以VioF(普罗威登斯菌O30中O抗原基因簇内)编码的甲酰基转移酶酶促反应产物为研究对象,采用碰撞诱导解离(CID)负离子模式基质辅助激光解吸电离-飞行时间质谱(MALDI-TOF MS)技术建立简单、高效的罕见单糖合成酶监测方法。本方法首先直接质谱分析0.3μL未经色谱分离、除盐处理的酶促反应混合物,随后应用CID串联质谱技术对酶活反应产物进行结构表征,实现酶活反应的快速监测。结果表明:高能CID MALDI-TOF/TOF-MS平台适用于新建克隆的单糖合成酶酶活反应的监测,与现有的监测方法相比,在速度、灵敏度、重现性、自动化和溶剂消耗方面具有绝对优势。 相似文献

The vanadium trapping effect of Mg and La containing additives in cracking catalyst contaminated with 2300 ppm Ni and 4700 ppm V has been analyzed by microactivity test (MAT) and imaging secondary ion mass spectrometry (SIMS). The results of SIMS imaging are consistent with cracking activity data and show that the La/spinel is a superior vanadium trap for the fluid catalytic cracking of hydrocarbons (FCC) operation. La/spinel serves as a dual function additive for both vanadium trap and SOx removal. The optimum amount of La/spinel added to the cracking catalyst is about 15% by weight. This results in an increased catalytic activity, an increase in gasoline yield, and a decrease in coke and gas factors. The MgAl2O4 phase of Mg/Al2O3 additive is found capable of trapping vanadium while its MgO of Mg/Al2O3 phase can migrate to zeolite particles (the active component of the FCC catalyst) that, in turn, causes a decline in the activity of that catalyst. 相似文献

合成了带有荧光基团的分子探针,光谱显示探针对糖类物质具有较强的荧光响应.通过条件优化,选取328 nm作为荧光激发波长,p H 10.0的缓冲溶液为最佳测定介质,当测试溶液中葡萄糖浓度为0.05~1 mmol/L时,显示良好的响应线性.方法对果糖中总糖含量测定的检测限为6.2μmol/L,回收率在76.4%~90.0%,RSD达到3.18%. 相似文献

近年来,以RDX为代表的痕量爆炸物检测已经是反恐斗争和国土安全领域的重要课题[1]。本实验将醋酸作为辅助试剂直接添加到待分析水样中,采用电喷雾电离直接进样,在正离子检测模式下,建立了直接快速测定水样中衡量RDX的电喷雾质谱分析方法。在样品流量为10.0μL/m in,喷雾电压为5.0 kV,毛细管温度为200℃条件下获得的RDX的检出限为0.001μg/L,线性范围为0.005~100μg/L。对天然湖水、矿泉水和自来水中添加的衡量RDX的回收率分别在92%~108%之间。单个样品分析(含串联质谱分析)所需时间不超过2 m in。 相似文献

Amyloid fibrils are self‐assembled protein structures with important roles in biology (either pathogenic or physiological), and are attracting increasing interest in nanotechnology. However, because of their high aspect ratio and the presence of some polymorphism, that is, the possibility to adopt various structures, their characterization is challenging and basic information such as their mass is unknown. Abstract FAB-MS/MS methods are used to quantify the neuropeptide leucine enkephalin (LE = YGGFL) in synthetic solutions. Maximum molecular specificity is provided by monitoring two metastable transitions from the LE (M + H)+, 556 → 425 and 556 → 336, in a forward geometry (E, B) mass spectrometer using a B/E linked-field selected reaction monitoring technique. Obtained sensitivity is 40 pg LE, which equals 72 fmol. The statistics of the best-fit straight lines are, for m/z 425: y = 34 + 166 (r = 0.999), and for m/z 336: y = 2.5x + 17.2 (r = 0.996). 相似文献

Neuropeptides are essential cell-to-cell signaling messengers and serve important regulatory roles in animals. Although remarkable progress has been made in peptide identification across the Metazoa, for some phyla such as Echinodermata, limited neuropeptides are known and even fewer have been verified on the protein level. We employed peptidomic approaches using bioinformatics and mass spectrometry (MS) to experimentally confirm 23 prohormones and to characterize a new prohormone in nervous system tissue from Strongylocentrotus purpuratus, the purple sea urchin. Ninety-three distinct peptides from known and novel prohormones were detected with MS from extracts of the radial nerves, many of which are reported or experimentally confirmed here for the first time, representing a large-scale study of neuropeptides from the phylum Echinodermata. Many of the identified peptides and their precursor proteins have low homology to known prohormones from other species/phyla and are unique to the sea urchin. By pairing bioinformatics with MS, the capacity to characterize novel peptides and annotate prohormone genes is enhanced. 相似文献

目的:测定白砂糖和红糖中硒含量为研制富硒糖提供科学依据。方法采用微波消解-原子荧光光谱法测定白砂糖和红糖中硒含量。结果在确定的最佳仪器条件下,原子荧光光谱法测定白砂糖和红糖中硒的检出限为0.063μg · L-1;白砂糖硒的RSD为2.28%,红糖的RSD为2.13%;白砂糖回收率为103.4%,红糖回收率为99.7%。结论该法具有方法简便、快捷、准确等优点,适用于白砂糖和红糖中硒含量测定。 相似文献
