Two-carbon homologation of ketones via sily ketene acetals: Synthesis of α,β-unsaturated acids and α-trimethylsilyl δ-ketoacids

The reaction of C,O,O-tris(trimethylsilyl)ketene acetal 1 with saturated, cyclic and aromatic ketones 2 proceeds smoothly in the presence of titanium chloride to give (E)-α,β-unsaturated carboxylic acids 3 with fairly good stereoselectivity. With α,β-unsaturated ketones 4, α-trimethylsilyl δ-ketoacids 5 (syn + anti) are obtained according to Michael-type 1,4 addition. These diastereoisomers are separated and the configurations of 5a are achieved by X-ray molecular analysis.     

The Darzens condensation of α,β-unsaturated aldehydes and ketones

The one-pot Darzens condensation of α,β-unsaturated aldehydes and ketones with enolates of an α-bromo ester or ketone is described.     

Modular approach to saturated and α,β-unsaturated ketones

2-Fluoro-6-pyridinyloxy derivatives of 2-ethoxyvinyl carbinols react with radicals derived from xanthates by an addition-fragmentation pathway to give highly functionalized ketones after acid hydrolysis. 1,4-Diketones are readily accessible by this approach. α,β-Unsaturated ketones can be obtained by starting with geminal acetoxy xanthates prepared by addition of a simpler xanthate to vinyl acetate.     

Synthesis of β-carbolines from aldehydes and ketones via the α-siloxy α,β-unsaturated esters

We report an efficient method for the synthesis of β-carbolines from α-siloxy α,β-unsaturated esters, which are accessible from a variety of aldehydes and ketones.     

Palladium-catalyzed 1,4-addition of diarylphosphines to α,β-unsaturated aldehydes

A highly stereoselective asymmetric 1,4-addition of diarylphosphines to α,β-unsaturated aldehydes catalyzed by a bis(phosphine) pincer-Pd complex has been developed for the synthesis of chiral phosphines with excellent stereoselectivity (up to 98% ee) under mild conditions. The application of the current method to the synthesis of enantiopure bisphosphine and its pincer-Pd complex has also been demonstrated.     

Organocatalytic oxy-Michael addition of alcohols to α,β-unsaturated aldehydes

1,4-Addition of alcohols to α,β-unsaturated aldehydes was found to be efficiently promoted by biphenyldiamine-based catalyst 3 without formation of the acetals.     

Cobalt-catalyzed conjugate addition of silylacetylenes to α,β-unsaturated ketones

Catalytic addition of silylacetylenes to α,β-unsaturated ketones proceeded in the presence of a cobalt complex coordinated with a bisphosphine ligand to give high yields of β-alkynylketones.     

Addition of esters of nitroacetic acid to α,β-unsaturated ketones

Summary The addition reaction of esters of nitroacetic acid to, -unsaturated ketones was studied, and new esters of ketonitroacids were prepared.     

DABCO catalyzed dimerization of α,β-unsaturated ketones and nitriles

α,β-Unsaturated ketones dimerize in the presence of catalytic amount of 1,4-diazabicyclo[2.2.2]octane(DABCO) to produce the corresponding 2-methylene-1,5-diketones in good yields. Acrylonitrile provides the corresponding dimerized product.     

Catalytic asymmetric aziridination of α,β-unsaturated aldehydes

The development, scope, and application of the highly enantioselective organocatalytic aziridination of α,β-unsaturated aldehydes is presented. The aminocatalytic azirdination of α,β-unsaturated aldehydes enables the asymmetric formation of β-formyl aziridines with up to >19:1 d.r. and 99% ee. The aminocatalytic aziridination of α-monosubstituted enals gives access to terminal α-substituted-α-formyl aziridines in high yields and up to 99% ee. In the case of the organocatalytic aziridination of disubstituted α,β-unsaturated aldehydes, the transformations were highly diastereo- and enantioselective and give nearly enantiomerically pure β-formyl-functionalized aziridine products (99% ee). A highly enantioselective one-pot cascade sequence based on the combination of asymmetric amine and N-heterocyclic carbene catalysis (AHCC) is also disclosed. This one-pot three-component co-catalytic transformation between α,β-unsaturated aldehydes, hydroxylamine derivatives, and alcohols gives the corresponding N-tert-butoxycarbonyl and N-carbobenzyloxy-protected β-amino acid esters with ee values ranging from 92-99%. The mechanisms and stereochemistry of all these catalytic transformations are also discussed.     

Organocatalytic asymmetric nitrocyclopropanation of α,β-unsaturated aldehydes

A novel organocatalytic highly enantioselective nitrocyclopropanation reaction of α,β-unsaturated aldehydes is presented. The 1-nitro-2-formylcyclopropane derivatives synthesized from this catalytic transformation were converted to the corresponding β-nitromethyl-acid esters, which are excellent precursors of GABA analogues such as Baclofen, by subsequent organocatalytic chemoselective ring-opening.     

Asymmetric organocatalytic oxy-Michael addition of alcohols to α,β-unsaturated aldehydes

A 1,4-addition of alcohols to α,β-unsaturated aldehydes was found to be efficiently promoted by biphenyldiamine-based catalyst 3 without formation of the acetals. An asymmetric variant of this reaction has also been performed by designing a novel axially chiral organocatalyst (R)-10c.     

Green, efficient and practical Michael addition of arylamines to α,β-unsaturated ketones

The aza-Michael addition of aromatic amines to α,β-unsaturated ketones was carried out effectively at room temperature in good to excellent yields without any catalyst or solvent. It was significant that part of adducts could be collected in almost quantitative yield without column chromatography. This procedure offered a green, efficient, and practical approach for the synthesis of β-amino ketones.     

Structure of N,N-disubstituted α,β-unsaturated α-amino aldehydes

Study of the IR and¹H and¹³C NMR spectra of N,N-disubstituted ,-unsaturated -amino aldehydes showed that in the nature of the electron density distribution these compounds occupy an intermediate position between -unsubstituted amino olefins and ,-unsubstituted aldehydes, being significantly closer to the latter.Translated from Izvestiya Akademii Nauk SSSR, Seriya Khimicheskaya, No. 9, pp. 2031–2034, September, 1991.     

CuCl/bipyridine-catalyzed addition reactions of arylboroxines with aldehydes, α,β-unsaturated ketones, and N-tosyl aldimines
