4-Methyl-1, 2, 4-triazoline-3, 5-dione was produced by lead tetraacetate oxidation of 4-methylurazole and allowed to react with phencyclone, 1-The resulting Diels-Alder adduct, 2, has been characterized by one-and two-dimensional 1H and 13C NMR at 300 and 75 MHz, respectively, at ambient temperatures in different solvents. The NMR data are consistent with hindered rotation of the bridgehead unsubstituted phenyl groups about the C(sp2)-C(sp3) bonds, based on numbers of absorption signals in the 1H and 13C NMR aryl region, together with magnetic anisotropic effects in the 1H spectrum. The spectral simplicity suggests further that stereochemistry at the ring junction nitrogens involves only a single isomer or very rapidly interconverting “exo”/“endo” isomers (if the ring junction nitrogens are pyramidalized).  相似文献

The Diels-Alder adduct of phencyclone and N-n-propylmaleimide has been studied in CDCl3 solution at ambient temperatures by one-and two-dimensional 1H NMR (300 MHz) and 13C NMR (75 MHz) techniques. Clear evidence is presented from slow exchange limit (SEL) spectra for hindered rotation of the bridgehead phenyls in the adduct. Full 1H spectral assignments have been made via selective homonuclear decoupling and high resolution COSY experiments. The number of signals in the aryl region of 13C NMR spectra also indicated slow rotation about the C(sp2)-C(sp3) bond to the unsubstantiated bridgehead phenyls. Striking evidence of magnetic anisotropic effects, seen from 1H NMR, permits stereochemical assignment of the adduct as endo.  相似文献

The Diels-Alder adduct of phencyclone and N-n-butylmaleimide has been prepared, and NMR studies have been carried out in CDCl3 solution at ambient temperatures by one-and two-dimensional 1H NMR (300 MHz) and 13C NMR (75 MHz) techniques. The resulting spectra appear to be consistent with slow rotation about the hindered C(sp2)-C(sp3) bonds to the bridgehead unsubstituted phenyls, i.e., slow exchange limit (SEL) spectra. Full rigorous 1H spectral assignments have been made via high-resolution COSY experiments. The number of signals in the 13C NMR aryl region were also consistent with hindered phenyl rotations; preliminary 13C assignments are given. Striking evidence for magnetic anisotropic effects due to the phenanthrene moiety, bridging ketone carbonyl, and bridgehead phenyls are discussed, supporting endo stereochemical assignment of the adduct.  相似文献

The bridged ruthenium cluster-polypyridine dimer [Ru3O(CH3COO)6(py)2(tmbpy)Ru(bpy)2(Cl)](PF6)2 (py = pyridine, = 2, 2′-bipyridine and tmbpy = 4, 4′-trimethylenedipyridine) has been synthesized and structurally characterized based on 1H and 13C NMR spectroscopy. This species exhibits a complex pattern of NMR signals due to the presence of a paramagnetic [Ru3O] core and seven non-equivalent aromatic rings. 2D NMR (COSY, HMQC and HMBC) correlation techniques have been required for the total assignment of the 1H and 13C NMR spectra.  相似文献

The potent Diels-Alder diene, phencyclone, 1, reacts with N-pentafluorophenylmaleimide, 2, to form an adduct, 3, characterized by 1H, 13C, and 19F NMR at 300, 75 and 282 MHz, respectively. The one-dimensional (1D) and two-dimensional (2D) 1H and 13C NMR spectra of 3 at ambient temperatures imply a slow exchange limit (SEL) regime with respect to rotation of the unsubstituted bridgehead phenyl groups about severely hindered C(sp2)-C(sp3) bonds. Major non-bonded interactions are expected between the ortho protons of the C6H5 groups and H-1, 8 of the phenanthrenoid moiety of 3. 19F 1D and 2D (COSY) NMR spectra show that the SEL regime also obtains for rotation about the N-C6F5 bond of 3, with five separate fluorine signals seen, consistent with a preferred conformation in which the C6F5 may lie roughly perpendicular to the plane of the pyrrolidinedione moiety, and may be in the mirror symmetry plane of 3. The results are considered relevant to hindered aryl rotations in numerous Pharmaceuticals. Selected spectral data for 2 and precursors are also presented.  相似文献

Abstract

Phencyclone, 1, reacted with N‐(2,6‐dimethylphenyl)maleimide, 2a; with N‐(2,6‐diethylphenyl)maleimide, 2b; and with N‐(2,6‐diisopropylphenyl)maleimide, 2c, respectively, to yield the corresponding Diels–Alder adducts, 3a–c. The adducts were extensively characterized by NMR (7 T) at ambient temperatures using one‐ and two‐dimensional (1D and 2D) proton and carbon‐13 techniques for assignments. Slow exchange limit (SEL) spectra were observed, demonstrating slow rotations on the NMR timescales, for the unsubstituted bridgehead phenyl groups [C(sp3)–C(aryl sp2) bond rotations] and for the 2,6‐dialkylphenyl groups [N(sp2)–C(aryl sp2) bond rotations]. Substantial magnetic anisotropic shifts were seen in the adducts. For example, in the N‐(2,6‐dialkylphenyl) moieties of the adducts, one of the alkyl groups is directed “into” the adduct cavity, toward the phenanthrenoid portion, and these “inner” alkyl proton NMR signals were shifted upfield. Thus, in CDCl3, the “inner” methyl of adduct 3a exhibits a proton resonance at ?0.13 ppm, upfield of tetramethylsilane (TMS); the “inner” ethyl group signals from 3b appear at 0.026 ppm (CH2, quartet), and ?0.21 ppm (CH3, triplet); and the “inner” isopropyl group from 3c is seen at ?0.06 ppm (methine, approx. septet) and ?0.39 ppm (CH3, doublet). Proton NMR of the crude N‐(2,6‐dialkylphenyl)maleamic acids (used as precursors of the maleimides, 2a–c) exhibited two sets of AB quartet signals, suggesting possible conformers from hindered rotation in the amide groups about the HN–C?O bonds.  相似文献

The Diels-Alder adduct of phencyclone with p-benzoquinone has been examined by 1H NMR at 300 MHz in CDCl3 at ambient temperatures for evidence of hindered rotation of the unsubstituted bridgehead phenyl groups. The nearly first-order spectrum exhibits four approximate doublets and five approximate triplets in the aromatic region, of roughly equal intensity (ca. 2H). This is consistent with a slow-exchange limit (SEL) spectrum of the hindered phenyls. For rapidly rotating phenyls, the predicted fast-exchange limit (FEL) spectrum would have shown two 2H doublets and one 4H doublet, in addition to three 2H triplets and one 4H triplet, in the aryl region. Full 1H assignments have been made, based on the two-dimensional 1H-1H homonuclear chemical shift correlation spectrum (COSY) and expected magnetic anisotropy effects.  相似文献

13C NMR chemical shift assignments were obtained for the Diels-Alder adduct of phencyclone with norbornadiene in CD2Cl2 and in CDCl3 solution. The 13C spectrum at 50.3 MHz, as well as the 1H spectrum at 200.1 MHz, show evidence for hindered rotation of the two unsubstituted bridgehead phenyl rings of the adduct at ambient temperatures. In CD2Cl2 solution, all 19 of the unique 13C nuclei of this molecule give rise to individual 13C resonances. The 1H assignments which were made earlier, together with one-bond and long-range 2D heteronuclear correlation experiments, allowed the assignment of all 13C chemical shifts in the molecule.  相似文献

The skeletal muscle relaxant, aflogualone, 1, has been studied by 1H NMR in CDC13 at 60 and 300 MHz in the presence of added chiral [Eu(HFC)3] or achiral [Eu(FOD)3] lanthanide shift reagents (LSR). With Eu(HFC)3, significant enantiomeric shift differences, ΔΔδ, can be induced for most of the nuclei of 1, with near-baseline resolution obtainable for the H-5 signals of each enantiomer, indicating excellent analytical potential for direct determination of enantiomeric excess of samples of 1. Observation of ΔΔδ directly confirms hindered rotation about the bond between N (3) and the 2-methylphenyl group, i.e., the N (sp2) - C(sp2) bond, leading to axial chirality in 1 with slow rotation on the NMR timescale. Assignments are supported by 2D COSY spectra at 300 MHz. Relative lanthanide-induced shift magnitudes for the protons of 1 are compared.  相似文献

Nitrogen-14 (spin I = 1) has always been a nucleus difficult to observe in solid-state NMR and until recently its observation was restricted to one-dimensional (1D) spectra. We present here the first 3D 1H–13C–14N NMR correlation spectrum. This spectrum was acquired on a test sample l-histidine·HCl·H2O using a recently developed technique, which consists in indirectly observing 14N nuclei via dipolar recoupling with an HMQC-type experiment.  相似文献

13C NMR and 2D (H,H and H,C COSY) spectra of selected examples of sugar (5H-1,2,4-triazino[5,6-b]-indol-3-yl) hydrazones, peracetylated sugar-1-acetyl -1- (5-acetyl-1,2,4-triazino[5,6-b]indol-3-yl)hydrazones, and 10-acetyl-3-(per-0-acetylalditol-1-yl)-1,2,4-triazolo[4′, 3′:2,3][1,2,4]triazino[5,6-b]indole have been reported. The conformation of the latter C-nucleoside analogues have been determined by analysis of their 1H NMR spectra. The D-galacto, D-manno and L-arabino isomers are preponderantly existing in the planar zigzag arrangement of carbon atoms.  相似文献

Abstract

N‐Phenylmaleimide, 2, and N‐(2‐trifluoromethylphenyl)maleimide, 3, were separately added to phencyclone, 1, to yield the corresponding phencyclone Diels–Alder adducts, 4 and 5. The resulting adducts (and some precursors) have been characterized by one‐ and two‐dimensional 1H and 13C NMR at 300 and 75 MHz, and by 19F NMR at 282 MHz, at ambient temperatures. The NMR data are consistent, for both adducts, with: (a) hindered rotation of the bridgehead unsubstituted phenyl groups about the C(sp2)–C(sp3) bonds, based on slow exchange limit (SEL) spectra and (b) endo adduct configuration based on magnetic anisotropic effects in the 1H NMR. The NMR spectra of the phencyclone adduct, 4, of N‐phenylmaleimide, indicate free rotation on the NMR timescales (fast exchange limit, FEL spectra) about the N‐phenyl bond. The spectra for the adduct, 5, of N‐(2‐trifluoromethylphenyl)maleimide are interpreted as consistent with SEL regimes, for the N‐aryl rotations, with a single rotamer present in which the trifluoromethyl group is directed “out of” the adduct cavity, and away from the phenanthrenoid moiety. This conclusion is based, in part, on NMR data suggesting the apparent slow N‐aryl bond rotation in a pair of atropisomers corresponding to the acetic acid addition products from the N‐(2‐trifluoromethylphenyl)maleimide. Evidence of magnetic anisotropic effects due to the phenanthrenoid moiety and proximal carbonyls is discussed. 1H, 13C, and 19F assignments are presented and interpreted. Molecular modeling calculations at the Hartree–Fock level, 6‐31G* basis set, were performed to provide geometry optimizations for energy‐minimized structures of selected compounds.  相似文献

The compound 2,10-dibromo-3-chloro-8-hydroxy-β-chamigrene was analysed in detail by NMR Spectroscopy. the complete assignment of the signals in the 1H and 13C NMR spectra and the determination of the relative configurations were achieved by 2D NMR techniques, AM1 data and 1H spectrum simulation. The 15N NMR chemical shifts and 1(15N-1H) coupling constants of a series of imidazolidine-2,4-dichalcogen (O, S) derivatives are reported.The 15N NMR chemical shifts show a linear correlatlon wlth the vNH stretchlng vlbratlons. The influence of the substitution of the oxygen at C2 and/or C4 with the sulphur, and of the hydrogen at C5 wlth the methyls and phenyls has been considered. Abstract

Inverse secondary kinetic isotope effects are determined for the dimerization of all‐cis‐cyclononatetraenyl radical, 1, to its corresponding dimer, all‐cis‐9,9′‐bicyclonona‐1,3,5,7‐tetraene, 2, (step 1, k H/k D=0.5), and cyclization of the latter to 9,9′‐bisbicyclo[4.3.0]cyclonona‐2,4,7‐triene, 3 (step 2, k H/k D=0.75). These results are obtained by comparison of 1H‐ and 2D‐NMR spectra of 3 and employment of a simple statistical method for acquiring kinetic data. This new strategy appears superior to conventional methods in being fast, simple, and less expensive.  相似文献

1H, 13C, 19F and 29Si NMR chemical shifts and coupling constants for Si-substituted silatranes, XSi(OCH2CH2)3N, and triethoxysilanes, XSi(OCH2CH3)3, where X = H, CH3, and F have been studied. Abstract

The 1H‐ and 13C‐NMR spectra of some substituted stilbenes and chalcones were assigned unambiguously on the basis of a combination of homo‐ (COSY) and heteronuclear (HETCOR) two‐dimensional methods, the chemical shifts, as well as spin‐coupling constants. The Aik empirical parameters of the –O–C(S)–N(CH3)2, –S–C(O)–N(CH3)2, and –SH group were calculated to help predict the chemical shifts of substituted stilbenes, 4′‐nitrostilbenes, and chalcones. The 1H‐ and 13C‐NMR spectra have been shown to be able to differentiate between the isomers of O‐stilbenyl (4, 5) and S‐stilbenyl N,N‐dimethylthiocarbamates (7, 8) as well as O‐chalconyl (6) and S‐chalconyl N,N‐dimethylthiocarbamates (9).  相似文献

A suite of 3D NMR experiments for measuring15N–{1H} NOE,15NT1, and15NTvalues in large proteins, uniformly labeled with15N and13C, is presented. These experiments are designed for proteins that exhibit extensive spectral overlap in the 2D1H–15N HSQC spectrum. The pulse sequences are readily applicable to perdeuterated samples, which increases the spectral resolution and signal-to-noise ratio, thereby permitting the characterization of protein dynamics to be extended to larger protein systems. Application of the pulse sequences is demonstrated on a perdeuterated13C/15N-labeled sample of the 44 kDa ectodomain of SIV gp41.  相似文献
