A tungsten-rhodium coating on the integrated platform of a transversely heated graphite atomiser (THGA) was used as a permanent chemical modifier for the determination of lead in biological materials by slurry sampling in electrothermal atomic absorption spectrometry (ETAAS). Slurries were sonicated during 20 s before being delivered to the previously W-Rh treated platform. The number of particles of biological materials introduced into the atomiser for delivering 20 microL slurry aliquot ranged from 5,100 to 39,000. The permanent W-Rh modifier remained stable during approximately 300 analytical measurements when 20 microL of slurries containing up to 1.5% m/v were delivered into the atomiser. In addition, the permanent modifier increases the tube lifetime by approximately 100% when compared to untreated integrated platforms. Also, there is less decrease of sensitivity during the atomiser lifetime when compared with the conventional modifiers, resulting in a decreased need of re-calibration during routine analysis and consequently increasing the sample throughput. The atomiser lifetime was limited to the THGA wall durability, because the W-Rh treated platform was intact after more than 650 analytical firings in a medium containing up to 1.5% m/v slurry of biological material. The detection limit based on integrated absorbance was 20 ng g(-1) Pb for 1.50% m/v slurries. Results from the determination of lead in slurries of biological materials using the W-Rh permanent modifier were in agreement with those obtained with digested solutions using Pd + Mg(NO3)2. 相似文献

A flow system was coupled to a graphite furnace with a platform coated with tungsten-rhodium permanent chemical modifier for in-line separation and preconcentration of copper by employing a minicolumn loaded with 1-(2-tiazolylazo)-2-naphthol (TAN) immobilized on C18-bonded silica fixed in the tip of the autosampler arm. Elution was made by sampling 35 μl of 0.50 mol l−1 HCl with further delivering into a coated platform. Remarkable improvements in both selectivity and sensitivity were observed. Copper(II) was effectively separated from solutions containing up to 20 g l−1 Na+; 10 g l−1 K+, Ca2+ and Mg2+; 1.0 g l−1 Fe3+ and Zn2+. For a sample flowing at 3.0 ml min−1 and a loading of 60 s, the detection limit was estimated as 5 ng l−1 Cu(II) at the 99.7% confidence level, and an enrichment factor of 33 was calculated. Coefficient of variation was estimated as 4% for a 0.30 μg l−1 copper solution (n=20). The W-Rh permanent chemical modifier was used to improve system stability, analytical performance and atomizer lifetime. More than 1500 firings were carried out with the same atomizer without significant variations in sensitivity and precision. On account of the reagent immobilization, its consumption was lower than 0.2 μg per determination. In addition, TAN purification was unnecessary. 相似文献

The direct determination of chromium in urine by electrothermal atomic absorption spectrometry (ETAAS) using graphite tubes modified with tungsten is proposed. Modification of the graphite is made by tungsten electrodeposition over the whole surface atomizer followed by carbide formation by heating the tube inside its own furnace. For tungsten electrocoating, the graphite tube and a platinum electrode were connected to a power supply as cathode and anode, respectively, and immersed in a solution containing 2 mg of W in 0.1% v/v HNO3. Then, 5 V was applied between the electrodes during 20 min for tungsten electrodeposition over the whole atomizer. A SpectrAA 220 Varian atomic absorption spectrometer equipped with a deuterium background corrector was used throughout. Undiluted urine (20 μl) was delivered over the tungsten-treated tube and the chromium-integrated absorbance was measured after applying a suitable heating program with maximum pyrolysis at 1300 °C and atomization at 2500 °C. With electrodeposited tungsten modifier, the tube lifetime increased up to four times when compared to previous published methods for Cr determination in urine by ETAAS, reaching 800 firings. Method detection limit (3 S.D.) was 0.10 μg l−1, based on 10 integrated absorbance measurements of a urine sample with low Cr concentration. Two reference materials of urines (SRM 2670) from National Institute of Standards and Technology (NIST) were analyzed for method validation. For additional validation, results obtained from eight human urine samples were also analyzed in a spectrometer with Zeeman effect background correction. 相似文献

In the present work, scandium was determined in sediment slurry samples (from three different rivers) by electrothermal atomic absorption spectrometry (ETAAS). Slurries were prepared by weighting 100 mg of dry sediment samples (≤53 μm particle sizes) and adding 6 ml of HCl:HNO3:HF (3:1:2, v/v). Accurate results were only possible due to the synergetic effect between Rh as permanent chemical modifier and acetylacetone (Acac) as complexing agent. The same platform was used for 400 heating cycles. The performance of the chemical modification was evaluated by using scanning electron microscopy (SEM), synchrotron radiation X-ray fluorescence (SRXRF) and some figures of merit (precision and detectability). The best analytical conditions were attained using 1500 and 2550 °C as pyrolysis and atomization temperatures. The scandium content in the liquid phase of the slurries ranged from 61 to 73%, thus indicating, in this study, that both liquid and solid phases play an important role in slurry analyses. An amount of 5.0–20.0 μg l−1 Sc linear range as well as LOD and LOQ of 0.19 and 0.62 μg l−1, respectively, were obtained under these conditions. The accuracy was checked by using microwave-assisted decomposition, and the results compared to those obtained with the proposed methodology (slurry analysis). By checking both sets of the results, there is no statistical difference at the 95% confidence levels. 相似文献

Cloud point extraction for ultra-trace Cd determination in microwave-digested biological samples by ETAAS 总被引:1,自引:0,他引:1

Cloud point extraction (CPE) has been used for the preconcentration of cadmium, after the formation of a complex with 2-(5-bromo-2-pyridylazo)-5-(diethylamino)-phenol (5-Br-PADAP), and further determination by graphite furnace atomic absorption spectrometry (ETAAS) using polyethyleneglicolmono-p-nonyphenylether (PONPE 7.5) as surfactant. The chemical variables that affect the cloud point extraction were optimized. The separation of the two phases was easily accomplished by cooling the mixture in order to make more viscous the surfactant-rich phase. In order to establish the optimum conditions for the determination of Cd by ETAAS, Pd + Mg, Pt, Ir, Rh and Ru were studied as chemical modifiers. The best thermal stabilization was obtained with Pd + Mg, with a maximum pyrolysis temperature of 1100 °C. Under the optimum conditions i.e., pH 9.0, [5-Br-PADAP] = 2.0 × 10−5 mol L−1, [PONPE 7.5] = 0.02% (w/v), an enhancement factor of 22-fold was reached. The lower limit of detection (LOD) obtained under the optimal conditions was 0.008 μg L−1. The precision for 10 replicate determinations at 0.2 μg L−1 Cd was 3.5% relative standard deviation (R.S.D.). The calibration graph using the preconcentration method was linear with a correlation coefficient of 0.9984 at levels close to the detection limit up to at least 1.0 μg L−1. The method was successfully applied to the determination of cadmium in urine samples and in a water standard reference material. 相似文献

Ruthenium (Ru), thermally deposited on a integrated platform graphite furnace, was investigated as a permanent modifier for the determination of Aluminum (Al) in blood serum and urine by electrothermal atomic absorption spectrometry (ETAAS). The platform was treated with 500 μg of Ru as previously described. The pyrolysis and atomization temperatures for each material were of 1300 and 2300 °C, for serum sample and of 1000 and 2400 °C, for urine. The characteristic mass were of 31 and 33 pg for Al in serum sample and urine, respectively (recommended of 31 pg for Al in nitric acid 0.2% (v/v)). For this reason, the calibration was made with aqueous solutions for both the samples. Calibration curves presented r of 0.99145 and 0.99991 for serum and urine, respectively. With the optimized temperatures, being analyzed eight spiked blood serum samples, the recovery was between 95.90 and 113.50%. Two certified urines samples were analyzed with good agreement between experimental and reference values. In both the samples the R.S.D. were <5% (n=3). The detection limit (k=3, n=10) was of 0.40 μg of Al per liter for both the samples. The absorption pulses obtained were symmetrical, with very low background and without interferences. The life time of the tube-platform was higher than 600 cycles of atomizations for both the urine and serum samples. 相似文献

The present work describes an analytical procedure for the determination of the total iodine content in biological materials (serum, milk, plants, tissues etc.). Liquid samples can be directly analyzed after dilution, if necessary, by ICP-MS. Milk powder, plants and tissues were dissolved by using a modified simple Schöninger combustion, subsequently the residue was taken up in 0.1 mol/l NaOH and this solution was analyzed by ICP-MS. The detection limit is in the range of 0.01 g/l. The method was tested using different CRMs certified for the iodine content. 相似文献

Cadmium is determined in urine samples collected from patients with age-related diseases. The urine is simply diluted 1:1 with water and placed on a tungsten coil electrothermal vaporizer treated with 200 μg of a permanent Pd modifier. A straightforward vaporization program is used to deliver the Cd vapor to an inductively coupled plasma atomic emission spectrometer. A high resolution spectrometer and a charge coupled device detector provide spectra across a 4.8 nm window encompassing two separate Cd emission lines: 226.5 and 228.8 nm. The limit of detection is 0.2 μg/L at each wavelength, and the linear dynamic range spans three orders of magnitude. The accuracy as measured with a urine standard reference material is 94%. The Pd modifier continues to be effective even after 150 vaporization cycles. Direct analysis of urine with the Pd modifier using simple aqueous calibration solutions provides results that are comparable to those observed after a much more complex method: chelation, extraction, and internal standardization without the modifier. The mean concentrations found by the two techniques differ by only 9%. The permanent Pd modifier allows direct analysis of limited sample volumes with decreased risks of contamination. 相似文献

This present work reports the development and evaluation of a method for the direct determination of manganese in waters extracted during petroleum exploitation by Electrothermal Atomic Absorption Spectrometry (ET AAS) using Ir-W as permanent modifier. These waters, usually called produced waters, contain a wide range of organic and inorganic substances and are characterized by their high salinity. In order to achieve suitable experimental conditions for the method application, studies about the effect of operational variables (chemical modifier, pyrolysis and atomization temperatures) were performed, as well as the establishment of convenient calibration strategy. The best results were verified when the temperatures of pyrolysis and atomization were 1000 °C and 2300 °C, respectively, and using Ir-W as permanent modifier. The results showed that manganese can be determined by the standard addition method or employing external calibration with standard solutions prepared in the same salinity of the samples (with NaCl). Three real samples with salinities varying between 74 and 84‰ were successfully analyzed by the developed procedure. The limits of detection and quantification were 0.24 and 0.80 μg L−1, respectively, in purified water, and 0.34 and 1.1 μg L−1, respectively, in 0.4 mol L−1 NaCl medium (approximately 23‰ salinity). 相似文献

Determination of cadmium and lead in biological samples by Zeeman ETAAS using various chemical modifiers 总被引:1,自引:0,他引:1
The electrothermal atomic absorption spectrometric determination of cadmium and lead in biological certified reference materials (CRMs) has been carried out by using NH(4)H(2)PO(4), Ni, Pd, Ni+NH(4)H(2)PO(4), Pd+NH(4)H(2)PO(4) and Ni+Pd+NH(4)H(2)PO(4) as chemical modifiers. A comprehensive comparison was made among the modifiers in 1% Triton X-100 plus 0.2% nitric acid as diluent and without modifier. Zeeman background correction and graphite tubes inserted with platforms were used. Comparison was made in terms of pyrolysis and atomization temperatures, atomization and background absorption profiles. Ni+Pd+NH(4)H(2)PO(4) modifier mixture was found to be preferable for the determination of Cd and Pb. Pyrolysis temperatures of analytes were increased up to 900 degrees C for Cd and 1250 degrees C for Pb by using Ni+Pd+NH(4)H(2)PO(4) in 1% Triton X-100 plus 0.2% nitric acid diluent solution. Biological CRMs were analyzed to verify the accuracy and precision of this method. Depending on the biological sample type, the percent recoveries were increased from 62 to 102% for Cd and from 58 to 106% for Pb by using the proposed modifier mixture. The detection limits of Cd and Pb were found to be 0.04, 0.92 mug l(-1), respectively. 相似文献

The feasibility of Ru as a permanent modifier for the determination of Cd in biological samples treated with tetramethylammonium hydroxide (TMAH) by ET AAS was investigated. The tube treatment with Ru was carried out only once and lasted for about 300 atomization cycles. The pyrolysis and atomization temperatures, 750 °C and 1300 °C, respectively, were chosen from the temperature curves. The sample dissolution procedure was very simple: a sample aliquot was mixed with a small volume of a 25% m/v TMAH solution, the volume was made up to 50 ml and the mixture was kept at 60 °C for 1 h. Six certified biological reference materials were analyzed and the obtained Cd concentrations are within the 95% confidence interval of the certified values, proving the accuracy of the proposed procedure for a variety of biological samples. The calibration curve, with correlation coefficient higher than 0.99, was established for a working range up to10 μg l−1. The precision was good as demonstrated by relative standard deviations below 3%, except for one sample. The limit of detection (3σ) was 0.05 μg l−1 and the characteristic mass was 1.30 pg, obtained in the presence of the Ru modifier. 相似文献

Cloud point extraction of mercury with PONPE 7.5 prior to its determination in biological samples by ETAAS 总被引:1,自引:0,他引:1
Cloud point extraction (CPE) has been used for the pre-concentration of mercury, after the formation of a complex with 2-(5-bromo-2-pyridylazo)-5-(diethylamino)-phenol (5-Br-PADAP), and later analysis by electrothermal atomic absorption spectrometry (ETAAS) using polyethyleneglycolmono-p-nonyphenylether (PONPE 7.5) as surfactant. The chemical variables affecting the separation step were optimized. Under the optimum conditions, i.e, pH 8.5, cloud point temperature 80 degrees C, 5-Br-PADAP=4x10(-5) mol L(-1), PONPE 7.5=0.2%, sample volume=1.0 mL, an enhancement factor of 22-fold was reached. The lower limit of detection (LOD) obtained under the optimal conditions was 0.01 microg L(-1). The precision for 10 replicate determinations at 2.0 microg L(-1) Hg was 4.0% relative standard deviation (R.S.D.). The calibration graph using the pre-concentration system for mercury was linear with a correlation coefficient of 0.9994 at levels near the detection limits up to at least 16 microg L(-1). The method was successfully applied to the determination of mercury in biological samples and in certified reference material (QC METAL LL3). 相似文献

The use of boric acid as a modifier for the determination of trace amount of indium in high purity antimony by electrothermal atomic absorption is described. It was found that the negative influence of the hydrofluoric acid, used for the digestion could not be eliminated by using stabilized temperature platform furnace (STPF) alone. Due to the high dissociation energy (D0 = 506 kJ mol−1) of indium fluoride, it is difficult to dissociate in the gas phase and hence is lost. In presence of HF (used for the dissolution of antimony), the universal Pd-Mg modifier does not work satisfactorily. Additionally, rising corrosion and reduced tube lifetime were observed when the acid digested (HF-HNO3) antimony solution was injected in to the platform. Improvement in platform life and elimination of interferences were achieved by the addition of boric acid as a chemical modifier together with ruthenium coating of the platform. Corrosive changes of the transversely heated graphite atomizer (THGA) platform surface were examined by scanning electron microscopy. The standard addition method was applied. A characteristic mass of 36 pg was obtained. The detection limit of the proposed method is around 0.04 μg g−1. The developed method was applied to the determination of indium in real samples. The data obtained by this method were in good agreement with those obtained by ICP-MS. 相似文献

Rhodium (Rh) was investigated as a permanent modifier for the atomization of Pb from biological fluids in W-filament atomic absorption spectrometry (AAS). Heating the W-filament with a Rh solution provided a protective coating for subsequent determinations of Pb in blood and urine matrices. The W-filament AAS instrumentation used was based on a prototype design that utilized self-reversal background correction scheme and peak area measurements. We found that Rh not only stabilized Pb during the pyrolysis step, but also facilitated the removal of carbonaceous residues during the cleaning step, requiring much less power than with phosphate modifier. Thus, the filament lifetime was greatly extended to over 300 firings. Periodic reconditioning with Rh was necessary every 30 firings or so. Conditioning the filament with Rh also permitted direct calibration using simple aqueous Pb standards. The method detection limit for blood Pb was approximately 1.5 μg dl−1, similar to that reported previously. Potential interferences from concomitants such as Na, K, Ca and Mg were evaluated. Accuracy was verified using lead reference materials from the National Institute of Standards and Technology and the New York State Department of Health. Blood lead results below 40 μg dl−1 were within ±1 μg dl−1 of certified values, and within ±10% above 40 μg dl−1; within-run precision was ±10% or better. Additional validation was reported using proficiency test materials and human blood specimens. All blood lead results were within the acceptable limits established by regulatory authorities in the US. When measuring Pb in urine, sensitivity was reduced and matrix-matched calibration became necessary. The method of detection limit was 27 μg l−1 for urine Pb. Urine lead results were also validated using an acceptable range comparable to that established for blood lead by US regulatory agencies. 相似文献

Summary The determination of thallium in biological materials sometimes cause problems because of the low concentrations of this toxic element. In the present work a method is described which optimizes the parameters affecting the specificity and sensitivity of the radiochemical NAA of thallium in biological samples. High thermal neutron flux, complete decomposition of the organic matter by pressurized digestion, TlI precipitations, liquid extraction of HTlBr4 and La(OH)3 scavenging purification are the steps leading to the final homogeneous preparation of Tl2CrO4 for -activity measurement. The method was applied to various materials as bovine liver, bone and nails. Good agreement was found between certified and determined thallium concentrations of the reference material CRM 176. The chemical yield comes to about 80%, with low deviations. The sensitivity of the method is about 10–3 g/g, the standard deviations being in the range of 3.6% (CRM 176), 14% (bovine liver), and 17% (bone). Detailed working instructions are given. 相似文献

Summary A new radiochemical neutron activation analysis (RNAA) method has been developed for low-level determination of Si in biological materials, which is based on the 30Si(n,γ)31Si nuclear reaction with thermal neutrons. The radiochemical separation consists of an alkaline-oxidative decomposition followed by distillation of SiF4. Nuclear interferences, namely that of the 31P(n,p)31Si with fast neutrons, have been examined and found negligible only when irradiation is carried out in an extremely well-thermalized neutron spectrum, such as available at the NIST reactor. The RNAA procedure yields excellent radiochemical purity of the separated fractions, which allows the measurement of the β--activity of the 31Si by liquid scintillation counting. Results for several reference materials, namely Bowen’s Kale, Bovine Liver (NIST SRM 1577b), Non-Fat Milk Powder (NIST SRM 1549) and several intercomparison samples, Pork Liver-1, Pork Liver-2 and Cellulose Avicel, are presented and compared with literature values. 相似文献
