Raman spectroscopy was applied for the direct non-destructive analysis of amiodarone hydrochloride (ADH), the active ingredient of the liquid formulation Angoron®. The FT-Raman spectra were obtained through the un-broken as-received ampoules of Angoron®. Using the most intense vibration of the active pharmaceutical ingredient (API) at 1568 cm−1, a calibration model, based on solutions with known concentrations, was developed. The model was applied to the Raman spectra recorded from three as-purchased commercial formulations of Angoron® having nominal strength of 50 mg ml−1 ADH. The average value of the API in these samples was found to be 48.56 ± 0.64 mg ml−1 while the detection limit of the proposed technique was found to be 2.11 mg ml−1. The results were compared to those obtained from the application of HPLC using the methodology described in the European Pharmacopoeia and found to be in excellent agreement. The proposed analytical methodology was also validated by evaluating the linearity of the calibration line as well as its accuracy and precision. The main advantage of Raman spectroscopy over HPLC method during routine analysis is that it is considerably faster and no solvent consuming. Furthermore, Raman spectroscopy is non-destructive for the sample. However, the detection limit for Raman spectroscopy is much higher than the corresponding for the HPLC methodology.  相似文献

A novel sol-gel 3-mercaptopropyltrimethoxysilane (MPTS) modified silica coating was developed for capillary microextraction (CME) of trace Cu, Hg and Pb prior to their on line determination by inductively coupled plasma-atomic emission spectrometry (ICP-AES). This organic-inorganic hybrid coating was in situ created on the inner walls of fused silica capillary using a sol solution containing TMOS (tetramethoxysilane) as a precursor, MPTS as a co-precursor, ethanol as the solvent and hydrochloric acid as a catalyst. The structure of the capillary coating was characterized by FT-IR spectroscopy, Raman spectroscopy, SEM and TEM. The factors affecting on the capillary microextraction of analytes such as pH, sample flow rate and volume, elution solution and interfering ions had been investigated, and the optimized experimental parameters were obtained. Under the optimized conditions, the absorption capacity of MPTS-silica coated capillary was found to be 1.17, 1.96 and 1.19 μg m−1 for Cu, Hg and Pb, and the limits of detection were as low as 0.17 0.22 and 0.52 ng mL−1, respectively. With a sampling frequency of 12 h−1, the relative standard deviations (R.S.D.s) were 4.2, 2.6 and 3.8% (C=4 ng mL−1, n = 7, sample volume = 1 mL) for Cu, Hg and Pb, respectively. The proposed method had been successfully applied to the determination of Cu, Hg and Pb in human urine, human serum and preserved egg. To validate the proposed method, certified reference materials of BCR151 milk powder, GBW07601 (GSH-1) human hair, GSBZ 50016-90 and GSB 07-1183-2000 water samples were analyzed and the determined values were in a good agreement with the certified values.  相似文献

A novel amperometric uric acid (UA) sensor has been developed by coating the surface of a gold electrode with a polystyrene (PS) membrane formed by 30 μL of a 30 mg mL−1 PS chloroform solution combined with 30 μL of a 5 mg mL−1 polymaleimidostyrene (PMS) solution as a dispersant for enzyme, uricase; this membrane has been successfully employed as an immobilization support for uricase. In the PS membrane, PMS forms micelle-like structures containing uricase in an active state. This immobilized uricase membrane permits the permeation of oxygen, which is consumed by the uricase reaction. A good linear relationship is obtained over the concentration range of 5-105 μM. The concentration of uric acid was determined at a negative potential based on the decrease in the reduction current of oxygen and the interference of l-ascorbic acid can be completely eliminated.  相似文献

A novel simple, fast and efficient ultra-high performance supercritical fluid chromatography (UHPSFC) method was developed and validated for the separation and quantitative determination of eleven illegal dyes in chili-containing spices. The method involved a simple ultrasound-assisted liquid extraction of illegal compounds with tetrahydrofuran. The separation was performed using a supercritical fluid chromatography system and CSH Fluoro-Phenyl stationary phase at 70 °C. The mobile phase was carbon dioxide and the mixture of methanol:acetonitrile (1:1, v/v) with 2.5% formic acid as an additive at the flow rate 2.0 mL min−1. The UV–vis detection was accomplished at 500 nm for seven compounds and at 420 nm for Sudan Orange G, Butter Yellow, Fast Garnet GBC and Methyl Red due to their maximum of absorbance. All eleven compounds were separated in less than 5 min. The method was successfully validated and applied using three commercial samples of chili-containing spices – Chili sauce (Indonesia), Feferony sauce (Slovakia) and Mojo sauce (Spain). The linearity range of proposed method was 0.50–9.09 mg kg−1 (r ≥ 0.995). The detection limits were determined as signal to noise ratio of 3 and were ranged from 0.15 mg kg−1 to 0.60 mg kg−1 (1.80 mg kg−1 for Fast Garnet) for standard solution and from 0.25 mg kg−1 to 1.00 mg kg−1 (2.50 mg kg−1 for Fast Garnet, 1.50 mg kg−1 for Sudan Red 7B) for chili-containing samples. The recovery values were in the range of 73.5–107.2% and relative standard deviation ranging from 0.1% to 8.2% for within-day precision and from 0.5% to 8.8% for between-day precision. The method showed potential for being used to monitor forbidden dyes in food constituents. The developed UHPSFC method was compared to the UHPLC-UV method. The orthogonality of Sudan dyes separation by these two methods was demonstrated. Benefits and drawbacks were discussed showing the reliability of both methods for monitoring of studied illegal dyes in real food constituents.  相似文献

An imidazole derivative, 2-(2′-pyridyl)imidazole (PIMH), was developed as a colorimetric probe for the qualitative analysis of Fe2+ in aqueous solution. PIMH was then used to post-functionalize poly(vinylbenzyl chloride) (PVBC) nanofibers after electrospinning so as to afford a solid state colorimetric probe. Upon treatment with Fe2+ the probe displayed a distinctive color change both in liquid and solid platforms. The linear dynamic range for the colorimetric determination of Fe2+ was 0.0988–3.5 μg mL−1. The ligand showed a high chromogenic selectivity for Fe2+ over other cations with a detection limit of 0.102 μg mL−1 in solution (lower than the WHO drinking water guideline limit of 2 mg L−1), and 2 μg mL−1 in the solid state. The concentration of Fe2+ in a certified reference material (Iron, Ferrous, 1072) was found to be 2.39 ± 0.01 mg L−1, which was comparable with the certified value of 2.44 ± 0.12 mg L−1. Application of the probe to real samples spiked with Fe2+ achieved recoveries of over 97% confirming accuracy of the method and its potential for on-site monitoring.  相似文献

In this paper, the conversion of azoxystrobin in a strongly fluorescent degradation product by UV irradiation with quantitative purposes and its fluorimetric determination are reported for the first time. A multicommuted flow injection-solid phase spectroscopy (FI-SPS) system combined with photochemically-induced fluorescence (PIF) is developed for the determination of azoxystrobin in grapes, must and wine. Grape samples were homogenized and extracted with methanol and further cleaned-up by solid-phase extraction on C18 silica gel. Wine samples were solid-phase extracted on C18 sorbent using dichloromethane as eluent. Recoveries of azoxystrobin from spiked grapes (0.5-2.0 mg Kg−1), must (0.5-2.0 μg mL−1) and wine (0.5-2.0 μg mL−1) were 84.0-87.6%, 95.5-105.9% and 88.5-111.2%, respectively. The quantification limit for grapes was 0.021 mg Kg−1, being within European Union regulations, and 18 μg L−1 and 8 μg L−1 for must and wine, respectively.  相似文献

A sensitive and selective column adsorption method is proposed for the preconcentration and determination of atrazine and simazine. Atrazine and simazine were preconcentrated on heat-treated diatomaceous earth as an adsorbent and then determined by high-performance liquid chromatography (HPLC). Several parameters on the recoveries of the analytes were investigated. The experimental results showed that it was possible to obtain quantitative analysis when the solution pH was 2 using 100 mL of validation solution containing 1.5 μg of triazines and 5 mL of ethanol as an eluent. Recoveries of atrazine and simazine were 95.7 ± 4.2% and 75.0 ± 1.9% with a relative standard deviation for seven determinations of 4.7% and 2.7% under optimum conditions. The maximum preconcentration factor was 100 for triazines when 500 mL of sample solution volume was used. The linear ranges of calibration curves for atrazine and simazine were 1-150 ng mL−1 and 1-300 ng mL−1, respectively, with correlation coefficients of 0.999 and the detection limits (3Signal-to-Noise) were 0.24 ng mL−1 and 0.21 ng mL−1 for atrazine and simazine. The capacity of the adsorbent was also examined and found to be 0.8 mg g−1 and 1.3 mg g−1 for atrazine and simazine, respectively. The proposed method was successfully applied to the determination of triazines in river water and tap water samples with high precision and accuracy.  相似文献

The paper proposes a simple and portable approach for the surface enhanced Raman scattering (SERS) spectroscopy in situ determination of carboxylated single walled carbon nanotubes (SWNTs) in river water samples. The method is based on the subsequent microfiltration of a bare gold nanoparticles solution and the water sample containing soluble carbon nanotubes by using a home-made filtration device with a small filtration diameter. An acetate cellulose membrane with a pore size of 0.2 μm first traps gold nanoparticles to form the SERS-active substrate and then concentrates the carbon nanotubes. The measured SERS intensity data were closely fit with a Langmuir isotherm. A portable Raman spectrometer was employed to measure SERS spectra, which enables in situ determination of SWNTs in river waters. The limit of detection was 10 μg L−1. The precision, for a 10 mg L−1 concentration of carbon nanotubes, is 1.19% intra-membrane and 10.5% inter-membrane.  相似文献

Matrix-assisted refolding is an excellent technique for performing refolding of recombinant proteins at high concentration because aggregation during refolding is partially suppressed. The autoprotease Npro and its engineered mutant EDDIE can be efficiently refolded on cation-exchangers. In the current work, denatured fusion proteins were loaded at different column saturations (5 and 50 mg mL−1 gel), and refolding and self-cleavage were initiated during elution. The contact time of the protein with the matrix significantly influenced the refolding rate and yield. On POROS 50 HS, the refolding rate was comparable to a batch refolding process, but yield was substantially higher; at a protein concentration of 1.55 mg mL−1, an almost complete conversion was observed. With Capto S, the rate of self-cleavage increased by a factor of 20 while yield was slightly reduced. Processing the autoprotease fusion protein on Capto S at a high protein loading of 50 mg mL−1 gel and short contact time (0.5 h) yielded the highest productivity.  相似文献

In this study, we compare near-infrared (NIR) and Raman spectroscopy for the determination of the density of linear low density polyethylene (PE) (in a pellet form). As generally known, Raman spectral features are more selective than those of NIR for most chemical samples. NIR spectroscopy has been more extensively used for the quantitative analysis of polymers, but Raman spectroscopy is the better choice as long as the problem of reproducibility of Raman measurements (especially for solid samples), mostly resulting from insufficient sample representation due to probing only localized chemical information and the sensitivity of sample placement with regard to the focal plane, can be overcome. To improve sample representation and reproducibility of Raman measurements, we have employed the wide area illumination (WAI) Raman scheme, capable of illuminating a laser onto a large sample area (28.3 mm2) for Raman spectral collection (a 6-mm laser spot with a focal length of 248 mm). Diffuse reflectance NIR spectra of PE pellets were collected using a sample moving system which allowed for the scanning of large areas. The prediction error was 0.0008 g cm−3 for Raman spectroscopy and 0.0011 g cm−3 for NIR spectroscopy. The harmonization of inherently selective Raman features and a reproducible spectral collection with correct sample representations using the WAI scheme led to an accurate determination of the density of the PE pellets.  相似文献

The present article reports on the application of modified multiwalled carbon nanotubes (MMWCNTs) as a new, easily prepared and stable solid sorbent for the preconcentration of trace rhodium ion in aqueous solution. Rhodium ions were complexed with 1-(2-pyridylazo)-2-naphthol (PAN) in the pH range of 3.2-4.7 and then the formed Rh-PAN complex was adsorbed on the oxidized MWCNTs. The adsorbed complex was eluted from MWCNTs sorbent with 5.0 mL of N,N-dimethylformamide (DMF). The rhodium in eluted solution was determined by flame atomic absorption spectrometry (FAAS). Linear range for the determination of rhodium was maintained between 0.16 ng mL−1 and 25.0 μg mL−1 in initial solution. Relative standard deviation for the 10 replicated determination of 4.0 μg mL−1 of rhodium was ±0.97%. Detection limit was 0.010 ng mL−1 in initial solution (3Sbl, n = 10) and preconcentration factor was 120. Sensitivity for 1% absorbance of rhodium (III) was 0.112 μg mL−1. The sorption capacity of oxidized MWCNTs for Rh (III) was 6.6 mg g−1. The effects of the experimental parameters, including the sample pH, flow rates of sample and eluent solution, eluent type, breakthrough volume and interference ions were studied for the preconcentration of Rh3+. The proposed method was successfully applied to the extraction and determination of rhodium in different samples.  相似文献

An easy to automate flow injection-pervaporation method for monitoring urea and ammonia in must and wine was developed. The method is based on separation of the ammonia from the sample matrix by pervaporation followed by its reaction with salicylate, hypochlorite and nitroprusside to form a diazonium salt with maximum absorption at 647 nm. Conversion of urea into ammonia catalysed by urease was mandatory before pervaporation. After optimisation by either the univariate or multivariate approaches as required, the linear range was established (between 0 and 25 mg l−1) for both analytes. Then, the assessment of the proposed method versus a reference one for urea and ammonia was studied in terms of repeatability (0.52 and 0.43 mg l−1, respectively), reproducibility (1.34 and 1.21 mg l−1, respectively), detection and quantification limits (LOD=0.9 and 0.6 mg l−1, LQ=1.02 and 0.67 mg l−1, respectively) and traceability. The sample throughput was 16 samples h−1. The method can be applied to the monitoring of the target analytes in must and young wine in order to control their contents, preventing formation of ethyl carbamate.  相似文献

A selective molecularly imprinted polymer (MIP) has been synthesized for isoxicam pre-concentration, followed by its spectrophotometric determination based on hydrogen bonding interactions between examined drug and alizarin yellow GG. This method is able to evaluate isoxicam in range of 1.0 × 10−3 to 20.0 μg mL−1, with a limit of determination of 1.0 ng mL−1. The retention capacity and pre-concentration factor of prepared sorbent are 18.5 mg g−1 and 200, respectively; and the prepared MIPs can be reused at least for five times. The MIP capability for isoxicam selection and extraction from the solution is higher than non-imprinted polymer (NIP). Under optimum conditions, this procedure can be successfully applied to assay trace amounts of isoxicam in pharmaceutical and biological samples.  相似文献

A suitable method for the gas chromatographic determination of 10 characteristic carbonyls in biological and oil samples based on the in-drop formation of hydrazones by using 2,4,6-trichlorophenylhydrazine (TCPH), has been developed. The derivatisation-extraction procedure was optimized separately for aqueous and oil samples with respect to the appropriate organic drop solvent, drop volume, in-drop TCPH concentration, sample stirring rate, temperature during single-drop microextraction (SDME), reaction time and headspace-to-sample volume ratio. The optimization showed differentiation of optimum values between the studied matrices. The limits of detection were found to range from 0.001 to 0.003 μg mL−1 for the aqueous biological samples and from 0.06 to 0.20 μg mL−1 for the oil samples. The limits of quantification were in the range of 0.003-0.010 μg mL−1 and 0.020-0.059 μg mL−1 for aqueous and oil samples, respectively. The overall relative standard deviations of the within-day repeatability and between-day reproducibility were <4.4% and <8.2% for the aqueous biological samples and <3.9% and <7.4% for the oxidized oil samples.  相似文献

Although there are a number of existing assays for monitoring the activity of both isopenicillin N synthase (IPNS) and deacetoxycephalosporin C synthase (DAOCS), none have demonstrated the qualities required for screening a mutant library. Hence, enzyme-linked immunosorbent assays (ELISAs) for IPNS and DAOCS were developed based on the detection of the catalytic turnover products isopenicillin N and cephalexin/phenylacetyl-7-aminodeacetoxycephalosporanic acid (G-7-ADCA), respectively. These assays are relatively fast compared to existing assays, such as the hole-plate bioassay, and are amenable with high-throughput screening. Both the IPNS and DAOCS-ELISAs were optimised for use with crude protein extracts rather than purified protein, thereby eliminating any additional time required for purification. The ELISA developed for the detection of cephalexin had an IC50 value of 154 ± 9 ng mL−1 and LOD of 7.2 ± 2.2 ng mL−1 under conditions required for the assay. Good recoveries and correlation was observed for spiked samples when the concentration of crude protein was kept below 1 mg mL−1. The DAOCS-ELISA was found to have increased sensitivity compared to the hole-plate bioassay (10.3 μg mL−1). The IPNS-ELISA did not significantly increase the sensitivity (approximately 5 μg mL−1) compared to that of the hole-plate bioassay (16 μg mL−1) for isopenicillin N. The minimum amount of crude protein extract required for producing detectable amounts of product for both assays was below 0.5% of the maximum amount of protein that the assay could contain without any effect on the ELISA. This suggests that when screening a mutant library, mutants producing low amounts of the product could still be detected using these assays.  相似文献

A gas chromatography–mass spectrometric method was developed in this study in order to determine fluoride in plasma and urine after derivatization with 2-(bromomethyl)naphthalene. 2-Fluoronaphthalene was chosen as the internal standard. The derivatization of fluoride was performed in the biological sample and the best reaction conditions (10.0 mg mL−1 of 2-(bromomethyl)naphthalene, 1.0 mg mL−1 of 15-crown-5-ether as a phase transfer catalyst, pH of 7.0, reaction temperature of 70 °C, and heating time of 70 min) were established. The organic derivative was extracted with dichloromethane and then measured by a gas chromatography–mass spectrometry. Under the established condition, the detection limits were 11 μg L−1 and 7 μg L−1 by using 0.2 mL of plasma or urine, respectively. The accuracy was in a range of 100.8–107.6%, and the precision of the assay was less than 4.3% in plasma or urine. Fluoride was detected in a concentration range of 0.12–0.53 mg L−1 in six urine samples after intake of natural mineral water containing 0.7 mg L−1 of fluoride.  相似文献

We demonstrate the potential of Raman spectroscopy to detect cocaine concealed inside transparent glass bottles containing alcoholic beverages. A clear Raman signature of cocaine with good signal-to-noise was obtained from a ∼300 g solution of adulterated cocaine (purity 75%) in a 0.7 L authentic brown bottle of rum with 1 s acquisition time. The detection limit was estimated to be of the order of 9 g of pure cocaine per 0.7 L (∼0.04 moles L−1) with 1 s acquisition time. The technique holds great promise for the fast, non-invasive, detection of concealed illicit compounds inside beverages using portable Raman instruments, thus permitting drug trafficking to be combated more effectively.  相似文献

In this paper we use surface-enhanced Raman spectroscopy (SERS) on 3D metallic structures for label-free detection and characterization of proteins of interest at low concentrations. The substrates are prepared via nanopatterning with latex nano/microparticles and Cr and Ag sputtering, yielding stable, tunable, and mechanically flexible plasmonic structures. The nanovoids generate a SERS signal of the proteins of interest that is background free and independent of the protein charge. Concentrations as low as 0.05 μg mL−1 could be detected for 4 different proteins. The proteins also exhibit significantly different SERS spectra on these substrates, which is an important feature for future label-free direct detection schemes.  相似文献

Molecularly imprinted polymers (MIP) have been elucidated to work as artificial receptors. In our present study, a MIP was applied as a molecular recognition element to a chemical sensor. We have constructed a creatinine sensor based on a MIP layer selective for creatinine and its differential pulse, cathodic stripping voltammetric detection (DPCSV) on a hanging mercury drop electrode (HMDE). The creatinine sensor was fabricated by the drop coating of dimethylformamide (DMF) solution of a creatinine-imprinted polymer onto the surface of HMDE. The modified-HMDE, preanodised in neutral medium at +0.4 V versus Ag/AgCl for 120 s, exhibited a marked enhancement in DPCSV current in comparison to the less anodised (≤+0.3 V) HMDE. The creatinine was preconcentrated and instantaneously oxidised in MIP layer giving DPCSV response in the concentration range of 0.0025-84.0 μg mL−1 [detection limit (3σ) 1.49 ng mL−1]. The sensor was found to be highly selective for creatinine without any response of interferents viz., NaCl, urea, creatine, glucose, phenylalanine, tyrosine, histidine and cytosine. The non-imprinted polymer-modified electrode did not show linear response to creatinine. The imprinting factor as high as 9.4 implies that the imprinted polymer exclusively acts as a recognition element of creatinine sensor. The proposed procedure can be used to determine creatinine in human blood serum without any preliminary treatment of the sample in an accurate, rapid and simple way.  相似文献

The presented experiments show the model of expectation of equine spleen ferritin extraction in a new aqueous two-phase system which was formed by mixing polyethylene glycol (PEG) and hydroxyethyl starch (HES). The tendency of the protein to migrate in the analyzed systems was dependent on the concentrations of HES and PEG as well as PEG molecular weight. The highest concentration of ferritin in the top phase (rich in PEG) was recorded in the system composed of 6% PEG 3000 and 3% HES. The obtained concentration was 0.88 mg mL−1. The lowest concentration was 0.42 mg mL−1 in the system composed of 5% PEG 6000 and 1% HES.  相似文献
