  相似文献

In methanol or chloroform/methanol solutions, reactions of Cltpy or MeOtpy (Rtpy = 4'-R-2,2':6',2'-terpyridine) with CoX(2)·xH(2)O (X(-) = Cl(-), [OAc](-), [NO(3)](-) or [BF(4)](-)) result in the formation of equilibrium mixtures of [Co(Rtpy)(2)](2+) and [Co(Rtpy)X(2)]. A study of the solution speciation has been carried out using (1)H NMR spectroscopy, aided by the dispersion of signals in the paramagnetically shifted spectra; on going from a low- to high-spin cobalt(II) complex, proton H(6) of the tpy ligand undergoes a significant shift to higher frequency. For R = Cl and X(-) = [OAc](-), increasing the amount of CD(3)OD in the CD(3)OD/CDCl(3) solvent mixture affects both the relative proportions of [Co(Cltpy)(2)](2+) and [Co(Cltpy)(OAc)(2)] and the chemical shifts of the (1)H NMR resonances arising from [Co(Cltpy)(OAc)(2)]. When the solvent is essentially CDCl(3), the favoured species is [Co(Cltpy)(OAc)(2)]. For the 4'-methoxy-2,2':6',2'-terpyridine, the speciation of mono- and bis(terpyridine)cobalt(II) complexes depends upon the anion, solvent and ligand:Co(2+) ion ratio. The (1)H NMR spectrum of [Co(MeOtpy)(2)](2+) is virtually independent of anion and solvent. In contrast, the signals arising from [Co(MeOtpy)X(2)] depend on the anion and solvent. In the case of X(-) = [BF(4)](-), we propose that the mono(tpy) complex formed in solution is [Co(MeOtpy)L(n)](2+) (L = H(2)O or solvent, n = 1-3). The formation of mono(tpy) species has been confirmed by the solid state structures of [Co(Cltpy)(OAc-O)(OAc-O,O')], [Co(MeOtpy)(OAc-O)(OAc-O,O')], [Co(MeOtpy)(NO(3)-O)(2)(OH(2))] and [Co(MeOtpy)Cl(2)]. The single crystal structure of the cobalt(III) complex [Co(Cltpy)Cl(3)]·CHCl(3) is also reported.  相似文献

The homoleptic magnesium carbamato complex Mg6(Et2NCO2)12, 1 (Et2NCO2- = diethylcarbamato anion), was prepared by the reaction of dibutylmagnesium with diethylamine, followed by carboxylation using gaseous carbon dioxide. Crystallographic characterization demonstrated that 1 has the standard M6(R2NCO2)12 structure and is a double helix of MgO(x)(x = 5, 6) coordination polyhedra with Delta or Lambda stereochemistry arising from the configuration around the six-coordinate Mg2+ cations. It crystallized in the orthorhombic space group Ccca with two molecules of Delta1 and two of Lambda1 per unit cell (a = 21.548 A, b = 25.094 A, c = 15.4485(11) A, alpha = beta = gamma = 90 degrees ). Extensive solution characterization of 1 by 1-dimensional proton and 13C NMR spectroscopy and by two-dimensional 1H-[13C] NMR correlation techniques verified that the helical structure is maintained in solution. Moreover, these measurements indicated that the intramolecular dynamics of 1 relating to motions of the ethyl groups was substantially hindered in solution. Correlation of the crystallographic and NMR structural studies indicated that this arises from a combination of hindered rotation about the carbamato C-N bond and efficient packing of the ethyl groups around the Mg6O24 core. The result is an inverted-micelle-like structure for 1 in which the hydrophobic ethyl groups form a sheath largely restricting access to the hydrophilic Mg6O24 core.  相似文献

栝楼蛋白(Trichobitacin)是从栝楼(Trichosanthes kirilowiiMaxim, Cucurbitaceae)中新发现的核糖体失活蛋白, 分子量为27,228; pI为9.6。应用基质辅助的激光解析飞行时间质谱(MALDI-TOF-MS)和快原子轰击质谱法(FAB-MS)分别测定胰蛋白酶酶解栝楼蛋白和天花粉蛋白(Trichosanthin)的混合肽质谱, 通过比较发现了一些分子量相同的肽。由于这两种蛋白质都来源于栝楼块根, 同源性比较强, 所以这些肽序列在两种蛋白质中基本一样; 再结合蛋白N-端自动顺序仪测定栝楼蛋白N-端的结果, 确定了栝楼蛋白N-端38个氨基酸的顺序, 栝楼蛋白经胰蛋白酶酶解后所得肽段用HPLC分离纯化, 再用蛋白质自动顺序仪, DABITC/PITC双偶合手工法和质谱法共确定了栝楼蛋白N-端, C-端等100多个氨基酸残基的序列。  相似文献

To determine the surface coverage of exfoliated carbon nanotubes by surfactant molecules, we propose four experimental methods based on thermogravimetric analysis, UV-vis spectroscopy, surface tension measurements, and a variant of Maron's titration. We apply all four methods to aqueous mixtures of carbon nanotubes and the surfactant sodium dodecyl sulfate and consistently find a surface coverage of between 1.5 and 2 g of surfactant per gram of carbon nanotubes. This corresponds to slightly more than two SDS molecules per square nanometer, which is comparable to the known maximum packing density of SDS at the air-water interface.  相似文献

A new monometallic complex [Co(phen)2tpphz]3+ (where tpphz is tetrapyrido [3,2-a: 2',3'-c: 3″,2″-h: 2'″,3'″-j] phenazine) was synthesized by the reaction of 5,6-diamino-1,10-phenanthroline with [Co(phen)2(phendione)]3+. It was characterized by elemental analysis, molar conductivity, IR, 1H NMR, ultraviolet and fluorescence spectroscopy. The interaction of the complex with DNA was also investigated. The complex shows the absorption hypochromicity, fluorescence enhancement, the specific viscosity increased when bound to calf thymus DNA. The cyclic voltammetry (CV) measurement showed a change in peak current with the addition of DNA. All the results provide the support for the intercalative binding mode of the mononuclear complex.  相似文献

Using the method of ab initio molecular dynamics, we examine the structural fluctuation and the low-frequency dynamics of beta-ribofuranose puckering in aqueous solution. Our analysis suggests that the distance between the anomeric and hydroxymethyl oxygens is a simple relevant geometrical parameter that dynamically correlates with the phase angle in the north region. The time-frequency analysis using the Hilbert-Huang transform also confirms the correlation, and most of the instantaneous frequencies for the phase angle and the above distance are found to be concentrated on the region below about 100 cm(-1). Our analysis of ab initio molecular dynamics trajectories suggests that the molecular origin of the hydration effects on the low-frequency dynamics of beta-ribofuranose puckering is closely related to this correlation and thus primarily attributed to the relatively local interactions among the anomeric and hydroxymethyl oxygens and the surrounding water molecules near them. Additionally, we discuss the difference in the low-frequency dynamics of beta-ribofuranose puckering between two hydroxymethyl rotamers.  相似文献
