Abstract

The resonance light scattering (RLS) spectra of ethyl rhodamine B with nucleic acid (calf thymus DNA and herring sperm DNA) have been studied. The effective factors and the optimum conditions have been studied, and the enhanced intensity of RLS is in proportion to the concentration of nucleic acids in the range 0~5.00 µg mL?1 for calf thymus DNA (ctDNA) and 0~3.50 µg mL?1 for herring sperm DNA (hsDNA). The limits of detection are 3.42 and 3.14 ng mL?1, respectively. Based on this, a RLS method for the determination of nucleic acids sensitized by emulsifier OP‐micelle was accordingly established. The binding mode concerning the interactions of ethyl rhodamine B with nucleic acids was also studied, and this method has good selectivity and high sensitivity and it has been applied to the determination of DNA in synthetic samples and real samples with satisfactory results.  相似文献

ABSTRACT

Aqueous glutathione-capped cadmium/tellurium quantum dots with a diameter of about 3 nm were synthesized. The fluorescence was quenched in the presence of protoporphyrin IX disodium salt, with the excitation wavelength at 320 nm. Under the optimal conditions, the quenched fluorescence intensity was linear in the range of 0.096–16 µg · mL?1 with a concentration of protoporphyrin IX disodium salt, and the detection limit (3σ) was 2.8 × 10?2 µg · mL?1. The proposed method has been applied to the determination of protoporphyrin in serum samples with satisfactory results. The interaction mechanism was investigated.  相似文献

ABSTRACT

Trace amounts of cerium were analyzed by flow injection kinetic spectrophotometry, based on the decolorization reaction between arsenazo III and Ce(IV) in sulfuric acid medium at room temperature. The absorbance difference (ΔA) of decolorization was linear with the concentration of Ce(IV). The flow injection technique was used to precisely control the timing. Under the optimum conditions, the determination of Ce(IV) in the range 0.0–8.0 µg mL?1 with a correlation coefficient (r) of 0.9982, the regression equation was ΔA = 0.0014 + 0.0406c (µg mL?1). The detection limit (3σ) of 0.2 µg mL?1 was achieved at a sampling frequency of 60 h?1. The proposed method was applied to the analyses of Ce in soil successfully.  相似文献

ABSTRACT

Rhizoma cyperi (tuberal part of Cyperus rotundus Linn) obtained from 15 different zones of China was studied to determine the contents of 16 trace elements such as 4 minor (Ca, K, Mg, and Na), 9 trace (Co, Cr, Cu, Fe, Mn, Mo, Ni, V, and Zn), and 3 toxic (Ag, Cd, and Pb) elements. The concentration determination of 16 elements was performed by atomic absorption spectrophotometry (AAS) after microwave-assisted digestion. A microwave-assisted digestion procedure based on the mixture nitric acid–hydrogen peroxide was evaluated. The method was successfully validated with the good recoveries (97–105%) against CRM GBW07603 (bush twigs and leaves). The calibration curve furnished good linear correlation coefficients (r = 0.9956–0.9999), excellent recoveries (99.35–103.7%), and limits of detection (LOD = 1–50 ng·mL?1) suitable to determine in Rhizoma cyperi. The results showed that K, Ca, Mg, and Na were the most abundant of the major elements in Rhizoma cyperi with average concentrations of K, 26,221 µg·g?1; Ca, 1097 µg·g?1; Mg, 714 µg·g?1, and Na, 293 µg·g?1, respectively. K element was determined for the first time in this plant.  相似文献

ABSTRACT

Surface-enhanced Raman spectroscopy spectra of dimethoate and phosmet pesticides were recorded using a Klarite substrate. Significant enhancements were achieved with dimethoate over a concentration range of 0.5–10 µg mL?1 and phosmet over a concentration range of 0.1–10 µg mL?1. The best prediction model for dimethoate pesticide was achieved with a correlation coefficient of 0.940 and a root mean square error of prediction of 0.864 µg mL?1, with the first derivative and standard normalized variate data preprocessing, and the best prediction model of phosmet pesticide was achieved with a correlation coefficient of 0.949 and a root mean square error of prediction of 0.741 µg mL?1 with the first derivative data preprocessing. Our study shows that pesticides, including dimethoate and phosmet, could be quantitatively measured at as low as 0.5 µg mL?1 level using surface-enhanced Raman technology coupled with a Klarite substrate and the results indicated that surface-enhanced Raman spectroscopy with a Klarite substrate has potential for the analysis of dimethoate and phosmet residues.  相似文献

We found that the fluorescence intensity of the lanthanum (La3+)-curcumin (CU) complex can be highly enhanced by proteins in the presence of sodium dodecyl benzene sulphonate (SDBS). Based on this finding, a new fluorimetric method for the determination of protein was developed. Under optimized conditions, the enhanced intensities of fluorescence are quantitatively in proportion to the concentrations of proteins in the range 0.0080?C20.0 ??g·mL?1 for bovine serum albumin (BSA) and 0.00080?C20.0 ??g·mL?1 for human serum albumin (HSA) with excitation of 425 nm, and 0.00020?C20.0 ??g·mL?1 for bovine serum albumin (BSA) and 0.00080?C20.0 ??g·mL?1for human serum albumin (HSA) with excitation of 280 nm, while corresponding qualitative detection limits (S/N????3) are as low as 5.368, 0.573, 0.049, 0.562 µg·mL?1, respectively. Study on reaction mechanism reveals that proteins can bind with La3+, CU and SDBS through self-assembling function with electrostatic attraction, hydrogen bonding, hydrophobic interaction and van der Waals forces, etc. The proteins form a supermolecular association with multilayer structure, in which La3+-CU is clamped between BSA and SDBS. The unique high fluorescence enhancement of CU is resulted through synergic effects of favorable hydrophobic microenvironment provided by BSA and SDBS, and efficient intermolecular energy transfer among BSA, SDBS and CU. In energy transfer process, La3+ plays a crucial role because it not only shortens the distance between SDBS and CU, but also acts as a ??bridge?? for transferring the energy from BSA to CU.  相似文献

ABSTRACT

A sensitive and simple method for the determination of luteolin (LTL) was developed based on the fluorescence quenching effect of LTL for thioglycolic acid–capped (TGA-capped) CdTe/CdS quantum dots (QDs). Under optimum conditions, a good linear relationship was obtained from 0.3 to 20.0 µg · mL?1 with a correlation coefficient of 0.9972, and the detection limit was 7.2 ng · mL?1. The fluorescence quenching mechanism has been proposed on the basis of electron transfer supported by ultraviolet-visible (UV-Vis) absorption, fluorescence (FL) spectroscopy. The proposed method was successfully applied to the determination of LTL in commercial capsules and human urine samples. It manifested several advantages such as high sensitivity, short analysis time, low cost, and ease of operation.  相似文献

Abstract

The effects of three systems on the chemiluminescence (CL) intensity have been studied in this paper, such as leucogen–potassium permanganate–rhodamine B, leucogen–cerium (IV)–rhodamine B, and leucogen–luminol–hydrogen peroxide (called system 1, system 2, and system 3, respectively). The mechanism of these reactions is also discussed. Surfactant (CTMAB) has a remarkably sensitive effect on these systems mentioned above. Therefore, three new flow injection chemiluminescence methods for the determination of leucogen have been established. For system 1, the linear range is 8.0×10?8 to 4.0×10?5 g mL?1, with limits of detection 2×10?8 g mL?1; the relative standard deviation is 2.5% (n=11, Cs=4.0×10?6 g mL?1). For system 2, the linear range is 1.0×10?8 to 5.0×10?6 g mL?1, with limits of detection 3×10?9 g mL?1; the relative standard deviation is 5.1% (n=11, Cs=1.0×10?6 g mL?1). For system 3, the linear range is 4.0×10?8 to 2.0×10?6 g mL?1, with limits of detection 1×10?8 g mL?1; the relative standard deviation is 1.3% (n=11, Cs=1.0×10?7 g mL?1). Compared with the three methods above, system 3 is confirmed as the best method. This method has been applied to the determination of leucogen with satisfactory results.  相似文献

Amitriptyline.HCl (AMI) and clomipramine.HCl (CMI) react with eosin Y (EY) in pH 3.8 NaAc-AcH buffer solution to form ion association complex which results in quenching of fluorescence of EY and appearance of a new resonance Rayleigh scattering (RSS) spectrum at 620 nm. The spectral characteristics of absorption, fluorescence and RSS spectra have been investigated. The factors influencing the reaction were studied and optimum conditions for the reaction have been determined. Based on fluorescence quenching, a simple and sensitive spectrofluorimetric method for determination of AMI and CMI has been developed. The fluorescence quenching intensity was measured at 550 nm using an excitation wavelength of 310 nm. The calibration graph was found to be rectilinear in the range 0.08–2.0 μg?mL?1 with detection limit of 0.017 μg?mL?1 for AMI and 0.06–2.0 μg?mL?1 with detection limit of 0.015 μg?mL?1 for CMI. The method can be satisfactorily applied to the determination of AMI and CMI in tablets without interference from commonly occurring exicipients. The recovery and RSD values obtained indicate good accuracy and precision of the method. The mechanism of the reaction and fluorescence quenching has also been discussed.  相似文献

Abstract

An automated flow injection system was developed for monitoring cyanide concentration in effluents from petroleum refineries. The method takes advantage of the reaction of cyanide ions with ninhydrin in basic medium in a flow injection system. A linear range of 0.01 to 0.04 µg mL?1 was obtained with a detection limit of 1.5 ng mL?1 by using 500 µL sample injection, with an analytical throughput of 30 samples hr?1, excluding sample pretreatment by distillation if required. Regarding interferences, cyanide can be determined in the presence of 100 mg L?1 of thyocianate and sulfide, both species normally found in industrial effluents. For total cyanide determination, strong acid distillation is recommended due to the presence of cyano‐metallic complexes in the refinery effluents. The method was validated by analyte addition and results compared with the standard methodology proposed by the American Public Health Association (APHA). The more significant advantage of the proposed method is the lack of use of carcinogenic reagent such as pyridine and psychotropic compound such as barbituric acid, both used in the recommended method by APHA. Thus, the proposed method is really a friendly analytical procedure.  相似文献

ABSTRACT

A new catalytic kinetic fluorescent quenching method for the determination of trace gold(III) was investigated. The method was based on the catalytic effect of gold on oxidation of 3-(3′-methylphenyl)-5- (2′-arsenoxylphenylazo) rhodanine by hydrogen peroxide in potassium hydrogen phthalate–hydrochloric acid (pH = 3.4). Under the optimum conditions, the great decrease of fluorescence intensity has a linear relationship against the concentration of gold in the range of 0 to 12.0 µg·L?1 with a detection limit of 6.0 × 10?10g·L?1. The coexistent metal ions can be separated, and gold can be enriched by TBP resin of solid-phase extraction, which greatly improves the selectivity and sensitivity of the system. The method can be used to determine trace amounts of gold in ore samples successfully with satisfactory results.  相似文献

A simple, rapid, and convenient method based on ion association for the determination of acetazolamide has been developed. Acetazolamide can react with palladium(II) to form cationic chelate at sodium acetate–acetic acid buffer solution, which can further react with eosin Y to form an ion association complex. As a result, the resonance Rayleigh scattering, second-order scattering, and frequency doubling scattering intensities are enhanced. Analytical wavelengths of resonance Rayleigh scattering, second-order scattering, and frequency doubling scattering located at 298, 558, and 338?nm, and the linearity range are 0.014–2.5, 0.034–2.5, and 0.119–2.4?µg?mL?1 respectively. In addition, the optimum conditions and the effect of coexisting substances are investigated. Besides, the structure of ion association complex, the reaction mechanism and the reasons for the enhancement of scattering are discussed through infrared spectra, absorption spectra, and quantum chemical calculations.  相似文献

Abstract

A new flow‐injection chemiluminescence (CL) method is described for the determination of carbendazim. The method is based on the CL reaction of luminol and hydrogen peroxide (H2O2). Carbendazim can greatly enhance the chemiluminescence intensity in sodium hydroxide–sodium dihydrogen phosphate (NaOH–NaH2PO4) medium (pH=12.6). Under the optimum conditions, the linear range for the determination of carbendazim is 2.00×10?8 to 2.00×10?6 g mL?1 with a detection limit (S/N=3) of 7.24×10?9 g mL?1. The relative standard deviation is 1.8% for 1.0×10?7 g mL?1 carbendazim (n=8). The proposed method has been applied to the determination of carbendazim in tap‐water samples. Furthermore, the possible enhanced CL mechanism is discussed by examining the CL spectra and fluorescence spectra.  相似文献

ABSTRACT

The interaction of pyronine Y (PY) with nucleic acids was studied by resonance Rayleigh scattering (RRS) for nucleic acid detection. The enhanced RRS intensity of nucleic acids reacted with PY was proportional to the concentration of nucleic acids in the ranges of 27.0–625 ng ml?1 for fish sperm DNA, 39.0–500 ng ml?1 for calf thymus DNA, and 59.0–375 ng ml?1 for yeast RNA. The limits of determination were 0.2 ng ml?1 for fish sperm DNA, 0.6 ng ml?1 for calf thymus DNA, and 0.7 ng ml?1 for yeast RNA. The method had been successfully applied to the quick determination of nucleic acids in synthetic and natural samples.  相似文献

Abstract

An automated sequential‐injection online preconcentration system was developed for the determination of lead by inductively coupled plasma–atomic emission spectrometry (ICP‐AES). The preconcentration of lead was performed with a minicolumn containing a lead‐selective resin, Analig Pb‐01, which was installed between a selection and a switching valve. In an acidic condition (pH 1), lead could be adsorbed on the resin. The concentrated lead was afterward eluted with 25 µL of 0.06 M nitrilotriacetic acid (NTA) solution (pH 9) and was subsequently transported into the nebulizer of ICP‐AES for quantification. The selectivity of the resin toward lead was examined using a solution containing a mixture of 61 elements. When a sample volume of 5 mL was used, the quantitative collection of lead (≥97%) was achieved, along with an enrichment factor of 19, a sampling frequency of 12 samples hr?1, a detection limit of 70 pg mL?1, and a lowest quantification limit of 100 pg mL?1. The linear dynamic range was 0.1 to 5 ng mL?1, and the relative standard deviation (n=9) was 0.5% at a 5 ng mL?1 Pb level. The detection limit of 30 pg mL?1 and lowest quantification limit of 50 pg mL?1 could be achieved when 10 mL of sample volume was used. The accuracy of the proposed method was validated by determining lead in the standard reference material of river water (SLRS‐4), and its applicability to the determination of lead in environmental river water samples was demonstrated.  相似文献

A novel fluorimetric method based on diazotization-coupling reaction (DCR) for the determination of clenbuterol is described. In acidic solution, clenbuterol was first diazotized with sodium nitrite, followed by coupling with bisphenol A to produce an azo-compound in NH3- NH4Cl buffer. It has found the diazotized clenbuterol- bisphenol A- NH3- NH4Cl (DCBN) system has strong fluorescence efficiency compare with the bisphenol A solution. There is a linear relationship between the increased intensity of the fluorescence emission spectra (λexem?=?276 nm/306 nm) and the concentration of clenbuterol. The effects of the amount of sodium nitrite, diazo reaction time, the amount of bisphenol A, coupling reaction time and coupling reaction temperature have been examined. Under the optional conditions, clenbuterol can be determined over the concentration range of 0.02 to 2.0 μg mL?1 with a correlation coefficient of 0.9953. The detection limit is 0.01 μg mL?1 at a signal-to-noise ratio of 3. The relative standard deviation (RSD) for 11 repetitive determinations of 0.9 μg mL?1 clenbuterol is 0.22 %. The utility of this method was demonstrated by determining clenbuterol in meat samples.  相似文献

ABSTRACT

An automatic fluorescence method was developed for the determination of ketorolac tromethamine. The method is based on the direct oxidation of the analyte by an acidic solution of permanganate, being the resulting fluorescence measured at 227 nm/320 nm (λexem). The reaction is carried out online due to the use of the Sequential Injection Analysis methodology. A detection limit of 0.12 µ g mL?1 and a R.S.D. lower than 3% (n = 10) were obtained under optimum conditions. The analyte was satisfactorily determined in pharmaceuticals and urine samples. Recovery experiments were carried out on human urine, with recoveries in the range 92–108%.  相似文献

Abstract

A new flow‐injection online reduction electrochemical hydride generation system for the determination of Se(IV) and Se(VI) by atomic fluorescence spectrometry (AFS) was developed. In the system, an electromagnetic induction oven was used as heating resource to reduce Se(VI) to Se(IV) and a homemade tubular electrolytic cell as hydride generator. All analytical procedures were automatically controlled by a computer. The conditions of online reduction, including temperature, HCl concentration, and reduction time, have been studied in detail. The detection limits (3σ) of Se(IV) and Se(VI) in aqueous solution were 0.26 µg L?1 and 0.23 µg L?1, respectively. The precision for 11 replicate measurements of 50 µg L?1 Se(IV) and Se(VI) was 2.2% and 2.5%. This proposed method has been applied to the determination of Se(IV) and Se(VI) in springwater samples.  相似文献

A new simple, rapid, selective and sensitive analytical procedure based on chemiluminescence (CL) detection is described for the determination of free fluoride at sub‐nanogram levels by use of controlled‐reagent‐release technology in a flow injection system. The analytical reagents involved in the CL reaction, including luminol and periodate, were both immobilized on anion‐exchange resins in a flow injection system. Through water injection luminol and periodate were eluted from the anion exchange column to generate the chemiluminescence, which was enhanced in the presence of fluoride. The increased CL intensity was linear with fluoride concentration in the range from 0.1 to 10 ng·mL? 1. The limit of detection was 20 pg·mL? 1 (3σ) and the relative standard deviation (RSD) was 1.02% (n = 5) for a 1.0 ng·mL? 1 fluoride. At a flow rate of 2.0 mL·min? 1, including sampling and washing, a typical analytical procedure could be performed in 0.5 min with a RSD of less than 3.0%. The proposed method was successfully applied to determine the free fluoride in water and human urine, and the results were in good agreement with those obtained by ion chromatography.  相似文献
