3-Substituted-4- and 6-azaindoles were prepared from ortho-methyl-nitropyridines in a practically convenient, one-pot process based on the Leimgruber-Batcho reaction. The procedure comprises a sequence of (a) condensation of an ortho-methyl-nitropyridine with N,N-dimethylformamide dimethyl acetal; (b) alkylation or acylation of the enamine intermediate; (c) reduction of the nitro group to an aniline with in situ cyclization and elimination of dimethylamine to generate the 3-substituted azaindole heterocycle.  相似文献

The ortho-lithiation of a phenyl phosphorodiamidothioate derived from an enantiopure C2-symmetric diamine is studied. It is shown that the migration of the diaminophosphoryl group from sulfur to carbon, leading to an ortho-sulfanylated phenyl phosphonodiamidate, only occurs in the presence of an alkylating agent or a Lewis acid as BF3·Et2O. The influence of the chiral diaminophosphoryl group on the stereoselectivity of the oxidation of the ortho-sulfanyl or alkylsulfanyl group is also examined.  相似文献

A library of 15 1,6,7,9-tetrasubstituted 6,7,9,9a-tetrahydro-5H-pyrazolo[1,2-a]pyrrolo[3,4-c]pyrazole-1,3,5(2H,3aH)-triones was prepared by combinatorial stereoselective cycloadditions of (1Z,4R,5R)-1-arylmethylidene-4-benzoylamino-5-phenylpyrazolidin-3-on-1-azomethine imines to N-substituted maleimides. Stereochemistry was controlled by the stereodirecting phenyl group at position-3 and by the ortho-substituents at the aromatic ring at position-1′ in azomethine imines. Consequently, two sets of diastereomeric cycloadducts were obtained, one set from the ortho-unsubstituted dipoles and the other set from the ortho-disubstituted dipoles.  相似文献

The zwitterionic intermediates (2a) in the oxidation of ortho-alkylphenols (1) and bis(sulfonium ylide) 3 form reasonably stable 2:1-complexes (4), in which the ortho-quinone methide (oQM) moieties are not present in quinoid form with the exocyclic in-plane methylene group, but as zwitterionic, aromatic conformer having an out-of-plane exocyclic methylene group. The complex 7 derived from the α-tocopherol model compound PMC (5) was comprehensively characterized. As exemplarily demonstrated, the adducts can be advantageously employed in organic synthesis as ‘stabilized oQMs’.  相似文献

A combinatorial library of 4-(2-hydroxyaryl)-3-nitro-4H-chromenes was synthesized in high yield by C4-SMe substitution in N-alkyl/phenyl 4-(methylthio)-3-nitro-4H-chromen-2-amines with a variety of phenols. The reaction always provided C2 substitution in the phenol ring, dictated by hydrogen bond interactions between the phenolic hydroxyl group and the nitro group in 3-nitro-4H-chromenes. Reduction of the nitro group with concomitant hydrolysis of the enamine in 4-(2-hydroxyaryl)-3-nitro-4H-chromenes with Zn, Ac2O in AcOH furnished hybrid amino-acid lactone incorporating ortho-tyrosine and phenyl alanine moieties.  相似文献

Cycloadditions of (1Z,4R*,5R*)-4-benzoylamino-5-phenylpyrazolidin-3-on-1-azomethine imines to olefinic dipolarophiles were studied. Stereochemistry of cycloadditions to azomethine imines 3 was found to be controlled by stereodirecting phenyl group at position 3, as well as by the ortho-substituents at the aromatic ring at position 1′. The structures of dipoles and products were confirmed by NMR and X-ray diffraction.  相似文献

2-(4-Chlorophenyl)-2-methyl-1,3-dioxolane 2a was lithiated ortho to the ketal group by treatment with butyllithium in THF at 0°C. Related 2-aryl-2-methyl-1,3-dioxolanes possessing a chlorine substituent at the meta position of the aryl group 2b,c were lithiated with butyllithium in THF at −78°C at the position between the two directing groups. The lithio species thus generated were treated with various electrophiles to give ortho-functionalized acetophenone derivatives.  相似文献

Molecular and crystal structure of a series of derivatives of N,N-dimethyl-4-nitroaniline has been studied by both X-ray diffraction method and high-level ab initio calculations. According to these data, the dimethylamino groups were found to have a trigonal-pyramidal configuration and are considerably turned with respect to the ring plane in all molecules having a substituent in the ortho-position; on the contrary, this group is planar in the meta-substituted molecules. Topological analysis of the electron density function for all molecules studied within the framework of Bader's ‘atoms in molecules’ (AIM) theory revealed that introduction of a substituent into the ortho- or meta-position of the ring results in increasing of the contribution of the resonance forms different from the quinoid one. Contribution of the latter form is predominant for the structure of N,N-dimethyl-4-nitroaniline (1). Topological analysis of the electron density distribution was used to explain a decreasing of the molecular hyperpolarisabilites of the ortho- and meta-substituted compounds as compared with those for 1.  相似文献

The metalation of (quasi)alkoxy-substituted dibromobenzenes C6H3(OR)Br2 with lithium diisopropylamide (LDA) has been investigated. For 1-(quasi)alkoxy-3,5-dibromobenzenes (R = Me, TMS), different selectivities were observed depending on reaction conditions and the size of the alkoxy group. The methoxy group was an effective ortho-director whereas this was not the case for the bulky trimethylsilyloxy group. The metalation of related 2,5-dibromoanisole was also examined showing a significant meta-directing effect by the methoxy group. The thermal stability of aryllithium intermediates is significantly lower when lithium is flanked by a bromine and a methoxy group, whereas 4-(quasi)alkoxy-2,6-dibromoaryllithiums are less labile.  相似文献

A facile one-pot synthesis of 3-aminoquinolines from ortho-aminobenzaldehydes was developed. Ethyl 6,7-dimethoxy-3-aminoquinoline-2-carboxylate, a key intermediate for the preparation of a 4-anilino-benzo[b][1,5]-naphthyridine-3-carbonitrile, was efficiently prepared by this method. A new and efficient method for the preparation of ortho-N-Boc-arylmethyl ketone derivatives is reported. The reaction involves the intermediacy of a 4-methylene-3,1-benzoaxin-2-one moiety which smoothly converts to the target compounds under acidic conditions. The ortho-N-Boc-arylmethyl ketone derivatives can be formed in a one-pot reaction or alternatively the 4-methylene-3,1-benzoaxin-2-ones can be isolated and subsequently transformed into the desired products.  相似文献

Dibenzylphenylphosphine in the reaction with CoMe(PMe3)4 afforded complex [(Me3P)3Co((ortho-C6H4)∩P(C6H5)(CH2C6H5))] (1) by Csp2-H activation via ortho-metalation with P atom as anchoring group. An unexpected dinitrogen iron(II) complex [(Me3P)2(N2)Fe((ortho-C6H4)2∩P(C6H5))] (2) stabilized by two five-membered chelate rings as [CPC]-pincer ligand was formed through the reaction of dibenzylphenylphosphine with FeMe2(PMe3)4 via double Csp2-H activation. The reactions of complexes 1 and 2 with carbon monoxide delivered carbonyl complexes [(Me3P)(CO)2Co((ortho-C6H4)∩P(C6H5)(CH2C6H5))] (3) and [(Me3P)2(CO)Fe((ortho-C6H4)2∩P(C6H5))] (4). An iodo methyl cobalt(III) complex [(Me3P)2(Me)(I)Co((ortho-C6H4)∩P(C6H5)(CH2C6H5))] (5) was isolated through the reaction of 1 with iodomethane. The structures of 1, 2, 3, 4 and 5 were determined by X-ray diffraction.  相似文献

Cleavage of ortho-(dodeca-1,3-diynyl)triazenes in HCl or HBr medium and subsequent cyclization of the resulting diazonium salts is investigated. In the absence of a strong electron-withdrawing substituent, the reaction affords 3-alkynyl-4-bromo(chloro)cinnolines as the only product. A methoxycarbonyl group promotes hydrolysis of 4-halocinnolines which results in the formation of by-products: furo[3,2-c]cinnoline and cinnolinone. Substitution of bromine in 3-(alk-1-ynyl)-4-bromocinnolines is achieved with methylamine, Na2S and ethynylbenzene affording pyrrolo[3,2-c]cinnoline, thieno[3,2-c]cinnoline and 3,4-diethynylcinnoline, respectively.  相似文献

Application of our original photooxidation-reduction methodology to prenylated dihydroxycoumarin and trihydroxyxanthone compounds led to the corresponding ortho-(2-hydroxy-3-methylbut-3-enyl)phenol derivatives with yields ranging from 8 to 65%. In most of the reported experiments, the oxidation products distribution, after the photooxygenation step, was controlled by the competition between the large group effect and the stabilising phenolic assistance effect. We also showed that ortho-(3-hydroxy-3-methylbut-1-enyl)phenol derivatives could be considered as biogenetic precursors of 2,2-dimethylbenzopyranic structures.  相似文献

3-Aroylquinoxalin-2(1H)-ones were found to be hetero analogues of α-diketones for the efficient, one-pot, three component synthesis of 2,4,5-trisubstituted imidazoles and imidazo[1,5-a]quinoxalin-4(5H)-ones in boiling methanol. The key advantages of this process are high yields, ready availability and low cost of 3-aroylquinoxalin-2(1H)-ones and easy work-up and separation of the products by non-chromatographic methods. Furthermore, the presence of an ortho-iminoanilide fragment at position 4 of the imidazoles obtained has made it possible to produce 2-(imidazol-4-yl)benzimidazoles in almost quantitative yields.  相似文献

Copper-catalyzed tandem reaction of ortho-bromobenzamides and isothiocyanates is described, which provides an efficient and practical route for the synthesis of 2-thioxo-2,3-dihydroquinazolin-4(1H)-ones. The optimal condition involved the following parameters: CuI as precatalyst, Cs2CO3 as base, N,N′-dimethylethane-1,2-diamine as ligand, and toluene as solvent, with reaction temperature at 120 °C.  相似文献

Oxidation of (3,4-dihydroxyphenyl)acetonitrile gives (3-hydroxy-4-oxo-cyclohexa-2,5-dienylidene)acetonitrile via the isomeric ortho-quinone ((3,4-dioxo-cyclohexa-2,5-dienylidene)acetonitrile). This para-quinomethane product is formed as a mixture of diastereoisomers and with an initial composition of Z/E 4:1. Over 24 h this mixture equilibrates to a composition of Z/E 5:4. The para-quinomethane reacts with morpholine to give (3,4-dihydroxyphenyl)-morpholin-4-yl-acetonitrile. Similar oxidation of (3,4-dihydroxynaphthalen-1-yl)acetonitrile also gives a para-quinomethane derivative and not the 1,2-naphthoquinone as previously described. The reactivity of this para-quinomethane derivative as a 1,3-dienophile in a key step of the Gates' morphine synthesis is attributed to formation of its conjugate acid.  相似文献

Novel fused azecine ring systems were synthesized via the microwave-assisted thermal isomerization of terphenyl or biphenyl-pyridazine compounds possessing a vinyl and a tert-amino group, through application of a new extension of the tert-amino effect. Substrates for the ring closure were prepared from ortho-dihalobenzene or pyridazinone by consecutive Suzuki couplings with ortho-sec-amino- and formylphenylboronic acids, followed by Knoevenagel condensation of the aldehydes obtained.  相似文献

An unprecedented intramolecular Friedel-Crafts ipso-alkylation at the ortho-alkyl group of the N-arylamino moiety of 1-allyl-1-N-arylaminocycloalkanes to give alkyl substituted dihydrospiro[(1H) quinoline-2,1-cycloalkanes] is reported. Unexpected results were explained in the context of intramolecular ipso-substitution of alkyl groups and their 1,2-rearrangement. A plausible mechanism for this type of Friedel-Crafts alkylation by an alkene moiety promoted by a Brönsted acid (H2SO4) is proposed.  相似文献
