The particles of CPV of silkworm contain double-stranded RNA polymerase and methyltransferase. It was reported in a previous paper that the genome-enzyme complex could be isolated. The genome-enzyme complex shows high enzyme activity of RNA polymerase and methyltransferase in spite of the fact that it consists of only 5 percent of the protein. In order to clarify the protein subunits of the RNA polymerase and methyltransferase, two methods were adopted. The SDS-polyacrylamide gel electrophoretogram showed that the ~(125)I-labeled genome-enzyme complex of CPV contained three protein components in molccular weight of 33 K, 67 K and 142 K daltons respectively and each protein component of them consisted of more than two protein subunits with different isoelectric points in 2-dimensional electrophoretogram. The antibody to the five protein components (P1, P2, P3, P4, P5) was prepared and used to inhibit the enzyme activities of RNA polymerase and mthyltransferase. To enhance the relative movement of domains, we inserted a random sequence of fifteen-peptide into the three domains of L-aspartase. By means of directed screening, the three isoforms of monomeric, dimmeric and tetrameric enzymes were obtained. Compared to the wild-type tetrameric L-asparease, these mutants remained 19. 7%, 42.3%, and 92% of the enzyme activity, respectively. Moreover, the examination of enzyme properties revealed that their kcat, and KM changed in varying degrees, and the optimum pH shifted towards acidic pH, while the dependence of the activity of enzyme on Mg^2 concentration and thermostability increased. Therefore this strategy provides a novel approach to directed evolution of enzymes.  相似文献

A novel choline oxidase electrode was constructed by entrapping choline oxidase into polyaniline-polyacrylonitrile composite film,The enzyme film was prepared by in situ electropolyme-ritztion of aniline into porous polyacrylonitrile-coated platinum electrode in the presence of choline oxidase ,the enzyme electrode exhibited sensitive and stable electrochemical response to choline ,The kinetics analysis,showed that the mass transport is partially rate-liniting.The influences of pH,applied potential and temperature on the response of the enzyme electrode were also desribed.  相似文献

Tryptophan(Trp)residues in pullulanase have been chemically modified with N-bromossuccinimide(NBS). The results of ultraviolet spectra indicated that there are 18 Trp residues in pullulanase and nine of them are located on the surface of the enzyme. Three of these Trp residues are none-essential residues which showed the fastest reaction speed by Zhou's plot. Two of the seven relative faster reacting residues are essential for the activity of the enzyme. The other eight are none-reactive residues with lowest reaction speed.  相似文献

Reported here is a protocol to fabricate a biocatalyst with high enzyme loading and activity retention, from the conjugation of electrospun nanofibrous membrane having biomimetic phospholipid moiety and lipase. To improve the catalytic efficiency and activity of the immobilized enzyme, poly(acrylonitrile-co-2-methacryloyloxyethyl phosphorylcholine)s(PANCMPCs) were, respectively, electrospun into nanofibrous membranes with a mean diameter of 90 nm, as a support for enzyme immobilization. Lipase from Candida rugosa was immobilized on these nanofibrous membranes by adsorption. Properties of immobilized lipase on PANCMPC nanofibrous membranes were compared with those of the lipase immobilized on the polyacrylonitrile(PAN) nanofibrous and sheet membranes, respectively. Effective enzyme loading on the nanofibrous membranes was achieved up to 22.0 mg/g, which was over 10 times that on the sheet membrane. The activity retention of immobilized lipase increased from 56.4% to 76.8% with an increase in phospholipid moiety from 0 to 9.6%(molar fraction) in the nanofibrous membrane. Kinetic parameter Km was also determined for free and immobilized lipase. The Km value of the immobilized lipase on the nanofibrous membrane was obviously lower than that on the sheet membrane. The optimum pH was 7.7 for free lipase, but shifted to 8.3-8.5 for immobilized lipases. The optimum temperature was determined to be 35 ℃ for the free enzyme, but 42-44℃ for the immobilized ones, respectively. In addition, the thermal stability, reusability, and storage stability of the immobilized lipase were obviously improved compared to the free one.  相似文献

Study on biotinylation strategies for competitive immunoassay of estradiol (E2) was carried out. Two types of competitive enzyme immunoassay (EIA) with Biotin-Avidin amplification system were established and optimized.The E2-Biotin conjugate was used as a tracer in one assay, and biotinylated antibody was used as a tracer in the other. In both of EIAs, horseradish peroxidase-labelled Avidin (Avidin-HRP) was used with a spectrometric determination of enzyme activity. The precision, sensitivity and specificity were measured and compared. The results showed that although both were satisfactory in specificity, the EIA with hapten-Biotin showed to be superior to the EIA with biotinylated antibody in sensitivity and precision. The limit of detection of serum E2 was 8 and 50 pg/mL with E2-Biotin and biotinylated antibody as tracer, respectively.  相似文献

Tryptophan residues in hyaluronidase (HAase) were modified by N-bromosuccinimide (NBS), the results indicated that there were eleven tryptophan residues in HAase, one of which was exposed and essential for the activity of the enzyme. The study on fluorescence quenching showed that KI could not quench all of the fluorescence from Trp residues in HAase, while acrylamide (Acr) could quench almost all of the fluorescence from Trp residues in HAase. The collisional quenching constants (KD) of HAase at different concentrations of Acr were calculated in terms of Stern-Volmer equation. The results implied that some of the Trp residues were buried in the interior of HAase and the Trp residue on the surface of HAase was not located in the hydrophobic pocket.  相似文献

Studies on the symmetry of quaternary structure of PEP carboxylase from sorghum leaves were carried out by means of chemical crosslinking with bifunctional reagents (diimidoesters) and subsequent SDS polyacrylamide gel electrophoresis.Results showed that the four identical subunits of the enzyme are associated isologously: i.e. the quaternary structure of the enzyme has a D_2 symmetry.The patterns of PEP carboxylase crosslinked with suberic dimethylimidate varied in the presence of various ligands.Substrate PEP, Mg~(2+) (required for catalysis of the enzyme), PEP plus Mg~(2+) and the inhibitor malate increased the extent of crosslinking of the enzyme in varying degrees, indicating that binding of these ligands to the enzyme gave rise to changes in the subunit interactions.However,the extent of crosslinking of PEP carboxylase with suberic dimethylimidate decreased greatly in the presence of the activator G6P. Meanwhile, the enzyme remains a tetrameric form either in the absence or in the presence of G6P.  相似文献

The preparation and characterization of an immobilized L-glutamic decarboxylase(GDC) were studied.This work is to develop a sensitive method for the determination of L-glutamate using a new biosensor,which consists of an enzyme column reactor of GDC immobilized on a novel ion exchange resin(carboxymethyl-copolymer of allyl dextran and N.N‘-methylene-bisacrylamide CM-CADB) and ion analyzer coupled with a CO2 electrode.The conditions for the enzyme immobilization were optimized by the parameters:buffer composition and concentration,adsorption equilibration time,amount of enzyme,temperature,ionic strength and pH.The properties of the immobilized enzyme on CM-CADB were studied by investigating the initial ate of the enzyme reaction,the effect of various parameters on the immobilized GDC activity and its stability.An immobilized GDC enzyme column reactor matched with a flow injection system-ion analyzer coupled with CO2 electrode-data collection system made up the original form of the apparatus of biosensor for determining of L-glutamate acid.The limit of detection is 1.0×10^-5M.The linearity response is in the range of 5×10^-2-5×10^-5M.The equation of linear regression of the calibration curve is y=43.3x+181.6(y is the milli-volt of electrical potential response,x is the logarithm of the concentration of the substrate of L-glutamate acid).The correlation coefficient equals 0.99.The coefficient of varioation equals 2.7%.  相似文献

The Langevin paramagnetic theory can’t describe the relation between magnetization of ferrofluids and applied magnetic field. The structuralization of ferrofluids, which is considered the main influence factor of the magnetization, is regarded. The part of magnetization works is deposited when the structure is forming. This action influences the magnetization of ferrofluids directly or indirectly. On the base of the “compressing” model, the Langevin function that usually describes the magnetization of ferrofluid is modified, and a well-fitted curve is obtained. An equation of the relation between the equivalent volume fraction after being “compressed” and the intensity of magnetic field is discovered, which approximately describes the process of magnetization. The relation between the approximate initial susceptibility and the volume fraction can be obtained from modified formula.  相似文献

The highly regioselective Buchwald–Hartwig amination at C-2 of the cheap and readily accessible reagent, 2,4-dichloropyridine with a range of anilines and heterocyclic amines is described. This new methodology is robust and provides a facile access to 4-chloro-N-phenylpyridin-2-amines on 0.25 mol scale. These intermediates undergo a further Buchwald–Hartwig amination at higher temperature to enable rapid exploration of the chemical space at C-4 and to provide a library of 2,4-bisaminopyridines.  相似文献

KMnO4-mediated oxidative CN bond cleavage of tertiary amines producing secondary amine was introduced, which was trapped by electrophiles (acyl chloride and sulfonyl chloride) to form amides and sulfonamides. Regiospecific and direct imidation of the methyl C(sp3)–H bond of thioanisoles is realized under mild and metal-free conditions with N-fluorobis(benzenesulfonyl)imide as an oxidant and nitrogen source. Proposed mechanism suggests that thionium ion intermediates and a Pummerer-type reaction are involved. The Rh(II)-catalyzed reaction of 2-carbonyl-substituted 2H-azirines with ethyl 2-cyano-2-diazoacetate or 2-diazo-3,3,3-trifluoropropionate provides an easy access to 2H-1,3-oxazines and 1H-pyrrol-3(2H)-ones. These compounds can be selectively prepared from the same starting material using temperature as the only varied parameter. The 2-azabuta-1,3-diene intermediate, a common precursor for both heterocyclic products, isomerizes into 2H-1,3-oxazine under kinetic control, while 1H-pyrrol-3(2H)-one is the sole product of the reaction at elevated temperatures. According to DFT-calculations a one-atom oxazine ring contraction involving ring-opening to a 2-azabuta-1,3-diene intermediate, followed by a 1,5- and 1,2-prototropic shift leads to the consecutive formation of imidoylketene and azomethine ylide, which then further undergo cyclization to the pyrrole derivative.  相似文献

Scope of the copper catalyzed/mediated selenium-nitrogen coupling reaction has been studied for the synthesis of isoselenazolones. It is noticed that the 2-chloro, 2-bromo-, and 2-iodo-aryl amides substrates can be exploited in the selenium-nitrogen coupling reaction by employing 25-100 mol % of CuI/1,10-phenanthroline (L) and potassium carbonate as a base in DMF. Furthermore, electron rich 2-chloro-arylamides also underwent selenium-nitrogen coupling reaction to give biologically important selenium-nitrogen heterocycles. Also, copper-catalyzed selenium-nitrogen coupling reaction has been meticulously applied for the synthesis of diaryl diselenides having methoxy, amine, and amide functionality from respective aryl iodides in the presence of stoichiometric amount of succinimide as an external Se-N coupling partner.  相似文献
