A series of derivatives of tetrandrine and fangchinoline was designed and synthesized to find more active anti-cancer compounds. Their anti-cancer activities were tested against human hcpatocellular carcinoma BEL-7402 and PLC/PRF/5 cells, human lung adenocarcinoma A549 cells as well as human leukaemia K562 cells, and the structure-activity relationship(SAR) was also studied. All the compounds except BI exhibited superior inhibitory ac- tivities against PLC/PRF/5 cells with half maximal inhibitory conccntration(ICs0) values of less than 15 μmol/L, and compounds A2, A4, B2 and B4 showed IC50 values of less than 9 pmol/L. Compounds A2, A6, B2 and B4 showed potent anti-cancer activities against all the tested cells with 1C5o values of less than 10 pmol/L. The results show that terandrine and fangchinoline derivatives are potential suppressors to human cancer.  相似文献

在已知具有氧化作用的2-苯基-1,2-苯并异硒唑-3(2H)-酮(Ebselen)的基础上, 将其2-位苯环更换成氨基酸乙酯, 设计并合成了9个苯并异硒酮的氨基酸衍生物3~11. 这类化合物的抗氧化作用药理研究表明,它们对Fe^2^+/半胱氨酸及维生素C诱发的大鼠肝微粒体脂质过氧化有抑制活性, 部分化合物活性比母体化合物Ebselen更优.  相似文献

75%aqueous ethanol extract from the whole herbs of Selaginella doederleinii was isolated,and two new apigenin derivatives,doederflavones A(1) and B(2),together with ten known compounds(3-12) were characterized.Their structures were assigned by extensive spectroscopic methods including 1D/2D NMR and HR-ESIMS.Compounds 1-6 bear an aryl substituent at the C-8 or C-6 positions in ring A of apigenin skeleton.Compounds 1 and 2 were evaluated for their in vitro cytotoxicity against four human cancer cell lines A549,MCF-7,SMMC-7721,and LoVo,both of which exhibited significant cytotoxicity against A549 with IC_(50) values of 0.82 μmol/L and 1.32 μmol/L,respectively.  相似文献

Phytochemical investigation on the roots and rhizomes of Notopterygium incisum led to the isolation of a new polyacetylene, notopolyenol A (1), along with thirteen known analogues (2-14). Their structures were elucidated by extensive analyses of NMR and HRMS data, and the absolute configuration of 1 was unambiguously determined as 3 R by comparison of its retention time and ECD curve with those of synthetic enantiomers (-)-1 and (+)-1, whose absolute configurations were established by using the modified Mosher's method. Subsequent activity screening revealed that (3 S)-1 exhibited the most significant cytotoxicity against MCF-7, H1299, and HepG2 cancer cells with IC_(50) values of 1.3 μmol/L,0.6μmol/L and 1.4μmol/L, respectively.  相似文献

One new 6,7-seco-enr-kaurane diterpenoid,sculponin T(1),was isolated from the aerial parts of Isodon sculponeatus,along with four known analogs,sculponeatin J(2),sculponeatin K(3),sculponeatin C(4),and sculponeatin Q(5).Their structures were elucidated by extensive spectroscopic analysis and by comparison with data reported in the literature.Significant cytotoxic activity was observed for compound 2 against five human tumor cell lines with IC_(50) values ranging from 1.8 μmol/L to 3.3 μmol/L,and it also inhibited NO production in LPS-stimulated RAW264.7 cells,with IC_(50) value of 3.3 μmol/L.  相似文献

An efficient synthesis of novel 3-(piperidin-4-yl)isoxazolo[4, 5-d]pyrimidine scaffold has been designed and deveopled. A series of 5-phenylurea derivatives was synthesized using this method. Their cytotoxic activities against breast cancer cell line BT-474 were evaluated by CCK-8 assay. Most of them showed potent anti-proliferative activities, of which compound 20 and 21 exhibited IC50s of 1.565 μmol/L and 1.311 μmol/L, respectively. Furthermore, compound 20 and 21 also showed potent inhibitory activities against PI3Kδ with IC50s of 0.286 μmol/L and 0.452 μmol/L, respectively. These results indicate that these 3-(piperidin-4-yl)isoxazolo[4, 5-d] pyrimidine derivatives are novel antitumor agents through the inhibition of PI3Kδ.  相似文献

Nine ethyl N-hydroxy-N-(substituted)phenyloxamates(2a—2i), based on the structure of the KARI inhibitor IpOHA, were synthesized. Their structures were established on the bases of 1H NMR, IR, ESI-MS and elemental analyses. Among the nine compounds, four show in vitro inhibitory activity on rice KARI, with 2c and 2g[Ki values of (35.2±4.9) and (49.4±6.3) μmol/L] having similar activity to the precursor of IpOHA(A, Ki=34.1±6.7 μmol/L). The results will be helpful to further molecular design of more potent KARI inhibitors.  相似文献

A new triterpenoid saponin.3 β-O-{ β-D-glucopyranosyl-(1—4)-[ β-D-glucopyranosyl-[1-2)]- α-L-arabinopyranosyl}-16α, 28-dihydroxyolean-12-en-30-oic acid 30-O- β-D-glucopyranosyl ester(ardisicrenoside N.1),together with two known saponins, ardisicrenoside C(2) and D(3),were isolated from the roots of Ardisia crenata Sim.Their structures were elucidated by extensive spectral analysis and chemical evidences.Saponins 1 showed cytotoxicity against MCI-7 and NCI-H460 cancer cell lines at 11.0μmol/L and 22.1μmol/L in vitro.  相似文献

Four new diterpenoid alkaloids,nagaconitines A-D(1-4),were isolated from the roots of Aconitum nagarum var.heterotrichum.Their structures were elucidated as 8-(4'-hydroxylbutyryl) aconitine(1),15-oxo-13-deoxyludaconitine(2),15-hydroxyldelphisine(3),and 14-hydroxyl-2- acetoxyspiradine C(4)based on extensive UV,IR,MS,1D NMR and 2D NMR analyses.Compounds 3-4 showed inhibition of cancer cell line SK-OV-3 with 31%(IC_(50) 43.78 μmol/L) and 24%(IC_(50) 32.14 μmol/L),respectively.  相似文献

根据活性亚结构拼接原理,通过紫罗兰酮与(取代)苯甲醛反应合成了紫罗兰酮基双查尔酮,然后经与氨基硫脲缩合得到一系列未见报道的新型含紫罗兰酮、查尔酮及氨基硫脲3种优势结构单元的杂化体,它们的化学结构经傅里叶变换红外光谱(FT-IR)、核磁共振波谱(~1H NMR、~(13)C NMR)、元素分析及质谱(MS)等测试技术所证实。采用溴化噻唑蓝四氮唑(MTT)法初步测定其体外抗肿瘤活性(乳腺癌细胞(MCF-7),肝癌细胞(Hep G2),肺癌细胞(A549)),结果表明,对于不同类型的肿瘤细胞,化合物展现较好的增殖抑制活性。尤其是化合物3a与3b对MCF-7细胞展现较强的抗增殖活性,半数致死量(IC_(50))值分别为10.83和7.62μmol/L,化合物3e对A549细胞显示一定的增殖抑制活性效果(IC_(50)值为13.36μmol/L),化合物3f对Hep G2细胞表现了高效的抗增殖活性(IC_(50)值为8.55μmol/L)。目标物的抗增殖活性与紫罗兰酮结构及查尔酮环上不同电子效应的取代基有关。  相似文献

通过人工发酵培养,从南海红树林内源真菌Fusarium sp.ZZF60的培养液中分离得到:1种新蒽醌衍生物,6,8-二甲氧基-1-甲基-2-(3-氧丁基)蒽醌(1),以及5种已知化合物:7-羟基-3-(4-甲氧基苯基)苯并-γ-吡喃酮(2),2,4-二羟基-6-[(1′E,3′E)-1′,3′-戊二烯基]苯甲醛(3),(E)-4-羟基肉桂酸甲酯(4),4-(4-羟基苯基)-2-丁醇(5),4-羟基苯甲酸(6)。它们的结构通过MS、NMR等波谱分析推导确定。初步药理活性显示,化合物1抑制体外培养人喉癌细胞Hep2和人肝癌细胞HepG2的IC50分别为16和23μmol/L。  相似文献

正A series of 2,6-disubstituted-4-morpholinothieno[3,2-d]pyrimidine derivatives were synthesized and their cytotoxic activity against H460,HT-29,MDA-MB-231,U87MG and H1975 cancer cell lines were evaluated in vitro.Most of the target compounds exhibited moderate to excellent activity to the tested cell lines.The most promising compound 23(0.84μmol/L,0.23μmol/L, 2.52μmol/L,1.80μmol/L) was 1.0,2.9,29.3 and 4.3 times more active than GDC-0941(0.87μmol/L,0.66μmol/L,73.8μmol/L, 7.77μmol/L) against H460,HT-29,MDA-MB-231 and U87MG cell lines,respectively.  相似文献

Periconones B-E(1-4),four new polyketide-terpenoid hybrid molecules were isolated from the endophytic fungus Periconia sp.F-31.Their structures and absolute configurations were established by extensive spectroscopic data analysis and electronic circular dichroism(ECD).Compound 4 exhibited in vitro cytotoxic activity against the human MCF-7 tumor cell line with an IC_(50) value of 4.2 μmol/L,and compound 1 displayed anti-HIV activity with an IC_(50) value of 18.0 μmol/L.  相似文献

A new sesquilignan, 7',8'-didehydroherpetotriol (1), and a new lignan glycoside, (+)-isolariciresinol-9'-O-α-L-rhamnopyra- nosy1-(1→6)-[3-D-glucopyranoside (2), were isolated from the branches of Dipteronia dyeriana. An ultrasensitive electrochemical biosensor was fabricated for electroanalytical determination of ascorbic acid(AA), dopamine(DA) and uric acid(UA) individually and simultaneously based on polypyrrole hollow nanotubes loaded with Au and Fe3O4 nanoparticles(NPs) uniformly(PPy@Au-Fe3O4). The PPy@Au-Fe3O4 nanotubes were synthesized in one-pot using MoO3 nanorods as templates and the polymerization of Py, the formation of Au and Fe3O4 NPs and the removel of MoO3 templates took place stimultaneously. Electrochemical studies reveal that PPy@Au-Fe3O4modified glassy carbon electrode(GCE) possesses excellent electro-catalytic activities toward the oxidation of AA, DA and UA. Their oxidation peak currents increase linearly in the concentration ranges of 1-2000 μmol/L for AA, 0.01-25 and 25-300 μmol/L for DA and 0.1-300 μmol/L for UA. Their detection limit values(S/N=3) were calculated as 0.45, 0.0049, and 0.051 μmol/L for AA, DA and UA in the individual detection. By changing the concentrations simultaneously, the calibration curves showed linearity to 1000, 200, and 200 μmol/L with detection limit of 0.39, 0.0060, and 0.060 μmol/L for AA, DA, and UA, respectively. Finally, the obtained biosensor was successfully applied to the detection of AA, DA, and UA with satisfactory results on actual samples.  相似文献

合成了两种稀土高氯酸盐与L 脯氨酸配合物的晶体.经热重、差热、化学分析及对比有关文献,知其组成是[Pr2(L Pro)6(H2O)4](ClO4)6和[Er2(L Pro)6(H2O)4](ClO4)6,质量分数为99.24%和98.20%.选用RE(NO3)3•6H2O(RE=Pr,Er)、L Pro、NaClO4•H2O和NaNO3作辅助物,使用具有恒温环境的反应热量计,以2 mol•L-1 HCl作溶剂,分别测定了[2RE(NO3)3•6H2O+6L Pro+6NaClO4•H2O]和{[RE2(L PrO)6(H2O)4](ClO4)6+6NaNO3}在298.15 K时的溶解热.设计一热化学循环求得化学反应的反应焓ΔrHm分别是:63.904 kJ•mol-1和91.017 kJ•mol-1,经计算得配合物[RE2(L Pro)6(H2O)4](ClO4)6(s)在298.15 K时的标准生成焓ΔfHm(298.15 K)分别是-6 594.78 kJ•mol-1和-6 532.87 kJ•mol-1.  相似文献

3种香叶基黄烷酮是从毛泡桐(Paulownia tomentosa)水果中分离出的3种具有相似结构的黄酮类化合物。我们以廉价的2,4,6-三羟基苯乙酮,羟基苯甲醛为原料经过取代、氯甲基甲基醚保护、羟醛缩合、分子内的麦克加成反应以及脱甲基甲基醚合成了3个香叶基取代黄烷酮。其结构经过核磁共振氢谱及碳谱(1H NMR,13C NMR)和高分辨质谱(HRMS)等技术手段进行了表征,并采用四氮唑蓝(MTT)法测试了合成化合物对肺癌细胞A459的体外抗肿瘤活性。结果表明,所合成的化合物均具有一定的抗肺癌细胞活性,其中化合物2抗肺癌活性明显优于化合物1和3,其半数抑制浓度为25.6μmol/L,为临床疾病的治疗和药物的研发提供了参考。  相似文献
