A series of novel 2,4-diaminopyrimidine-modified compounds was designed and synthesized. Compound 14 showed micromolar dual inhibitory effect on both FP-2 and PfDHFR, and potential inhibition to the proliferation of P. falciparum 3D7 strain and chloroquine-resistant P. falciparum Dd2 strain.  相似文献

Thiosemicarbazones of 2-amino-5-chlorobenzophenone and 3-aminobenzophenone(L1-L4) have been synthesized and their Cu(Ⅱ) complexes(1-4) were afforded via coordination with cupric chloride.All these compounds were characterized by UV-vis and IR spectroscopy together with CHN elemental analysis.NMR spectroscopy was also applied to characterize the ligands.In vitro chohnesterase inhibitory assays for the complexes(1-4) showed IC50 values less than 10μmol/L,with complex 1 exhibiting the most activity,IC50=2.15μmol/L and 2.16μmol/L for AChE and BuChE,respectively. Molecular modeling simulation revealed the binding interaction template for complex 1 with the AChE and BuChE receptors.In DPPH assay,the complexes also showed more in vitro antioxidant activities in comparison to their parent ligands.  相似文献

The chemical investigation of the antimalarial plant Phyllanthus fraternus G. L. Webster (Phyllanthaceae) resulted in the discovery of the Securinega alkaloid (+)-allonorsecurinine (1), previously reported as a synthetic compound, together with the known ent-norsecurinine (2), nirurine (3), bubbialine (4), epibubbialine (5) and the lignan phyllanthin (6). The structure and absolute configuration of the new compound were elucidated on the basis of extensive spectroscopic analysis, optical rotation, and GIAO NMR shift calculation followed by CP3 analysis. The antiplasmodial activity of these compounds was evaluated against chloroquine-resistant (W2) and -sensitive (3D7) strains of Plasmodium falciparum. Among them, ent-norsecurinine (2) and (+)-allonorsecurinine (1) showed the strongest activity (IC50: 1.14 ± 0.32 and 2.57 ± 0.53 µM) respectively, against W2 but one of the weakest against 3D7.  相似文献

A series of flavonoids were isolated from Dodonaea Viscosa and tested for inhibition of human neutrophil elastase (HNE), enzyme involved in inflammatory disorders. Isolated compounds were identified as a novel flavonol (1) along with eight known flavonoids (29). Novel flavonol, visconata (1) has a very rare skeleton having odd numbered long chain (C19) fatty acid, which was completely identified by mass fragmentation and 2D NMR analysis. All compounds (19) inhibited HNE in dose dependent manner with IC50s ranging between 2.4 and 150 μM. Visconata (1) emerged to be the most potent compound with 2.4 μM of IC50. In kinetic studies, compound (1) was observed to be reversible, noncompetitive inhibitor having Ki = 1.8 μM, whereas other flavonoids (29) displayed mixed type inhibition.  相似文献

Eight new water-soluble amino acid derivatives of chlorin p6 ethers 6a-h were designed and synthesized using purpurin-18 (2) as key intermediate. All target compounds exhibited better phototoxicity than talaporfin and the most phototoxic compound 6d showed IC50 values of 0.20 μmol/L against A549 cell and 0.41 μmol/L against B16-F10 cell, which represented 31- and 24-fold increase of PDT antitumor efficacy compared to talaporfin.  相似文献

Bistachybotrysins D and E (1 and 2), one stereoisomeric pair of phenylspirodrimane dimers, were isolated from Stachybotrys chartarum CGMCC 3.5365. They represent novel phenylspirodrimane dimers with a central [6,5,6]-tricyclic carbon scaffold containing a cyclopentanone core. The structures of 1 and 2 were elucidated through extensive spectroscopic data analysis, and their absolute configurations were characterized by calculated electronic circular dichroism (ECD). Compounds 1 and 2 displayed potent cytotoxic activity against the four human tumor cell lines HCT116, BGC823, Daoy and HepG2 with IC_ (50)values ranging from 6.7 μmol/L to 11.6 μmol/L. Furthermore, a plausible biogenetic pathway for 1 and 2 is proposed.  相似文献

Twelve-membered cyclic cis- and trans-β-lactams 1b and 2b and the corresponding cyclic cis- and trans-β-amino acid enantiomers, 1a, 1c and 2a, 2c were prepared through the CAL-B-catalysed enantioselective ring cleavage of racemic cis-13-azabicyclo[10.2.0]tetradecan-14-one, (±)-1, and trans-13-azabicyclo[10.2.0]tetradecan-14-one, (±)-2. High enantioselectivities (E >200) were observed for the ring opening of both the cis- and trans-β-lactams when the Lipolase-catalysed reactions were performed with 0.5 equiv of H2O in i-Pr2O at 70 °C. The resolved β-lactams 1b and 2b (yield ⩾47%) and β-amino acids 1a and 2a (yield ⩾32%) could be easily separated.  相似文献

Two xanthanolide-type dimmers, namely Mogolide D (4) and Mogolide E (5) are isolated from the aerial parts of Xanthium mogolium Kitag plant collected from Heilongjiang province, China. The dimmers’ relative structures are elucidated on the basis of HRESIMS, IR spectrum and NMR spectroscopy. The absolute configurations of them are assigned with ECD spectra. The biosynthetic pathway of the two dimmers is also proposed. Besides, Mogolide D (4) and Mogolide E (5) are tested against MDA-MB-231 breast cancer cell line with IC50 8.46 μM and 19.20 μM, respectively.  相似文献

Mass spectrometry-guided isolation of the lipophilic extract of Trichodesmium bloom material led to the isolation and structure characterization of a new thiazole-containing di-chlorinated polyketide (1). The structure of 1 was deduced using 1D and 2D NMR analysis, high-resolution mass spectrometry analysis and complementary spectroscopic procedures. Trichothiazole A possesses interesting structural features, such as a terminal alkyne, two vinyl chlorides and a 2,4-disubstituted thiazole. Trichothiazole A showed moderate cytotoxicity to Neuro-2 A cells (EC50: 13.3 ± 1.1 μM).  相似文献

Oxidative addition of H–R (H--Ph and H2) to trans-Ir(--Ph)(CO)(PPh3)2 (2) gives the initial products, cis, cis-Ir(H)(--Ph)2(CO)(PPh3)2 (3a) and cis, cis-Ir(H)2(--Ph)(CO)(PPh3)2 (3b), respectively. Both cis-bis(PPh3) complexes, 3a and 3b undergo isomerization to give the trans-bis(PPh3) complexes, trans, trans-Ir(H)(--Ph)2(CO)(PPh3)2 (4a) and cis, trans-Ir(H)2(--Ph)(CO)(PPh3)2 (4b). The isomerization, 3b4b is first order with respect to 3b with k1=6.37×10−4 s−1 at 25°C under N2 in CDCl3. The reaction rate (k1) seems independent of the concentration of H2. A large negative entropy of activation (ΔS=−24.9±5.7 cal deg−1 mol−1) and a relatively small enthalpy of activation (ΔH=14.5±3.3 kcal mol−1) were obtained in the temperature range 15∼35°C for the isomerization, 3b4b under 1 atm of H2.  相似文献

Four new natural products, including three arylbenzofurans named heterophyllenes A-C (13), and one stilbene named heterophyllene D (4), together with twenty-one known compounds were isolated from the twigs of Artocarpus heterophyllus and their structures elucidated by spectroscopic methods, mainly 1D and 2D NMR spectroscopy. The cytotoxic activity of selected compounds against KB, MCF-7 and NCI-H187 cell lines was evaluated. Heterophyllene C (3) exhibited cytotoxicity against the MCF-7 cell line with an IC50 value of 12.56 μM. Additionally, the known compounds norartocarpin and artocarpin showed cytotoxic activity against MCF-7 and KB cell lines with IC50 values of 10.04 and 13.57 μM, respectively. Both compounds also displayed cytotoxicity against the NCI-H187 cell line with values of 14.78 and 14.21 μM, respectively.  相似文献

Two acridones, paratrimerins C (1) and D (2), and two coumarins, paratrimerins E (3) and F (4), were isolated from the CHCl3 and EtOAc extracts of Paramignya trimera (Rutaceae), together with twelve known compounds (516). Their structures were elucidated on the basis of spectroscopic data. All isolated compounds possessed significant α-glucosidase inhibitory activity in a concentration-dependent manner, and showed more potent inhibitory activity, with IC50 values ranging from 14.6 to 112.2 μM, than the positive control acarbose (IC50, 214.5 μM). The biosynthesis of the isolated coumarins and acridones was proposed.  相似文献

In an effort to develop novel antiamoebic scaffolds having better efficacy than the standard drug metronidazole (IC50 = 1.80 μM) used against Entamoeba histolytica, quinazolin-4(3H)-one Schiff base conjugates were synthesized and evaluated against HM1: IMSS strain of E. histolytica. Out of the thirteen compounds (S2-S14), six compounds (S2, S3, S4, S5, S6 and S11) were found to be better inhibitors than metronidazole and showed low cytotoxicity on HeLa cells, a cervical cancer cell line. The structure of intermediate compound S1 was confirmed by crystal structure studies.  相似文献

A series of novel quinazolin-4(3H)-one derivatives (6a6y) containing a 1,2,4-triazolo[3,4-b][1,3,4]thiadiazole moiety were designed and synthesized, and their structures were fully characterized by 1H NMR, 13C NMR, HRMS and IR spectra. Among them, the structure of compound 6u was unambiguously confirmed via single crystal X-ray diffraction analysis. The obtained bioassay results showed that compounds 6h, 6k, 6l and 6y had the EC50 (half-maximal effective concentration) values of 34.8, 28.2, 41.5 and 42.5 μg/mL against the phytopathogenic bacterium Xanthomonas oryzae pv. oryzae (Xoo), respectively, which were significantly better than commercial bactericide Bismerthiazol (EC50 = 95.8 μg/mL). Additionally, compounds 6a and 6b exhibited the strong inhibition activity against the pathogen Xanthomonas axonopodis pv. citri (Xac).  相似文献

Insect chitinase and N-acetyl-β-d-hexosaminidases (Hex) are potential targets for developing new pesticides. Here, a series of thiazolylhydrazones I (with substituted group R1 at N3) and II (with substituted group R1 at N2) were designed, synthesised and evaluated as competitive inhibitors of OfHex1 and OfChi-h, from the agricultural pest Ostrinia furnacalis. Derivatives I-3d and II-3d, with phenoxyethyl group at R1, demonstrated the best inhibitory activities against OfHex1 and OfChi-h. Molecular docking analysis indicated that the branched conformation compound II-3d (Ki = 1.5 μmol/L) formed more hydrogen bonds with OfHex1 than the stretched conformation compound I-3d (Ki = 5.9 μmol/L). The differences in compounds’ binding conformations with OfChi-h explained differences in inhibitory activity of compounds I-3d (Ki = 1.9 μmol/L) and II-3d (Ki = 4.1 μmol/L). This work suggests a novel scaffold for developing specific Hex and Chi-h inhibitors.  相似文献

New luminescent mononuclear and dinuclear copper(II) (S = 1/2) complexes [Cu(HL)(H2O)2](ClO4)2 (1a) and [Cu2(HL)2(μ-SO4)2]·2H2O (1b) were synthesized with the acyclic tridentate pyridine-2-carboxaldehyde-2-pyridylhydrazone ligand, HL (1). Interestingly, the mononuclear complex 1a can be converted into the disulfate bridged dimeric copper(II) complex 1b by passing freshly prepared SO2 through the basic medium. On excitation at 290 nm, the ligand fluoresces at 364 nm due to an intraligand 1(π–π1) transition. Upon complexation with copper(II), the emission peak is slightly blue shifted (356 nm, F/F0 0.76 for 1a and 354 nm, F/F0 0.89 for 1b) with a little quenching in the emission intensity. The association constants (Kass (5.06 ± 0.004) × 104 for 1a and Kass (5.46 ± 0.006) × 104 for 1b at 298 K) and the thermodynamic parameters have been determined by UV–Vis spectroscopy. The molecular structure of the complex 1b (Cu?Cu 4.456 Å) has been determined by single crystal X-ray diffraction studies. The complex 1b exhibits a strong interaction towards DNA as revealed from the Kb (intrinsic binding constant) 6.3 × 104 M?1 and Ksv (Stern–Volmer quenching constant) 2.93 values.  相似文献

Four complexes of M(NO3)2(4NOPy-OMe)2, (4NOPy-OMe = 4-(N-tert-butyloxylamino)-2-(methoxymethylenyl)pyridine, and M = MnII, 1; CoII, 2; NiII, 3; CuII, 4), were prepared and fully characterized. X-ray single crystal analysis reveals that four complexes are isostructural. The molecular structures are distorted octahedral in which the methoxy oxygen atoms coordinate to the metal ion by trans-configuration while the pyridyl nitrogen atoms and the nitrate oxygen atoms coordinate by cis-configuration. The magnetic properties of all complexes were investigated by SQUID magneto/susceptometry. Temperature dependence of the molar magnetic susceptibilities in the temperature range of 2–300 K indicated that the magnetic coupling between aminoxyl radicals and metal ion was antiferromagnetic in the complex 1 and were ferromagnetic in the complexes 24. The quantitative analysis based on the spin Hamiltonian, H = −2J(S1SM + SMS2) yielded the best fit as J/kB = −13.4 ± 0.1 K, g = 1.94 ± 0.002, and θ = −0.78 ± 0.02 K for the complex 1, J/kB = 48.7 ± 2.1 K, g = 2.07 ± 0.02, and θ = −2.83 ± 0.41 K for the complex 3 (the data in the temperature range 300–50 K were used), and J/kB = 57.0 ± 1.2 K, g = 2.002 ± 0.004, and θ = −9.8 ± 0.1 K for the complex 4.  相似文献

Nodupetide (1), a new cyclodepsipeptide unique in its incorporation of a 3-hydroxy-4-methylhexanoic acid (HMHA) derived motif, was discovered from Nodulisporium sp. IFB-A163, a fungus residing in the insect (Riptortus pedestris) gut. The nodupetide structure was elucidated by its MS/MS and 2D NMR spectra, and its absolute configuration by the X-ray crystallography and modified Marfey’s method. Nodupetide is insecticidal against rice brown planthopper (Nilaparvata lugens) with an LD50 value of 70 ng/larva, and inhibitory towards the drug-resistant human pathogenic bacterium Pseudomonas aeruginosa with its MIC value (5.0 μM) comparable to that (3.2 μM) of ciprofloxacin, a prescribed antibacterial agent co-assayed equally.  相似文献
