A novel aldehyde dual-linker system has been developed for the solid phase synthesis of sterically hindered amides. The linker [5-(4-formyl-3-hydroxyphenoxy)pentanoic acid] exploits an intramolecular oxygen-nitrogen acyl transfer mechanism to prepare compounds that are unattainable with current commercially available linkers. A dual linker system, exploiting the hyper-acid labile Sieber amide linker as part of the construct, enabled the initial reductive alkylation reactions of hindered amines and their subsequent acylation with a range of carboxylic acids with varying stereoelectronic properties to be monitored. Simple acylation conditions (HBTU/HOBt/NMM) sufficed to provide near quantitative reaction of test acids with support-bound hindered amines, reaction conditions which failed when commercial linkers were used.  相似文献

In organic molecules carboxylic acid groups are among the most common functionalities. Activated derivatives of carboxylic acids have long served as versatile connection points in derivatizations and in the construction of carbon frameworks. In more recent years numerous catalytic transformations have been discovered which have made it possible for carboxylic acids to be used as building blocks without the need for additional activation steps. A large number of different product classes have become accessible from this single functionality along multifaceted reaction pathways. The frontispiece illustrates an important reason for this: In the catalytic cycles carbon monoxide gas can be released from acyl metal complexes, and gaseous carbon dioxide from carboxylate complexes, with different organometallic species being formed in each case. Thus, carboxylic acids can be used as synthetic equivalents of acyl, aryl, or alkyl halides, as well as organometallic reagents. This review provides an overview of interesting catalytic transformations of carboxylic acids and a number of derivatives accessible from them in situ. It serves to provide an invitation to complement, refine, and use these new methods in organic synthesis.  相似文献

A photocatalytic acyl lactonization of unsaturated carboxylic acids using simple and inexpensive acyl chlorides has been developed for a modular synthesis of acyl lactones. Significantly, the simple protocol could allow an efficient construction of biologically important phthalide framework. The transformation could be extended to a wide variety of unsaturated carboxylic acids, including substituted 2-vinyl benzoic acids and different types of alkenoic acids. Moreover, a series of aroyl, heteroaroyl chlorides could serve as coupling partners. Notably, functional groups including MeO, F, Cl and Br could survive. It is believed that acyl radicals generated from acyl chlorides under photoredox catalysis reacted with alkenes via atom-transfer radical addition (ATRA) and triggered subsequent lactonization in the process. Preliminary mechanistic investigations revealed that the transformation probably proceeded through a free radical pathway.  相似文献

The synthesis of 3-acyltetramic acids, the substructure of bioactive natural products, via O-acylation of tetramic acids with carboxylic acids followed by acyl migration, has been investigated. This acylation sequence is mediated by N,N'-dicyclohexylcarbodiimide (DCC) and 4-dimethylaminopyridine (DMAP) and is very sensitive to the nature of the nitrogen substituent (R(1)), the nature of the carboxylic acid (R(2)CO(2)H), and the amount of DMAP. Acylation of N-acyl tetramic acids with an alkyl carboxylic acid using 1.3 equiv of DMAP (with 1.1 equiv of DCC) unexpectedly gave the 3-acyltetramic acid directly as a result of acyl migration induced by excess amounts of DMAP. On the other hand, N-unsubstituted, N-alkyl, and N-acyl tetramic acids with alkyl and aromatic carboxylic acids gave the O-acyl tetramic acids by using only 0.1 equiv of DMAP (with 1.1 equiv of DCC); these could be further rearranged to the acyl product by treatment with excess DMAP. The tautomeric equilibrium of these 3-acyltetramic acids in solution was found to strongly depend on the nitrogen substituent group (R(1)) rather than the 3-acyl group.  相似文献

A new efficient and selective esterification reaction of carboxylic acids with chloroformates is described using silica-supported catalyst (PBGSiCl). Catalysts have been found for the selective reaction of aliphatic isocyanates with carboxylic acids giving amides after carbon dioxide extrusion. Magnesium and calcium salts lead to a dramatic increase in reaction rates while improving the selectivity when sterically hindered isocyanates and/or carboxylic acids are used.  相似文献

N,N-dimethylchlorosulfitemethaniminium chloride formed from thionyl chloride and dimethylformamide was found and efficient reagent for the synthesis of acyl azides from carboxylic acids and nitriles from oximes. The first general method for efficient electron transfer reduction of carboxylic acids has been developed. The protocol using SmI(2)-H(2)O-Et(3)N allows for reduction of a variety of carboxylic acids in excellent yields and provides an attractive alternative to processes mediated by reactive alkali metals, lithium aluminum hydride, and boron hydrides. Of broader significance, the method allows acyl radical equivalents to be generated from carboxylic acids under mild reaction conditions.  相似文献

In the ozone oxidation of thiolesters in 1,2-dichloroethane at room temperature, S-acylthiol S-oxides 3 are proposed as intermediates. The isolation of carboxylic acid anhydrides and dialkyl disulphides as the actual reaction products can be explained as resulting from the decomposition of the unstable. S-acyl S-acyloxysul-phonium salt 4 arising probably from the reaction of the intermediate S-acylthiol S-oxides with another mole of thiolester. However, in the case of hindered thiolesters, the S-acylthiol S-oxides formed rearrange, possibly via an S→ O acyl shift, to the corresponding acyl sulphenates 12 which on further oxidation lead to the mixed carboxylic acid sulphonic acid anhydrides 13.  相似文献

A chlorophosphite‐modified, Staudinger‐like acylation of azides involving a highly chemoselective, direct nucleophilic acyl substitution of carboxylic acids is described. The reaction provides the corresponding amides with analytical purity in 32–97 % yield after a simple aqueous workup without the need for a pre‐activation step. The use of chlorophosphites as dual carboxylic acid–azide activating agents enables the formation of acyl C? N bonds in the presence of a wide range of nucleophilic and electrophilic functional groups, including amines, alcohols, amides, aldehydes, and ketones. The coupling of carboxylic acids and azides for the formation of alkyl amides, sulfonyl amides, lactams, and dipeptides is described.  相似文献

羧酸选择加氢是合成醇的重要方法,廉价高效的催化体系仍然在探索中.我们利用地球上储量丰富的钴氧化物作为催化剂,通过控制催化反应过程,进而实现高选择性地催化羧酸加氢制备醇.一系列含有不同官能团的羧酸可以被选择加氢至相应的醇类化合物,反应选择性可以满足工业生产要求.通过一系列的谱学表征以及理论计算,我们证实了钴氧化物在羧酸选择加氢反应中的优选活性位点位为氧化亚钴,从而建立了催化剂与反应活性之间的构效关系,为催化剂的理性设计提供指导.首先,我们选取硬脂酸加氢反应作为模型反应,通过对地球上储量丰富的氧化镍、四氧化三铁和四氧化三钴的催化活性对比发现,四氧化三钴催化剂活性最高,在473 K,2 MPa氢气条件下,反应速率可以达到1.2 mmol/(h·g).对四氧化三钴催化剂进行不同温度的预还原处理,我们发现催化剂的活性得到显著提高,其中573 K还原的样品活性最高,反应速率可以达到7.3 mmol/(h·g),要远远高于贵金属催化剂Pd/C(0.6 mmol/(h·g))和Pt/C(1.8 mmol/(h·g)).XRD结果表明,随着还原处理温度的不断升高,催化剂由四氧化三钴变为氧化亚钴,最终变为金属态的钴.当还原温度为573 K时,催化剂的组成为单一相氧化亚钴.XPS测试结果表明,当还原温度为573 K时,样品中只含有Co~(2+)的信号峰,并且Co/O的比例为1/1,进一步证明样品是纯态的氧化亚钴.从TEM照片中可以发现,在原始的四氧化三钴样品中观察到晶面间距为0.467和0.244 nm,分别对应四氧化三钴的(111)和(311)晶面.而对于573 K还原的样品只观察到一种晶面间距(0.246 nm),对应氧化亚钴的(111)晶面.结合表征手段和硬脂酸催化加氢活性结果,我们得出氧化亚钴是573 K还原样品催化羧酸加氢反应的活性位点.理论计算结果进一步证实了这个实验结论.理论计算结果表明,在氧化亚钴(111)晶面,硬脂酸加氢转换为十八醇是非常快速和高效的,然而,对于氢解C-C键和C-O键,需要耗费更高的能量,能垒约为1.2 e V.因为硬脂酸的吸附远远强于十八醇的吸附,硬脂酸的存在会抑制十八醇氢解形成烯烃的反应,只有当硬脂酸酸完全转化为十八醇,才会发生随后的氢解反应.通过控制催化反应过程,可以实现在氧化亚钴(111)晶面高选择性催化酸加氢至醇,也就是反应控制催化过程.基于氧化亚钴在硬脂酸加氢制备十八醇上的优异催化性能,我们进一步研究了一系列含有不同官能团的羧酸化合物的催化加氢,发现氧化亚钴表现出良好的官能团容忍度,可以实现高效、广谱的酸选择加氢至醇反应.  相似文献

Carboxylic acids are converted to the corresponding 4-methoxybenzyl (PMB) esters with 4-methoxybenzyl-2,2,2-trichloroacetimidate in the absence of an acid catalyst. This operationally simple procedure is a highly effective method for the formation of PMB esters. The reaction is promoted by the carboxylic acids themselves in excellent yields (72–99%). Sterically hindered carboxylic acids, which provide lower yields with other imidates, are esterified in higher yield with the more reactive PMB imidate. No racemization is observed in the case of carboxylic acids bearing an α-stereocenter, and no isomerization is observed with Z-α,β-unsaturated acids. This method may therefore find use in the esterification of complex or sensitive substrates where more common techniques lead to decomposition.  相似文献

As a result of studying the interaction of hindered amine stabilizers (2, 2, 6, 6-tetramethylpiperidines) with simple hydroperoxides, peroxy radicals, and acylperoxy radicals, the last two in AIBN-initiated oxidation experiments in chlorobenzene, the following conclusions have been reached:
  • 1 Hindered amines have multiple mechanisms of functioning as photostabilizers of polymers.
  • 2 Reactions between tetramethylpiperidines and simple hydroperoxides are too slow at moderate temperatures to make a significant contribution to polymer stabilization.
  • 3 Reactions between tetramethylpiperidines and alkylperoxy radicals at moderate temperatures occur at varying rates with varying effectiveness for stabilization. With favorable alignment among reaction rates for oxidation propagation and termination, reactions between tetramethylpiperidines and alkylperoxy radicals can play a significant role in oxidation inhibition.
  • 4 Hydrocarbon polymer photooxidation proceeds by two major paths - the usually accepted alkyl radical/alkylperoxy radical/hydroperoxide route and the usually neglected aldehyde/acyl radical/acylperoxy radical/peracid route.
  • 5 Hindered amine stabilizers are able to participate in inhibiting both photooxidation reactions - they trap acylperoxy radicals, converting them to carboxylic acids and are converted to nitroxyl radicals in the process; the nitroxyl radicals trap alkyl radicals and the hindered amines trap alkylperoxy radicals to inhibit the other oxidation pathway.
  • 6 Nitroxyls are regenerated from N-alkyloxy hindered amines in a fast, efficient reactions with acylperoxy radicals and in slow reactions with alkylperoxy radicals. We postulate neither reaction yields peroxides: carboxylic acids and oxidized alkyloxy substituents are obtained from the first reaction; alcohols and oxidized alkyloxy substituents are obtained from the second reaction.

Acylphosphonates, which are easily available from carboxylic acids, are potent acyl anion precursors and undergo cyanide ion promoted phosphonate-phosphate rearrangement to provide the corresponding acyl anion equivalents as reactive intermediates. The protonation of these acyl anion equivalents furnished cyanohydrin O-phosphates in good yields. For the high yield formation of cyanohydrin O-phosphates from arylphosphonates THF should be used and from alkylphosphonates DME was used.  相似文献

A method for the simultaneous determination of non-esterified short-, medium- and long-chain fatty acids and other types of metabolically relevant carboxylic acids such as hydroxy, keto, aromatic and dicarboxylic acids in biological material by capillary gas chromatography of benzyl ester derivatives is described. Sample preparation avoiding incomplete isolation of carboxylic acids consisted of deproteinization and extraction with ethanol, fixation of carboxylic acids as carboxylates, removal of interfering compounds such as neutral lipids by hexane extraction and amino acids, acyl carnitines and other cations by cation-exchange chromatography, derivatization of keto groups of ketocarboxylic acids into O-methyl oximes and benzyl ester formation by reaction of the potassium carboxylates with benzyl bromide via crown ether catalysis. The sample preparation conditions were investigated, showing the usefulness of this method for quantitative determinations. Chromatograms obtained from human serum, human urine and rat heart ventricle and concentrations of carboxylic acids in these specimens are presented.  相似文献
