In this work, we report the development and validation of an analytical method for the trace level determination of 14 selected (EU-directive) priority organic pollutants (namely, 1,2,3-trichlorobenzene (1,2,3-TCB), 1,2,4-trichlorobenzene, 1,3,5-trichlorobenzene, hexachloro-1,3-butadiene, pentachlorobenzene, hexachlorobenzene, alachlor, α-hexachloro-cyclohexane (α-HCH), β-HCH, γ-HCH (lindane), δ-HCH, tetra-brominated diphenyl ether (tetra-BDE), penta-brominated diphenyl ether and hepta-brominated diphenyl ether) in wastewater samples from 5 different sewage treatment plants (STPs) located in Spain. The proposed methodology is based on liquid-liquid extraction with n-hexane followed by identification and confirmation of the selected pollutants by gas chromatography high-resolution mass spectrometry in selected ion recording acquisition mode. Recovery studies performed with spiked wastewater samples at two different concentration levels (0.1 and 1 μg L−1) gave mean recoveries in the range 80-120% (except for trichlorobenzenes, ca. with 50%) with RSD values below 10% in most cases, thus confirming the usefulness of the proposed methodology for the analyses of this kind of complex samples. The obtained detection limits in effluent wastewater matrices were in the low nanogram per liter range, with values as low as 0.09 ng L−1 for tetra-BDE and 0.3 ng L−1 for hexachlorobenzene. Finally, the proposed methodology was successfully applied to a monitoring study intended to characterize wastewater effluents of 5 different sewage treatment plants with different major activities (Industrial, Coastal, Urban). Most of the compounds targeted were detected in the ng L−1 range at concentrations ranging from 0.19 ng L−1 to 135 ng L−1 (hexachlorobenzene).  相似文献

A simple, fast, sensitive and robust analytical method using gas chromatography (GC)-isotope dilution mass spectrometry (MS) was developed and validated for the identification and quantification of 1,4-dichlorobenzene (p-DCB) residues in honey samples. The proposed methodology is based on steam-distillation using a Clevenger-type apparatus followed by gas chromatography-mass spectrometry (GC-MS) in the selected ion monitoring (SIM) mode employing the isotopically labeled analogue d4-1,4-dichlorobenzene (d4-p-DCB) as internal standard (IS). Validation of the method was performed in two different GC-MS systems, using quadrupole MS (QMS) and ion-trap MS (ITMS) detectors, with no statistically significant differences between two. Recoveries were better than 91% with percent relative standard deviations lower than 12%. The instrumental limits of detection were 1 μg kg−1 in the GC-ITMS system and 0.6 μg kg−1 in the GC-QMS system. The expanded uncertainty was estimated as 17% at the currently accepted “action level” of 10 μg kg−1. The method was applied to the analysis of 310 honey samples in an extensive national monitoring study. A quality control (QC) system applied during the assays has demonstrated a good performance and long-term stability over a period of more than 8 months of continuous operation.  相似文献

A platinum plate coated with single-walled carbon nanotubes (SWCNTs@Pt) was prepared by means of electrophoretic deposition. Using the SWCNTs@Pt plate, an electrosorption-enhanced solid-phase microextraction (EE-SPME) technique was proposed for the extraction of trace anions in water, described as follows: a positive potential was applied to the SWCNTs@Pt plate to extract F, Cl, Br, NO3 and SO42− from water using electrosorption, and then a negative potential was applied to the plate placed in ultra-pure water for the desorption of the absorbed anions, and finally the desorbed anions were analyzed using ion chromatography (IC). The EE-SPME parameters, including extraction potential and time as well as desorption potential and time, were investigated. An analytical method based on the above procedures, i.e., EE-SPME-IC, was established and used for the analysis of trace anions in water. The results showed that the application of potential on the SWCNTs@Pt plate significantly enhanced the ion extraction efficiency, and an enrichment factor of 15-38 was achieved. The SWCNTs@Pt plate could be used more than 50 times without significant decay. The linear range, the limit of detection (S/N = 3), the limit of quantification (S/N = 10) and repeatability (n = 7) of our EE-SPME-IC method were 1.0-150.0 μg/L, 0.06-0.26 μg/L, 0.19-0.85 μg/L and 2.1-8.0%, respectively. The proposed method was successfully applied for the analysis of trace anions in deionized water, and acceptable recoveries between 65.3 and 121.1% were obtained for the spiked deionized water samples.  相似文献

In this work, a straightforward, reliable and effective automated method has been developed for the direct determination of monoaromatic volatile BTEXS group (namely benzene, toluene, ethylbenzene, o-, m- and p-xylenes, and styrene) in olives and olive oil, based on headspace technique. Separation, identification and quantitation were carried out by headspace-gas chromatography-mass spectrometry (HS-GC-MS) in selected ion monitoring (SIM) mode. Sample pretreatment or clean-up were not necessary (besides olives milling) because the olives and olive oil samples are put directly into an HS vial, automatically processed by HS and then injected in the GC-MS for chromatographic analysis. The chemical and instrumental variables were optimized using spiked olives and olive oil samples at 50 μg kg−1 of each targeted species. The method was validated to ensure the quality of the results. The precision was satisfactory with relative standard deviations (RSD (%)) in the range 1.6-5.2% and 10.3-14.2% for olive oil and olives, respectively. Limits of detection were in the range 0.1-7.4 and 0.4-4.4 μg kg−1 for olive oil and olives, respectively. Finally, the proposed method was applied to the analysis of real olives and olive oil samples, finding positives of the studied compounds, with overall BTEXS concentration levels in the range 23-332 μg kg−1 and 4.2-87 μg kg−1 for olive oil and olives, respectively.  相似文献

Ochratoxin A (OTA) is a secondary fungal metabolite produced by several moulds, mainly by Aspergillus ochraceus, A. carbonarius, A. niger and by Penicillium verrucosum. The present work shows the results of comparative studies using different procedures for the analysis of OTA in maize bread samples. The studied analytical methods involved extraction with different volumes of PBS/methanol, different extraction apparatus, and clean-up through immunoaffinity columns. The separation and identification were carried out by high-performance liquid chromatography with fluorescence detection. The optimized method for analysis of OTA in maize bread involved extraction with PBS:methanol (50:50), and clean-up with IAC column. The limit of quantification was 0.033 ng g−1. Recoveries ranged from 87% to 102% for fortifications at 2.000 and 0.500 ng g−1, respectively, within-day R.S.D. of 1.4% and 4.7%. The proposed method was applied to 15 samples and the presence of OTA was found in nine samples at concentrations ranging from nd to 2.650 ng g−1.  相似文献

This work reports a rapid, specific and sensitive multi-residue method based on the Quick Easy Cheap Effective Rugged and Safe (QuEChERS) sample preparation method and gas chromatography with mass spectrometric detection in the selected ion monitoring mode (GC-MS-SIM) using one quantification ion and two identification ions for the routine analysis of 203 pesticides in rice paddies. Analyses of fortified rice paddy samples were performed at different levels (0.05, 0.20 and 0.50 mg kg−1). Mean recoveries from five replicates ranged from 75% to 115%, with coefficients of variation lower than 17%. The limit of quantification was in the range of 0.002-0.05 mg kg−1 for the pesticides. 1040 rice paddy samples were analyzed for method application.  相似文献

A new IC-ESI-MS/MS method, with simple sample preparation procedure, has been developed for quantification and confirmation of perchlorate (ClO4) anions in water, fresh and canned food, wine and beer samples at low part-per-trillion (ng l−1) levels. To the best of our knowledge, this is the first time an analytical method is used for determination of perchlorate in wine and beer samples. The IC-ESI-MS/MS instrumentation consisted of an ICS-2500 ion chromatography (IC) system coupled to either an API 2000™ or an API 3200™ mass spectrometer. The IC-ESI-MS/MS system was optimized to monitor two pairs of precursor and fragment ion transitions, i.e., multiple reaction monitoring (MRM). All samples had oxygen-18 isotope labeled perchlorate internal standard (ISTD) added prior to extraction. Chlorine isotope ratio (35Cl/37Cl) was used as a confirmation tool. The transition of 35Cl16O4 (m/z 98.9) into 35Cl16O3 (m/z 82.9) was monitored for quantifying the main analyte; the transition of 37Cl16O4 (m/z 100.9) into 37Cl16O3 (m/z 84.9) was monitored for examining a proper isotopic abundance ratio of 35Cl/37Cl; and the transition of 35Cl18O4 (m/z 107.0) into 35Cl18O3 (m/z 89.0) was monitored for quantifying the internal standard. The minimum detection limit (MDL) for this method in de-ionized water is 5 ng l−1 (ppt) using the API 2000™ mass spectrometer and 0.5 ng l−1 using the API 3200™ mass spectrometer. Over 350 food and beverage samples were analyzed mostly in triplicate. Except for four, all samples were found to contain measurable amounts of perchlorate. The levels found ranged from 5 ng l−1 to 463.5 ± 6.36 μg kg−1 using MRM 98.9 → 82.9 and 100 μl injection.  相似文献

Two highly sensitive and selective methods based on gas chromatography coupled to mass spectrometry (GC-MS) in the selected ion monitoring (SIM) mode have been developed for the quantification of 2,6-dichlorophenol (2,6-DCP), a sex pheromone of the tick females of Anocentor nitens. Standard addition method and calibration curve techniques using 5-bromine-4-hydroxy-3-methoxybenzaldehyde (5-BrV) as internal standard (IS) afforded detection limit of 0.1 ng ml−1. The calibration curve was linear over the concentration range from 0.5 to 500 ng ml−1 for 2,6-DCP. Results show that the concentration range of sex pheromone in the extracts samples was 1.08-10.35 ng ml−1. The methods developed provided reliable procedures to determine amounts of 2,6-DCP present in ticks.  相似文献

A simple and sensitive method for the determination of patulin in fruit juice and dried fruit samples was developed using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography–mass spectrometry (LC–MS). Patulin was separated within 5 min by high-performance liquid chromatography using a Synergi MAX-RP 80A column and water/acetonitrile (80/20, v/v) as the mobile phase. Electrospray ionization conditions in the negative ion mode were optimized for MS detection of patulin. The pseudo-molecular ion [M−H] was used to detect patulin in selected ion monitoring (SIM) mode. The optimum in-tube SPME conditions were 25 draw/eject cycles of 40 μL of sample using a Carboxen 1006 PLOT capillary column as an extraction device. The extracted patulin was readily desorbed from the capillary by passage of the mobile phase, and no carry-over was observed. Using the in-tube SPME LC–MS with SIM method, good linearity of the calibration curve (r = 0.9996) was obtained in the concentration range of 0.5–20 ng/mL using 13C3-patulin as an internal standard, and the detection limit (S/N = 3) of patulin was 23.5 pg/mL. The in-tube SPME method showed >83-fold higher sensitivity than the direct injection method (10 μL injection volume). The within-day and between-day precision (relative standard deviations) were below 0.8% and 5.0% (n = 6), respectively. This method was applied successfully for the analysis of fruit juice and dried fruit samples without interference peaks. The recoveries of patulin spiked into apple juice were >92%, and the relative standard deviations were <4.5%. Patulin was detected at ng/mL levels in various commercial apple juice samples.  相似文献

A sensitive sandwich type immunoassay has been proposed with the detection by inductively coupled plasma mass spectrometry (ICP-MS) in a single particle mode (time resolved analysis). The signal induced by the flash of ions (197Au+) due to the ionization of single Au-nanoparticle (Au-NP) label in the plasma torch can be measured by the mass spectrometer. The frequency of the transient signals is proportional to the concentration of Au-NPs labels. Characteristics of the signals obtained from Au-NPs of 20, 45 and 80 nm in diameters were discussed. The analytical figures for the determination of Au-labeled IgG using ICP-MS in conventional integral mode and single particle mode were compared in detail. Rabbit-anti-human IgG was used as a model analyte in the sandwich immunoassay. A detection limit (3σ) of 0.1 ng mL−1 was obtained for rabbit-anti-human IgG after immunoreactions, with a linear range of 0.3-10 ng mL−1 and a RSD of 8.1% (2.0 ng mL−1). Finally, the proposed method was successfully applied to spiked rabbit-anti-human IgG samples and rabbit-anti-human serum samples. The method resulted to be a highly sensitive ICP-MS based sandwich type immunoassay.  相似文献

Terbium sensitized fluorescence was used as a post-column detection system to develop a simple, sensitive and rapid high-performance liquid chromatographic method for the simultaneous determination of catecholamines norepinephrine (NE), epinephrine (E) and dopamine (DA).Catecholamines were separated by an ion-pair reversed-phase chromatography on a BDS-Hypersil analytical column with a mobile phase of methanol and 50 mmol l−1 acetate buffer (pH 4.7) containing 1.1 mmol l−1 SOS and 0.11 mmol l−1 EDTA (15+85 v/v).Catecholamines and the internal standard (3,4-dihydroxybenzylamine, DHBA) were post-column derivatized by the addition to the eluent of an alkaline solution containing a stoichiometric mixture of terbium(III) chloride and EDTA. Fluorescence detection (λex=300 nm, λem=545 nm) is based on the sensitization of terbium ion fluorescence after complexation with catecholamines.The chemical compatibility between the eluent and the post-column reagent was studied and the analytical characteristics of the method were established. Detection limits found were 1.0×10−8, 4.0×10−8 and 7.0×10−8 mol l−1 for NE, E and DA, respectively. The method has been successfully applied to the determination of catecholamines in urine samples after solid-phase extraction (SPE) pre-treatment. Recoveries from urine spiked with NE (4.0×10−7, 2.0×10−6 and 4.0×10−6 mol l−1), E (8.2×10−8, 4.1×10−7 and 8.2×10−7 mol l−1) and DA (1.0×10−6, 5.0×10−6 and 1.0×10−5 mol l−1) varied from 98 to 100% (mean=99.3%), from 106 to 107% (mean=106.3%) and from 98 to 101% (mean=99.3%), respectively. The between-run precision (relative standard deviation, R.S.D.) for the method for three urine samples at different concentration levels of each catecholamine varied from 3.6 to 7.0%.  相似文献

A selective and sensitive isotope dilution–high performance liquid chromatography–linear ion trap mass spectrometry (Isotope Dilution–HPLC–LIT-MS3) method was developed for the simultaneous determination of 19 triazine pesticides and their degradation products in processed cereal samples from Chinese total diet study (TDS). The method integrated the addition of isotope internal standards, liquid–liquid extraction (LLE), clean-up with MCX solid-phase extraction (SPE) cartridges and HPLC–LIT-MS3 analysis with selected reaction monitoring (SRM) mode. Matrix-matched calibration curves showed good linearity (R2 ≥ 0.9940) verified by applying the Mandel's fitting test (p > 0.087) performed at the 95% confidence level. Decision limits (CCαs) and detection capabilities (CCβs) of the 19 triazine pesticides and their degradation products fell in the ranges of 0.0020–0.4200 μg kg−1 and 0.0024–0.4500 μg kg−1, respectively. Recoveries ranged from 70.1% to 112.8%, with the relative standard deviations (RSDs) ranging from 1.5% to 13.5%. Furthermore, the proposed method was applied to analyzing the proposed cereal samples from the fourth Chinese TDS. Eleven triazines were detected in six cereal samples with the concentrations ranging from 0.013 to 0.987 μg kg−1. This method can also be used for the further determination of the triazines in other food group composites, and ultimately served as a methodological foundation for assessing the triazines in the average Chinese diet in the general population.  相似文献

A simple and rapid spectrophotometric method is proposed for the determination of transparent exopolymer particles (TEP) in freshwater samples. In this method, TEP reacts with excess of alcian blue solution yielding a low solubility dye-TEP complex. After centrifugation, the concentration of the remaining dye in the supernatant was determined at 602 nm and its concentration was related to the concentration of TEP in freshwater. The effect of alcian blue concentration from 1.5×10−3 to 9.0×10−3% (m/v), solution pH from 2.5 to 6.9 and stirring time from 20 to 120 s on the analytical curve was investigated. Under the optimum conditions established, such as alcian blue concentration of 3.0×10−3% (m/v); pH of 4.0 (0.2 mol l−1 acetate buffer solution) and stirring time of 1 min, the analytical curve was linear from 0.50 to 10 μg ml−1 (A=0.34−0.037[GX]; r2=0.9999; where A is the absorbance and [GX] the gum xanthan concentration in μg ml−1) with a detection limit of 0.10 μg ml−1. The recovery of TEP (as gum xanthan) for two samples ranged from 95.3 to 108 and the relative standard deviations (R.S.D.s) were lower than 0.8% for gum xanthan solutions at concentrations of 1.0 and 1.5 μg ml−1 (n=8). The results obtained for TEP in freshwater samples using the proposed spectrophotometric method and those obtained using a literature method are in agreement at the 95% confidence level and within an acceptable range of error.  相似文献

The use of ion chromatography (IC) in conjunction with ultrasonic extraction is described for the routine analysis of water-soluble major inorganic ions and organic acids in atmospheric fine particles (PM2.5). Both the extraction method and the IC analysis were validated using NIST SRM 1648 (urban particulate matter). In addition, the reliability of the IC method was established by intercomparison of results obtained with those from suitable alternative analytical techniques (atomic absorption spectrometry (AAS), proton-induced X-ray emission (PIXE) spectrometry, and UV-Visible spectrophotometry). The validated IC method was successfully applied for field monitoring of PM2.5 particles collected in Singapore over an extended period of time. The IC analysis revealed that the concentrations of individual ions were in the order, SO42− > NH4+ > NO3 > Na+ > K+ > Cl, respectively. Among the major ionic components, SO42− contributed 50% to the measured water-soluble aerosol mass followed by NH4+ (16.5%) and NO3 (9.0%). The cations Na+, K+, Mg2+, and Ca2+ accounted for 24% of the total water-soluble mass. The IC analysis was performed to quantify the organic acids, which typically account for a small fraction of water-soluble organic compounds in PM2.5. Oxalate was found to be the dominant species among the organic acids measured in this work.  相似文献

A simple, efficient and environmentally friendly analytical methodology is proposed for extracting and preconcentrating pyrethroids from water samples prior to gas chromatography-negative ion chemical ionization mass spectrometry (GC-NCI-MS) analysis. Fourteen pyrethroids were selected for this work: bifenthrin, cyfluthrin, λ-cyhalothrin, cypermethrin, deltamethrin, esfenvalerate, fenvalerate, fenpropathrin, τ-fluvalinate, permethrin, phenothrin, resmethrin, tetramethrin and tralomethrin. The method is based on ultrasound-assisted emulsification-extraction (UAEE) of a water-immiscible solvent in an aqueous medium. Chloroform was used as extraction solvent in the UAEE technique. Target analytes were quantitatively extracted achieving an enrichment factor of 200 when 20 mL aliquot of pure water spiked with pyrethroid standards was extracted. The method was also evaluated with tap water and river water samples. Method detection limits (MDLs) ranged from 0.03 to 35.8 ng L−1 with RSDs values ≤3–25% (n = 5). The coefficients of estimation of the calibration curves obtained following the proposed methodology were ≥0.998. Recovery values were in the range of 45–106%, showing satisfactory robustness of the method for analyzing pyrethroids in water samples. The proposed methodology was applied for the analysis of river water samples. Cypermethrin was detected at concentration levels ranging from 4.94 to 30.5 ng L−1.  相似文献

This work describes an on-line molecularly imprinted solid-phase extraction (MISPE) method for spectrophotometric determination of nicotine in urine samples of smokers. This method is based on manganese (VII) to manganese (VI) reduction in an alkaline medium, promoted by nicotine. Two wash solutions (1:4 (v/v) acetonitrile:sodium hydroxide - pH 11.4, and nitric acid - pH 2.5) were employed to circumvent interferences. Aqueous solutions containing nicotine plus different possible concomitants (cotinine, anabasine, norcotinine and caffeine) were tested individually. The analytical calibration curve was prepared in urine samples collected from non-smokers and spiked with nicotine standard from 1.1 to 60 μmol L−1 (r2 > 0.998). The limit of quantification and the analytical frequency were 1.1 μmol L−1 and 11 h−1, respectively. The precision, evaluated using 3, 10 and 30 μmol L−1 nicotine in urine, was 10, 10 and 4% (intra-day precision) and 12, 13 and 5% (inter-day precision), respectively. Accuracy was checked through high performance liquid chromatography and the results did not present significant differences at the 95% confidence level according to the Student's t-test.  相似文献

Simultaneous determination of putrescine, cadaverine, spermidine, spermine, N1-acetylputrescine, N1-acetylcadaverine, N1-acetylspermidine, N8-acetylspermidine and N1-acetylspermine in aqueous samples was achieved as their N-ethoxycarbonyl-N-pentafluoropropionyl derivatives by gas chromatography-mass spectrometry (GC-MS) in selected ion monitoring (SIM) mode. The overall GC-SIM-MS method was linear (r ≥ 0.9987), repetitive (% relative standard deviation = 1.3-6.4), and accurate (% relative error = −2.1-8.6). As compared to normal subjects, the levels of putrescine, N1-acetylspermine and spermine were significantly elevated while the levels of N1-acetylputrescine, N1-acetylcadaverine and N1-acetylspermidine were markedly reduced in all four Alzheimer's disease (AD) patients. When star symbol plotting was applied to the normalized levels of nine polyamines relative to the corresponding normal mean values, each star pattern of the AD patients was deformed, thus being readily distinguishable from the nonagon shape of the normal group average.  相似文献

A sensitive and reliable method was developed and validated for detection and confirmation of melamine in egg based on gas chromatography-mass spectrometry (GC-MS) and ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Trichloroacetic acid solution was used for sample extraction and precipitation of proteins. The aqueous extracts were subjected to solid-phase extraction by mixed-mode reversed-phase/strong cation-exchange cartridges. Using ultra-performance liquid chromatography and electrospray ionization in the positive ion mode, melamine was determined by LC-MS/MS, which was completed in 5 min for each injection. For the GC-MS analysis, extracted melamine was derivatized with N,O-bis(trimethylsilyl)trifluoracetamide prior to selected ion monitoring detection in electron impact mode. The average recovery of melamine from fortified samples ranged from 85.2% to 103.2%, with coefficients of variation lower than 12%. The limit of detection obtained by GC-MS and UPLC-MS/MS was 10 and 5 μg kg−1, respectively. This validated method was successfully applied to the determination of melamine in real samples from market.  相似文献

A method has been developed to determine acrylamide in aqueous matrices by using direct immersion solid-phase microextraction (SPME) coupled to gas chromatography-positive chemical ionization tandem mass spectrometry (GC-PCI-MS-MS) in the selected reaction monitoring (SRM) mode. The optimized SPME experimental procedures to extract acrylamide in water solutions were: use of a carbowax/divinylbenzene (CW/DVB)-coated fiber at pH 7, extraction time of 20 min and analyte desorption at 210 °C for 3 min. A detection limit of 0.1 μg L−1 was obtained. The linear range was 1-1000 μg L−1. The relative standard deviation was 10.64% (n = 7). The proposed analytical method was successfully used for the quantification of trace acrylamide in foodstuffs such as French fries (1.2 μg g−1) and potato crisps (2.2 μg g−1).  相似文献

A method based on immersed solid-phase microextraction (SPME) and gas chromatography mass spectrometry detection (GC-MS) for the determination of organophosphorous pesticides (OPPs) in aqueous samples was developed. A sol-gel based coating fiber was prepared using 3-(trimethoxysilylpropyl) amine as precursor. The synthesized fiber was prepared in a way to impart polar moiety into the coating network and would be more suitable for extracting polar and semi-polar organic pollutants. Important parameters influencing the extraction process were optimized and an extraction time of 40 min at 30 °C gave maximum peak area, when NaCl (20% w/v) was added to the aqueous sample. The linearity for disulfoton, phorate and sulfotep was in the concentration range of 0.01 to 5 ng mL− 1 and for parathion and O,O,O-triethylthiphosphate was in the range of 0.01 to 50 ng mL− 1. Limits of detection ranged from 1 ng L− 1, for parathion, to 0.05 ng L− 1, for disulfoton using time-scheduled selected ion monitoring (SIM) mode, and the RSD% values were all below 10.5% at the 1 ng mL− 1 level. The developed method was successfully applied to real water samples while the relative recovery percentages obtained for the spiked water samples were from 80 to 115%.  相似文献
