To control the depolymerization process of poly(l-lactic acid) into l,l-lactide for feedstock recycling, the racemization of l,l-lactide as a post-depolymerization reaction was investigated. In the absence of a catalyst, the conversion to meso-lactide increased with increase in the heating temperature and time at a higher rate than the conversion into oligomers. The resulting high composition of meso-lactide suggests that the direct racemization of l,l-lactide had occurred in addition to the known racemization mechanism that occurs on the oligomer chains. In the presence of MgO, the oligomerization rapidly proceeded to reach an equilibrium state between monomers and oligomers. The equilibrium among l,l-, meso-, and d,d-lactides was found to be a convergent composition ratio l,l-:meso-:d,d-lactides = 1:1.22:0.99 (wt/wt/wt) after 120 min at 300 °C. This composition ratio also indicates that in addition to the known racemization reaction on the oligomer chains, direct racemization among the lactides is also a frequent occurrence.  相似文献

Speed of sound and density values for ternary systems (amino acid + salt + water): l-isoleucine/l-proline/l-glutamine in aqueous solutions of 1.5 M KCl, 1 M KNO3, and 0.5 M K2SO4 have been measured for several concentrations of amino acids at different temperatures (303.15, 308.15, 313.15, 318.15, and 323.15 K). Using speed of sound and density data, the thermodynamic parameters such as isentropic compressibility (κs), change in isentropic compressibility (Δκs) and relative change (Δκs/κ0) in isentropic compressibility have been computed. The isentropic compressibility values decrease with increase in the amino acid concentration as well as with temperature. The decrease in κs values with increase in concentration of l-isoleucine/l-proline/l-glutamine in 1.5 M KCl, 1 M KNO3, and 0.5 M K2SO4 has been ascribed to an increase in the number of incompressible zwitterions in solutions, and the formation of ‘zwitterions-ions’ and ‘zwitterions-water dipole’ entities in solutions. The decrease in κs values with increase in temperature has been attributed to the corresponding decrease of κrelax (a relaxational part of compressibility), which is dominant over the corresponding increase of κ (an instantaneous part of compressibility). The trends of variation of Δκs and Δκs/κ0 with variations in solute concentration and temperature have also been discussed in terms of solute-solute and solute-solvent intermolecular/interionic interactions operative in the systems.  相似文献

Low molecular weight poly(lactic acid) was synthesized by direct polycondensation of lactic acid. The oligomers were characterized by viscometry, light scattering, and gel permeation chromatography (GPC). The swelling behaviour of tablets made of the above polymer immersed in buffer solutions at 37 °C was studied. In the same experiments, the hydrolytic stability of d,l-PLA was assessed by measuring the weight loss after drying the tablets. In order to inhibit any degradation due to bacteria, formaldehyde was added in the solution as biostatic factor. The effect of an incorporated drug on the swelling behaviour of d,l-PLA tablets was also considered. It was found that the incorporation of drug in d,l-PLA tablets increases their swelling index, probably due to the creation of additional porosity in the specimens or other interaction between drug and polymeric matrix.  相似文献

A method for the quantification of total d-gluconate by flow-injection analysis was developed using an immobilized-enzyme reactor and fluorescence detection. d-Gluconate was quantified using co-immobilized gluconate kinase (GK) and 6-phosphogluconate dehydrogenase (PGDH) reactor. d-Gluconate was phosphorylated to 6-phospho-d-gluconate by GK in the presence of ATP, and then the 6-phospho-d-gluconate produced was oxidized by PGDH with NADP+. The NADPH produced by the GK-PGDH reactor was monitored fluorometrically at 455 nm (excitation at 340 nm). A linear relationship between the responses and concentrations of d-gluconate was obtained in the ranges of 1.0 × 10−6-1.6 × 10−4 M. The relative standard deviation for 10 successive injections was 0.57% at the 0.1 mM level. This analytical method was applied to the quantification of d-gluconate in honeys, vinegars and noble rot wines, and the results showed good agreement with those obtained using the conventional F-kit method.  相似文献

Films of poly(l-lactic acid) (PLLA) with different number-average molecular weights (Mn) and d-lactide unit contents (Xd) were made amorphous and the effects of molecular weight and small amounts of d-lactide units on the hydrolytic degradation behavior in phosphate-buffered solution at 37 °C of PLLA were investigated. The degraded films were investigated using gravimetry, gel permeation chromatography, polarimetry, differential scanning calorimetry, X-ray diffractometry, and tensile testing. To exclude the effects of crystallinity on the hydrolytic degradation, the films were made amorphous by melt-quenching. The incorporation of small amounts of d-lactide units drastically enhanced the hydrolytic degradation of PLLA. In the period of 0-32 weeks, the hydrolytic degradation rate constant (k) of PLLA films increased with increasing Xd, while the k values did not depend on Mn. This means that the effects of Xd on the hydrolytic degradation rate of the films are higher than those of Mn. In contrast, in the period of 32-60 weeks neither Xd nor Mn was a crucial parameter to determine k values, probably because in addition to these parameters the differences in the amount of catalytic oligomers accumulated in films and crystallinity affect the hydrolytic degradation behavior of the films. The initially amorphous PLLA films remained amorphous even after the hydrolytic degradation for 60 weeks.  相似文献

The predominant mechanism of the hydrolytic degradation of oligo(d,l-lactide)-grafted dextrans in phosphate buffer was followed by quantifying both released dextran and lactic acid from the copolymers. The studied amphiphilic copolymers, with well-defined structure, exhibited various oligo(d,l-lactide) weight fractions (FOLA) while having a quite high extent of free hydroxyl groups (>90%). Depending on their FOLA, oligo(d,l-lactide)-grafted dextrans were soluble either in water or in organic solvents (THF, toluene, …) and different prevailing mechanisms of hydrolytic degradation were observed. The copolymer soluble in THF, with longer oligo(d,l-lactide) grafts and higher FOLA, was found to degrade via a particular mechanism by which the greatest part of dextran was released into buffer medium during the first two weeks of degradation. During the initial stage of degradation, the hydrophilicity of dextran backbone was considered to be the main driving force for the hydrolytic cleavage of the ester linkage between backbone and grafts. Released oligo(d,l-lactide) grafts were found to be degraded via chain-end degradation or random degradation depending on their solubility in buffer medium. In case of water-soluble copolymers with shorter oligo(d,l-lactide) grafts and lower FOLA, the chain-end degradation was exclusively observed.  相似文献

Water-soluble CdTe quantum dots (QDs) with five sizes (2.25, 2.50, 2.77, 3.12, and 3.26 nm) were synthesized with the hydrothermal method. The electrochemiluminescence (ECL) of CdTe QDs was investigated in detail in air-saturated solution without adding foreign oxidant. It was found that the ECL of CdTe QDs displayed a size-dependent property. With the increasing in the particle size of the CdTe QDs, the ECL intensity was gradually increased, in addition, both ECL peak potentials and ECL onset potentials of CdTe QDs were shifted positively. Influences of some factors on the ECL intensity were investigated. Under the optimal conditions, the ECL intensity had a linear relationship with the concentration of l-cysteine (l-Cys) in the range from 1.3 × 10−6 to 3.5 × 10−5 mol L−1 (R2 0.996) with a detection limit of 8.7 × 10−7 mol L−1 (S/N = 3). The proposed method was applied to the determination of l-Cys in real samples with satisfactory results. Compared with previous reports, it has better selectivity for the determination of l-Cys.  相似文献

5-Deoxy-5-phospho-d-ribonohydroxamic acid, a mimic of the 1,2-cis-enediolate high-energy intermediate species of the allose-6-phosphate isomerase reaction, was obtained by a six-step synthesis from d-erythronolactone. In contrast to the known competitive ribose-5-phosphate isomerase (Rpi) inhibitors 4-deoxy-4-phospho-d-erythronohydroxamic acid, 4-deoxy-4-phospho-d-erythronate, and 4-deoxy-4-phosphonomethyl-d-erythronate, the new hydroxamic acid selectively inhibits Mycobacterium tuberculosis RpiB (Ki = 0.40 mM, Km/Ki = 4.5) versus Spinacia oleracea RpiA, and hence appears as a promising lead for the design of potent species-specific inhibitors of the bacterial enzyme.  相似文献

Screening of a combinatorial CTV-based artificial, synthetic receptor library 1 {1-13, 1-13, 1-13} for binding of a variety d-Ala-d-Ala and d-Ala-d-Lac containing ligands (6-11) was carried out in phosphate buffer (0.1 N, pH=7.0). After screening and Edman sequencing, synthetic receptors were found containing amino acid sequences, which are either characteristic for binding dye labeled d-Ala-d-Ala or d-Ala-d-Lac containing ligands. For example, receptors capable of binding d-Ala-d-Ala containing ligands 6, 7, 9 and 11 contained—almost in all cases—at least one basic amino acid residue—predominantly Lys—in their arms. This was really a striking difference with the arms of the receptors capable of binding d-Ala-d-Lac containing ligands 8 and 10, which usually contained a significant number of polar amino acids (Gln and Ser), especially in ligand 8, but hardly any basic amino acids. Use of different (fluorescent) dye labels showed that the label has a profound, albeit not decisive, influence on the binding by the receptor. A hit from the screening of the CTV-library with FITC-peptidoglycan (6) was selected for resynthesis and validation.  相似文献

Ring-opening polymerization of d,l-lactide initiated with new chlorotris(bis(trimethylsilyl) amido) tin(IV), tetrakis(bis(trimethylsilyl)amido) tin(IV) and titanium(IV) was investigated. New complexes reveal practically quantitative conversion degrees and produced polymers with higher molecular weight with respect to reference alkoxo-species.The X-ray crystal structure of chlorotris(bis(trimethylsilyl)amido) tin (IV) was investigated. Axial enantiomerism of [SnCl{N(SiMe3)2}3] molecules in solution was studied by high-field dynamic NMR, the value of Gibbs activation energy ΔG = 59.5 kJ/mol.Field emission SEM investigation of bulk polymer samples and thin films on conductive Al foil revealed a stratified fibrous textures in the bulk polymers, as well as nanoscaled topographical features due to coils and entanglements of individual macromolecules in thin films.  相似文献

Trace amounts of l-cysteine can function as a trigger, i.e., reaction initiator, in the autocatalytic sodium sulfite/hydrogen peroxide reaction system. Rapidly changing of pH after induction time is visually confirmed by color changing of bromothymol blue in this autocatalytic reaction. Based on this finding, μg L−1 levels of l-cysteine were measured over time using the autocatalytic reaction system. The determination range using the above method was 5.0 × 10−8-2.5 × 10−6 M, the detection limit (3σ) was 1.8 × 10−8 M (1.94 μg L−1), and the relative standard deviation was 2.41% at an l-cysteine concentration of 5 × 10−7 M (n = 5). This method was also applied to length detection-flow injection analysis. The determination range for the flow injection analysis was 2.0 × 10−7-1.0 × 10−5 M. The detection limit (3σ) was 1.4 × 10−7 M (17.0 μg L−1), and the relative standard deviation was 0.91% at an initial l-cysteine concentration of 10−6 M (n = 5).  相似文献

A rapid method for the identification and quantification of l-ascorbic acid in wines by direct injection liquid chromatography equipped with a UV detection was developed. The levels of ascorbic acid were determined using a polymeric PLRP-S 100 A (5 μm) column (150 mm × 4.6 mm) with a mobile water/trifluoroacetic acid (99/1, v/v) phase. The method is rapid (less than 5 min) and sensitive (LOQ of 5 mg L−1). The calibration curve of ascorbic acid was linear (r = 0.999) over a concentration range between 1 and 200 mg L−1. Repeatability was less than 2.5% and the recovery over 95%.  相似文献

Ultrasonic velocity and density values have been measured for ternary systems (amino acid/di-peptide + salt + water): l-leucine/l-asparagine/glycylglycine each in 1.5 M aqueous solutions of NaCl or NaNO3 or KNO3 used as solvents for several concentrations of amino acids/di-peptide at different temperatures in the range of 298.15-323.15 K. The ultrasonic velocity values have been found to increase with increase in amino acids/di-peptide concentration and temperature in all the systems. The increase in ultrasonic velocity with increase in concentration has been discussed in terms of electrostatic interactions occurring between terminal groups of zwitterions (NH4+ and COO) and Na+, K+, Cl, NO3 ions. The interactions of water dipoles with cations/anions and with zwitterions have also been taken into consideration. It has been observed that the ion-zwitterion and ion-dipole attractive forces are stronger than those of ion-hydrophobic repulsive forces. These interactions comprehensively introduce the cohesion into solutions under investigation. The cohesive forces are further enhanced on successive increases in solute concentration. Using ultrasonic velocity and density data, the parameters such as isentropic compressibility (κs), change (Δκs) and relative change (Δκs/κ0) in isentropic compressibility, specific acoustic impedance (Z) and relative association (RA) have been computed. The isentropic compressibility values decrease with increase in the concentration of solutes as well as with temperature. The decrease in κs values with increase in concentration of l-leucine, l-asparagine and glycylglycine in 1.5 M aqueous solutions of NaCl, NaNO3 and KNO3 have been explained in terms of an increase in the number of incompressible molecules/zwitterions in solutions and the formation of compact zwitterions-water dipole and zwitterions-ions structures in solutions. The decrease in κs values with increase in temperature has been attributed to the corresponding decrease of κrelax. (relaxational part of compressibility), which is dominant over the corresponding increase of κ (instantaneous part of compressibility). Poly(d,l-lactide) (PDLLA) degraded at processing temperature under air and nitrogen. A random chain scission model was established and used to determine the activation energy Ea, and FT-IR, 1H and 13C NMR were used to elucidate the degradation behavior under different atmospheres. Results showed that there were two to three stages. The 1st stage was dominated by the oligomers containing carboxylic acid groups and hydroxyl groups, during which oxygen and nitrogen had little effect on the degradation, thus they share similar Ea. When the oligomers were consumed over or evaporated, the 2nd stage began, and oxygen had a promoting effect on the thermo-oxidation process, resulting in the great decrease in Ea. The third stage of PDLLA was observed when it degraded under nitrogen over 200 °C, which was caused by the appearance of carboxylic acid substance.  相似文献

Triptolide (TP), which has immunosuppressive effect, anti-neoplastic activity, anti-fertility function and severe toxicities on digestive, urogenital, blood circulatory system, was used as a model drug in this study. TP-loaded poly (d,l-lactic acid) (PLA) nanoparticles were prepared by the modified spontaneous emulsification solvent diffusion method (modified-SESD method). Dynamic light scattering system (DLS), transmission electron microscope (TEM), atomic force microscopy (AFM), differential scanning calorimetry (DSC), X-ray powder diffractometry and Fourier transform infra-red spectroscopy (FT-IR) were employed to characterize the nanoparticles fabricated for size and size distribution, surface morphology, the physical state of drug in nanoparticles, and the interaction between the drug and polymer. Encapsulation efficiency (EE) and the in vitro release of TP in nanoparticles were measured by the reverse phase high-performance liquid chromatography (RP-HPLC). The produced nanoparticles exhibited a narrow size distribution with a mean size of approximately 150 nm and polydispersity index of 0.088. The morphology of the nanoparticles exhibited a fine spherical shape with smooth surfaces without aggregation or adhesion. TP-entrapped in nanoparticles was found in the form of amorphous or semicrystalline. It was found that a weak interaction existed between the drug and polymer. In all experiments, more than 65% of EE were obtained. The in vitro release profile of TP from nanoparticles exhibited a typical biphasic release phenomenon, namely initial burst release and consequently sustained release. In this case, the particle size played an important role for the drug release. The modified-SESD method was a potential and advantage method to produce an ideal polymer nanoparticles for drug delivery system (DDS).  相似文献

Poly(l-lactic acid) (PLLA) nonwoven fabric was obtained by using a carbon dioxide laser-thinning method. The obtained PLLA nonwoven fabric was made of endless microfibers with a uniform diameter without droplets. The fiber diameter can be varied by controlling an airflow rate supplied to the air jet, a supplying speed of an original fiber into a laser-irradiating point, and laser intensity. When the microfiber prepared by irradiating the laser operated at a laser intensity of 66 W cm−2 to the original fiber supplied at Ss = 0.1 m min−1 was dragged at an airflow rate of 30 L min−1, the thinnest microfiber with an average diameter of 3.4 μm was obtained. The obtained microfiber had a degree of crystallinity of 45%, and the degree of crystal orientation of 84%. The existence of highly oriented crystallites suggests that a flow-induced crystallization occurred during the laser-thinning.  相似文献

A selective and sensitive amperometric method of analysis has been developed for determination of the trace amounts of mercury in waters at a platinum electrode based on the effect of the presence of mercury ions on the current due to oxidation of l-tyrosine. A decrease of signal was observed due to the formation of a complex of tyrosine with the Hg(II) ion adsorbed on the electrode surface. Several parameters were varied, such as applied potential, pH and concentration of tyrosine. The calibration plot was linear in the range from 0.02 to 3 μmol l−1 Hg(II) with r=0.997 and the detection limit (3σ) was 0.014 μmol l−1; the relative standard deviation was 2.2%. The study of interferences from other metal ions revealed a good selectivity of this method towards mercury(II). The stoichiometry of the mercury-tyrosine complex was determined to be 1:2 and the formation constant 627±19. Formation of complexes with mercury ions was also demonstrated with several catechol compounds and other amino acids. The method was applied to the analysis of contaminated waters.  相似文献

A study was performed to assess the performance of aminoacids immobilized on carbon nanotubes (CNTs) for their employment as a sorbent for solid phase extraction systems. An immobilization method is introduced and the aminoacid l-tyrosine was chosen as a case study. A spectrophotometric study revealed the amount of aminoacid immobilizated on CNTs surface, and it turned to be of 3174 μmol of l-tyr g−1. The material was tested for Co retention using a minicolumn inserted in a flow system. At pH 7.0, the amount of Co retained by the column was of 37.58 ± 3.06 μmol Co g−1 of CNTs. A 10% (v/v) HNO3 solution was chosen as eluent. The pH study revealed that Co binding increased at elevated pH values. The calculation of the mol ratio (moles of Co bound at pH 9 to moles of l-tyr) turned to be 3:1. The retention capacity was compared to other bivalent cations and showed the following tendency: Cu2+ > Ni2+ > Zn2+ ? Co2+. The analytical performance was evaluated and an enrichment factor of 180 was obtained when 10 mL of 11.37 μg L−1 Co solution was loaded onto the column at pH 9.0; reaching a limit of detection (LoD) of 50 ng L−1. The proposed system was successfully applied to Co determination in QC-LL2 standard reference material (metals in natural water).  相似文献

d-Ribono-1,4-lactone was treated with ethylamine in DMF to afford N-ethyl-d-ribonamide 9a in quantitative yield. Bromination of amide 9a by the system SOBr2 in DMF or PPh3/CBr4 in pyridine led, after acetylation, to epoxide 7. However, treatment of amide 9a with acetyl bromide in dioxane followed by acetylation gave 2,3,4-tri-O-acetyl-5-bromo-5-deoxyl-N-ethyl-d-ribonamide 10a. Methanolysis of 10a, with sodium methoxide, afforded the N-ethyl-d-ribonolactam 11a in 51% overall yields. Using this method, N-butyl, N-hexyl, N-dodecyl, and N-benzyl-d-ribonolactams 11b-e were obtained in good yields (48-53%).  相似文献
