The effect of different alternative techniques to the traditional aging on lees on the low molecular weight phenolic compounds of red wines was study as well as their evolution during the aging in oak wood barrels for six months. The study was carried out with Tempranillo red grapes from two consecutive vintages. The techniques assayed were the traditional aging on lees with or without the addition of exogenous β-glucanase enzymes, the use of yeast derivative preparations also with or without the addition of exogenous β-glucanase enzymes, the micro-oxygenation applied together with the aging on lees, and the use of non-toasted oak wood chips. 相似文献

Polyphenolic composition of two Syrah wines aged during 6 or 12 months in medium toasting acacia and oak 225 L barrels was studied by LC–DAD-ESI/MS. A total of 43 nonanthocyanic phenolic compounds were found in all wines, and other 15 compounds only in the wines from acacia barrels. Thus, the nonanthocyanic phenolic profile could be a useful tool to identify the wines aged in acacia barrels. Among all of them the dihydrorobinetin highlights because of its high levels, but also robinetin, 2,4-dihydroxybenzaldehyde, a tetrahydroxydihydroflavonol, fustin, butin, a trihydroxymethoxydihydroflavonol and 2,4-dihydroxybenzoic acid were detected at appreciable levels in wines during aging in acacia barrels, and could be used as phenolic markers for authenticity purposes. Although longer contact time with acacia wood mean higher concentrations of phenolic markers found in wines, the identification of these wines will also be easy after short aging times due the high levels reached by these compounds, even after only 2 months of aging. 相似文献

The evolution of low molecular weight polyphenols in a red wine aging in different systems (barrels, oak chips and oak staves) made of different oak species was studied. All were placed under the same aging conditions in a wine cellar participating the Cigales Appellation of Origin programme (Spain). The wine aged in contact with oak chips experienced a quicker aging and loss of certain compounds, and a greater number of polymerisations than the wine aged in barrels. The discriminant analysis showed that samples of the same source wine aged in barrels, oak chips and oak staves tended to group together according to the aging system when wood from different botanical species is used (American, French or Hungarian). 相似文献

The aim of this paper is to evaluate the ability of the mesoporous silica SBA-15 to adsorb polyphenols from red wine. The mesoporous molecular sieve silica SBA-15 was hydrothermally synthesized in acidic media and characterized by SAXRD, BET, EDX and SEM. The adsorption behavior of mesoporous silica SBA-15 was investigated at 5 °C for 24 h using an adsorbent dose of 8 g SBA-15 L−1 red wine. The total polyphenols content expressed as mg of gallic acid equivalents (GAE L−1) was estimated from the standard curve of gallic acid (absorbance at 280 nm). HPLC chromatograms of methanolic extract from mesoporous SBA-15 at 256, 280, 324, and 365 nm exhibits the strong retention of quercetin and cis-resveratrol and a reasonable retention of trans-resveratrol, catechin, epicatechin, rutin, and phenolic acids (meta- and para-hydroxybenzoic, vanillic, caffeic, syringic, salicylic and para-coumaric acids). 相似文献

Near infrared (NIR) spectroscopy was used to simultaneously predict the concentrations of malvidin-3-glucoside (M3G), pigmented polymers (PP) and tannins (T) in red wine. A total of 495 samples from 32 commercial scale red wine fermentations over two vintages using two grape varieties (Cabernet Sauvignon and Shiraz), and also including as additional variables two types of fermenters, two different yeasts, and three fermentation temperatures were used. Samples were scanned in transmission mode (400-2500 nm) using a monochromator instrument (NIRSystems6500). Calibration equations were developed from high performance liquid chromatography (HPLC) and NIR data using partial least squares (PLS) regression with internal cross validation. Using PLS regression, very good calibration statistics (Rcal2>0.80) were obtained for the prediction of M3G, PP and T with standard deviation (S.D.)/standard error in cross validation (SECV) ratio (residual predictive deviation, RPD)) ranging from 1.8 to 5.8. It was concluded that near infrared spectroscopy could be used as rapid alternative method for the prediction of the concentration of phenolic compounds in red wine fermentations. 相似文献

A continuous method, easy to automate, for the determination of anthocyanins in wine based on the coupling of continuous liquid–solid extraction, evaporation, HPLC individual separation and photometric detection is proposed. The target analytes are removed from the wine in a continuous fashion using a C18 minicolumn and eluted with an aqueous solution (pH 2) with 16% acetonitrile. The eluted fraction is concentrated by solvent evaporation assisted by heat and dragging off the vapour using a flow of N2. For in-line preconcentration, a continuous evaporation module was designed and located in the manifold between the solid-phase minicolumn and the injection valve of the chromatograph. In this way, injection of the sample into the dynamic system leads the plug through it for liquid–solid extraction of the anthocyanins, partial evaporation of the eluent (with a preconcentration factor as required) and transport to the high-pressure injection valve of the chromatograph, where individual separation and subsequent photometric detection take place. The method thus developed for the determination of malvidin-3-glucoside, cyanidin-3-glucoside and peonidin-3-glucoside anthocyanins in Spanish red wines is more sensitive than the batch manual method based on the same steps, has better linearity of the calibrations curves with lower detection limits and much wider determination range for the most abundant anthocyanins in wine. Solid-phase extraction cartridges (SPE)-GC/MS method was used to analyse red wines aromas. The matrix effect was studied with chemicals standard prepared in synthetic wines with water/alcohol solutions (12% ethanol, v/v) following the procedure proposed. The method offers good reproducibility since the relative standard deviations (RSD%) for the volatile compounds levels were less than 9%. This method was used to differentiate the aroma of one hundred mono-varietal young, crianza, reserva and gran reserva La Mancha D.O. wines (cv. Tempranillo) on the basis of oak barrel contact period. Samples were checked at ten time points over 36 months. Sixty important wine odorants, such as volatile phenols, vanillin derivatives, lactones, norisoprenoids, benzene compounds, esters and terpenols, can be quantitatively determined in a single run. Results showed significant quantitative differences in the volatile profiles of wines depending on the length of time in contact with wood, even in wines belonging to the same commercial category. Stepwise multiple linear regression (SMLR) was used to obtain a model that predicted the time of barrel ageing to which wines were submitted in relation with the wine volatile composition. A successful function based on eight compounds with a mean deviation of 0.37 months in the predictions, was obtained. 相似文献

Nowadays, micro-oxygenation is a very important technique used in aging wines in order to improve their characteristics. The techniques of wine tank aging imply the use of small doses of oxygen and the addition of wood pieces of oak to the wine. Considering the low dissolved oxygen (DO) levels used by micro-oxygenation technique it is necessary to choose the appropriate measurement principle to apply the precise oxygen dosage in wine at any time, in order to assure its correct assimilation. This knowledge will allow the oenologist to control and run the wine aging correctly.This work is a thorough revision of DO measurement main technologies applied to oenology. It describes the strengths and weaknesses of each of them, and draws a comparison of their workings in wine measurement. Both, the traditional systems by electrochemical probes, and the newest photoluminescence-based probes have been used. These probes adapted to red wines ageing study are then compared.This paper also details the first results of the dissolved oxygen content evolution in red wines during a traditional and alternative tank aging. Samples have been treated by three different ageing systems: oak barrels, stainless-steel tanks with small oak wood pieces (chips) and with bigger oak pieces (staves) with low micro-oxygenation levels. Moringa oleifera leaves are cultivated in warm regions of Brazil for commercial ends due to their nutritional and biological properties. This study evaluated, by chemometric tools, the influence of seasonality (winter, spring, autumn, and summer), growing area in Brazil (South, Southeast, and Northeast), and regrowth age (40 and 80 days) in antioxidant potential, and biochemical markers determined by HPLC-DAD. The results obtained in the present study showed that all the parameters evaluated were affected by seasonality, regrowth age, and growing region. The content of phenolic compounds and antioxidant activity using the ABTS method was higher in samples cultivated in the southeast (59.4 mg GAE g?1 and 346 µmol TEAC g?1, respectively). The highest antioxidant activities evaluated by DPPH and FRAP methods were obtained from leaves of the southeast and northeast. In addition, young leaves presented higher antioxidant potential and total phenolic content. The cultivation region significantly influenced the content of chlorogenic acid, isoquercitrin, and astragalin, which ranged from 4.2 to 7.2 mg g?1, 8.0 to 10.7 mg g?1, and 2.2 to 3.8 mg g?1, respectively. In addition, a positive correlation between solar radiation and temperature with caffeic acid, rutin, phenolic compounds, and antioxidant assay from the DPPH method was observed. Additionally, the RGB pattern of the images of these leaves was correlated with the levels of compounds with antioxidant activity. Models generated through machine learning showed good performances, and ABTS and rutin analyses developed the best models with a coefficient of determination above 75 %. Thus, color patterns can be used to measure the antioxidant activity by the ABTS method and to determine the rutin concentration in M. oleifera leaves. 相似文献

Dendrobium is a large genus of orchid plants containing several bioactive polyphenols, including bibenzyls, phenanthrenes, and flavanones that have been used as antioxidants in nutraceutical and pharmaceutical products. Several polyphenols including (2S)-eriodictyol, (2S)-homoeriodictyol, moscatilin, gigantol, chrysotoxine and crepidatin are primarily found in Dendrobium species and can serve as bioactive markers of orchid plants. In the present study, a simple UPLC-UV method for the simultaneous determination of six polyphenols was developed for evaluating the distribution of bioactive phytochemicals in orchid plants and validated according to the ICH Q2 (R1) guidance. The sample was prepared by extracting dendrobium powder with methanol and the supernatant was analyzed using the Waters ACQUITY UPLCTM H-Class system on an ACQUITY UPLCTM BEH C18 column (2.1 × 50 mm, 1.7 μm) with gradient elution of water and acetonitrile, containing 1% v/v trifluoroacetic acid (TFA) each, at a flow rate of 0.2 mL/min and UV detection at 280 nm. The chromatographic condition provided good peak shape and resolution. The method was linear over the specific ranges with the coefficient of determination (r2) > 0.995. Accuracy of the method expressed as %recovery ranged from 80 to 110%. The precision of the method demonstrated as %CV was < 7.3%. The method is simple, accurate, precise and robust and is recommended for routine quality control analysis of orchid plants containing the six polyphenols as the main principles in the herb. The proposed method was successfully applied for simultaneous quantification of the 6 polyphenolic compounds in 11 samples from 3 Dendrobium species, suggesting that the method was suitable for quality assessment of orchid herbal raw materials. 相似文献

Organic acids, sugars, phenolic compositions and antioxidant capacities of orange juice and orange wine obtained from the cv. Kozan of Turkey were determined. High-performance liquid chromatographic methods were used to identify and quantify of these compounds. Three organic acids (citric, malic and ascorbic acids) and three sugars (sucrose, glucose and fructose) were determined. The major organic acid was found as citric acid. With regard to sugars, sucrose was present in the largest amounts for orange juice and wine. A total of 13 phenolic compounds were identified and quantified in orange juice and wine, including hydroxybenzoic acids (2), hydroxycinnamic acids (5), and flavanones (6). Hesperidin, narirutin and ferulic acid were the most abundant phenolic compounds in orange juice and wine. Antioxidant activities of orange juice and wine were measured using the DPPH• (2,2-diphenyl-1-picrylhydrazyl) assay, and the antioxidant capacity of orange juice was found to be higher than that of orange wine. 相似文献

A simple and rapid HPLC method with UV and fluorescence detection (FLD) for the separation of ten phenolic compounds including gallic acid, catechin, epicatechin, caffeic acid, coumaric, trans-piceid, cis-piceid, trans-resveratrol, cis-resveratrol and quercetin is reported. The UV and fluorescence detector in series provided a high selectivity for the determination of these compounds. Precisions, recoveries and LODs achieved for all the analytes were satisfactory. Beverages, especially wines are well-known to contain a variety of health-beneficial bioactive substances, mainly of phenolic nature which frequently exhibit antioxidant activity. Significant information is available about the separation and identification of polyphenols from some beverages by chromatographic and spectroscopic techniques, but considerably poor is chemical data related to the polyphenolic content in rums. In this paper, a method involving the all-liquid chromatographic technique of high-speed countercurrent chromatography (HSCCC) combined with high-performance liquid chromatography coupled with diode-array detection and electrospray ionization mass spectrometry (HPLC-DAD-ESI-MS(n)) has been successfully applied for separation and identification of phenolic compounds in an aged rum. Besides, the phenolic fraction (PF) was assayed for its antioxidant effects using three different free radical in vitro assays (DPPH·, RO(2)· and spontaneous lipid peroxidation (LPO) on brain homogenates) and on ferric reducing antioxidant power (FRAP). Results showed that PF potently scavenged DPPH and strongly scavenged peroxyl radicals compared to ascorbic acid and butylated hydroxytoluene (BHT); and almost equally inhibited LPO on brain homogenates subjected to spontaneous LPO when compared to quercetin. Moreover, PF also exhibited strong reducing power. This chemical analysis illustrates the rich array of phenols in the aged rum and represents a rapid and suitable method for the isolation and identification of phenolic compounds from mixtures of considerable complexity, achieving high purity and reproducibility with the use of two separation steps. 相似文献

The phenolic potential of Tannat, Cabernet-Sauvignon and Merlot grapes was evaluated in 2001 and 2002 and its correspondence with the colour and composition of the respective wines was established. Three vineyards of each variety, situated in the south of Uruguay were considered. Two samples of each vineyard were taken at the moment of the harvest. Phenolic richness, extractable anthocyanins contents and total potential in anthocyanins of the grapes were estimated. Two fermentations on skins were carried out for each vineyard using 50 kg of grape in each one. The anthocyanic and total polyphenols contents of the musts were analysed every 24 h, and skins extractions were carried out in parallel in the laboratory. The duration of the maceration for each variety was decided in function of the analytical results in the grapes, musts and skins extractions. Wines were analysed 2 months after the alcoholic fermentation, determining its phenolic composition and colour. Tannat grapes presented anthocyanic and total polyphenols contents significantly higher in both years. Therefore, wines from this variety presented colour intensity and phenolic contents statistically higher than Cabernet-Sauvignon and Merlot. The correlations between the phenolic contents of the grapes, skins, musts and wines were very significant. Colour intensity and phenolic contents of the wines were highly correlated with the total polyphenols of the grapes and with anthocyanins of the grapes, skins, musts and wines. The estimate of the phenolic potential of the grapes and the extractability of the pigments allows to manage more adequately the fermentation on skins and is an interesting tool to predict the colour and the composition of the wines. 相似文献

Hibiscus manihot L. is a kind of healthy plant with edible value and health benefits, which possesses multiple pharmacological activities that are closely related to antioxidant and anti-inflammatory activities. The dynamic changes of main active components and biological activities in Hibiscus manihot L. flower (HMLF) during its flowering period were systematically studied to determine the appropriate harvest time. Chemopreventive efficacies of the investigated HMLF extracts, by means of their anti-inflammatory and antioxidant activities, were assessed. The sample harvested on early August had the supreme total flavonoid content, total phenolic content and the strongest antioxidant activity (DPPH radical scavenging activity (IC50 0.160 mg/mL), ABTS radical scavenging activity (1.570 mmol/g Trolox), reducing power (IC50 0.101 mg/mL) and FRAP (3.644 mmol FeSO4/g)). The results of principal component analysis indicated that the primary active components included hyperin, isoquercetin, hibifolin and quercetin-3′-O-glucoside, which were strongly associated with the antioxidant activity in the early August sample, while neochlorogenic acid, chlorogenic acid and caffeic acid were associated with the anti-inflammatory activity. The extracts significantly inhibited lipopolysaccharide-induced nitric oxide production in RAW264.7 cells, especially the samples harvested around August, which was only 3.569 μΜ with the inhibition ratio of>50%. This study indicated that HMLF harvested on the early August possessed the highest antioxidant and anti-inflammatory potential and could be used as high bioactive resources for healthy production. 相似文献

The insect Blaps japanensis have been used as an ethnomedicine in China for the treatment of several disorders such as cancer and inflammation. Our investigation with this insect led to the isolation of eight new and two known phenolic compounds. The structures of the new compounds, blapsins C?J (1–8), were identified by spectroscopic data. The inhibitory activities of all the compounds except of 9 against human cancer cells (A549, K562 and Huh-7), COX-2, ROCK1, and JAK3 were evaluated. High-performance liquid chromatography with diode array and electrospray ionization mass spectrometric detection was used to carry out the comprehensive characterization of a lemon verbena extract with demonstrated antioxidant and antiinflammatory activity. Two different MS techniques have been coupled to HPLC: on one hand, time-of-flight mass spectrometry, and on the other hand, tandem mass spectrometry on an ion-trap. The use of a small particle size C18 column (1.8 μm) provided a great resolution and made possible the separation of several isomers. The UV–visible spectrophotometry was used to delimit the class of phenolic compound and the accurate mass measurements on time-of-flight spectrometer enabled to identify the compounds present in the extract. Finally, the fragmentation pattern obtained in MS–MS experiments confirmed the proposed structures. This procedure was able to determine many well-known phenolic compounds present in lemon verbena such as verbascoside and its derivatives, diglucuronide derivatives of apigenin and luteolin, and eukovoside. Also gardoside, verbasoside, cistanoside F, theveside, campneoside I, chrysoeriol-7-diglucuronide, forsythoside A and acacetin-7-diglucuronide were found for the first time in lemon verbena. 相似文献
