We have developed a system that couples an on-line microdialysis (MD) system with flow injection high-performance liquid chromatography (HPLC)-fluorescence detection for simultaneous measurement of the concentrations of malondialdehyde (MDA) and ofloxacin (OFL) in whole blood samples. The sample matrix was first cleaned with an MD system using an MD probe. A continuously flowing dialysate stream was derivatized on-line and auto-injected into a separation column. MDA and OFL were separated through a reverse-phase C18 column (250 mm × 4.6 mm) at a flow rate of 0.8 mL min−1 and then detected using a fluorescence detector (excitation: 532 nm; emission: 553 nm); the system's components were connected on-line using a valve control. Validation experiments demonstrated good linearity, precision, accuracy, and recovery. The precisions for the determinations of MDA and OFL, measured in terms of relative standard deviations, were 6.5% and 4.6%, respectively, for intra-day assays and 7.5% and 8.7%, respectively, for inter-day assays. The average recoveries of MDA and OFL spiked in plasma were each close to 100%. The use of this on-line MD-HPLC system permitted continuous monitoring of MDA and OFL in OFL-treated whole blood subjected to UV-A irradiation. Based on our results, the UV-A irradiation markedly increased the level of MDA in the OFL-treated whole blood.  相似文献

Diamond-like carbon (DLC) films are gaining big interest in electrochemistry research area. DLC electrodes made with different ratio of sp3/sp2 carbon hybridization or doped with different percentages of nickel were characterized electrochemically by cyclic voltammetry and by amperometric measurements towards hydrogen peroxide. SiCAr1 and SiCNi5% were chosen as sensitive transducers for the elaboration of amperometric glucose biosensors. Immobilization of glucose oxidase was carried out by cross-linking with glutaraldehyde. Measurements were made at a fixed potential + 1.0 V in 40 mM phosphate buffer pH 7.4. SiCAr1 seems to be more sensitive for glucose, 0.6875 μA/mM, than SiCNi5%, 0.3654 μA/mM. Detections limits were 20 μM and 30 μM, respectively. Apparent Michaelis-Menten constants were found around 3 mM. Forty-eight percent and 79% of the original response for 0.5 mM glucose remained after 10 days for both biosensors, respectively.  相似文献

Heroin metabolites including morphine, codeine, and 6-acetylmorphine were determined by cation-selective exhaustive injection and sweeping micellar electrokinetic chromatography (CSEI–sweep-MEKC). Liquid–liquid extraction was used for urine pretreatment. An uncoated fused silica capillary (Ld = 30 cm, 50 μm ID) was filled with phosphate buffer (50 mM, pH 2.5) containing 30% methanol, then high conductivity buffer (100 mM phosphate, 41.3 kPa for 18 s) was followed. Samples were injected electrokinetically (20 kV, 300 s). The sweeping and separation were performed at −25 kV using phosphate buffer (20 mM, pH 2.5) and 80 mM sodium dodecyl sulfate. The baseline separation was done within 10 min. During method validation, the calibration curves were linear over a range of 50–500 ng/mL (r ≧ 0.994). The RSD and RE values in intra-day and inter-day assays were all below 20%, which showed good precision and accuracy. Their detection limits were 10 ng/mL (S/N = 3). The optimized method was applied to determine real urine samples from addicts. These samples were confirmed by liquid chromatography/mass spectrometry.  相似文献

Two methods, based on the use of capillary electrophoresis (CE) and high-performance liquid chromatography (HPLC), respectively, were developed for the analysis of the atypical antipsychotic aripiprazole in plasma of schizophrenic patients for therapeutic drug monitoring purposes. Good analytical performances were obtained with the CE method, using uncoated fused silica capillaries and a background electrolyte composed of 50 mM phosphate buffer at pH 2.5. With 20 kV voltage, aripiprazole was detectable at 214 nm within 5 min. The second analytical method, based on HPLC with diode array detection, employed a C8 reversed-phase column and a mixture of a 12.5 mM phosphate buffer, pH 3.5, containing triethylamine and acetonitrile as the mobile phase. Aripiprazole was detected at 254 nm and a complete chromatographic run lasted about 10 min. For both analytical methods loxapine was used as the internal standard and the same plasma sample pre-treatment by means of solid-phase extraction on cyano cartridges was carried out, with extraction yield values always higher than 91.3%. Linear responses for aripiprazole were obtained between 70 and 700 ng mL−1 and precision assays (expressed as relative standard deviation values) were lower than 7.0%. After validation, both methods were successfully applied to human plasma samples drawn from schizophrenic patients undergoing therapy with Abilify® tablets. Accuracy was satisfactory, with recovery value higher than 91.0%.  相似文献

A rapid and simple method was demonstrated for the analysis of atropine, anisodamine, and scopolamine by nonaqueous capillary electrophoresis (NACE) coupled with electrochemiluminescence (ECL) and electrochemistry (EC) dual detection. The mixture of acetonitrile (ACN) and 2-propanol containing 1 M acetic acid (HAc), 20 mM sodium acetate (NaAc), and 2.5 mM tetrabutylammonium perchlorate (TBAP) was used as the electrophoretic buffer. Although a short capillary of 18 cm was used, the decoupler was not needed and the separation efficiency was good. The linear ranges of atropine, anisodamine, and scopolamine were 0.5–50, 5–2000, and 50–2000 μM, respectively. For six replicate measurements of 100 μM scopolamine, 15 μM atropine, and 200 μM anisodamine, the RSDs of ECL intensity, EC current, and migration time were less than 3.6%, 4.5%, and 0.3%, respectively. In addition, because the organic buffer was used, the working electrode (Pt) was not easily fouled and did not need reactivation. The method was also applied for the determination of these three alkaloids in Flos daturae extract.  相似文献

A simple, fast, sensitive and accurate reversed-phase ion-pair HPLC method for simultaneous determination of nitrite and nitrate in atmospheric liquids and lake waters has been developed. Separations were accomplished in less than 10 min using a reversed-phase C18 column (150 mm × 2.00 mm i.d., 5 μm particle size) with a mobile phase containing 83% 3.0 mM ion-interaction reagent tetrabutylammonium hydroxide (TBA-OH) and 2.0 mM sodium phosphate buffer at pH 3.9 and 17% acetonitrile (flow rate, 0.4 mL/min). UV light absorption responses at 205 nm were linear over a wide concentration range from 100 μg/mL to the detection limits of 10 μg/L for nitrite and 5 μg/L nitrate. Quantitation was carried out by the peak area method. The relative standard deviation for the analysis of nitrite and nitrate was less than 3.0%. This method was applied for the simultaneous determination of nitrite and nitrate in dew, rain, snow and lake water samples collected in southeast Massachusetts. Nitrate was found being present at 4.79-5.99 μg/mL in dew, 1.20-2.63 μg/mL in rain, 0.32-0.60 μg/mL in snow and 0.12-0.23 μg/mL in lake water. Nitrite was only a minor species in dew (0.62-0.83 μg/mL), rain (<0.005-0.14 μg/mL), snow (0.021-0.032 μg/mL) and lake water (0.12-0.16 μg/mL). High levels of nitrite and nitrate observed in dew water droplets may constitute an important source of hydroxyl radicals in the sunny early morning.  相似文献

In this work, the rapid detection of cholesterol using poly(dimethylsiloxane) microchip capillary electrophoresis, based on the coupling of enzymatic assays and electrochemical detection, was developed. Direct amperometric detection for poly(dimethylsiloxane) (PDMS) microchip capillary electrophoresis was successfully applied to quantify cholesterol levels. Factors influencing the performance of the method (such as the concentration and pH value of buffer electrolyte, concentration of cholesterol oxidase enzyme (ChOx), effect of solvent on the cholesterol solubility, and interferences) were carefully investigated and optimized. The migration time of hydrogen peroxide, product of the reaction, was less than 100 s when using 40 mM phosphate buffer at pH 7.0 as the running buffer, a concentration of 0.68 U/mL of the ChOx, a separation voltage of +1.6 kV, an injection time of 20 s, and a detection potential of +0.5 V. PDMS microchip capillary electrophoresis showed linearity between 38.7 μg/dL (1 μM) and 270.6 mg/dL (7 mM) for the cholesterol standard; the detection limit was determined as 38.7 ng/dL (1 nM). To demonstrate the potential of this assay, the proposed method was applied to quantify cholesterol in bovine serum. The percentages of recoveries were assessed over the range of 98.9-101.8%. The sample throughput was found to be 60 samples per hour. Therefore, PDMS microchip capillary electrophoresis, based on the coupling of enzymatic assays and electrochemical detection, is very rapid, accurate and sensitive method for the determination of cholesterol levels.  相似文献

This paper describes a robust and sensitive HPLC method for determination of glyphosate in water and plant material after pre-column derivatisation with 9-fluorenylmethyl chloroformate (FMOC-Cl) using single and coupled polymeric amino columns, respectively. New findings about optimisation of the derivatisation reaction and chromatographic behaviour of glyphosate and AMPA on polymeric amino column are also presented.The best HPLC conditions for the separation of glyphosate and AMPA in water samples were achieved using a single polymeric amino column and mobile phase at pH 10 which contained 55% (v/v) acetonitrile and 50 mM phosphate buffer. The method was validated by analysing 10 fortified rainwater samples at a level of 1 μg/l. The average recovery was 94% with relative standard deviation (R.S.D.) of 5.4% and the method detection limit of 0.16 μg/l.The HPLC separation of glyphosate (only) in plant material entailed the coupling of two polymeric amino columns via a switching valve to separate interfering products from the desired analyte. The method was validated using grass samples spiked at the level of 1 mg/kg and gave a method detection limit of 0.3 mg/kg for glyphosate and a recovery of 82.4% with R.S.D. of 10.3%.  相似文献

A bienzyme reactor sensor system with amperometric detection was developed for the determination of ornithine. The system based on the immobilized enzymes (ornithine carbamyl transferase and pyruvate oxidase) consisted of a buffer tank, a peristaltic pump, an enzyme reactor, an oxygen electrode and a recorder. Then, 0.1 M MOPS buffer, containing pyruvic acid (0.5 mM) and carbamyl phosphate (0.5 mM), was continuously transferred into the system at 35 °C. Phosphate ion was formed enzymatically by transformation of ornithine in the presence of carbamyl phosphate. Pyruvate oxidase is activated by the presence of phosphate. Therefore, ornithine was determined from the oxygen consumed upon oxidation of pyruvic acid catalyzed by pyruvate oxidase in the presence of phosphate ion. The limit of detection was 0.05 mM and the response was linear to 3 mM (R2=0.9905). The variation coefficients were 4.9 (n=15) and 3.9% (n=15) for 1.1 and 3.0 mM standard ornithine, respectively. Good comparative results (R2=0.9238) were observed between ornithine contents in prawn muscle determined by the proposed system and by the HPLC. One assay was completed within 4 min. The immobilized enzymes were stable for 2 months at 4 °C and more than 150 samples could be continually determined using this enzyme reactor.  相似文献

A simple sample preparation method was developed by using a centrifugal ultrafiltration (CF-UF) device with hollow fiber (HF) for the determination of cefaclor in plasma by HPLC. Samples were placed into a homemade device, which was consisted of a glass tube and a U-shaped hollow fiber. The filtrate was withdrawn from the hollow fiber into a syringe after centrifugation and 20 μL was directly injected into the HPLC for analysis. The HPLC method had a linear calibration curve in the concentration range of 6.00 × 10−2–30.7 μg mL−1(r = 0.9996). The limit of detection (LOD) and limit of quantitation (LOQ) were 0.02 and 0.06 μg mL−1, respectively. The intra and inter-day precisions (RSD) were 1.7%, 1.2%, 1.0% and 3.6%, 2.5%, 1.9%, respectively, for three concentrations. Assay accuracy was higher than 99.2% and the absolute recovery was 86.8–92.5%. It is feasible to use this novel and low cost device for sample pretreatment for the analysis of cefaclor in plasma.  相似文献

Microemulsion electrokinetic chromatography (MEEKC) was developed for quantitative analysis of avermectins, such as abamectin, doramectin and ivermectin, in commercial formulations, using the microemulsion buffer containing a 50 mM phosphate buffer at pH 2.5, 1.1% (v/v) n-octane as oil droplets, 180 mM sodium dodecylsulphate as surfactant, 890 mM 1-butanol as co-surfactant and 30% (v/v) ethanol as organic co-solvent. High accuracy and precision of the method were obtained. The contents of avermectins in commercial formulations determined by MEEKC were found to be insignificantly different with those determined by high performance liquid chromatography (HPLC). Therefore, MEEKC can be used an alternative method to HPLC for quantitative determination of avermectins.  相似文献

In this study, a new biosensor based on the inhibition of tyrosinase for the determination of fluoride is described. To construct the biosensor tyrosinase was immobilized by using gelatine and cross-linking agent glutaraldehyde on a Clark type dissolved oxygen (DO) probe covered with a teflon membrane which is sensitive for oxygen. The phosphate buffer (50 mM, pH 7.0) at 30 °C were established as providing the optimum working conditions. The method is based on the measurement of the decreasing of dissolved oxygen level of the interval surface that related to fluoride concentration added into reaction medium in the presence of catechol. Inhibitor effect of fluoride results in decrease in dissolved oxygen concentration. The biosensor response depends linearly on fluoride concentration between 1.0 and 20 μM with a response time of 3 min.In the characterization studies of the biosensor some parameters such as reproducibility, substrate specificity and storage stability were carried out. From the experiments, the average value (x), Standard deviation (S.D) and coefficient of variation (C.V %) were found as 10.5 μM, ± 0.57 μM, 5.43%, respectively for 10 μM fluoride standard.  相似文献

In this study, micellar electrokinetic chromatographic (MEKC) methods were developed for the detection of traces of melamine and its related by-products (ammeline, ammelide, and cyanuric acid). Two on-line sample concentration steps namely reversed electrode polarity stacking mode (REPSM) and cation-selective injection (CSI) were used for improving the detection sensitivity. For REPSM, a borate-NaOH buffer (pH 10, 35 mM) composed of 60 mM SDS and 10% (v/v) methanol, was used as carrier electrolyte, and samples were prepared in an aqueous solution of 10 mM NaOH. In CSI, a phosphate buffer (pH 2, 50 mM) containing 41 mM SDS was used as the carrier electrolyte, and samples were prepared with an aqueous solution of 10 mM NaOH and a phosphate buffer (pH 2.0, 25 mM) in a volume ratio of 1:9. The results indicated that REPSM enhanced all analyte signals except for melamine, which could be concentrated only by the CSI. The detection limit was reduced from 1.7 mg L−1 to 2.8 μg L−1 for melamine by the optimal CSI step, and from 0.23-1.2 mg L−1 to 2.4-5.0 μg L−1 for the other three analytes by the optimal REPSM step. Tableware made of melamine and samples of flour were used as test samples, and the results indicated that the proposed MEKC methods can successfully determine contaminations from melamine. The study also indicated that when the plastic made of melamine was exposed only once to an acidic solution (acetic or phosphoric acid) at 80 °C for 30 min, melamine continuously leached out from the test sample even without any further treatment with an acidic solution.  相似文献

Multi-walled carbon nanotube (CNT) was mixed with epoxy to fabricate microdisc electrode used as a detector for a specially designed miniaturized capillary electrophoresis (CE)-amperometric detection system for the separation and detection of several bioactive thiols. The end-channel CNT amperometric detector offers favourable signal-to-noise characteristics at a relatively low potential (0.8 V) for detecting thiol compounds. Factors influencing the separation and detection processes were examined and optimized. Four thiols (homocysteine, cysteine, glutathione, and N-acetylcysteine) have been separated within 130 s at a separation voltage of 2000 V using a 20 mM phosphate running buffer (pH 7.8). Highly linear response is obtained for homocysteine, cysteine, glutathione, and N-acetylcysteine over the range of 5-50 μM with detection limits of 0.75, 0.8, 2.9, and 3.3 μM, respectively. Good stability and reproducibility (R.S.D. < 5%) are obtained reflecting the minimal adsorption of thiols at the CNT electrode surface. The new microchip protocol should find a wide range of bioanalytical applications involving assays of thiol compounds.  相似文献

A new capillary electrophoresis (CE) method for the determination of quinolizidine alkaloids in Sophora medicinal plants was developed. A total of seven alkaloid components (cytisine, sophocarpine, matrine, lehmannine, sophoranol, oxymatrine and oxysophocarpine) were separated within 15 min. The running buffer was a 50 mM phosphate buffer containing 1%HP-β-CD and 3.3% isopropanol. The linear calibration ranges were 5.50-88.0 μg ml−1 for cytisine and lehmannine, 5.00-88.0 μg ml−1 for sophocarpine and sophoranol, 5.60-89.6 μg ml−1 for matrine and oxysophocarpine, and 24.0-384 μg ml−1 for oxymatrine. The recoveries of the seven alkaloids were 96.0-102.9% with relative standard deviations from 1.50 to 3.00% (n = 5). The method was successfully applied to different Sophora medicinal plants including Sophora flavescens, Sophora tonkinensis and Sophora alopecuroides.  相似文献

Summary A sensitive HPLC method has been developed for determination of ofloxacin (OFL) in biological fluids. Sample preparation was performed by adding phosphate buffer (pH 7.4, 0.1m) then extraction with trichloromethane. OFL and the internal standard, sarafloxacin (SAR), were separated on a reversed-phase column with aqueous phosphate solution-acetonitrile, 80∶20, as mobile phase. The fluorescence of the column effluent was monitored at λex 338 and λem 425 nm. The retention times were 2.66 and 4.24 min for OFL and SAR, respectively, and the detection and quantitation limits were 8 and 15 ng mL−1, respectively. Plots of response against ofloxacin concentration were linear in the range 8 to 2000 ng mL−1. Recovery was 92.9% for OFL.  相似文献

A novel high-performance liquid chromatographic (HPLC) method using pre-column derivatization and UV detection at 275 nm for the determination of clarithromycin in rat plasma has been validated. Clarithromycin was extracted from plasma sample spiked with internal standard (erythromycin) under alkaline condition with ethyl ether and derivatizated with trimethylbromosilane. The analyses were run on a C18 column, maintained at 40 °C during elution, using a mobile phase comprised of potassium dihydrogen phosphate (50 mM, pH 6.8, contained 0.7% triethylamine), acetonitrile, and methanol (30:45:25, v/v/v). The standard calibration curve for clarithromycin was linear (r2 = 0.9998) over the concentration range of 0.1-10 μg ml−1 in rat plasma. The limit of detection (LOD) and limit of quantitation (LOQ) was 30 ng ml−1 and 0.1 μg ml−1 respectively. The intra- and inter-day assay variability range was 2.6-7.4% and 3.3-8.5%, respectively. This method has been successfully applied to a pharmacokinetic study of clarithromycin in rats.  相似文献

Fluorescence and terbium-sensitised luminescence properties of new quinolone garenoxacin have been studied. The fluorimetric method allows the determination of 0.060-0.600 μg ml−1 of garenoxacin in aqueous solution containing HCl/KCl buffer (pH 1.5) with λexc=282 nm and λem=421 nm. Micellar-enhanced fluorescence was also studied, leading to a higher than 400% increase in analytical signal in presence of 12 mM sodium dodecyl sulphate (SDS), allowing the determination of 0.020-0.750 μg ml−1 of garenoxacin. The terbium-sensitised luminescence method allows the determination of 0.100-1.500 μg ml−1 of garenoxacin in 12 mM SDS solution containing 0.08 M acetic acid/sodium acetate buffer (pH 4.1) and 7.5 mM Na2SO3 (chemical deoxygenation agent), with λexc=281 nm and λem=546 nm. Relative standard deviation (R.S.D.) values for the three methods were in the range 1.0-2.0%. The proposed procedures have been applied to the determination of garenoxacin in spiked human urine and serum.  相似文献

A wide size range of SiO2 particles were synthesized and were used as enzyme immobilization carriers to fabricate glucose biosensors. The size of the particles was in the range of 17-520 nm. These biosensors could be operated under physiological conditions (0.1 M phosphate buffer, pH 7.2). Particle size could affect the performance of SiO2 modified glucose biosensors drastically. The smaller particles had higher performance. The smallest SiO2 modified biosensor could work well in the glucose concentration range of 0.02-10 mM with a correlation coefficient of 0.9993. Its sensitivity was 2.08 μA/mM and the detection limit was 1.5 μM glucose.  相似文献

A quantitative method of capillary electrophoresis with sample stacking induced by moving reaction boundary (MRB) was developed for sensitive determination of oxymatrine (OMT) and matrine (MT) in rat plasma. The experimental conditions were optimized firstly. Below are the optimized experimental conditions: 20 mM sodium formate solution (HCOONa, adjusted to pH 10.70 by ammonia) as sample solution, 3 min 14 mbar sample injection, 40 mM formic buffer (HCOOH-HCOONa, pH 2.60) as stacking buffer, 7 min 14 mbar injection of stacking buffer, 100 mM HCOOH-HCOONa (pH 4.80) as separation buffer, 73 cm capillary (effective length 64 cm), 21 kV voltage, 210 nm wavelength. Under the optimized conditions, higher than 60-fold sensitivity improvement of the stacking was simply achieved as compared with capillary zone electrophoresis, and the detectable limits obtained for OMT and MT were 0.26 and 0.19 μg mL−1, respectively. Then, numerous demonstrations were carefully performed for the methodological validations of OMT and MT in rate plasma, including high specificity of method, good linearity (r = 0.9993 for OMT, r = 0.9991 for MT), fair wide linear concentration range (1.30-65.00 μg mL−1 for OMT, 0.84-42.00 μg mL−1 for MT), low limit of detection (1.03 μg mL−1 for OMT, 0.38 μg mL−1 for MT), less than 5% intra- and inter-day variance value, and higher than 96% recovery of OMT and MT in plasma. The developed method could be used for the trace analyses of OMT and MT in plasma and was finally used for the investigation on pharmacokinetic study of OMT in rat plasma.  相似文献
