[reaction: see text] A general process for the efficient synthesis of sulfinyl transfer agents has been developed using cinchona alkaloids quinine and quinidine as chiral auxiliaries. The importance of these new and unique sulfinyl transfer agents is exemplified by the expedient synthesis of several sulfoxides in excellent enantiopurities and high yields.  相似文献

A three step efficient strategy for the synthesis of substituted 5-nitropiperidones in high de, employing Michael addition of N-p-tolylsulfinyl beta-nitroamines to alpha,beta-unsaturated esters, hydrolysis of the sulfinyl group, and cyclization of the resulting free amines, has been developed. A very simple experimental procedure involving mild conditions and only one chromatographic purification are the main features of the process.  相似文献

[reaction: see text] An efficient method has been developed to prepare all four isomers of the hydroxyl derivatives of sibutramine by addition of Grignard reagents (R)- or (S)-5 to a single enantiomer of sulfinyl imine (R)-1 simply by tuning the reaction solvent. The phenomenon of the reversed diastereoselectivity in CH(2)Cl(2) and THF implied that the reaction may proceed through a chelated cyclic transition state in CH(2)Cl(2) and nonchelated acyclic transition state in THF.  相似文献

Diastereomerically pure tridentate heteroorganic ligands containing hydroxyl, sulfinyl and aziridine moieties as nucleophilic centers, capable of binding to various organometallic reagents, have been proven to be highly efficient catalysts in the enantioselective addition of phenylethynylzinc to aldehydes to give the desired products in very high yields (up to 95%) and with ee’s up to 90%. The influence of the stereogenic centers located on the sulfinyl sulfur atom and in the aziridine moiety on the stereochemical course of the reaction is also discussed.  相似文献

Thiolsulfinates react with trifluoro- or trichloroacetic anhydride to give equimolar mixtures of the corresponding disulfides and sulfinyl trifluoro- or trichloroacetates which are in equilibria with sulfenyl carboxylates. Although the equilibrium lies far toward sulfinyl carboxylates at room temperature, addition of olefins to the mixed solution of sulfinyl carboxylate and a corresponding disulfide affords the adducts which are formed in the reaction between the corresponding sulfenyl carboxylates and the olefins. Meanwhile, treatment of carboxylic acid silver salts with sulfinyl chlorides also gives sulfinyl carboxylates, however, sulfinyl carboxylates have not been successfully isolated yet.  相似文献

A remarkable perturbation of the azo system by the stereogenic sulfinyl moiety has been observed by CD in both the trans and the cis azobenzenes resulting by photoisomerization. After five irradiation cycles, the configurational integrity of these chiral switches remains unchanged.  相似文献

A simple, environmentally friendly, and very efficient procedure for the synthesis of optically pure N-(tert-butylsulfinyl)imines has been developed with microwave-promoted condensation of aldehydes and ketones using (R)-2-methylpropane-2-sulfinamide in the presence of Ti(OEt)(4), under solvent-free conditions. This procedure allows for the preparation of a variety of sulfinyl aldimines with excellent yields and purities in only 10 min, making any further purification of the imines unnecessary. Several sulfinyl ketimines have also been prepared in good yields by extension of the reaction times to 1 h. This methodology has proved to be equally efficient for the synthesis of aromatic, heteroaromatic, and aliphatic N-(tert-butylsulfinyl)imines. Conventional heating has also been shown to be useful to promote these reactions, especially for the synthesis of aldimines.  相似文献

The highly selective base-promoted cyclization of enantiopure sulfinyl dienols affords allylic sulfinyl dihydropyrans. The scope of this methodology, including the preparation of seven-membered rings, has been studied in depth. The reactivity of our sulfinyl dihydropyrans toward oxidation, imination, and dihydroxylation has been explored, and thus several routes to densely functionalized pyran derivatives have been outlined. The reactivity of allylic dihydropyranyl sulfones and sulfoximines in S(N)2' processes with organocuprates has been examined. The displacement products were obtained with good regio- and stereoselectivity and fair to good yields. The reactivity of these products to dihydroxylation opens new possibilities to access enantiopure polyhydroxylated tetrahydropyrans that could be of interest for the synthesis of natural products.  相似文献

2,3-Dihydro-2,2,4-trimethylthieno[3,2-b]pyridinium 1-oxide iodide ( 1 ) has been reacted with various inorganic and organic hydride donors. It has been found that the stereochemistry of reaction is controlled by the orientation of the sulfinyl dipole, and the relative bulkiness of substituents plays no role in determining the reaction face: a reactive hydride donor prefers to attack at the anti-face with respect to the sulfinyl dipole, whereas a less reactive hydride donor prefers to attack at the syn-face.  相似文献

New chiral ligands, N-diphenylphosphano nitrogen-containing five-membered aromatic compounds bearing chiral sulfinyl groups as the sole chiral source has been developed. The structure of a palladium intermediate derived from the new chiral sulfoxide ligand was determined as a palladium complex with a five-membered chelate ring formed by coordination of the phosphano group and the sulfinyl sulfur atom involved. The stereoelectronic effects of substituents on the aromatic rings are discussed.  相似文献

A novel route to enantiopure densely functionalized epoxy sulfinyl tetrahydrofurans, based on the unexpected and highly stereoselective remote nucleophilic epoxidation of hydroxy 1-sulfinyl butadienes with t-BuOOK, followed by ring closure and subsequent epoxidation of the resulting sulfinyl dihydrofurans, is described. Alternatively, the treatment of these dienes with m-CPBA followed by acid-catalyzed cyclization gives rise to related sulfonyl dihydrofurans in high yields but with low selectivity. The stereochemical outcome of the nucleophilic epoxidation of these substrates has also been studied.  相似文献

The design and preparation of a novel class of ligands based on the sulfinyl imine functionality is described. In particular, an efficient and modular synthesis of bis(sulfinyl)imidoamidine (siam) ligands is reported. The versatility of the synthetic sequence is demonstrated by the preparation of various analogues to explore the effect of substitution about the ligand framework on catalytic activity. The utility of the siam ligands in asymmetric catalysis is demonstrated in the Cu(II)-catalyzed Diels-Alder reaction where highly enantio- and diastereoselective reactions are reported for a range of N-acyloxazolidinone dienophile and diene substrate combinations. Of particular note is the efficiency of these asymmetric catalysts for reactions involving challenging and relatively unreactive acyclic diene substrates. Finally, structural data are provided for several ligands as well as metal-ligand complexes.  相似文献

An efficient synthesis of peri-hydroxy aromatic compounds has been accomplished via a strong-base-induced [4+2] cycloaddition of homophthalic anhydrides with alpha-sulfinyl-substituted derivatives of enolizable enones. The unsubstituted enones did not undergo an efficient [4+2] cycloaddition reaction with homophthalic anhydrides, presumably due to their enolization under the basic reaction conditions. The sulfinyl group not only promotes the cycloaddition reaction but also undergoes in situ elimination under the reaction conditions to afford the peri-hydroxy aromatic compounds in a single step. The application of this methodology for the synthesis of a key intermediate of antitumor antibiotic fredericamycin A is described. PM3 calculations of various 2-substituted cyclopentenones as well as the mechanism of the cycloaddition are also discussed.  相似文献

[reactions: see text] Readily available sulfinyl and sulfonyl tetrahydrofuran methanol derivatives have been transformed efficiently into a variety of substituted tetrahydrofuryl alcohols by treatment with (PhSe)2 in the presence of an excess of NaBH4. Alternatively, oxirane cleavage with MgI2 produces the related ketones, amenable to stereocontrolled reduction. This reductive cleavage methodology has been applied to short formal syntheses of trans-Kumausyne and Kumausallene.  相似文献
