The electrochemiluminescence(ECL) behavior of N-(4-aminobutyl)-N-ethylisoluminol(ABEI)-functionalized graphene composite(ABEI-GC) modified on an indium tin oxide(ITO) electrode was studied. ABEI-GC exhibited excellent ECL activity. On this basis, a label-free ECL immunosensor was developed for the sensitive detection of human immunoglobulin G(h Ig G) by using ABEI-GC as the ECL nano-interface via a layer-by-layer assembly technique. ABEI-GC was first assembled onto an ITO electrode. Positively charged chitosan was then electrostatically adsorbed to the modified electrode. Finally, negatively charged antibody-coated gold nanoparticles were attached to the surface to form the ECL immunosensor. In the presence of h Ig G, h Ig G was captured by its antibody. In addition, an ECL signal was detected in the presence of H2O2 when a double potential was applied. The ECL immunosensor for the determination of h Ig G showed a linear range of 1.0×10-13–1.0×10-8 g/mL with a detection limit of 5.0×10-14 g/m L. This immunosensor has high sensitivity, wide linearity and good reproducibility. The superior sensitivity of the proposed ECL immunoassay mainly derives from the incorporation of ABEI-GC, which not only improves the ECL intensity, response speed, and stability, but also provides a large specific surface for high levels of protein loading. This work reveals that ABEI-GC is good nano-interface for the construction of ECL biosensors. Our strategy is promising for protein detection and may open up a new avenue for ultrasensitive label-free immunoassays.  相似文献

A simple and feasible potentiometric immunosensing platform based on enzymatic biocatalytic precipitation technique was designed for the sensitive detection of thyroid-stimulating hormone(TSH;a typical kind of biomarkers for thyroid carcinoma),using horseradish peroxidase(HRP)-loaded liposome for the signal amplification.To construct such an assay system,a sandwich-type immunoreaction was readily carried out on monoclonal anti-TSH capture antibody-coated electrode by using polyclonal antiTSH secondary antibody-conjugated HRP-loaded liposome.Accompanying the formation of sandwichtype immunocomplex,the carried liposome was lysed through the added Triton X-100 to release the entrapped HRP molecules,which catalyzed the oxidation of 4-chloro-1-naphthol to produce an insoluble and uncharged organic precipitation on the electrode surface,thereby causing the change of the local electrical potential.Two labeling protocols with and without the liposome were investigated for detection of target TSH,improved analytical features were achieved with HRP-entrapped liposome.Under optimal conditions,the potentiometric immunosensor had good responses for TSH detection within the linear range of 0.01-30 p IU/mL at a detection limit of 0.0067 p IU/mL.Good reproducibility,high specificity and long-time stability were acquired during the assay procedure.Importantly,a wellmatched accuracy between the potentiometric immunosensor and commercial human TSH ELISA kit was gave for the analysis of human serum samples.  相似文献

A piezoelectric immunosensor has been developed for the detection of complement C4. Anti-C4 antibody was immobilized onto the gold electrodes of a 9 MHz AT-cut piezoelectric crystal. The coated crystal with the physical adsorption method to immobilize antibody showed the better results than the polyethyleneimine adhesion, glutaraldehyde cross-linking method with respect to sensitivity and reproducibility. The antibody-bound crystal with the physical adsorption method was successfully used for the detection of human complement C4 in the concentration range of 0.1-10 μg/mL for 40 min incubation time. The immunosensor system had good selectivity, and other materials in human serum did not interfere the detection remarkably. The crystal could be regenerated nearly 15 times when the bound materials on the crystal surface were eluted by strong acid and strong alkali solutions and subsequently cleaned in a ultrasonic cleaner.  相似文献

A novel, direct noncompetitive flow injection enzyme immunoassay for α-fetoprotein (AFP) was developed by enhanced chemiluminescence detection. The method was based on off-line incubation of AFP and horseradish peroxidase (HRP)-labeled anti-AFP, and then trapping of the unbound enzyme conjugate by an immunoaffinity column filled with AFP-modified Sepharose. The immunocomplex formed in incubation passed through the column and then was directly detected by a postcolumn chemiluminescence technique. The optimal conditions for the immunoassay procedure and chemiluminescence detection were established. At a 1:10 dilution of enzyme conjugate solution, the linear range for chemiluminescence detection of AFP was from 2.0 to 75 ng/mL with a correlation coefficient of 0.993 and a coefficient of variation of 2.67% at 30 ng/mL. The detection limit was 0.5 ng/mL. This method was flexible, sensitive, and rapid. The immunoaffinity column of 200 μL could be repeatedly used 100 times without a single decrease. The whole assay time including the preincubation step was only 30 min for one sample.  相似文献

In this work,a fluorescent probe(TPEBe-I)was developed for adenosine triphosphate(ATP)detection based on the synergetic effect of aggregation-induced emission and counterion displacement.TPEBe-I gave weak emission in aqueous solution due to the heavy-atom effect of counter iodide ion.However,upon the addition of ATP,the new aggregate complex(TPEBe-ATP)was formed between the cationic unit of TPEBe-I and ATP through electrostatic interactions,which not only restricted the intramolecular motion of luminogen but also eliminated the quenching effect of iodide ion.As a result,the fluorescent light-up detection for ATP was successfully achieved.Moreover,TPEBe-I exhibited high selectivity towards ATP and showed a wide linear detection region towards the logarithm of ATP concentration(5—600μmol/L)with a detection limit of 1.0μmol/L,enabling TPEBe-I as a promising probe for ATP quantitative analysis.  相似文献

The toxic organic dye contaminants in wastewater are extremely harmful to the ecosystem.Surface enhanced Raman scattering(SERS) is a technique with high sensitivity and chemical specificity which fulfills the requirements for monitoring dye contaminants in wastewater.However,as one of the common dye contaminants,methyl orange(MO) has very weak affinity to metallic surfaces and is difficult to be detected by SERS at low concentrations.Therefore,a new type of SERS substrate with Ag nanoparticle monolayer functionalized by mono-6-deoxy-6-thio-β-cyclodextrin(β-CD-SH) was prepared to efficiently capture and detect MO in wastewater with a limit of detection of 5×10-7 mol/L.The hydrophobic cavity of β-CD is responsible for the efficient trap and enrichment of MO on the Ag NPs surface,achieving a strong SERS signal of MO at low concentrations and at different pH values.This study provides new insight into designing a well-performed adsorbent for the capture and detection of organic contaminants.  相似文献

The residues of organophosphorus pesticide(OPs)on fruits and vegetables pose a threat to human health,so it is very meaningful to explore simple and fast detect methods for OPs residual.In this work,nickel ferrite/nickel oxide nanoparticles co-loaded three-dimensional reduced graphene oxide(3DRGONiFe2O4/NiO NPs),as a new low cost nanocomposite,was prepared.Based on its high performance mimetic peroxidase activity,a colorimetric method for the detection of OPs has been developed.Dichlorvos was chosen as model compounds to evaluate the detection performance.The detection linear range for dichlorvos is from 50μg/mL to 2.5×10^4μg/mL with a detection limit of 10μg/mL.Furthermore,a test paper can be developed based on the 3 DRGO-NiFe2O4/NiO NPs for visual detection of dichlorvos,and the image information of the paper sensor can be converted into digital signal and quantitative detection by a smartphone.Notably,this method can also be used to detect dichlorvos in real samples,including vegetables and fruits.Thus,the developed naked assay holds great potential in simple,inexpensive and rapid detection of OPs in fruit and vegetable samples.  相似文献

Electrochemical sensing of carcinoembryonic antigen(CEA)on a gold electrode modified by the se- quential incorporation of the mediator,thionine(Thi),and gold nanoparticles(nano-Au),through co- valent linkage and electrostatic interactions onto a self-assembled monolayer configuration is de- scribed in this paper.The enzyme,horseradish peroxidase(HRP),was employed to block the possible remaining active sites of the nano-Au monolayer,avoid the non-specific adsorption,instead of bovine serum albumin(BSA),and amplify the response of the antigen-antibody reaction.Electrochemical ex- periments indicated highly efficient electron transfer by the imbedded Thi mediator and adsorbed nano-Au.The HRP kept its activity after immobilization,and the studied electrode showed sensitive response to CEA and high stability during a long period of storage.The working range for the system was 2.5 to 80.0 ng/mL with a detection limit of 0.90 ng/mL.The model membrane system in this work is a potential biosensor for mimicking the other immunosensor and enzyme sensor.  相似文献

The application of a new fluorogenic probe-based PCR assay (PCR duplex scorpion primer assay) to the detection of Hepatitis B virus (HBV) DNA in human sera was described. Duplex scorpion primer is a modified variant of duplex Amplifluor, and the incorporation of a PCR stopper between probe and primer sequences improve the detection specificity and sensitivity. Combined with PCR amplification, this probe can give unambiguous positive results for the reactions initiated with more than 20 HBV molecules. In addition, the particular unimolecular probing mechanism of this probe makes the use of short target-specific probe sequence possible, which will render this probe applicable in some specific systems.  相似文献

A novel sensing chip was designed for MALDI-MS quantitation of acid phosphatase(ACP).The ACP sensing chip was constructed through non-covalent interaction of streptavidin and biotin for the assembly of biotinylated peptide substrate on biotinylated polyethylene-glycol(PEG)modified indium-tin oxide(ITO)slide.In the presence of ACP,the peptide substrate was dephosphorylated under acidic condition to generate a new mass signal.The quantitative assay of ACP was achieved with the mass signal ratio of product to the sum of product and left peptide substrate.Under optimal detection conditions,the ratio was linearly correlated with the concentration of ACP in the range of 0.05–12 g/L with a detection limit(LOD)of 0.04 g/L.The designed ACP sensing chip has been used to analyze ACP in complex clinical samples,which exhibited high selectivity,good repeatability,and admirably anti-interference ability.This work further demonstrates the concept of MS sensing and the application of MALDI-MS in quantitative analysis,and provides a convenient method for the quantitation of proteases in clinical diagnosis.  相似文献

An improved enzyme-linked immunosorbent assay (ELISA) for the determination of southern bean mosaic virus (SBMV) with linear sweep voltammetry based on a new system of p-aminophenol (PAP)-H2O2-horseradish peroxidase (HRP) has firstly been developed. The enzymatic product 3-[(4-hydrox-yphenyl) amino]-4-(2-amino-5-hydroxyphenyl)-6-[ (4-hydrox-yphenyl)imino]-2,4-cyclohexadiene-l-one, produced from the oxidation of PAP with H2O2 catalyzed by HRP, yielded a sensitive linear sweep voltammetric response at - 0.45 V ( vs. SCE) in Britton-Robinson (BR) buffer solution. Based on the voltammetric peak, HRP can be measured with a detection limit of 0.4 mU/L and a linear range of 1.0-1.0 × 102 mU/ L. The detection limit for the SBMV is 8.0 ng/mL and the highest dilution ratio for the detection of infected leaf sap is 1: 1.5×105.  相似文献

<正>In this work,we describe a new strategy for the electrochemical determination of captopril(CA) using ferrocenemonocarboxylic acid as a mediator and multiwall carbon nanotubes as sensors in aqueous solution at pH 7.0.The diffusion coefficient(D),and the kinetic parameters such as electron transfer coefficient(α).and heterogeneous rate constant(k_h),for CA were also determined using electrochemical approaches.Under the optimized conditions,the electrocatalytic oxidation peak current of captopril showed two linear dynamic ranges with a detection limit of 0.3×10~(-6) mol L~(-1) captopril.The linear calibration range was 0.8×10~(-6) to 65×10~(-6) mol L~(-1) using cyclic voltammetry.Finally,this modified electrode was also examined as a selective,simple and precise new electrochemical sensor for the determination of captopril in real samples such as drug and patient human urine.  相似文献

In a pH 3.6―5.0 HAc-NaAc buffer solution, when sodium tanshinon ⅡA silate (STSⅡA) reacts with La(Ⅲ) to form a chelate, the resonance Rayleigh scattering (RRS) intensity can be en-hanced greatly and a new RRS spectrum will appear. The maximum RRS peak is located at 306 nm and the RRS intensity is proportional to the concentration of STSⅡA in a certain range. The method is very sensitive and the detection limit for STSⅡA (3σ/K) is 82.12 ng·mL?1. The optimum reaction condi-tions and the effect of coexisting substances have been investigated. A new, simple and fast method for the determination of STSⅡA based on RRS method is developed. It can be applied to the deter-mination of STSⅡA in the synthesis samples and Nuoxinkang injection. Combined with infrared ab-sorption and NMR spectra, the structure of the chelate and the reasons of RRS enhancement are also discussed.  相似文献
