A biflavonoids, morelloflavone (1) and a prenyltated xanthone, camboginol (2), isolated from the fruits of Garcinia dulcis (Roxb.) Kurz., exhibited strong antioxidation effects in both Fe2+ -mediated and non-metal induced human low-density lipoprotein (LDL) oxidations. However, a well-known antioxidant, alpha-tocopherol (vitamin E), was found less potent than both compounds based on the same test systems.  相似文献

冠心病患者血清钙、镁、铁、铜、铬、铝变化的临床观察   总被引:2,自引:0,他引:2  
观察血清钙、镁、铁、铜、铬、铝与不同类型冠心病的关系,将冠心病患者分为三组:(1)冠状动脉痉挛30例;(2)稳定型心绞痛(SAP)患者27例;(3)急性冠脉综合症(ACS)患者35例,以34例健康者为对照组(NC),利用ICP-AES法测定了其血清钙、镁、铁、铜、铬、铝。结果表明,与正常对照组比较,钙水平在冠状动脉痉挛组和稳定型心绞痛组偏低(P<0.05或P<0.01),男性急性冠脉综合症组血清铝偏低(P<0.05)。提示血清钙水平下降可能与稳定型心绞痛及冠状动脉痉挛有关,而男性血清铝水平下降可能与急性冠脉综合症有关。  相似文献

本文对非创伤性血管成像技术(CTA)定量评估稳定型心绞痛患者的冠状动脉斑块进展及其在心血管事件中的预测价值进行了分析。选取2017年1月~2017年12月我院心血管科142例因新发症状或症状加重随访行两次冠状动脉CT血管造影(CCTA)的稳定型心绞痛患者作为研究对象,根据后期随访患者是否有主要不良心脏事件发生进行分组,其中82例发生主要不良心脏事件的患者作为观察组,60例未发生主要不良心脏事件的患者作为对照组,定量评估稳定型心绞痛患者斑块的进展情况。结果显示:观察组患者有糖尿病、高血脂、吸烟史的比例均高于对照组,差异有统计学意义(P<0.05)。两组随访CCTA总体斑块负荷率、冠状动脉直径狭窄率比较差异有统计学意义(P<0.05);两组斑块进展方面,脂质斑块负荷率、纤维斑块负荷率、钙化斑块负荷率、总体斑块负荷率、冠状动脉直径狭窄率比较差异有统计学意义(P<0.05)。通过多因素Logisitic回归分析,脂质斑块负荷进展与基线高血脂有关(P<0.05),总体斑块负荷进展与基线高血脂具有相关性(P<0.05),冠状动脉直径狭窄率进展与糖尿病、高血脂、吸烟史具有相关性(P<0.05)。本文证实了CCTA检查是检测稳定型心绞痛患者斑块负荷进展的一种有效方法,且心血管危险因素能加快稳定型心绞痛患者负荷进展。  相似文献

A method of sample clarification and high-performance liquid chromatography specifically developed to permit precise and rapid determination of vitamin A (retinol) and vitamin E (alpha-tocopherol) in serum and plasma is reported. Serum proteins were denatured by the addition of acetonitrile containing alpha-tocopherol acetate, the internal standard; the vitamins were subsequently extracted into an organic matrix consisting of ethyl acetate-butanol (1:1); no solvent evaporation step was required. The three vitamins of interest were eluted from a reversed-phase C18 column with an isocratic mobile phase methanol-water (95:5); detection was accomplished by measuring ultraviolet absorption at 280 nm. Recoveries of retinol, alpha-tocopherol and alpha-tocopherol acetate from spiked aqueous samples averaged 100.0, 100.0 and 98.8%, respectively. Recoveries of retinol, alpha-tocopherol and alpha-tocopherol acetate from plasma and serum relative to water were 102.6, 96.9 and 96.5%, respectively. Retinol and alpha-tocopherol were stable in the extraction matrix for up to 3.5 h, and were stable in heparinized plasma stored at room temperature for two days. Oxalate, citrate and EDTA caused significant losses of retinol and alpha-tocopherol, while vitamin levels in serum and heparinized plasma were similar. Limits of detection for retinol and alpha-tocopherol were 60 ng/ml and 0.9 micrograms/ml, respectively. Each run required 12 min. Same-day coefficients of variation were 3.5 and 3.6% for retinol and alpha-tocopherol, respectively (n = 11). Between-day coefficients of variation for retinol and alpha-tocopherol were 4.8 and 5.5%, respectively (n = 5). This method permits simple, rapid, sensitive, selective and precise determination of retinol and alpha-tocopherol using 0.5 ml of serum or heparinized plasma.  相似文献

The reaction of vitamin E (alpha-tocopherol) with linoleic acid containing peroxidized linoleic acid has been studied. No significant reaction was found in ethanol solution, whereas in sodium dodecyl sulfate micelles vitamin E reacted rapidly with peroxidized linoleic acid, and thereby induced the peroxidation of linoleic acid, leading to oxygen absorption. The reaction kinetics was studied in detail by u.v. spectroscopy, HPLC and ESR spectroscopy. It was found that the main product was alpha-tocopherone with alpha-tocopheroxy radical as the reaction intermediate. A mechanism involving two consecutive bimolecular reactions between peroxidized linoleic acid and alpha-tocopherol and between peroxidized linoleic acid and alpha-tocopheroxy radical, with rate constant 2.93 and 6.21 mol/L-1s-1 respectively is proposed. The micellar effect on the reaction is discussed.  相似文献

We used reversed-phase HPLC with diode array detection to simultaneously measure the major isoforms of vitamins A, E, and the carotenoids in serum from 55 healthy people with spinal cord injuries. Typically, the method measured retinol (vitamin A), alpha-tocopherol (vitamin E) and beta-carotene, alpha-carotene, lutein, lycopene, and cryptoxanthin (carotenoids). gamma-Tocopherol (vitamin E), 25-hydroxycalciferol (vitamin D), and the carotenoid zeaxanthin could also be measured when they were present in high concentrations. Healthy people with spinal cord injuries were more likely than similar people without injuries to have low concentrations of alpha-tocopherol, and to a lesser extent retinol and beta-carotene.  相似文献

应用微流控芯片电泳,以40 mmol/L Tricine(pH9.8)作为电泳缓冲体系,十二烷基硫酸钠(SDS)作为添加剂(0.1 mmol/L SDS样品溶液,0.02 mmol/L SDS分离缓冲液),分离血清小而密低密度脂蛋白(sdLDL)。研究荧光染料硝基苯并噁二唑-C6-酰基鞘胺醇(NBD C6-ceramide)与脂蛋白结合的特异性、饱和性以及血清保存和检测时间对脂蛋白电泳行为的影响;探讨SDS有效降低蛋白吸附,提高血清脂蛋白分辨率的作用。冠心病(CHD)组sdLDL检出率(75%)显著高于对照组(6%,P<0.01)。该法具有简易、快速、高效等优点,可望成为CHD危险性评估的常规分析手段。  相似文献

In this study, we focused on studying the changes in urine metabolites in hyperlipidemic rats using ultra-performance liquid chromatography coupled with quadrupole time-of-fight mass spectrometry (UPLC–Q-TOF/MS) and metabolomics, as well as the effect of Citri Reticulatae Chachiensis Pericarpium (CRCP) on hyperlipidemia. These urine samples were examined by UPLC–Q-TOF/MS to obtain MS data. The MS data were analyzed by principal component analysis and partial least squares-discriminant analysis to identify the differential metabolites. CRCP reduced the body weight and levels of triglycerides, total cholesterol and low-density lipoprotein cholesterol and abnormally decreased high-density lipoprotein cholesterol in hyperlipidemic rats, which were significantly raised by a high-fat diet. Twenty-seven potential biomarkers were identified within the complex sample matrix of urine. Fourteen biomarkers increased in the hyperlipidemia rats compared with normal rats. Meanwhile, 13 biomarkers decreased. CRCP reversed abnormal changes in biomarkers, including 5-l -glutamyl-taurine, 5-aminopentanoic acid, cis-4-octenedioic acid and 2-octenedioic acid. These biomarkers show that hyperlipidemia is related to the metabolic pathways of taurine and hypotaurine metabolism, fatty acid biosynthesis , and arginine and proline metabolism . CRCP mainly prevents hyperlipidemia by intervening in these metabolic pathways.  相似文献

目的观察枳实、枳壳用于复方治疗冠心病的临床疗效对比分析。方法观察260例冠心病心绞痛患者,随机分为对照组、复方组(分1~4小组)、阴性组(分a~d小组)、考察组(分Ⅰ~Ⅳ小组),疗程8周。结果各项评价指标显示,复方组(1~4)、考察组(Ⅰ、Ⅱ)明显优于对照组(P0.05),复方各小组与对应考察组比较无显著性差异(P0.05),但考察组(Ⅲ、Ⅳ)不良反应率高于对应的复方组。阴性组疾病疗效、心电图与对照组比较无显著性差异,中医症状改善明显优于对照组。复方组与对应阴性组比较有明显差异。结论枳实、枳壳用于复方中治疗冠心病心绞痛有重要作用,能明显缓解心绞痛症状,改善中医症状,提高疾病疗效,两者之间不可完全相互替代。  相似文献

Vitamin E (alpha-tocopherol) has been implicated in several cellular processes including signaling, transport, lipid membrane curvature, and several neurodegenerative disorders. Vitamin E imaging has been hindered by the inaccessibility of the molecule to traditional immunohistochemical methods. Using time-of-flight secondary ion mass spectrometry (ToF-SIMS), the distribution of major constituents in the cellular membrane of isolated neurons was investigated. There is a significant increase in the vitamin E signal at the soma-neurite junction compared to the cell as a whole (165 +/- 11% of that found across the cell, p = 0.004, n = 12). The observed membrane distribution suggests an important new role for vitamin E in neuronal function.  相似文献

A simple method is described which permits, avoiding saponification, alpha-tocopherol and alpha-tocopheryl acetate measurement in semi-synthetic diets for experimental animals by HPLC, with both UV and fluorescence detection. Phenyldodecane was chosen as internal standard with remarkable performances, and EDTA and BHT were added to prevent oxidation in aqueous and non-aqueous phases respectively. The mobile phase was methanol-water (94:6 v/v) at a flow-rate of 2 ml/min. Samples were homogenized and extracted twice with n-hexane by probe sonication. Extracts were evaporated to dryness and redissolved with chloroform-methanol (1:1, v/v). Validation parameters were studied between 25 ng and 6 micrograms for alpha-tocopherol and between 3 and 24.2 micrograms for alpha-tocopheryl acetate, which corresponds to the range of values in the existing diets. Results had correlation coefficients > 0.99; recoveries > 85%; R.S.D. < 6%, so the method is adequate to control vitamin E intake in animals as well as vitamin E stability in food during storage.  相似文献

A study on the regeneration of alpha-tocopherol (vitamin E) by phenolic co-antioxidants in homogeneous hydrocarbon solution is reported. The behavior of some relevant phenols such as BHA, BHT, and trans-resveratrol appears to be nicely predicted by a model based on the knowledge of kinetic and thermochemical data concerning the various reactants. Despite its good reputation as an antioxidant, trans-resveratrol was found only moderately effective (k(inh) = 2.0 x 10(5) M(-1) s(-1) in chlorobenzene at 303 K) and unable to recycle vitamin E.  相似文献

A simple HPLC method for vitamin E (alpha-tocopherol) measurement in the leaves of Rosmarinus officinalis has been developed and validated. It has enabled new data for alpha-tocopherol content to be established. The leaves, recently harvested, were dried in a microwave oven and crushed; then, alpha-tocopherol was directly extracted from portions of ground material with acetone, by probe sonication. After centrifugation the acetonic extract was analysed by HPLC with ergocalciferol (vitamin D2) added as internal standard and a gradient elution with a Nucleosil C18 column at 35 degrees C. Validation parameters of the method can be considered adequate. For standards: linearity is r=0.999, recovery is 100+/-2%, intra-assay precision has RSD=+/-3% and inter-assay precision has RSD=+/-6%. For samples: linearity is r=0.99, recovery: 93+/-7%, intra-assay precision has RSD=+/-4% and inter-assay precision has RSD=+/-7%.  相似文献

Several parameters and risk factors were compared between Korean male myocardial infarction (MI) patients (n = 10) and angina pectoris (AP) patients (n = 17) to search unique biomarkers for myocardial infarction (MI) in lipoprotein level. Individual serum and lipoprotein fractions (VLDL, LDL, HDL2, HDL3) were isolated and analyzed by lipid and protein determination and enzyme assay. The MI group was found to have a 25 and 30% higher serum cholesterol and triacylglycerol (TG) level than the AP group, respectively, however, their body mass index (BMI), LDL-cholesterol (C), HDL-C, and glucose levels fell within the normal range. MI patients were found to have an approximately two-fold higher level of serum IL-6 and an 18% lower serum apoA-I level than that of the AP group. LDL and HDL2 fraction of the MI group were more enriched with TG than those of AP group. The increased TG was correlated well with the increased level of apoC-III in the same fraction. Cholesteryl ester transfer protein (CETP) activity and protein level were greatly increased in MI patients in the LDL and HDL3 fractions. MI patients showed more severely oxidized LDL fraction than patients in the AP group, as well as the weakest antioxidant ability of serum. Conclusively, MI patients were found to have unique serum and lipoprotein characteristics including increased IL-6 and TG in serum, with CETP and apoC-III in the LDL and HDL fractions, as well as severely impaired antioxidant ability of HDL.  相似文献

The antioxidant effect of resveratrol (3,4',5-trihydroxy-trans-stilbene) and its analogues, that is, 4-hydroxy-trans-stilbene (4-HS), 3,5-dihydroxy-trans-stilbene (3,5-DHS), 4,4'-dihydroxy-trans-stilbene (4,4'-DHS), 3,4-dihydroxy-trans-stilbene (3,4-DHS), 3,4,5-trihydroxy-trans-stilbene (3,4,5-THS) and 3,4,4'-trihydroxy-trans-stilbene (3,4,4'-THS), against the peroxidation of linoleic acid has been studied in sodium dodecyl sulfate (SDS) and cetyltrimethyl ammonium bromide (CTAB) micelles. The peroxidation was initiated thermally by a water-soluble azo initiator 2,2'-azobis(2-methylpropionamidine) dihydrochloride (AAPH), and the reaction kinetics were studied by monitoring the formation of linoleic acid hydroperoxides. The synergistic antioxidant effect of these compounds with alpha-tocopherol (vitamin E) was also studied by following the decay kinetics of alpha-tocopherol and the reaction intermediate, the alpha-tocopheroxyl radical. Kinetic analysis of the antioxidant process demonstrates that these compounds are effective antioxidants in micelles used either alone or in combination with alpha-tocopherol. The antioxidative action involves trapping the propagating lipid peroxyl radical and reducing the alpha-tocopheroxyl radical to regenerate alpha-tocopherol. It was found that the antioxidant activity of resveratrol analogues depends significantly on the position of the hydroxyl groups, the oxidation potential of the molecule and the reaction medium. Molecules with ortho-dihydroxyl and/or para-hydroxyl functionalities possess high activity.  相似文献

(1) Background: Apolipoprotein E(ApoE) plays a critical role in lipid transport. The specific allele of APOE being expressed is associated with the development of coronary heart disease (CHD), however the specific mechanisms by which ApoE drives disease are unclear. In this study, we investigated the relationship between APOE allele, lipoprotein metabolome, and CHD severity to provide evidence for the efficacy of clinical cholesterol-lowering therapy; (2) Methods: Blood samples were collected from 360 patients with CHD that were actively being treated with statins. The lipoprotein profile, including particle numbers, particle size, and lipoprotein composition concentrates, was measured by nuclear magnetic resonance (NMR) spectroscopy. The severity of CHD was determined by quantifying coronary angiography results using the Gensini scoring system; (3) Results: We found there was no significant difference in low-density lipoprotein cholesterol (LDL-C) levels among ε2+ (ε2 allele carriers, consisting of ε2/ε2 and ε2/ε3 genotypes), ε3 (consisting of ε3/ε3 and ε2/ε4 genotypes), and ε4+ (ε4 allele carriers, consisting of ε3/ε4 and ε4/ε4 genotypes) participants receiving statin treatment. Compared with the ε3 group, patients with the ε2+ genotype showed lower concentrations of total low-density lipoprotein (LDL), small-LDL, and middle-LDL particles, as well as a larger LDL size, higher very low-density lipoprotein (VLDL) composition concentrates, and higher intermediate density lipoprotein (IDL) composition concentrates. The ε4+ group showed higher concentrations of total LDL, small LDL particles, and LDL compositions with smaller LDL size. The higher level of small LDL concentration was associated with a high Gensini score (B = 0.058, p = 0.024). Compared with the ε3 group, the risk of increased branch lesions in the ε2+ group was lower (OR = 0.416, p = 0.027); (4) Conclusions: The specific allele of APOE being expressed can affect the severity of CHD by altering components of the lipoprotein profile, such as the concentration of small LDL and LDL size.  相似文献

The peroxidation was initiated thermally by the water-soluble initiator 2,2'-azobis(2-methylpropionamidine) dihydrochloride, and the reaction kinetics were studied by monitoring the formation of linoleic acid hydroperoxides. The synergistic antioxidant effect of the flavonols with alpha-tocopherol (vitamin E) was also studied by following the decay kinetics of alpha-tocopherol and the alpha-tocopheroxyl radical. Kinetic analysis of the antioxidative process demonstrates that the flavonols are effective antioxidants in solution and in micelles, either alone or in combination with alpha-tocopherol. The antioxidative action involves trapping the initiating radicals in solution or in the bulk-water phase of the micelles, trapping the propagating lipid peroxyl radicals on the surface of the micelles, and regenerating alpha-tocopherol by reducing the alpha-tocopheroxyl radical. It was found that the antioxidant activity of the flavonols and their glycosides depends significantly on the position and number of the hydroxy groups, the oxidation potential of the molecule, and the reaction medium. The flavonols bearing ortho-dihydroxy groups possess significantly higher antioxidative activity than those without such functionalities, and the glycosides are less active than their parent aglycones. The activity of the flavonols is higher in micelles than in solution, while the activity of alpha-tocopherol is lower in micelles than in solution. This is because the predominant factor for controlling the activity is the hydrogen-bonding interaction of the antioxidant with the micellar surface in the case of hydrophilic flavonols, while it is the inter- and intramicellar diffusion in the case of lipophilic alpha-tocopherol.  相似文献

The effect of pretreatment with vitamin E on cytotoxicity, DNA single strand breaks, and chromosomal aberrations as well as on mutation induced by ultraviolet-B light (UV-B) was investigated in Chinese hamster V-79 cells. Cellular pretreatment with non-toxic levels of 25 microM alpha-tocopherol succinate (vitamin E) for 24 h prior to exposure resulted in a 10-fold increase in cellular levels of alpha-tocopherol. Using a colony-forming assay, this pretreatment decreased the cytotoxicity of UV-B light. However, alkaline elution assays demonstrated that pretreatment with vitamin E did not affect the number of DNA single strand breaks caused by UV-B light. In addition, UV-B exposure produced a dose-dependent induction of chromosomal aberrations and mutations at the HGPRT locus, and neither of these actions of UV-B was influenced by pretreatment with the vitamin. These results suggest that vitamin E protects cells from UV-B-induced cytotoxicity, possibly through its ability to scavenge free radicals. The results also suggest that the extent of genotoxicity induced by UV-B light may not correlate directly with the cytotoxic action of this wavelength region in sunlight.  相似文献

Lipoproteins are important cardiovascular (CV) risk biomarkers. This study aimed to investigate the associations of lipoprotein subclasses with micro- and macrovascular biomarkers to better understand how these subclasses relate to atherosclerotic CV diseases. One hundred and fifty-eight serum samples from the EXAMIN AGE study, consisting of healthy individuals and CV risk patients, were analysed with nuclear magnetic resonance (NMR) spectroscopy to quantify lipoprotein subclasses. Microvascular health was quantified by measuring retinal arteriolar and venular diameters. Macrovascular health was quantified by measuring carotid-to-femoral pulse wave velocity (PWV). Nineteen lipoprotein subclasses showed statistically significant associations with retinal vessel diameters and nine with PWV. These lipoprotein subclasses together explained up to 26% of variation (R2 = 0.26, F(29,121) = 2.80, p < 0.001) in micro- and 12% (R2 = 0.12, F(29,124) = 1.70, p = 0.025) of variation in macrovascular health. High-density (HDL-C) and low-density lipoprotein cholesterol (LDL-C) as well as triglycerides together explained up to 13% (R2 = 0.13, F(3143) = 8.42, p < 0.001) of micro- and 8% (R2 = 0.08, F(3145) = 5.46, p = 0.001) of macrovascular variation. Lipoprotein subclasses seem to reflect micro- and macrovascular end organ damage more precisely as compared to only measuring HDL-C, LDL-C and triglycerides. Further studies are needed to analyse how the additional quantification of lipoprotein subclasses can improve CV risk stratification and CV disease prediction.  相似文献
