In situ generated Ru(bpy)(3)(3+)-based chemiluminescence (CL) detection will be shown to be compatible with micellar electrokinetic chromatography (MEKC) providing significant advantages over other Ru(3+) generation protocols. The CL reagent, Ru(bpy)(3)(2+) is continuously added post-capillary to avoid precipitating the anionic surfactant used to enhance the separation of neutral analytes. Ru(bpy)(3)(3+) is then electrochemically generated in situ at the interface between the separation capillary and the working electrode, where it can react with specific analytes, for example amines and amino acids to produce chemiluminescent emission. With this scheme, the critical micelle concentration is not exceeded in the detection zone, freeing the analyte to react with the Ru(bpy)(3)(3+) CL reagent. The separation and detection of various underivatized amines will be demonstrated using this methodology. For triethylamine, 70 000 plates per meter are demonstrated with MEKC providing a limit of detection (S/N=2) of 1.5 fmol of injected mass. The experimental approach used to improve the limit of detection while maintaining high separation efficiency will be evaluated and discussed.  相似文献

Amino acids with different chemical structures have different abilities in terms of increasing the intensity of chemiluminescence (CL) of tris(2,2'-bipyridine)ruthenium(II) [Ru(bpy)3(2+)]. In a flow system, CL caused by the reaction between Ru(bpy)3(3+) and 15 amino acids was observed, but only tryptophan (Trp) and histidine (His) enhanced the intensity obviously, and so the CL of Trp and His and their molecular groups was studied. A calculation of the ionization potentials (IPs) of their N atom indicated that the CL intensities of these compounds depended on their IPs. In addition, the flow system was used for the determination of Trp and His, and the detection limits were 3 x 10(-8) mol L(-1) for His and 2.5 x 10(-9) mol L(-1) for Trp. The calibration curves for the two amino acids were 1.0 x 10(-7) to 5.0 x 10(-3) mol L(-1) for His and 1.0 x 10(-8) to 1.0 x 10(-4) mol L(-1) for Trp. The proposed approach was applied to the determination of His in Ganoderma.  相似文献

We demonstrate here for the first time the detection of electrogenerated chemiluminescence (ECL) upon selective, electrochemically triggered tris(2,2'-bipyridyl)ruthenium(II) (Ru(bpy)(3)(2+)) transport across a supported liquid membrane doped with a lipophilic cation-exchanger. This approach allows one to separate the location of ECL generation from the sample compartment, thereby avoiding the contamination of the sample with compounds necessary for ECL.  相似文献

Anodic Ru(bpy)32+ electrogenerated chemiluminescence (ECL) can be enhanced by polyphenols in alkaline solution. Spin trapping-electron spin resonance (ESR) experiments verified that reactive oxygen species (ROS) were generated during the electrolysis of Ru(bpy)32+ in alkaline solution, and oxidation of quercetin enhanced Ru(bpy)32+ ECL at anodic potential by producing additional ROS. This ECL enhancement can be used to analyze real sample and evaluate antioxidant activity of polyphenols.  相似文献

The electrochemical and electrogenerated chemiluminescence of Ru(bpy)(3)(2+) immobilized in [clay/Ru(bpy)(3)(2+)](n) multilayer films by layer-by-layer assembly were investigated. The stable multilayer films of clay and Ru(bpy)(3)(2+) were assembled by alternate adsorption of negatively charged clay platelets and positively charged Ru(bpy)(3)(2+) from their aqueous dispersions. UV-vis spectroscopy, quartz crystal microbalance (QCM), cyclic voltammetry, and electrogenerated chemiluminescence (ECL) were used to monitor the immobilization of Ru(bpy)(3)(2+) and the regular growth of the [clay/Ru(bpy)(3)(2+)](n) multilayer films. The multilayer films modified electrode was used for the ECL detection of tripropylamine (TPA) and oxalate. The proposed novel immobilized method exhibited good stability, reproducibility and high sensitivity for the determination of TPA and oxalate, which mainly resulted from the contributing of clay nanoparticles with appreciable surface area, special structural features and unusual intercalation properties. Detection limits were 20 and 100 nM for TPA and oxalate, respectively and the linear concentration range extended from 60 nM to 0.66 mM for TPA.  相似文献

The electrochemiluminescent (ECL) response of allopurinol was studied in aqueous media over a wide pH range (pH 2–13) using flow injection (FI) analysis. It was revealed that allopurinol itself had no ECL activity, but could greatly enhance the ECL of Ru(bpy)32+ in alkaline media giving rise to a sensitive FI-ECL response. The effects of experimental conditions including the mode of applied voltage signal, the potential of working electrode, pH value, the flow rate of carrier solution, and the concentration of Ru(bpy)32+ and allopurinol on the ECL intensity were investigated in detail. The most sensitive FI-ECL response of allopurinol was found at pH 12.0, where the FIA-ECL intensity showed a linear relationship with concentration of allopurinol in the range 1 × 10−8 mol L−1 to 5 × 10−7 mol L−1, and the detection limit was 5 × 10−9 mol L−1.  相似文献

Eleven different Ru(2)(4+) and Ru(2)(3+) derivatives are characterized by thin-layer FTIR and UV-visible spectroelectrochemistry under a CO atmosphere. These compounds, which were in-situ electrogenerated from substituted anilinopyridine complexes with a Ru(2)(5+) core, are represented as Ru(2)(L)(4)Cl where L = 2-CH(3)ap, ap, 2-Fap, 2,3-F(2)ap, 2,4-F(2)ap, 2,5-F(2)ap, 3,4-F(2)ap, 3,5-F(2)ap, 2,4,6-F(3)ap, or F(5)ap. The Ru(2)(5+) complexes do not axially bind CO while mono- and bis-CO axial adducts are formed for the Ru(2)(4+) and Ru(2)(3+) derivatives, respectively. Six of the eleven investigated compounds exist in a (4,0) isomeric form while five adopt a (3,1) geometric conformation. These two series of compounds thus provide a large enough number of derivatives to examine trends and differences in the spectroscopic data of the two types of isomers in their lower Ru(2)(4+) and Ru(2)(3+) oxidation states. UV-visible spectra of the Ru(2)(4+) derivatives and IR spectra of the Ru(2)(3+) complexes under CO are both isomer dependent, thus suggesting that these data can be used to reliably predict the isomeric form, i.e., (3,1) or (4,0), of diruthenium complexes containing four unsymmetrical substituted anilinopyridinate bridging ligands; this was confirmed by X-ray crystallographic data for seven compounds whose structures were available.  相似文献

A novel high performance liquid chromatography-chemiluminescence(HPLC-CL) method for investigation of in vitro metabolism of repaglinide in pig liver microsomes with microdialysis sampling technique was developed.The chromatographic separation was performed on a Hypersil BDS-C18 column with an isocratic mobile phase consisting of methanol and 0.01 mol/L KH2PO4(pH 3.0)(volume ratio 75:25) at a flow rate of 1.0 mL/min.The detection was based on the chemiluminescence reaction of repaglinide with acidic potassium permanganate(KMnO4) and tris(2,2’-bipyridyl)ruthenium(Ⅲ)(Ru(bpy)33+),which was immobilized on the cationic ion-exchange resin for obtaining high sensitivity and reducing consumption of expensive reagent.  相似文献

We explored the CE with (bpy) (2+) (3) electrochemiluminescence detection for the kinetic study of drug-enzyme interaction. Effects of four nonsteroidal anti-inflammatory drugs including aspirin, paracetamol, sodium salicylate and phenacetin on prolidase (PLD) activity in erythrocytes were investigated. Aspirin enhanced PLD activity whereas the other three had inhibiting effects. This may reveal their different effects on the collagen biosynthesis and catabolism that influence tumor invasiveness. Kinetic study of paracetamol on PLD showed that the value of Michaelis constant K(m) for PLD was 1.23 mM. The mechanism of PLD inhibition by paracetamol is noncompetitive inhibition, and the inhibitor constant K(i) value obtained in our research was 9.73 x 10(3) microg/L.  相似文献

An HPLC method is described for the electrochemiluminescence (ECL) detection of amino acids, following cycloaddition reaction of their amino groups with divinyl sulfone (DVS), using electrogenerated tris(bipyridine)ruthenium(III). The derivatization reaction conditions were examined, with the optimum conditions found to be 40 mM DVS (pH 8.0) at 50 degrees C for 15 min. Detection limits for the 15 amino acids examined varied greatly (0.04-8.0 pmol) using a standard solution by flow injection analysis (FIA). These optimized conditions were used for HPLC determination of the amino acids in human plasma. A linear relationship was obtained up to 100 pmol on a column for aromatic and branched-chain amino acids. Recoveries of Tyr, Met, Val, Leu, Ile, Phe and Trp when added to human plasma (1 micromol/10 ml plasma, n=5) were 101.5+/-1.1, 99.0+/-1.2, 98.0+/-1.4, 101.1+/-1.6, 95.1+/-1.6, 99.2+/-1.5 and 97.7+/-1.3 % (mean+/-S.D.) respectively. The concentrations of the amino acids in the plasma are in good agreement with other published data.  相似文献

In this work, a stable electrogenerated chemiluminescence (ECL) detector was developed. The detector was prepared by packing cation-exchanged resin particles in a glass tube, followed by inserting Pt wires (working electrode) in this tube and sealing. The leakage of Ru(bpy)32+ can be compensated by adding a small amount of Ru(bpy)32+ into solution phase. Coupled with high-performance liquid chromatography separation, the detector has been used for determination of itopride hydrochloride in human serum. Under the optimal conditions, the ECL intensity has a linear relationship with the concentration of itopride hydrochloride in the range of 1.0 × 10−8 g mL−1 to 1.0 × 10−6 g mL−1 and the detection limit was 3 × 10−9 g mL−1 (S/N = 3). The as-prepared ECL detector displayed good sensitivity and stability.  相似文献

Chemiluminescence emission intensity is modulated by modulating the potential of a working electrode which is used to generate a key species in the electrogenerated Chemiluminescence (ECL) reaction. The emission is monitored synchronously using a lock-in amplifier. The reactions used in the characterization are luminol with hydrogen peroxide and tris(2,2-bipyridyl)ruthenium (II) (or Ru(bpy) 3 2+ ) with oxalate. Modulation widths of ± 50 mV yield maximum signals for luminol when centered at 0.45 V (vs Ag/AgCl) and for Ru(bpy) 3 2+ when centered at 1.05 V. The resulting signal decreases with increasing modulation frequency and shows that luminol/H2O2 is a faster ECL system than Ru(bpy) 3 2+ /oxalate. Working curves for luminol and for oxalate have essentially the same linear range and slope with the modulated potential approach as with a DC electrode potential. This approach provides capability for differentiating the analytical signal from constant background emission or stray light.  相似文献

Melatonin and some of its important derivatives were found to be able to enhance the ECL of Ru(bpy)32+ in an alkaline Britton–Robinson buffer solution. The optimum conditions for the enhanced ECL, such as the selection of applied potential mode, type of buffer solution, pH effect and effect of Ru(bpy)32+ concentration have been investigated in detail in this paper. Under the optimum conditions, the enhanced ECL is linear with the concentration of melatonin and its derivatives over the wide range, and the detection limit for these compounds was found to be in the range of 5.0 × 10−8 to 1.0 × 10−10 mol L−1. The proposed procedure was applied for the determination of drug in tablets with recoveries of 85–93%. A possible mechanism for the enhanced ECL of Ru(bpy)32+ by melatonin and its derivatives was proposed, and the relationship between molecular structure of melatonin and its derivatives and the enhanced ECL behavior was also discussed.  相似文献

The use of cyclen (1,4,7,10-tetraazacyclododecane) as a blocking ligand enables assembly of the mixed-valence square complex [(cyclen)4Ru4(pz)4]9+ (pz = pyrazine). A crystal structure determination shows the molecule to possess a regular square geometry wherein each Ru atom has an equivalent coordination environment. Consistent with the presence of one RuIII and three RuII centers, cyclic voltammetry reveals a single reversible reduction wave and three successive oxidation waves. The separation between the first oxidation and reduction waves indicates a comproportionation constant of Kc = 108.9 for the [(cyclen)4Ru6(pz)4]9+ square, suggesting a greater extent of electron delocalization than that observed for the Creutz-Taube ion. The closer spacing between oxidation waves suggests a lesser degree of delocalization in the [(cyclen)4Ru6(pz)4]10+ (Kc = 102.0) and [(cyclen)4Ru6(pz)4]11+ (Kc = 103.0) species, bearing the higher average oxidation states of Ru2.5+ and Ru2.75+, respectively.  相似文献
