We investigated the signaling events involved in the wound response of the marine macroalga Dasycladus vermicularis, finding nitric oxide (NO) production in relation to injury. The addition of exogenous H2O2 to aliquots of injured algae accelerated the kinetics of NO production in the wounded region. Similarly, the addition of an NO donor caused an increase in detectable H2O2 around the site of injury. By wounding or incubating uninjured algae with an NO donor, peroxidase activity was enhanced. Based on the use of selected pharmacological probes, our results indicate that H2O2 production involves the upstream activation of signaling events similar to those observed in the physiology of higher plants.  相似文献

The kinetics of the oxidation of styrene, cinnamic acid, and some of their substituted derivatives by hexachloroiridate(IV) in dimethyl formamide–water mixtures and in the presence of perchloric acid have been investigated. The reactions appear to proceed via the formation of an unstable intermediate 1:1 complex between iridium(IV) and the substrate, followed by the decomposition of the complex in the rate‐determining step. Correlation with σ yielded ρ values of ?4.0 and ?3.5 which suggests the formation of a cationic intermediate in the rate‐determining step of the reaction. Subsequent cleavage of the carbon–carbon bond yielded the product aldehydes. Anthocyanins possess cancer chemopreventive properties in preclinical models. Their clinical pharmacology is only poorly understood. In this pilot study, anthocyanins and their metabolites were analysed in the urine of two patients with colorectal liver metastases. They received a single dose of 1.88 g standardized bilberry extract (mirtoselect) via either nasogastric or nasojejunal tube intra‐operatively during liver resection. HPLC‐MS/MS and HPLC‐UV analysis showed there were more anthocyanins and metabolites in the urine of the patient who received mirtoselect via the stomach than via the jejunum. This result is consistent with information obtained in rodents which suggests the stomach is the predominant site for anthocyanin absorption. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Inorganic nitrogen species (nitrate, nitrite and ammonium ions) were simultaneously determined by microcolumn ion chromatography. Nitrate and nitrite were determined by UV detection at 206 nm, whereas ammonium ion was determined by fluorescence detection at excitation 410 nm and emission 470 nm. The latter fluorescence detection is based on the postcolumn reaction of ammonium ion with o-phthalaldehyde in the presence of 2-mercaptoethanol. Effects of the reagent concentration, pH, and other reaction conditions on the signal intensity were examined, and the optimum condition was explored. The present method allowed simultaneous determination of nitrate, nitrite and ammonium ions in river water.  相似文献

Summary The reactivities of - and -glycerophosphates towards MnO 4 in HClO4 medium have been examined. The reactions between glycerophosphates and MnO 4 are first order with respect to [glycerophosphate], [MnO 4 ] and [H+]. There is no evidence for cleavage of the C–O–P bond rather than the terminal –CH2OH group being oxidized to the respective phosphoglyceraldehyde.  相似文献

A microchip electrophoresis method was developed for simultaneous determination of reactive oxygen species (ROS) and reduced glutathione (GSH) in the individual erythrocyte cell. In this method, cell sampling, single-cell loading, docking, lysing, and capillary electrophoretic separation with LIF detection were integrated on a microfluidic chip with crossed channels. ROS was labeled with dihydrorhodamine 123 in the intact cell, while GSH was on-chip labeled with 2,3-naphthalene-dicarboxaldehyde, which was included in the separation medium. On-chip electrical lysis, characterized by extremely fast disruption of the cellular membrane (<40 ms), was exploited to minimize enzymatic effects on analyte concentrations during the determination. The microfluidic network was optimized to prevent cell leaking from the sample reservoir (S) into separation during the separation phase. The structure of the S was modified to avoid blockage of its outlet by deposited cells. Detection limits of 0.5 and 6.9 amol for ROS and GSH, respectively, were achieved. The average cell throughput was 25 cells/h. The effectiveness of the method was demonstrated in the simultaneous determination of GSH and ROS in individual cells and the variations of cellular GSH and ROS contents in response to external stimuli.  相似文献

Oxidative and nitrosative stress induced by ROS/RNS play crucial roles in a wide range of physiological processes and are also implicated in various diseases, including cancer and neurodegenerative disorders. Sensitive and selective methods for the detection of ROS/RNS based on fluorescent and luminescent probes are of great use in monitoring the in vivo production of these species and elucidating their biological functions. This critical review highlights recent advances that have been made in the development of fluorescent and luminescent probes employed to monitor various ROS/RNS (132 references).  相似文献

Reactive oxygen and nitrogen species (ROS and RNS) play important roles in various physiological processes (e.g. phagocytosis) and pathological conditions (e.g. cancer). The primary ROS/RNS, viz., hydrogen peroxide, peroxynitrite ion, nitric oxide, and nitrite ion, can be oxidized at different electrode potentials and therefore detected and quantified by electroanalytical techniques. Nanometer-sized electrochemical probes are especially suitable for measuring ROS/RNS in single cells and cellular organelles. In this article, we survey recent advances in the localized measurements of ROS/RNS inside single cells and discuss several methodological issues, including optimization of nanoelectrode geometry, precise positioning of an electrochemical probe inside a cell, and interpretation of electroanalytical data.  相似文献

Genus Curcuma a wild species presents an important source of valuable characters for improving the cultivated Curcuma varieties. Based on the collected germplasms, herbariums, field surveys and other literatures, the ecogeographical diversity of Genus Curcuma and its potential distributions under the present and future climate are analysed by DIVA-GIS. The results indicate Genus Curcuma is distributed over 17 provinces in China, and particularly abundant in Guangxi and Guangdong provinces. The simulated current distributions are close to the actual distribution regions. In the future climate, the suitable areas for four Curcuma species will be extended, while for other three species the regions will be significantly decreased, and thus these valuable resources need protecting.  相似文献

In this study, we established a method for simultaneously determining twelve biogenic amines in serum by using reversed phase high performance liquid chromatography (RP-HPLC). The biogenic amines were first extracted from human serum by perchloric acid solution and derivatized by dansyl chloride. An ODS column was selected as separation column at 40 °C. The mobile phase solutions were consisted of A, 0.1 mol/L ammonium acetate and B, acetonitrile. A gradient elution was carried out with a flow rate at 1.0 ml/ml. The results show that the detection limit for twelve biogenic amines ranged between 0.0621 and 0.628 μg/L. All the correlation coefficients were above 0.999. The linearity was over the range from 0.001 to 20 mg/L depending on individual biogenic amine. The intra-day and inter-day coefficients of variations were from 0.53% to 7.50%,and from 1.10% to 7.25% respectively. The average analytical recovery in serum was from 92.02% to 107.65%. Moreover, the serum concentrations of tryptamine, tyramine and histamine in healthy females were found lower than that in healthy males significantly. The method is sensitive, convenient, and reliable, and suitable for simultaneous analysis of multiple biogenic amines in the clinical diagnosis and drug discovery.  相似文献

A microchip electrophoresis method coupled with laser-induced fluorescence (LIF) detection was established for simultaneous determination of two kinds of intracellular signaling molecules (reactive oxygen species, ROS, and reduced glutathione, GSH) related to apoptosis and oxidative stress. As the probe dihydrorhodamine-123 (DHR-123) can be converted intracellularly by ROS to the fluorescent rhodamine-123 (Rh-123), and the probe naphthalene-2,3-dicarboxaldehyde (NDA) can react quickly with GSH to produce a fluorescent adduct, rapid determination of Rh-123 and GSH was achieved on a glass microchip within 27 s using a 20 mM borate buffer (pH 9.2). The established method was tested to measure the intracellular ROS and GSH levels in acute promyelocytic leukemia (APL)-derived NB4 cells. An elevation of intracellular ROS and depletion of GSH were observed in apoptotic NB4 cells induced by arsenic trioxide (As(2)O(3)) at low concentration (1-2 microM). Buthionine sulfoximine (BSO), in combination with As(2)O(3) enhanced the decrease of reduced GSH to a great extent. The combined treatment of As(2)O(3) and hydrogen peroxide (H(2)O(2)) led to an inverse relationship between the concentrations of ROS and GSH obtained, showing the proposed method can readily evaluate the generation of ROS, which occurs simultaneously with the consumption of the inherent antioxidant.  相似文献

Nitric oxide (NO) is a messenger molecule implicated in a number of physiological processes. Nitrosation of selenoproteins has been suggested as playing an important role in NO-mediated cellular functions such as the inactivation of glutathione peroxidase (GPx), but no chemical information about Se-nitrosated species has been available to date. Here a stable Se-nitrososelenol (RSeNO), a new class of NO derivative, was synthesized and fully characterized by X-ray crystallography and spectroscopic methods. This Se-nitrososelenol can be formed by direct transnitrosation from an S-nitrosothiol to a selenol, as is the case in the proposed mechanism for the NO-mediated inactivation of GPx.  相似文献

Topical natural antioxidants are a useful strategy for the prevention of photoaging and oxidative stress mediated skin diseases. In view of this underlying principle, the screening of natural plant extracts with scavenging activity for pro-oxidant reactive species is a primary requirement for the development of new topical antioxidant formulations. In the present study, an ethanol:water (7:3) extract from Castanea sativa leaves and a ethanol:water (2:3) extract from Quercus robur leaves were evaluated for their putative in vitro scavenging effects on reactive oxygen species (ROS) namely superoxide radical (O(2)(-)), hydroxyl radical (HO()), peroxyl radical (ROO()), hydrogen peroxide (H(2)O(2)) and singlet oxygen ((1)O(2)) as well as on reactive nitrogen species (RNS) namely nitric oxide (()NO) and peroxynitrite (ONOO(-)). The extracts presented a high potency to scavenge the tested reactive species, all the IC(50)s being found at the microg/mL level. IC(50)s (mean+/-SE) for the ROS O(2)(-),HO(),H(2)O(2) and (1)O(2) were 13.6+/-1.8; 216+/-4; 410+/-8; 12.3+/-0.7 microug/mL, respectively, for C. sativa, and 11.0+/-0.5; 285+/-22; 251+/-32; 7.90+/-0.56 microg/mL, respectively, for Q. robur. The ORAC values obtained for ROO() were 1.24+/-0.13 for C. sativa and 1.09+/-0.06 for Q. robur. The IC(50)s (mean+/-SE) for ()NO and ONOO(-) were 3.10+/-0.14 and 1.49+/-0.10 microg/mL, respectively, for C. sativa and 3.13+/-0.11 and 0.95+/-0.02 microg/mL, respectively, for Q. robur. The content of total phenolics for C. sativa and Q. robur were 284+/-9 and 346+/-4 mg of gallic acid equivalents (GAE)/g of lyophilized extract respectively. The observed effects might be of relevance considering the putative interest of these extracts as topical antioxidants.  相似文献

The kinetics of oxidation of three alpha beta-unsaturated compounds, CH2CHX (X=CN, CONH2 and CO2-) by bis(dihydrogentellurato)metallate(III) ions ([MIII-(H2TeO6)2]5-, M=Cu or Ag) have been studied in an alkaline medium. The reactions take place according to the rate expression: The k3 values are 2.42*10-2, 1.67*10-2 and 1.10*10-2mol6dm-2s-1 for oxidations of the respective substrate by the copper(III) complex and 1.49*10-2, 1.07*10-2 and 0.80*10-2mol6dm-2s-1, respectively, in the oxidations by silver(III), all at 303K. [CuIII(H2TeO6)2]5- reacts faster than the corresponding reactions with [AgIII(H2TeO6)2]5-. The oxidation rates follow the order: CN CONH2CO2-. The oxidations of the substrates by [CuIII(H2TeO6)2]5- take place by an outer-sphere mechanism, unlike [AgIII-(H2TeO6)2]5-, which reacts by an inner-sphere mechanism. The substrates are oxidized to diols under kinetic control and a tentative reaction mechanism leading to the formation of oxidation products is suggested. Condensation of methyl 7-methylcoumarin-4-acetate ( 2 ) with primary amines and with anthranilic acid gave 7-methyl-2-oxo-N-aryl-2H-[1]-benzopyran-4-acetamide ( 4a—d ) and (7), respectively. Compound 7 underwent cyclization to give 2-(7-methyl-2-oxo-2H-[1]-benzopyran-4-yl)-methyl-4H-3,1-benzoxazin-4-one ( 3 ). The reaction of 3 with aromatic amines gave the corresponding quinazolone derivatives 5 which tautomerises to the thermodynamically more stable isomer 6 , whereas its reaction with Grignard reagents and aromatic aldehydes gave 8a, 8b , and 9a, 9b , respectively.  相似文献

A procedure is described for the preconcentration of 100 ml of estuarine and seawater into a solid sample using Chelex-100 resin. This solid sample weighs less than half a gram and contains the transition metals and many other elements of interest, but is essentially free from the alkali metals, the alkaline earth metals, and the halogens. The concentrations of Co, Cr, Cu, Fe, Mn, Mo, Ni, Sc, Th, U, V and Zn have been determined in seawater when this procedure was coupled to neutron activation analysis.  相似文献
