A non-aqueous capillary electrophoresis (NACE) method for quantifying the seven crocin metabolites that are the major biologically active ingredients of saffron was developed. Separation is done by using a fused silica capillary filled with a 12.5 mM H3BO3/37.5 mM sodium tetraborate methanolic solution as background electrolyte. The results obtained were compared with the total index "safranal value", widely used as a quality measure of saffron products. The comparison revealed that the proposed NACE method provides useful information not obtained in the safranal value. Infact, samples with a similar safranal value can contain crocin metabolites in different concentrations and relative proportions. This new method is very useful for quality control in commercial saffron samples.  相似文献

Saffron is a very high value-added ingredient used in the food supplement market and contains a high level of safranal. Adding synthetic safranal to saffron, which is significantly cheaper, and falsifying the origin of saffron may represent recurrent fraud. Saffron from different countries was analyzed to determine the stable isotope ratios δ13C and δ2H from safranal by gas chromatography coupled with isotope-ratio mass spectrometry (GC-C/P-IRMS) and the concentration of saffron metabolites with ultra-high performance liquid chromatography coupled with diode array detector (UHPLC-DAD). The isotopic analysis highlighted a higher ratio of δ2H in synthetic safranal than in natural safranal; the mean values were 36‰ (+/− 40) and −210‰ (+/− 35), respectively. The δ13C between Iranian, Spanish and other saffron was significantly different and represents median values of −28.62‰, −30.12‰ and −30.70‰, respectively. Moreover, linear and quadratic discriminant analyses (LDA and QDA) were computed using the two isotope ratios of safranal and the saffron metabolites. A first QDA showed that trans-crocetin and the δ13C of safranal, picrocrocin, and crocin C3 concentrations clearly differentiated Iranian saffron from other origins. A second model identified δ13C, trans-crocetin, crocin C2, crocin C3, and picrocrocin as good predictors to discriminate saffron samples from Iran, Spain, or other origins, with a total ability score classification matrix of 100% and a prediction matrix of 82.5%. This combined approach may be a useful tool to authenticate the origin of unknown saffron.  相似文献

A sensitive reagent-injection flow analysis method for the spectrophotometric determination of nitrate in marine, estuarine and fresh water samples is described. The method is based on the reduction of nitrate in a micro column containing zinc granules at pH 6.5. The nitrite formed is reacted with sulfanilamide and N-(1-naphthyl)ethylene diamine (Griess reagent), and the resulting azo compound is quantified spectrophotometrically at 520 nm. Water samples in the range of 3-700 μg L−1 NO3-N can be processed with a throughput of up to 40 samples per hour, a detection limit of 1.3 μg L−1 and reproducibility of 1.2% RSD (50 μg L−1 NO3-N, n = 10). The proposed method was successfully applied for the determination of nitrate in estuarine waters and the reliability was assessed by the analyses of certified reference materials and recovery experiments. The method is suitable for waters with a wide range of salinities, and was successfully used for more than 3200 underway nitrate measurements aboard SV Pelican1 in the “Two Bays” cruise in January 2010.  相似文献

Saffron is widely cultivated and used as a spice. Recently published data on the chemical composition and pharmacological potential of saffron determine its use in pharmacy and medicine. The proposed high-performance thin-layer chromatography (HPTLC) method allows good separation of 11 analytes. The saffron quality (Iran, Ukraine, Spain, Morocco samples) assessment was based on the European Pharmacopoeia monograph and ISO 3632. The HPTLC method for the safranal, crocin, and picrocrocin quantification was proposed and validated. The crocins content in Ukrainian saffron was from 17.80% to 33.25%. Based on qualitative and quantitative assessment results, the saffron sample from Zaporizhzhia (Ukraine) had the highest compounds content and was chosen to obtain the working standards of picrocrocin and crocins (trans-4GG, trans-2G, trans-3Gg) by preparative chromatography. The compounds were isolated from lyophilized extract of saffron using a Symmetry Prep C18 column (300 × 19 mm × 7 µm), and identified by spectroscopic techniques (HPLC-DAD, UPLC-ESI-MS/MS). The purity of crocins and picrocrocin was more than 97%. A novel method proposed to obtain working standards is simple and reproducible for the routine analysis of saffron quality control.  相似文献

A simple, fast, sensitive and robust analytical method using gas chromatography (GC)-isotope dilution mass spectrometry (MS) was developed and validated for the identification and quantification of 1,4-dichlorobenzene (p-DCB) residues in honey samples. The proposed methodology is based on steam-distillation using a Clevenger-type apparatus followed by gas chromatography-mass spectrometry (GC-MS) in the selected ion monitoring (SIM) mode employing the isotopically labeled analogue d4-1,4-dichlorobenzene (d4-p-DCB) as internal standard (IS). Validation of the method was performed in two different GC-MS systems, using quadrupole MS (QMS) and ion-trap MS (ITMS) detectors, with no statistically significant differences between two. Recoveries were better than 91% with percent relative standard deviations lower than 12%. The instrumental limits of detection were 1 μg kg−1 in the GC-ITMS system and 0.6 μg kg−1 in the GC-QMS system. The expanded uncertainty was estimated as 17% at the currently accepted “action level” of 10 μg kg−1. The method was applied to the analysis of 310 honey samples in an extensive national monitoring study. A quality control (QC) system applied during the assays has demonstrated a good performance and long-term stability over a period of more than 8 months of continuous operation.  相似文献

A new flow injection (FI) method is reported for the spectrophotometric determination of fosfestrol (diethyl-stilbestrol (DES) diphosphate) in pharmaceutical formulations. The proposed method is based on the on-line hydrolysis of the analyte by alkaline phosphatase (Al-Pase) using a “chasing zones” FI manifold. The orthophosphate ions, thus, generated are determined spectrophotometrically (λmax=690 nm) using the molybdenum blue approach. The chemical and FI variables affecting the enzymatic reaction were investigated. The proposed method is very precise (sr=1.1% at 1×10−4 mol l−1 fosfestrol, n=12), fast (allowing up to 40 samples h−1 to be analyzed) and has a determination range of 2×10−5 to 2×10−4 mol l−1, with a satisfactory 3σ detection limit of 5×10−6 mol l−1. The method was shown to provide accurate determinations of the fosfestrol concentration in a pharmaceutical formulation, giving relative errors, er, of +0.6 and −0.5% compared to the value stated by the supplier (Asta Medica Inc.) and the concentration derived using a method recommended by the United States Pharmacopoeia XXI, respectively. In addition, the average recoveries of known amounts of the analyte ranged between 99.2 and 101.2%.  相似文献

High-performance liquid chromatographic method (HPLC) with evaporative light scattering detection (ELSD) coupled with microwave-assisted extraction (MAE) as an efficient sample preparation technique has been developed for fingerprint analysis of Dioscorea nipponica. The samples were separated with an Agilent C8 column using water (A) and acetonitrile (B) under gradient conditions (0-10 min, linear gradient 20-40% B; 10-12 min, linear gradient 40-42% B; 12-25 min, isocratic 42% B) as the mobile phase at a flow rate of 1 mL min−1 within 22 min. The ELSD conditions were optimized at nebulizer-gas flow rate 2.7 L min−1 and drift tube temperature 90 °C. Precision experiments showed relative standard deviation (R.S.D.) of peak area and retention time were better than 2.5%; inter-day and intra-day variabilities showed that R.S.D. was ranged from 0.78% to 4.74%. Limit of detection was less than 50 μg mL−1 and limit of quantification was less than 80 μg mL−1. Accuracy validation showed that average recovery was between 97.39% and 104.07%. The method was validated to achieve the satisfactory precision and recovery. Relative retention time and relative peak area were used to identify the common peaks for fingerprint analysis. There are nine common peaks in the fingerprint. The quality of seven batches of D. nipponica samples was evaluated to be qualified or unqualified by the parameters “difference” and “total difference” of common peaks. Furthermore, the contents of important medicinal compounds (dioscin, prodioscin and gracillin) in different batches of D. nipponica samples were determined simultaneously using the developed HPLC-ELSD method. The results indicated variation of the herb quality which might be related to different producing area, growing condition, climate, harvest time, drug processing and so on. The developed analytical procedure was proved to be a reliable and rapid method for the quality control of D. nipponica.  相似文献

A flow injection hydride manifold was coupled to a 150 W tungsten coil electrothermal atomizer for in situ hydride collection followed by selenium and arsenic determination by ET AAS. Rhodium (200 μg), thermally reduced over the double layer tungsten atomizer, was very efficient at collecting selenium or arsenic hydrides. Prior to analysis, biological samples were digested in closed-vessels microwave digestion system. Prior to the hydride formation, both selenium and arsenic were reduced to valence state (IV) and (III), respectively. The detection limit was 35 ng L−1 for selenium and 110 ng L−1 for arsenic. Sample throughput was 70 h−1 using 30 s of hydride trapping time. Method accuracy was evaluated by analyzing biological-certified reference materials from the National Institute of Standard and Technology (SRM-1577a and SRM-1577b “bovine liver” and RM-8414 “bovine muscle powder”) and from the International Agency for Energy Atomic (A-13 “animal blood”) and one water-certified reference material from the National Institute of Standard and Technology (SRM-1640 trace elements in natural water). By applying a t-test, there was no significant difference at the 95% probability level between the results obtained with the proposed method and those certified values.  相似文献

In this study, thioglycolic acid capped-CdTe quantum dots (QDs) were modified by polyethylenimine (PEI), and then combined with fluorescein isothiocyanate (FITC) to fabricate FITC–CdTe conjugates. The self-assembly of FITC, CdTe and PEI was ascribed to electrostatic interactions in aqueous solution. The resulting conjugates were developed toward two routes. In route one, ratiometric photoluminescence (PL) intensity of conjugates (IFITC/IQDs) was almost linear toward pH from 5.3 to 8.7, and a ratiometric PL sensor of pH was favorable obtained. In route two, firstly added S2− induced remarkable quenching of QDs PL peak (at the “OFF” state), which was restored due to following addition of Cd2+ (at the “ON” state). In the conjugates, successive introduction of S2− and Cd2+ hardly influenced on FITC PL peaks. According to this PL “OFF-ON” mode, a ratiometric PL method for the detection of Cd2+ was achieved. Experimental results confirmed that the IFITC/IQDs exhibited near linear proportion toward Cd2+ concentration in the range from 0.1 to 15 μM, and the limit of detection was 12 nM. Interferential experiments adequately testified that the proposed sensors of pH and Cd2+ were practicable in real samples and complex systems. In comparison with conventional analytical techniques, the ratiometric PL method was simple, rapid, economic and highly selective.  相似文献

The application of comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC × GC-TOFMS) for the analysis of six anabolic agents (AAs) in doping control is investigated in this work. A non-polar–polar column configuration with 0.2 μm film thickness (df) second dimension (2D) column was employed, offering much better spread of the components on 2D when compared to the alternative 0.1 μm df2D column. The proposed method was tested on the “key” AA that the World Anti-Doping Agency (WADA) had listed at the low ng mL−1 levels (clenbuterol, 19-norandrosterone, epimethendiol, 17α-methyl-5α-androstane-3α,17β-diol, 17α-methyl-5β-androstane-3α,17β-diol and 3′-OH-stanozolol). The compounds were spiked in a blank urine extract obtained by solid-phase extraction, hydrolysis and liquid–liquid extraction; prior to analysis they were converted to the corresponding trimethylsilyl (TMS) derivatives. The limit of detection (LOD) was below or equal to the minimum required performance limit (MRPL) of 2 ng mL−1 defined by WADA, and the correlation coefficient was in the range from 0.995 to 0.999. The method allows choosing an ion from the full mass spectra which shows the least interference from the matrix and/or the best sensitivity; this can only be done if full scan mass spectral data are available. The advantage of GC × GC over classical one-dimensional GC (1D GC), in terms of separation efficiency and sensitivity, is demonstrated on a positive urine control sample at a concentration of 5 ng mL−1. The obtained similarity to the in-house created TOFMS spectra library at this level of concentration was in the range from 822 to 932 (on the scale from 0 to 999). Since full mass spectral information are recorded, the method allows the retro-search of non-target compounds or new “designer steroids”, which cannot be detected with established GC–MS methods that use selected ion monitoring (SIM) mode.  相似文献

Thermal analysis of the products resulted during crystallization of ETS-10 by using starting co gels with molar composition 5.0 Na2O-3.0 KF-TiO2-6.4 HCl-TAABr-7.45 SiO2-197.5 H2O, where tetralkylammonium (TAA) are tetramethyl (TMA), tetraethyl (TEA), tetrapropyl (TPA) and tetrabutylammonium (TBA), was performed. The effect of TAA+ cations (ionic radius in hydrated forms, shapes and hydrophilic/hydrophobic character) on the crystallization of ETS-10 is evident from the induction time, ti (TMA+ ? TEA+ < TPA+ < TBA+), the rate of crystallization, R (TMA+ < TEA+ < TPA+ < TBA+), morphology and size of crystallites. Organic cations play a “pore filling” role rather than as a “structure-directing” agent. The relatively flexible molecules of the symmetric tetraalkylammonium cations mixed with alkali cations (Na+, K+) participate directly at prenucleation and nucleation steps by their interaction with the silicate and titanate in aqueous colloidal dispersion.  相似文献

In this work, a flow analysis-hydride generation-gas phase derivative molecular absorption-(UV) spectrophotometric method has been developed for the direct determination of antimony in aqueous and hydro-alcoholic samples. Antimony (III) from undiluted samples is directly transformed into the gaseous stibine (SbH3) form by on-line reaction with sodium tetrahydroborate (NaBH4) in acidic medium (HCl). The gaseous phase generated is separated from the liquid phase using a commercial gas-liquid separator, and swept - with the help of a carrier gas (N2) stream - into a quartz gas cell (10 cm pathlength); where the corresponding absorption spectrum is acquired in a continuous mode over the 190-300 nm wavelength range, using a conventional spectrophotometer. A derivative strategy was selected in order to avoid the strong spectral interference of the ethanol vapor on the gaseous SbH3 absorption spectrum. In this way, the peak height at 223 nm of the second order derivative spectrum appears as a clear, clean and interference free analytical signal, which allows the direct determination of antimony. The recovery values obtained from homeopathic formulations (prepared in alcoholic medium) spiked with know amounts of antimony ranged between 97.5 and 103%. The method provides a dynamic range from 0.20 to 30 mg Sb l−1. The precision (RDS), evaluated by replicate analysis (n = 5) of samples and standard solution containing between 2.5 and 15 mg Sb l−1 was in all cases lower than 1.2%. The proposed method was applied to the determination of antimony in commercial homeopathic products (“Antimonium Tartaricum”) prepared in hydro-alcoholic medium; and showed to be simple, precise, and accurate.  相似文献

An automatic method for kinetics independent spectrometric analysis is proposed in this study. It uses a non-linear calibration model that explores concentration gradients generated by a flow-batch analyser (FBA) for the samples, dye, and the single standard solution. The procedure for obtaining the gradients of the dye and standard solution is performed once at the beginning of analysis. The same procedure is applied thereafter for each sample. For illustration, the proposed automatic methodology was applied to determine total protein and albumin in blood serum by using the Biuret and Bromocresol Green (BCG) methods. The measurements were made by using a laboratory-made photometer based on a red and green bicolour LED (Light-Emitting Diode) and a phototransistor, coupled to a “Z” form flow cell. The sample throughput was about 50 h−1 for albumin and 60 h−1 for total protein, consuming about 7 μL of sample, 2.6 mL of BCG and 1.2 mL of biuret reagents for each determination. Applying the paired t-test for results from the proposed analyser and the reference method, no statistic differences at 95% confidence level were found. The absolute standard deviation was usually smaller than 0.2 g dL−1. The proposed method is valuable for the determination of total protein and albumin; and can also be used in other determinations where kinetic effects may or may not exist.  相似文献

A simple and robust on-line sequential insertion system coupled with hydride generation atomic absorption spectrometry (HG-AAS) was developed, for selective As(III) and total inorganic arsenic determination without pre-reduction step. The proposed manifold, which is employing an integrated reaction chamber/gas-liquid separator (RC-GLS), is characterized by the ability of the successful managing of variable sample volumes (up to 25 ml), in order to achieve high sensitivity. Arsine is able to be selectively generated either from inorganic As(III) or from total arsenic, using different concentrations of HCl and NaBH4 solutions. For 8 ml sample volume consumption, the sampling frequency is 40 h−1. The detection limit is cL = 0.1 and 0.06 μg l−1 for As(III) and total arsenic, respectively. The precision (relative standard deviation) at 2.0 μg l−1 (n = 10) level is sr = 2.9 and 3.1% for As(III) and total arsenic, respectively. The performance of the proposed method was evaluated by analyzing the certified reference material NIST CRM 1643d and spiked water samples with various concentration ratios of As(III) to As(V). The method was applied for arsenic speciation in natural waters samples.  相似文献

The volatile components of Iranian saffron were extracted using ultrasonic solvent extraction (USE) technique and then were separated and detected by gas chromatography–mass spectrometry (GC–MS). Variables affecting the extraction procedure were screened by using a 25−1 fractional factorial design and among them; sample amount, solvent volume, solvent ratio and extraction time were optimized by applying a rotatable central composite design (CCD). The optimum values of factors were: 2.38 g sample, 29.04 mL solvent, 69.23% MeOH solvent ratio and 71.8 min for the extraction time. Forty constituents were identified for Iranian saffron by GC–MS representing 90% of the total peak area. The major components were 2,6,6-trimethyl-1,3-cyclohexadiene-1-carboxaldehyde, namely safranal (26.29%), bicyclo[3,2,0]hept-2-ene-,4-ethoxy-,endo (5.69%), linoleic acid (4.77%), 4-hydroxy-2,6,6-trimethyl-1-cyclohexene-1-carboxaldehyde, namely HTCC (4.44%), and nonadecanol (3.32%). Some new compounds were identified for the first time in saffron. In addition, the results of this study were compared with those of Greek saffron.  相似文献

A quantitative determination method of formic acid in apple juices is proposed by means of the proton nuclear magnetic resonance (1H NMR) technique. Formic acid gives a singlet signal at the 8.2-8.4 ppm interval of the spectrum, and its area is used to determine the concentration of the acid. 1,3,5-Benzenetricarboxylic acid is added to the juice as an internal standard. Since the chemical shift of both species varies with the pH, ascorbic acid is also added to adjust it at 2.74 and to avoid the overlapping of the signals. Recoveries between 95 and 109% are obtained when the standard addition method is applied to the juices of five different cider apple varieties. The coefficient of variation obtained is 3.9% for intra-day repeatability (n = 5), and 4.6% for inter-day repeatability (n = 10). The limit of detection is 1.49 mg/l, calculated from “3Sy/x + intercept”. The described method is direct and no previous derivatization is needed.  相似文献

RNA integrity is important in RNA studies because poor RNA quality may impact downstream methodologies. This study proposes a rapid and cost-effective method for the determination of RNA integrity based on CE-LIF in the presence of electroosmotic flow. The proposed method uses poly(ethylene) oxide (Mavg = 4,000,000 Da) as a sieving matrix for total RNA separation. Ethidium bromide (μg mL−1) was dissolved in a polymer solution as an interchelating dye for on-column fluorescent labeling. The 28S rRNA, 18S rRNA, 5.8S rRNA, 5S rRNA and tRNA from the total human RNA extracted from the cells were fully separated using the proposed method. The lowest detectable concentration of total RNA achieved was 100 pg μL−1 with a 6 min sample injection followed by on-column concentration. In addition, the temperature-induced degradation of total RNA was observed by CE-LIF. The electropherograms revealed more fragmentation of 28S and 18S rRNAs by temperature-induced hydrolysis compared with the 5.8S rRNA, 5S rRNA and tRNA. Therefore, the results indicated that RNA degradation should be considered for long-term, high-temperature incubations in RNA-related experiments involving RNA hybridization. The proposed method is furthermore, applied to the determination of 5S rRNA overexpressed in ovarian cancer cells as compared to the cervical cancer cells. Overall, CE-LIF is highly promising for rapid screening of ovarian cancers without tedious pre-amplification steps.  相似文献

We report the synthesis of a novel bistriazene, 4,4′-bis(3-(4-phenylthiazol-2-yl)triazenyl)biphenyl (BPTTBP), and its highly sensitive color reaction with Hg2+. The new reagent was synthesized in good yield by coupling 2-amino-4-phenylthiazole with 4,4′-biphenyldiamine bisdiazonium salt. Using a blend of surfactants N-cetylpyridinium chloride (CPC) and polyethylene glycol n-octanoic phenyl ether (OP) as a micelle sensitizer, the red colored reagent assembles with Hg2+ in pH 9.8 borate buffer according to a 1:1 stoichiometry, forming a blue oligomeric/polymeric chelating complex with a high apparent stability constant (1.1 × 108 M−1). Whereas the maximum absorption of reagent occurs at 510 nm with an extinct coefficient of 1.35 × 104 M−1 cm−1, the complex absorbs at 611 nm, with an apparent extinct coefficient of 1.04 × 105 M−1 cm−1. Beer's law is obeyed in the range of 0-15 μg/25 mL Hg2+, and Sandell's sensitivity is 1.92 × 10−3 μg/cm2. In the presence of thiourea and Na4P2O7 as masking agents, the method was found free from interferences of foreign ions commonly occurring with mercury. The optimized protocol has been successfully applied to spectrophotometric determination of mercury in waste water samples. The features of the new reagent associated with its special structure were discussed, and an unprecedented “domino effect” was proposed to account for its unique chelating stoichiometry with Hg2+.  相似文献

The response of a single TE102 and double TE104 rectangular cavity to the insertion of samples contained in tubes with variable wall thickness and a quartz Dewar into the cavity has been analyzed. A direct, indirect, and concurrent (positive or negative) “lens effect” inside the double TE104 rectangular cavity is discussed. The experimental dependence of the EPR signal intensity on the wall thickness of the sample tube, δ, for the line-like samples with identical length of the sample material column, L=30 mm, recorded in the microwave cavity showed a directly proportional increase of the relative “lens effect” with the increase of the wall thickness of the tube in the interval, δ∈<0.1 mm, >0.5 mm. The insertion of the variable-temperature double-wall quartz Dewar (home-built, resonant frequency shift, ca. −300 MHz) into the single TE102 rectangular cavity showed the same relative “lens effect”, with ca. 1.5-time increase of the EPR signal intensity, for a point-like sample and the line-like samples with material columns of diameter of 1 and 1.3 mm, and wall thickness of the sample tubes, δ∈<0.1 mm, >0.5 mm. The increased effect of the Dewar arises because the active volume of the quartz Dewar tube walls is always much more larger than the active volume of the sample tube wall. In the case of the double TE104 rectangular cavity, the insertion of the quartz Dewar: (i) into the same cavity, in which the sample is present, caused a direct “lens effect”, with ca. 1.8-fold increase of the EPR signal intensity; however, (ii) into the complementary cavity, in which the sample is absent, caused an indirect “lens effect”, with ca. 0.6-fold decrease of the EPR signal intensity. With the Dewar and sample in one cavity and a large empty sample tube in the complementary cavity, a concurrent (positive or negative) “lens effect” can be observed. Thus, the possible increase/decrease of the EPR signal intensity depends on the volume ratio of the quartz Dewar tube walls and large sample tube wall inserted into the double TE104 rectangular cavity. Each of the above phenomena may be a significant source error in quantitative EPR spectrometry unless the samples to be compared in the quantitative EPR analysis are contained in sample tubes having the same wall thickness and each EPR spectra should be recorded inside an identical quartz Dewar.  相似文献

Fourier Transform mid-infrared and Raman spectroscopies were used to investigate the cation/polymer, cation/urea bridge, cation/anion and hydrogen bonding interactions in poly(oxyethylene) (POE)/siloxane di-ureasil networks prepared by the sol–gel route and doped with lithium bis(trifluoromethanesulfonyl)imide (LiTFSI). Materials with compositions 200 ?n ? 5 (where n expresses the molar ratio OCH2CH2/Li+) were studied. The Li+ ions coordinate to the urea carbonyl oxygen atoms over the whole range of salt concentration considered. Bonding to the ether oxygen atoms of the POE chains occurs at n ? 40, although a significant fraction of the POE chains remain non-coordinated. In these high salt content samples, the cations interact with the anions forming contact ion pairs. “Free” ions are probably the main charge carriers at the room temperature conductivity maximum of these ormolytes.  相似文献
