In situ mercury film electrode produced in the presence of thiocyanate has been shown extremely useful for highly sensitive adsorptive stripping voltammetric measurements of atrazine down to sub-μg L−1 level. Operational parameters have been optimized and the stripping voltammetric performance has been investigated using square wave scans. The adsorptive stripping response is linear over the range of 0.5-60 μg L−1 atrazine, with a detection limit of 0.024 μg L−1. The method has been applied to the determination of atrazine in soil and water samples.  相似文献

A simple and rapid analytical method is presented in which the three veterinary antibiotics oxytetracycline (OTC), sulfachloropyridazine (SCP) and tylosin (TYL) are simultaneously extracted and determined in four different soils. Extractions were carried out by a combination of ultrasonic agitation and vortex mixing using a mixture of methanol, EDTA and McIlvaine buffer at pH 7 as the extractant solution. The extracts were then cleaned-up by a tandem solid phase extraction (SPE) method using an Isolute SAX anion exchange cartridge to remove natural organic matter and an Oasis HLB polymeric cartridge to retain the study compounds. Analysis was by HPLC-UV with additional fluorescence detection for SCP. Recoveries were in the range 68-85% for SCP in all soil types, 58-75% for OTC in sandy soils, 27-51% for OTC in clay containing soils, 74-105% for TYL and 47-61% in a clay soil. OTC and SCP were also extracted from liquid pig manure using a mixture of EDTA and McIlvaine buffer at pH 7 with ultrasonic agitation and vortex mixing with SPE clean-up and HPLC-UV analysis. Recoveries were greater than 77% and 58% for OTC and SCP, respectively. Limits of detection were 18 μg kg−1 for OTC and SCP and 40 μg kg−1 for TYL in soils and 70 μg L−1 for OTC and 140 μg L−1 for SCP in pig slurry.  相似文献

A gas chromatography–mass spectrometric (GC–MS) method has been established for the determination of hydrazine in drinking water and surface water. This method is based on the derivatization of hydrazine with ortho-phthalaldehyde (OPA) in water. The following optimum reaction conditions were established: reagent dosage, 40 mg mL−1 of OPA; pH 2; reaction for 20 min at 70 °C. The organic derivative was extracted with methylene chloride and then measured by GC–MS. Under the established condition, the detection and the quantification limits were 0.002 μg L−1 and 0.007 μg L−1 by using 5.0-mL of surface water or drinking water, respectively. The calibration curve showed good linearity with r2 = 0.9991 (for working range of 0.05–100 μg L−1) and the accuracy was in a range of 95–106%, and the precision of the assay was less than 13% in water. Hydrazine was detected in a concentration range of 0.05–0.14 μg L−1 in 2 samples of 10 raw drinking water samples and in a concentration range of 0.09–0.55 μg L−1 in 4 samples of 10 treated drinking water samples.  相似文献

The determination of atrazine in real samples (commercial pesticide preparations and water matrices) shows how the Fenton's reagent can be used with analytical purposes when kinetic methodology and multivariate calibration methods are applied. Also, binary mixtures of atrazine-alachlor and atrazine-bentazone in pesticide preparations have been resolved. The work shows the way in which interferences and the matrix effect can be modelled. Experimental design has been used to optimize experimental conditions, including the effect of solvent (methanol) used for extraction of atrazine from the sample. The determination of pesticides in commercial preparations was accomplished without any pre-treatment of sample apart from evaporation of solvent; the calibration model was developed for concentration ranges between 0.46 and 11.6 × 10−5 mol L−1 with mean relative errors under 4%. Solid-phase extraction is used for pre-concentration of atrazine in water samples through C18 disks, and the concentration range for determination was established between 4 and 115 μg L−1approximately. Satisfactory results for recuperation of atrazine were always obtained.  相似文献

Polyclonal antibodies against microcystin-LR (MC-LR), a cyclic heptapeptide toxin, were generated in rabbits using MC-LR-BSA. An enzyme-linked immunosorbent assay (ELISA) was developed for the characterization of the antibodies and their potential use for analytical purposes. The concentration of MC-LR that inhibits 50% of antibody-antigen binding (IC50) was 0.5 μg L−1 for the indirect ELISA format and 0.9 μg L−1 for the direct ELISA, using MC-LR-horseradish peroxidase conjugate. The limit of detection corresponding to IC80 was found to be 0.06 μg L−1, well below the Word Health Organization level for drinking water of 1 μg L−1. The direct competitive ELISA was applied to water samples and was shown useful for screening purposes. The developed anti-microcystin antibodies were immobilized on solid supports for use in selective solid phase extraction (SPE) systems, prior to liquid chromatography (LC) quantification. An immunoaffinity cartridge (IAC), a Sepharose®-based cartridge incorporating 2 mg of antibodies allowed the selective and quantitative recovery of a mixture of 0.2 μg of MCs showing potential use in sample preparation of real matrices. When applied to water and green algae samples, average recoveries from Sepharose®-based cartridges were in the range of 86-113% for water samples and 85-92% for blue-green algae samples. Selectivity of the IAC clean-up was proven by comparison with non-specific solid phase extraction using octadecylsilica (ODS) sorbent. Results obtained using LC/UV after IAC clean-up agreed well with results obtained using liquid chromatography and mass spectrometry detection (LC/MS and LC/MS/MS) after SPE-C18 clean-up, allowing therefore to validate the resulting technique.  相似文献

A single-drop microextraction (SDME) procedure was developed for the analysis of organophosphorus and pyrethroid pesticides in water by gas chromatography (GC) with flame ionization detection (GC-FID). The significant parameters that affect SDME performance, such as the selection of microextraction solvent, solvent volume, extraction time, and stirring rate, were studied and optimized using a tool screening factorial design. The limits of detection (LODs) in water for the four investigated compounds were between 0.3 and 3.0 μg L−1, with relative standard deviations ranging from 7.7 to 18.8%. Linear response data were obtained in the concentration range of 0.9-6.0 μg L−1 (λ-cyhalothrin), 3.0-60.0 μg L−1 (methyl parathion), 9.0-60.0 μg L−1 (ethion), and 9.0-30.0 μg L−1 (permethrin), with correlation coefficients ranging from 0.9337 to 0.9977. The relative recoveries for the spiked water ranged from 73.0 to 104%. Environmental water samples (n = 26) were successfully analyzed using the proposed method and methyl parathion presented concentration up to 2.74 μg L−1. The SDME method, coupled with GC-FID analysis, provided good precision, accuracy, and reproducibility over a wide linear range. Other highlights of the method include its ease of use and its requirement of only small volumes of both organic solvent and sample.  相似文献

Present study described a simple, sensitive, and viable method for the determination of bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol in water samples using temperature-controlled ionic liquid dispersive liquid-phase microextraction coupled to high performance liquid chromatography-fluorescence detector. In this experiment, 1-octyl-3-methylimidazolium hexafluorophosphate ([C8MIM][PF6]) was used as the extraction solvent, and bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol were selected as the model analytes. Parameters affecting the extraction efficiency such as the volume of [C8MIM][PF6], dissolving temperature, extraction time, sample pH, centrifuging time and salting-out effect have been investigated in detail. Under the optimized conditions, good linear relationship was found in the concentration range of 1.0-100 μg L−1 for BPA, 1.5-150 μg L−1 for 4-NP, and 3-300 μg L−1 for 4-OP, respectively. Limits of detection (LOD, S/N = 3) were in the range of 0.23-0.48 μg L−1. Intra day and inter day precisions (RSDs, n = 6) were in the range of 4.6-5.5% and 8.5-13.3%, respectively. This method has been also successfully applied to analyze the real water samples at two different spiked concentrations and excellent results were obtained.  相似文献

A multi-residue method for the determination of organochlorine pesticides in fish feed samples was developed and optimized. The method is based on a cleanup step of the extracted fat, carried out by liquid–liquid extraction on diatomaceous earth cartridge with n-hexane/acetonitrile (80/20, v/v) followed by solid phase extraction (SPE) with silica gel–SCX cartridge, before the identification and quantification of the residues by gas chromatography–triple quadrupole tandem spectrometry (GC–MS/MS). Performance characteristics, such as accuracy, precision, linear range, limits of detection (LOD) and quantification (LOQ), for each pesticide were determined. Instrumental LODs ranged from 0.01 to 0.11 μg L−1, LOQs were in the range of 0.02–0.35 μg L−1, and calibration curves were linear (r2 > 0.999) in the whole range of explored concentrations (5–100 μg L−1). Repeatability values were in the range of 3–15%, evaluated from the relative standard deviation of six samples spiked at 100 μg kg−1 of fat, and in compliance with that derived by the Horwitz's equation. No matrix effects or interfering substances were observed in fish feed analyses. The proposed method allowed high recoveries (92–116%) of spiked extracted fat samples at 100 μg kg−1, and very low LODs (between 0.02 and 0.63 μg kg−1) and LOQs (between 0.05 and 2.09 μg kg−1) determined in fish feed samples.  相似文献

Atrazine is a triazine herbicide which contains two secondary aliphatic amine groups. Previous studies have shown that aliphatic amines react with tris(2,2′-bipyridyl)ruthenium(III) to produce chemiluminescence. This paper describes the application of tris(2,2′-bipyridyl)ruthenium(III) to the detection of atrazine and related triazine herbicides in water by flow injection chemiluminescence analysis. The optimised experimental conditions were determined to be: sample and carrier flow rates of 4.6 mL min−1, sample at pH 9 buffered with 50 mM borax, and reagent concentration of 1 mM tris(2,2′-bipyridyl)ruthenium(III) in 20 mM H2SO4 (pH 1). Under these conditions, the logarithm of the chemiluminescence intensity versus concentration was linear in the range of 2.15-2150 μg L−1 for samples in MilliQ water, and the limit of detection of atrazine in water was determined to be 1.3 ± 0.1 μg L−1. Validation of the method was performed using direct injection HPLC. The presence of natural organic matter (NOM) significantly increased the chemiluminescence, masking the signal generated by atrazine. Isolating the target analyte via solid phase extraction (SPE) prior to analysis removed this interference and concentrated the samples, resulting in a greatly improved sensitivity with a detection limit of 14 ± 2 ng L−1.  相似文献

There is a need for simple and inexpensive methods to quantify potentially harmful persistent pesticides often found in our water-ways and water distribution systems. This paper presents a simple, relatively inexpensive method for the detection of a group of commonly used pesticides (atrazine, simazine and hexazinone) in natural waters using large-volume direct injection high performance liquid chromatography (HPLC) utilizing a monolithic column and a single wavelength ultraviolet-visible light (UV-vis) detector. The best results for this system were obtained with a mobile phase made up of acetonitrile and water in a 30:70 ratio, a flow rate of 2.0 mL min−1, and a detector wavelength of 230 nm. Using this method, we achieved retention times of less than three minutes, and detection limits of 5.7 μg L−1 for atrazine, 4.7 μg L−1 for simazine and 4.0 μg L−1 for hexazinone. The performance of this method was validated with an inter-laboratory trial against a National Association of Testing Authorities (NATA) accredited liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) method commonly used in commercial laboratories.  相似文献

A molecularly imprinted polymer (MIP) designed to enable the selective extraction of carbamazepine (CBZ) from effluent wastewater and urine samples has been synthesised using a non-covalent molecular imprinting approach. The MIP was evaluated chromatographically in the first instance and its affinity for CBZ also confirmed by solid-phase extraction (SPE). The optimal conditions for SPE consisted of conditioning of the cartridge using acidified water purified from a Milli-Q system, loading of the sample under basic aqueous conditions, clean-up using acetonitrile and elution with methanol. The attractive molecular recognition properties of the MIP gave rise to good CBZ recoveries (80%) when 100 mL of effluent water spiked with 1 μg L−1 was percolated through the polymer. For urine samples, 2 mL samples spiked with 2.5 μg L−1 CBZ were extracted with a recovery of 65%. For urine, the linear range was 0.05-24 mg L−1, the limit of detection was 25 μg L−1 and precision, expressed as relative standard deviation at 0.5 mg L−1 (n = 3), was 3.1% and 12.6% for repeatability and reproducibility between days, respectively.  相似文献

Bismuth and Sb were evaluated as internal standards (IS) to minimize matrix effects on the direct and simultaneous determination of As, Cu, and Pb in cachaça by graphite furnace atomic absorption spectrometry using W-coated platform plus Pd-Mg(NO3)2 as modifier. For 20 μL injected sample, calibration within the 0.5-10 μg L−1 As, 100-1000 μg L−1 Cu and 0.5-30 μg L−1 Pb intervals were established using the ratios As absorbance to Sb absorbance, Cu absorbance to Bi absorbance and Pb absorbance to Bi absorbance versus analytes concentration, respectively. Typical linear correlations of 0.998, 0.999 and 0.999 were, respectively, obtained. The proposed method was applied for direct determination of As, Cu and Pb in 10 commercial cachaça samples and results were in agreement with those obtained by inductively coupled plasma mass spectrometry at 95% confidence level. The found characteristic masses were 30 pg As, 274 pg Cu and 39 pg Pb. The useful lifetime of the graphite tube was around 760 firings. Recoveries of As, Cu and Pb added to cachaça samples varied, respectively, from 98% to 109%, 97% to 108% and 98% to 104% with internal standards and from 48% to 54%, 53% to 92% and 62% to 97% without internal standards. The limits of detection were 0.13 μg L−1 As, 22 μg L−1 Cu and 0.05 μg L−1 Pb. The relative standard deviations (n = 12) for a spiked sample containing 20 μg L−1 As, Pb and 500 μg L−1 Cu were 1.6%, 1.0%, and 1.8% with IS and 4.3%, 5.2%, and 5.5% without IS.  相似文献

Phthalates (PAEs) are ubiquitous toxic chemical compounds. During the last few years, some phthalate metabolites (MPAEs) have been proposed as appropriate biomarkers in human urine samples to determine PAE human intake and exposure. So, it is necessary to have fast, easy, robust and validated analytical methods to determine selected MPAEs in urine human samples. Two different instrumental methods based on gas (GC) and ultra-high performance liquid (UHPLC) chromatography coupled to mass spectrometry (MS) have been optimized, characterized and validated for the simultaneous determination of nine primary and secondary phthalate metabolites in urine samples. Both instrumental methods have similar sensitivity (detection limits ranged from 0.03 to 8.89 pg μL−1 and from 0.06 to 0.49 pg μL−1 in GC–MS and UHPLC–MS2, respectively), precision (repeatability, expressed as relative standard deviation, which was lower than 8.4% in both systems, except for 5OH-MEHP in the case of GC–MS) and accuracy. But some advantages of the UHPLC–MS2 method, such as more selectivity and lower time in the chromatographic runs (6.8 min vs. 28.5 min), have caused the UHPLC–MS2 method to be chosen to analyze the twenty one human urine samples from the general Spanish population. Regarding these samples, MEP showed the highest median concentration (68.6 μg L−1), followed by MiBP (23.3 μg L−1), 5cx-MEPP (22.5 μg L−1) and MBP (19.3 μg L−1). MMP (6.99 μg L−1), 5oxo-MEHP (6.15 μg L−1), 5OH-MEHP (5.30 μg L−1) and MEHP (4.40 μg L−1) showed intermediate levels. Finally, the lowest levels were found for MBzP (2.55 μg L−1). These data are within the same order of magnitude as those found in other similar populations.  相似文献

This paper presents the optimization of instrumental and solution parameters for determination of atrazine in river waters and formulation by square wave voltammetry (SWV) using a hanging mercury drop electrode. The best sensitivity (35.2±0.4 μA ml μg−1) was achieved using a frequency of 400 Hz and a medium composed of 40 mmol l−1 Britton-Robinson (BR) buffer at pH 1.9. The detection limit was 2 μg l−1 with a linear dynamic range between 10 and 250 μg l−1. Application of the method to real samples of river waters fortified with 10 μg l−1 of atrazine resulted recoveries between 92 and 116%. Additionally, good agreement was observed between results obtained by the proposed method and by HPLC for river water samples spiked with 25 μg l−1 of atrazine. The determination was not affected by the presence of humic acid at concentration of 5 mg l−1, indicating that interactions of the herbicide with this class of compounds are fully labile. The stability of the voltammetric signal for samples spiked with 250 μg l−1 atrazine was evaluated over a period of 14 days in four samples. For two samples, no systematic variation was observed, while for the other two, a decrease of peak current between 3 and 15% occurred, suggesting that the stability is dependent on the sample nature. HPLC analyses suggest formation of deethylatrazine during the second week of storage in the samples for which the SWV peak current had the more intense decrease.  相似文献

In this study a method for the determination of low concentrations of silver in waters using solid-phase extraction with a flow injection analysis system and detection by flame atomic absorption spectrometry (FAAS) was developed. Moringa oleifera seeds were used as a biosorbent material. Chemical and flow variables of the on-line preconcentration system such as sample pH and flow rate, preconcentration time, eluent concentration and sorbent mass were studied. The optimum preconcentration conditions were obtained using sample pH in the range of 6.0-8.0, preconcentration time of 4 min at a flow rate of 3.5 mL min− 1, 0.5 mol L− 1 HNO3 eluent at a flow rate of 4.5 mL min− 1 and 35 mg of sorbent mass. With the optimized conditions, the preconcentration factor, precision, detection limit and sample throughput were estimated as 35 (for preconcentration of 14 mL sample), 3.8% (5.0 μg L− 1, n = 7), 0.22 μg L− 1 and 12 samples per hour, respectively. The developed method was successfully applied to mineral water and tap water, and accuracy was assessed through analysis of a certified reference material for water (APS-1071 NIST) and recovery tests, with recovery ranging from 94 to 101%.  相似文献

In this study, the steroid hormone levels in river and tap water samples were determined by using a novel dispersive liquid-liquid microextraction method based on the solidification of a floating organic drop (DLLME-SFO). Several parameters were optimized, including the type and volume of the extraction and dispersive solvents, extraction time, and salt effect. DLLME-SFO is a fast, cheap, and easy-to-use method for detecting trace levels of samples. Most importantly, this method uses less-toxic solvent. The correlation coefficient of the calibration curve was higher than 0.9991. The linear range was from 5 to 1000 μg L−1. The spiked environmental water samples were analyzed using DLLME-SFO. The relative recoveries ranged from 87% to 116% for river water (which was spiked with 4 μg L−1 for E1, 3 μg L−1 for E2, 4 μg L−1 for EE2 and 9 μg L−1 for E3) and 89% to 102% for tap water (which was spiked with 6 μg L−1 for E1, 5 μg L−1 for E2, 6 μg L−1 for EE2 and 10 μg L−1 for E3). The detection limits of the method ranged from 0.8 to 2.7 μg L−1 for spiked river water and 1.4 to 3.1 μg L−1 for spiked tap water. The methods precision ranged from 8% to 14% for spiked river water and 7% to 14% for spiked tap water.  相似文献

A general and broad class-specific enzyme-linked immunosorbent assay was developed for the O,O-dimethyl organophosphorus pesticides, including malathion, dimethoate, phenthoate, phosmet, methidathion, fenitrothion, methyl parathion and fenthion. Three haptens with different spacer-arms were synthesized. The haptens were conjugated to bovine serum albumin (BSA) for immunogens and to ovalbumin (OVA) for coating antigens. Rabbits were immunized with the immunogens and six polyclonal antisera were produced and screened against each of the coating antigens using competitive indirect enzyme-linked immunosorbent assay for selecting the proper antiserum. The effect of hapten heterology on immunoassay sensitivity was also studied. The antibody-antigen combination with the most selectivity for malathion was further optimized and tested for tolerance to co-solvent, pH and ionic strength changes. The IC50 values, under optimum conditions, were estimated to be 30.1 μg L−1for malathion, 28.9 μg L−1 for dimethoate, 88.3 μg L−1 for phenthoate, 159.7 μg L−1 for phosmet, 191.7 μg L−1 for methidathion, 324.0 μg L−1 for fenitrothion, 483.9 μg L−1 for methyl parathion, and 788.9 μg L−1 for fenthion. Recoveries of malathion, dimethoate, phenthoate, phosmet and methidathion from fortified Chinese cabbage samples ranged between 77.1% and 104.7%. This assay can be used in monitoring studies for the multi-residue determination of O,O-dimethyl organophosphorus pesticides.  相似文献

A HPLC method using a coulometric electrode array detector (CEAD) to analyse 4-ethylcatechol in wine was established. The procedure does not require any sample preparation or analyte derivatisation and performs chromatographic separation in a short time. The assay method is linear up to 1520 μg L−1 and precise (R.S.D. < 3%), with limits of detection and quantitation of 1.34 μg L−1 and 2.2 μg L−1, respectively. Recoveries in spiked wine samples ranged from 95% to 104% with a median value of 102% and matrix effects were not observed. The method was applied to the evaluation of the concentration of 4-EC in 250 commercial Italian wines. The red wines analysed had median, 75° percentile and maximum values of 37 μg L−1, 89 μg L−1 and 1610 μg L−1, respectively. For Sangiovese-based wines the mean ratios of 4-EP and 4-EG to 4-EC were 3.7:1 and 0.7:1, respectively. The feasibility of a cheaper fluorimetric approach to 4-EC quantification was investigated.  相似文献

A highly sensitive flow analysis system has been developed for the trace determination of reactive phosphate in natural waters, which uses a polymer inclusion membrane (PIM) with Aliquat 336 as the carrier for on-line analyte separation and preconcentration. The system operates under flow injection (FI) and continuous flow (CF) conditions. Under optimal FI conditions the system is characterised by a linear concentration range between 0.5 and 1000 μg L−1 P, a sampling rate of 10 h−1, a limit of detection of 0.5 μg L−1 P and RSDs of 3.2% (n = 10, 100 μg L−1) and 7.7% (n = 10, 10 μg L−1). Under CF conditions with 10 min stop-flow time and sample solution flow rate of 1.32 mL min−1 the flow system offers a limit of detection of 0.04 μg L−1 P, a sampling rate of 5 h−1 and an RSD of 3.4% (n = 5, 2.0 μg L−1). Interference studies revealed that anions commonly found in natural waters did not interfere when in excess of at least one order of magnitude. The flow system, operating under CF conditions, was successfully applied to the analysis of natural water samples containing concentrations of phosphate in the low μg L−1 P range, using the multipoint standard addition method.  相似文献

In this study a new method for determination of cadmium in alcohol fuel using Moringa oleifera seeds as a biosorbent in an on-line preconcentration system coupled to flame atomic absorption spectrometry (FAAS) was developed. Flow and chemical variables of the proposed system were optimized through multivariate designs. The limit of detection for cadmium was 5.50 μg L−1 and the precision was below 2.3% (35.0 μg L−1, n = 9). The analytical curve was linear from 5 to 150 μg L−1, with a correlation coefficient of 0.9993.
