The micellar electrokinetic chromatography separation of a group of triazine compounds was optimized using a combination of experimental design (ED) and artificial neural network (ANN). Different variables affecting separation were selected and used as input in the ANN. A chromatographic exponential function (CEF) combining resolution and separation time was used as output to obtain optimal separation conditions. An optimized buffer (19.3 mM sodium borate, 15.4 mM disodium hydrogen phosphate, 28.4 mM SDS, pH 9.45, and 7.5% 1-propanol) provides the best separation with regard to resolution and separation time. Besides, an analysis of variance (ANOVA) approach of the MEKC separation, using the same variables, was developed, and the best capability of the combination of ED-ANN for the optimization of the analytical methodology was demonstrated by comparing the results obtained from both approaches. In order to validate the proposed method, the different analytical parameters as repeatability and day-to-day precision were calculated. Finally, the optimized method was applied to the determination of these compounds in spiked and nonspiked ground water samples.  相似文献

The prediction of migration time of electroosmotic flow (EOF) marker was achieved by applying artificial neural networks (ANN) model based on principal component analysis (PCA) and standard normal distribution simulation to the input variables. The voltage of performance, the temperature in the capillary, the pH and the ionic strength of background electrolytes (BGE) were applied as the input variables to ANN. The range of the performance voltage studied was from 15 to 27 kV, and that of the temperature in the capillary was from 20 to 30 °C. For the pH values studied, the range was from 5.15 to 8.04. The range of the ionic strength investigated in this paper was from 0.040 to 0.097. The prediction abilities of ANN with different pre-processing procedure to the input variables were compared. Under the same performance conditions, the average prediction error of the migration time of the EOF marker was 5.46% with RSD = 1.76% according to 10 parallel runs of the optimized ANN structure by the proposed approach, and that of the 10 parallel predictions of the optimal ANN structure for the different performance conditions was 12.95% with RSD = 2.29% according to the proposed approach. The study showed that the proposed method could give better predicted results than other approaches discussed.  相似文献

Artificial neural networks (ANNs) were successfully developed for the modeling and prediction of electrophoretic mobility of a series of sulfonamides in capillary zone electrophoresis. The cross-validation method was used to evaluate the prediction ability of the generated networks. The mobility of sulfonamides as positively charged species at low pH and negatively charged species at high pH was investigated. The results obtained using neural networks were compared with the experimental values as well as with those obtained using the multiple linear regression (MLR) technique. Comparison of the results shows the superiority of the neural network models over the regression models.  相似文献

A capillary zone electrophoresis method for determining cis- and trans-resveratrol isomers is proposed. Optimal conditions for the quantitative separation were investigated. A background electrolyte solution consisting of 40 mM borate buffer adjusted to pH 9.5, hydrodynamic injection and 5 kV of separation voltage were used. Good linearity and precision were obtained for the two isomers. Detection limits of 0.06 mg L-1 for trans-resveratrol and 0.08 mg L-1 for cis-reveratrol were obtained. The developed method is rapid and sensitive and it has been applied to determine cis- and trans-resveratrol in several red wines. The samples were purified and enriched by passing them through a preconditioned C18 cartridge and eluting the isomers with acetonitrile-water (3 + 7).  相似文献

Matrix solid-phase dispersion (MSPD) as a sample preparation method for the determination of two potential endocrine disruptors, linuron and diuron and their common metabolites, 1-(3,4-dichlorophenyl)-3-methylurea (DCPMU), 1-(3,4-dichlorophenyl) urea (DCPU) and 3,4-dichloroaniline (3,4-DCA) in food commodities has been developed. The influence of the main factors on the extraction process yield was thoroughly evaluated. For that purpose, a 3(4–1) fractional factorial design in further combination with artificial neural networks (ANNs) was employed. The optimal networks found were afterwards used to identify the optimum region corresponding to the highest average recovery displaying at the same time the lowest standard deviation for all analytes. Under final optimal conditions, potato samples (0.5 g) were mixed and dispersed on the same amount of Florisil. The blend was transferred on a polypropylene cartridge and analytes were eluted using 10 ml of methanol. The extract was concentrated to 50 μl of acetonitrile/water (50:50) and injected in a high performance liquid chromatography coupled to UV–diode array detector system (HPLC/UV–DAD). Recoveries ranging from 55 to 96% and quantification limits between 5.3 and 15.2 ng/g were achieved. The method was also applied to other selected food commodities such as apple, carrot, cereals/wheat flour and orange juice demonstrating very good overall performance.  相似文献

Quantitative capillary electrophoretic analysis of chiral compounds might be difficult or even impossible when baseline separation is not reached. In this work, the use of n-th derivative of the electropherogram was studied and examined on model and experimental data. The electropherograms should be first smoothed using Savitzky-Golay method and the quantitative analysis is then possible using either a graphical method or multivariate calibration applying a combination of experimental design (ED) and artificial neural networks (ANNs). The best results were obtained for the first derivative, higher derivatives are not suitable because of noise accumulation. The method was applied to real experimental data to quantify chiral amino acids from unresolved peaks, but it is applicable for quantitative analysis of any other chiral analytes from poorly resolved peaks. Precision of analysis from partially resolved peaks reached was about +/- 3.2% relative standard deviation.  相似文献

A method for the determination of cyclamate in food was developed using solid-phase extraction (SPE) and capillary electrophoresis (CE) with indirect ultraviolet (UV) detection. A 5-10 g sample in 0.1 mol/L hydrochloric acid was homogenized and made up to a volume of 50 mL with 0.1 mol/L hydrochloric acid. After the sample was centrifuged, 25 mL of supernatant was loaded into an Oasis HLB SPE cartridge. The cartridge was washed with 2 mL of demineralized water followed by 2 mL of 50% aqueous methanol, and cyclamate was eluted with 4.5 mL of 50% aqueous methanol. The eluate was added to a solution of sodium propionate (internal standard) for CE analysis. The cyclamate in the eluate was electrophoresed on a fused-silica capillary using 1 mmol/L hexadecyltrimethylammonium bromide and 10 mmol/L potassium sorbate as a running buffer. Detection and reference wavelengths of cyclamate determined with a UV detector were 300 and 254 nm, respectively. The calibration curves for cyclamate showed good linearity in the range of 2-1000 microg/mL and the limits of detection in beverage, fruit in syrup, jam, pickles and confectionary are sample dependent and ranged from 5-10 microg/g. The recovery of cyclamate added at a level of 200 microg/g to various kinds of foods was 93.3-108.3% and the relative standard deviation was less than 4.9% (n=3). A number of commercial samples were analyzed using the proposed method. Cyclamate was detected in one waume, two pickles, and two sunflower seeds. The quantitative values determined with CE correlated to those from high-performance liquid chromatography (HPLC) (the detected values of cyclamate in a sunflower seed measured by CE and HPLC were 3.40 g/kg and 3.51 g/kg, respectively). This analytical method for cyclamate using CE is especially suitable for use in the field.  相似文献

The potentiality of artificial neural networks for multicomponent analysis in unresolved peaks from capillary electrophoresis (CE) is evaluated. The system chosen consists of mixtures of three ebrotidine metabolites, which cannot be successfully separated by CE. Data selected for analysis consist of UV spectra taken at the maximum of the CE peak. The most dissimilar analyte, in terms of spectral differences, is accurately quantitated in any type of mixture with an overall prediction error of 5%. Because of the strong interference of the two most overlapped compounds, a preliminary procedure for spectral data filtering based on principal component analysis is performed to improve their quantitation.  相似文献

A rapid solid-phase extraction-capillary zone electrophoresis (CZE) method for determining 2,4-dichlorophenoxyacetic acid, 4-(2,4-dichlorophenoxy) butyric acid, and 2,4,5-trichlorophenoxyacetic acid in real water samples is described. Factors affecting the recoveries and detection of the targets are investigated. With samples being acidified to pH 2 and salted by sodium sulfate to 2% (w/w), an average recovery of greater than 85% is obtained using ethyl acetate as the eluent on an octadecylsilane-bonded silica cartridge. A running buffer of 5 mM sodium tetraborate in a water-acetonitrile mixture (70:30, v/v) adjusted to pH 9 is employed in the CZE analysis, and the targets can be analyzed within 7 min with good reproducibility and acceptable sensitivity. The method is suitable for detecting herbicide residues of sub-parts-per-billion levels in surface water. A local pond water is analyzed, and the concentrations of 2,4-dichlorophenoxyacetic acid and 4-(2,4-dichlorophenoxy) butyric acid are detected to be 0.27 +/- 0.03 ppb and 0.61 +/- 0.08 ppb, respectively.  相似文献

Summary Sulfonylurea herbicides in soil extracts were concentrated using off-line solid-phase extraction (SPE), and determined by capillary zone electrophoresis (CZE) and UV detection. The method involves extraction of soils with 0.1 M NaHCO3 solution and subsequent preconcentration by using C18 cartridges prior to separation of the pesticide using CZE. The results show that a C18 cartridge is suitable for the purification of sulfonylurea herbicides in soil extracts with the recoveries ranging from 65–103%. The separation conditions affecting the resolution and detection sensitivity was systematically investigated. The sulfonylureas were resolved well using 30 mM sodium acetate (NaAc)/acetic acid (HAc)+10% acetonitrile (ACN) buffer at pH 4.80. The calibration plots for the test solutes in the concentration of 0.2–50 mg L−1 were linear with detection limits in the range of 0.05–0.10 mgL−1. The proposed method has been successfully demonstrated for the determination of sulfonylurea herbicides in soil samples.  相似文献

Herba Epimedii (family Berberidaceae), Ying-Yang-Huo in Chinese, is a famous Chinese herbal medicine. Flavonoids are thought to be the major active components in it. A capillary zone electrophoresis (CZE) separation were developed for simultaneous determination of four flavonoids including icariin, epimedin A, epimedin B and epimedin C in Epimedium. The effects of the experimental variables on CZE had been optimized by using central composite design (CCD). The best separation of four flavonoids could be obtained using 50 mM borate buffer (pH 10.0) containing 22% acetontrile as modifier, while separation voltage was 15 kV and temperature was at 25 degrees C. The method developed is accurate, simple and reproducible, which could be used for quality control of Epimedium and its medical preparations.  相似文献

In this paper, we propose a new strategy for separation and determination of tobacco-specific N-nitrosamines (TSNAs), a group of strong carcinogens found only in tobacco products, by using CZE and CE-MS associated with SPE. Six TSNAs: N'-nitrosonornicotine, N'-nitrosoanatabine, N'-nitrosoanabasine, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol, and 4-(methylnitrosamino)-4-(3-pyridyl)-1-butanol were simultaneously separated by either of two CZE methods, one of which worked with ammonium formate buffer (pH 2.5) and another with citrate buffer (pH 2.4), as well as a CE-MS method. The CZE conditions including pH and concentration of running buffer, capillary length, applied voltage, and capillary temperature were systematically optimized. For CE-MS method, an optimized sheath liquid consisted of methanol-water was used at a flow rate of 10 muL/min. With SPE procedure, our proposed CE-MS method was successfully applied to determine TSNAs after 15 min metabolism in rabbits. A comparison study between CZE and CE-MS methods for quantitative purposes was carried out, showing that both methods provided similar separation efficiency, selectivity, repeatability, linearity, and recovery. However, CE-MS method was better suited for the analysis of TSNAs in complicated biological samples for its sensitivity and extra information on molecular structure. Having good accordance with our previous work by using LC-MS, the new CE-MS method is expected to be an alternative to the LC-MS method and applied to study the metabolism of TSNAs.  相似文献

Capillary zone electrophoresis (CZE) in an organic medium was used to analyse triazines at sub-ppb concentration levels in natural waters after a preconcentration step using conventional C18 cartridges and new Oasis HLB devices. With both sorbents, satisfactory results were obtained on analysing deionized water. However, on analysing natural waters, both sorbents showed very different types of behaviour. The different variables affecting the elution of both sorbents were studied, resulting in the choice of Oasis HLB as the most suitable for later separation by CZE in non-aqueous medium. Combination of a preconcentration step with electrokinetic injection revealed that capillary electrophoresis with simple UV detection can also be used satisfactorily for the quantification of micropollutants in natural waters. The detection limits obtained varied between 0.01 and 0.05 microg l(-1), depending on the type of matrix analysed. The day-to-day precision varied between 0.9% and 2.3%, expressed as the relative standard deviation.  相似文献

A magnetic solid-phase extraction (MSPE) method combined with capillary electrophoresis for the simultaneous determination of seven quinolones (QNs) (danofloxacin, ciprofloxacin, marbofloxacin, enrofloxacin, difloxacin, oxolinic acid, and flumequine), using (S)-(+)-6-methoxy-α-methyl-2-naphthaleneacetic acid as internal standard, in milk samples was developed. The variables involved in the preconcentration magnetic procedure were: the composition of the magnetic support composition, the sample pH, and the weight of magnetic adsorbent used. The variables were optimized using a simplex-lattice design. Different magnetite covered with octyl-phenyl silica adsorbents were synthesized by varying the molar ratio of phenyltrimethylsilane and octyltrimethoxysilane; the solids were evaluated for QN preconcentration. Under optimal conditions, a linear range was obtained from 27 to 1000 μg L(-1) with limits of detection ranging from 9 to 12 μg L(-1) for the seven QNs. The absolute recoveries of the seven QNs at three different spiked levels (40, 150, and 400 μg L(-1) ) ranged from 74% to 98% with a relative standard deviation less than 10% in all cases. The proposed method was applied to analyze 20 whole milk samples of different brands. All samples were positive for the presence of QN residues; in some cases, extract dilution was required. The concentrations found are in the range from 31.1 to 5047.3 μg L(-1) . Marbofloxacin was the most frequently found. The method proposed offers advantages in terms of simplicity, sensitivity, efficiency, cost, and analysis time making it an alternative for the analysis of QNs in whole milk samples.  相似文献

A gas chromatographic method to determine thymol, eucalyptol (cineole), menthol and camphor residues in honey and beeswax is proposed. To isolate the compounds, three methods involving liquid-liquid extraction with methylene chloride, distillation, or solid-phase extraction on octadecylsilica cartridges can be used. The GC separation is carried out on a 60 m x 0.53 mm Stabilwax DA capillary column, using a flame ionization detector. The method is applied to the analysis of natural honey and also honey and beeswax samples from beehives treated with the above compounds.  相似文献

Flavonoids are an important bioactive group in the commonly used herbal medicine Flos Lonicerae. A new method of capillary zone electrophoresis (CZE) coupled with solid-phase extraction (SPE) was developed for simultaneous assay of flavonoid aglycones and glycosides in Flos Lonicerae. Optimum CZE separation was achieved with a background electrolyte (BGE) solution consisting of 80 mM boric acid and 20 mM phosphate acid, adjusted to pH 8.1, with 15% acetonitrile (v/v) added, and applying a separation voltage of 28 kV. The SPE method was used for pretreating the complex matrix of botanical materials and good reproducibility was obtained when avicularin was used as internal standard. Linearity of the method was excellent with correlation coefficients (r2) in the range of 0.9995-0.9999 and detection limits were lower than 0.6 microg/mL for the four flavonoids. The obtained recoveries varied between 93 to 104% while the relative standard deviations (RSDs) were below 4.4% (n=3). The developed CZE method was successfully used for the separation of eight flavonoids and the quantification of the four flavonoids in five species of Flos Lonicerae.  相似文献

A simple and robust solid-phase extraction (SPE) procedure for the cleanup and sample preconcentration of antifungals (ketoconazole, clotrimazole, itraconazole, fluconazole, and voriconazole) and their metabolites after incubation with human liver microsomes, as well as a simplified capillary zone electrophoresis (CZE) method for their rapid analysis, have been developed to determine the stability of these compounds in in vitro samples. Three different sample pretreatment procedures using SPE with reversed-phase sorbents (100 mg C8, 100 mg C18, and 30 mg Oasis-HLB) were studied. The highest and most reproducible recoveries were obtained using a 30 mg Oasis-HLB sorbent and methanol containing 2% acetic acid as eluent. Enrichment by a factor of about four times was achieved by reconstituting the final SPE eluates to a small volume. For the CZE separation, good separations without interfering peaks due to the in vitro matrix were obtained with a simple running electrolyte using a fused-silica capillary. The best separation for all components originated by each tested drug after incubation with human liver microsomes (unmetabolized parent drug and its metabolites) was obtained using a 0.05 M phosphate running buffer (pH 2.2) without additives. The effect of the injection volume was also investigated in order to obtain the best sensitivity. Performance levels in terms of precision, linearity, limits of detection, and robustness were determined.  相似文献

The separation of three selective serotonin reuptake inhibitors (SSRIs) by capillary electrophoresis (CE) with fully integrated solid-phase extraction (SPE) is described. Polymeric monolithic SPE modules were prepared in situ within a fused silica capillary from either butyl methacrylate-co-ethylene dimethacrylate or 3-sulfopropyl methacrylate-co-butyl methacrylate-co-ethylene dimethacrylate. Using a 1 cm SPE module placed at the inlet of the capillary, a mixture of sertraline, fluoxetine and fluvoxamine was extracted from aqueous solution by applying a simple pressure rinse. Under pressure-driven conditions, efficient elution was possible from both SPE materials investigated using 50 mM phosphate buffer, pH 3.5 in acetonitrile (20/80, v/v). Two different strategies were investigated for the efficient elution and subsequent CE separation. Injection of an aqueous sample plug directly into the non-aqueous elution/separation buffer was found to be unsuitable with poor elution profiles observed in the electrodriven mode. Alternatively, a sample plug equivalent to several capillary volumes could be injected by pressure followed by filling the capillary with the non-aqueous elution/separation buffer from the outlet end using a combination of pressure and electrodriven flow. Using a neutral monolith, efficient elution/separation was not possible due to an unstable electroosmotic flow (EOF), however, by adding the ionisable monomer, 3-sulfopropyl methacrylate to the SPE module to increase and stabilise the EOF, it was possible to achieve efficient elution from the SPE module, followed by baseline separation by CE using a 200 mM acetate buffer, pH 3.5 in acetonitrile (10/90, v/v). With enrichment factors of over 500 achieved for each of the analytes this demonstrates the potential of in-line SPE-CE for the sensitive analysis of these drugs.  相似文献
