The synthesis of novel NOTA (1,4,7-triazacyclononane-1,4,7-triacetic acid) and DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) chelating groups bearing perfluorinated appendages is described. DOTA and NOTA groups are used in the production of radiopharmaceutical agents for PET and SPECT imaging (by chelation of radioactive metal ions), as well as MRI contrast agents (by chelation of lanthanide Ln3+ ions). The novel perfluorinated variants disclosed herein will enhance the synthesis and purification of such agents, as they are compatible with fluorous purification strategies. Moreover, the perfluorous tag is anticipated to be detectable by 19F-MRI, suggesting future applications in hybrid molecular imaging such as PET–MRI.  相似文献

Available online Development of water soluble AIE-active “light-up” bioprobes for the detection of biomacromolecules has drawn huge research interests in recent past. In this study, a series of ethylene glycol modified water soluble tetrameric tetraphenylethylene amphiphiles with pyridinium polar heads (TPE-xEG-Py, x = 3, 4, 6 or 1a-c) have been synthesized by varying the ethylene glycol spacer. Their unique structure allows them to form vesicles and other nanoaggregates in aqueous solutions. These amphiphiles were successfully utilized for fluorimetric detection and quantitation of BSA and DNA based on the electrostatic interactions to trigger AIE-emission from the TPE moiety. The electrostatic interaction was also proved very effective in wash-free imaging of both Gram-negative (E. coli) and Gram-positive (S. aureus) bacteria with up to 92 folds increase in fluorescence response within bacterial concentration 0–12 × 108 CFU mL?1. The strategy is advantageous due to cost-effective and easy synthesis, high water solubility, and fast response.  相似文献

Reliable methods for the determination of tryptophan and its metabolites are vital to the monitoring of biochemical states during the initiation and progression of cardiovascular disease. In the present study, a single‐run liquid chromatography–tandem mass spectrometry (LC‐MS/MS) method was developed for the simultaneous determination of tryptophan (Trp) and its metabolites, including kynurenine (Kyn), kynurenic acid (KA), xanthurenic acid (XA) and 5‐hydroxytryptamine (5‐HT), in human plasma. The plasma samples were prepared using a protein precipitation approach, and the chromatographic separation was performed by gradient elution on a C18 column within a total analysis time of 3.5 min. The calibration ranges were 40–20,000 ng/mL for Trp, 4–2000 ng/mL for Kyn, 0.2–100 ng/mL for KA, 0.4–200 ng/mL for XA and 1–500 ng/mL for 5‐HT, and the precision and accuracy were acceptable. The evaluation of recovery and internal standard‐normalized matrix effect proved that the sample preparation approach was effective and the matrix effect could be negligible. The newly developed method was successfully applied to the analysis of plasma samples from healthy individuals and myocardial infarction patients. The findings suggested that the plasma concentrations of Trp, Kyn, 5‐HT as well as the concentration ratios of Kyn/Trp and Trp/5‐HT might serve as biomarkers for the monitoring of acute myocardial infarction.  相似文献
