A series of 3-diphenylmethyl-6-substituted-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole derivatives (4aj and 5ad) were synthesized by condensation of 4-amino-5-diphenylmethyl-4H-1,2,4-triazole-3-thiol with various substituted aromatic acids and aryl/alkyl-isothiocyanates. The structures of synthesized compounds were characterized by elemental analysis, IR, 1H NMR, 13C NMR and mass spectroscopic studies. These compounds were tested in vivo for their anti-inflammatory activity. The compounds which showed activity comparable to the standard drug ibuprofen were screened for their analgesic, ulcerogenic, lipid peroxidation and hepatotoxic effects. Compounds 6-(4-chlorophenyl)-3-diphenylmethyl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole (4a) and 6-(2,4-dichlorophenyl)-3-diphenylmethyl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole(4c) emerged as the most active compounds of the series and were moderately more potent than the standard drug ibuprofen.  相似文献

A new series of 1H-pyrazole derivatives 5a-j bearing an aryl sulfonate moiety have been synthesized by a one-pot cyclo-condensation reaction of 2-(3-(dimethylamino)acryloyl)phenyl-4-methyl benzene sulfonates 4a-e and hydrazine hydrate or phenyl hydrazine in ethanol under reflux conditions.Some of the newly synthesized compounds were screened for their anti-inflammatory activity.All synthesized compounds were screened against Gram positive and Gram negative bacterial and fungal strains.The compound 5b was found to be a potent anti-inflammatory agent while the majority of the compounds were found to be active against microbial strains.  相似文献

A novel series of 5H-pyridazino[4,5-b]indoles were designed and synthesized in order to find novel potent anticancer compounds.The structures were confirmed by ~1H NMR and MS.Their antiproliferative activities against two cancer cell lines were tested by the MTT method in vitro.Three of compounds (1e,1g,and 1h) exhibited potent antiproliferative activities,especially compound 1h (with IC_(50) values of 5.2μmol/L and 1.9μmol/L against Bel-7402 and HT-1080,respectively).The preliminary structure-activity relationships of 5H-pyridazino[4,5-b]indole derivatives were discussed.  相似文献

A series of novel 1,2,3-thiadiazole derivatives were designed and synthesized. Their structures were characterized by 1H NMR, MS and HRMS. The bioactivity tests indicated that compound 9b exhibits a favorable KARI inhibition rate, and some of these novel compounds also showed moderate herbicidal activity against Brassica campestris.  相似文献

The cysteine protease, falcipain-2 is an important drug target in human malaria parasite Plasmodium falciparum. A new series of 2-(4-(substituted benzoyl)-1,4-diazepan-1-yl)-N-phenylacetamide derivatives 5(at) were designed as per pharmacophoric requirements of falcipain-2 inhibitors using ligand-based approach. The target compounds were synthesized from the key intermediate, 2-(1,4-Diazepan-1-yl)-N-phenylacetamide, by coupling it with appropriate carboxylic acids using carbodiimide chemistry. Structural features of target compounds were characterized by spectral data (1H NMR, and mass) and elemental analyses. The purity of the final compounds was confirmed by HPLC. The compounds were tested for their in vitro falcipain-2 inhibitor activity on recombinant falcipain-2 enzyme. Five compounds 5b, 5g, 5h, 5j, 5k showed good inhibitory activity (>60%), against falcipain-2 at 10 μM concentration, and fifteen compounds showed weak to moderate inhibitor activity. Compound 5g, the most potent compound from this series showed 72% inhibition at 10 μM concentrations.  相似文献

A series of novel naphtho[1,2-e][1,3]oxazines bearing arylsulfonamide moiety have been synthesized via a one-pot approach and in a green reaction medium. These new naphtho[1,2-e][1,3]oxazine derivatives have been characterized by their 1H NMR, 13C NMR and the X-ray single crystallography method for compound 7a. All the newly synthesized compounds were examined for their in vitro anticancer activity against breast (MCF-7), colon (HCT116), and B-CLL (Waco3-CD5) cancers. Some of these compounds such as 7j and 7l showed remarkable activities against MCF-7 (breast) and HCT116 (colon) cancers with comparable IC50 (The half maximal inhibitory concentration) values as that of known drugs such as 5-fluorouracil (5-FU). In vitro antimicrobial activities of all compounds were also evaluated against five human pathogenic fungi strains and two bacteria (one gram positive and one gram negative). The best MICs (Minimum Inhibitory Concentrations) were found against the C. albicans.  相似文献

A series of new benzimidazole derivatives was synthesized and characterized by IR,1H NMR,13C NMR,MS,and HRMS spectra.All the new compounds were screened for their antimicrobial activities in vitro by a twofold serial dilution technique.The bioactive evaluation showed that 3,5-bis(trifluoromethyl)phenyl benzimidazoles were comparably or even more strongly antibacterial and antifungal than the reference drugs Chloromycin,Norfloxacin,and Fluconazole.The combination of2,4-difluorobenzyl benzimidazole derivative 5l and its hydrochloride 7 respectively with the antibacterials Chloromycin,Norfloxacin,and the antifungal Fluconazole was more sensitive to methicillin-resistant MRSA and Fluconazole-insensitive A.flavus.In addition,the interaction of compound 5l with calf thymus DNA demonstrated that this compound could effectively intercalate into DNA to form a compound 5l-DNA complex that might block DNA replication and thereby exert good antimicrobial activity.  相似文献

In the current work, a novel series of substituted imidazole, triazine and metastable Oxazolone derivatives were synthesized. The performance of 4-arylideneoxazol-5(4H)-one (2a-c) towards amoniuma acetate, hydrazine hydrate, phenyhydrazine and primary amines has been discussed. DFT bases and calculations of HUMO, LUMO were used to predict the ability of binding between synthesized compound and cations. All the synthesized compounds were assessed as chemosensors for detection metals ions. Colorimetric investigation confirmed and showed high-selectivity and specificity of compounds 5b and 5c to detect cobalt ion, and copper ion respectively, so compounds 5b and 5c can be directed as a chemosensor for optical detection of Co2+and Cu2+ respectively. On the other hand, new organic chemosensor were established by, IR, 1H NMR, 13C NMR ms spectroscopy and elemental analysis.  相似文献

A number of novel urea and thiourea derivatives of 8-hydroxyquinoline have been designed, synthesized and evaluated for their anticancer activities. The structures of the new compounds were established by spectroscopic techniques, 1H NMR, 13C NMR, and mass spectrometry. The in vitro cytotoxicity against MCF7, and MDA-MB-231 cell lines were assessed by MTT assay. Six of the 11 compounds synthesized namely 5b, 5c, 5f, and 6b-d exhibited cytotoxicity with IC50 values ranged between 0.5 and 42.4 µM. Apoptotic features of cells treated with 5b compound were observed via florescent microscopy using DAPI and ethidium bromide/acridine orange staining against MCF-7 cells. Molecular docking of these molecules against 16 potential breast cancer protein revealed that these compounds could interact with the active site of poly (ADP-ribose) polymerase-1 (PARP1), B-cell lymphoma-extra large (Bcl-xL) and PARP5A (Tankyrase 1) by forming hydrogen bonds, π-π interactions and hydrophobic interactions. The docked poses of these molecules were observed to be similar in the active site of each of these targets.  相似文献

A series of 2,5-disubstituted-1,3,4-oxadiazole derivatives bearing pyrazole moiety were synthesized by reacting various substituted pyrazole-4-carboxylic acids with different hydrazides in POCl3. All the synthesized compounds (4a–n) were characterized by IR, NMR, mass spectra and elemental analyses. Synthesized 1,3,4-oxadiazole derivatives were screened for their antibacterial activity against three different strains, namely Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa, while antifungal activity was determined against three different strains Aspergillus flavus, Chrysosporium keratinophilum and Candida albicans. The investigation of antimicrobial screening revealed that compounds 4i and 4j exhibited excellent activity when compared with the standard drugs.  相似文献

In the present study, we have synthesized a series of fifteen nipecotic acid 1,3,4-oxadiazole based hybrids with significant (60–78%) yields. All the compounds were characterized by using different spectroanalytical techniques such as FT-IR, 1H NMR, 13C NMR, and elemental analysis. This design strategy was validated by using in vivo anti-epileptic and anti-depressant bioassay models. Anti-convulsant activity was evaluated using subcutaneous pentylenetetrazol (scPTZ) in mice and MES induced seizure. Among a spectrum of activities, three compounds (4i, 4m, and 4n) displayed significant activity against pentylenetetrazole (scPTZ) induced seizures. No disruptions in motor co-ordination were observed in mice pretreated with the test compounds in the rotarod test. Their influence on the safety profile of elevated serum levels of biochemical markers such as hepatic and renal toxicity has been found to be safe. The derivatives also show marked anti-depressant activity, devoid of serotonergic augmentation as assessed using the despair swim test, 5-hydroxytryptophan (5-HTP)-induced head twitch test and learned helplessness test. In silico docking studies targeted on homology modelled GABA transporter 1 (GAT1) protein shows the critical enzyme-ligand interactions leading to the inhibition of the GAT1 transporter. The compound 4m was found to be the most active compound among all the synthesized compounds.  相似文献

A series of novel arylidene derivatives of 5,6-dihydro-4H-cyclopenta[b]-thiophene-2-carboxylic acid and 4,5,6,7-tetrahydrobenzo[b]-thiophene-2-carboxylic acid were synthesized by reacting benzylidene derivatives of chloro aldehyde with 2-mercaptoacetic acid. Benzylidene derivatives of chloro aldehyde were prepared from Vilsmeier reaction of 2-benzylidenecyclopentanone and 2-benzylidenecyclohexanone derivatives, obtained from condensation of various aromatic aldehydes with cyclopentanone and cyclohexanone. All synthesized compounds were characterized by nuclear magnetic resonance (NMR), infrared (IR), and mass spectroscopy and X-ray single-crystal analysis. The synthesized compounds were screened for their in vitro antimicrobial and antifungal activity. Good antimicrobial activity, especially against methicillin-resistant Staphylococcus aureus, was observed for most of the compounds tested. In particular, compound 9f emerged as an effective antibacterial agent and may be a potential candidate for future drug discovery and development.  相似文献

New bis-hydrazonothioxothiazolidinone derivatives based on 2-thioxothiazolidin-4-one were synthesized in good yields using a simplified experimental condition. The structure of synthesized compounds was established with the help of common physico-chemical analysis and various spectroscopic techniques like FT-IR, mass and 1H NMR. The results of characterizations are in good agreement with the proposed structure of all the synthesized compounds. Further, the antimicrobial (antibacterial and antifungal) activities of all the synthesized derivatives were carried out against various species like Bacillus subtilis, Escherichia coli, Aspergillous niger and Aspergillous flavus by using agar-cup method. The results of antimicrobial screening showed that all the compounds have mild to moderate activity. However, some of the compounds (3a, 3b, 3d, 3e, 3f, 3g, 3i and 3j) have shown better activity than the other.  相似文献

A series of thiosemicarbazones of aromatic iodide derivatives were designed and synthesized. The structures of all the newly synthesized compounds had been identified by elemental analysis, 1H NMR and 13C NMR. Their biological activities were evaluated against Plasmodium falciparum. Among these compounds, at concentrations of 3, 9 and 27 mg/kg of mouse per day, 4e inhibited the growth of the malaria parasite in vivo test in mice with the respective percentages: 88.1%, 90.7% and 92.6%. The present work suggest that thiosemicarbazones of aromatic iodide may become a lead compound for anti-malaria medicine.  相似文献

A series of N-substituted 5-chloro-6-phenylpyridazin-3(2H)-one derivatives were synthesized based on our previous work; all compounds were characterized by spectral data and tested for in vitro insecticidal activity against Plutella xylostella. The results showed that the synthesized pyridazin-3(2H)-one compounds possessed good insecticidal activities, especially the compounds 4b, 4d, and 4h which showed > 90% activity at 100 mg/L. The structure-activity relationships (SAR) for these compounds were also discussed.  相似文献

A series of (E)-2-[5-chloro-1-(1H-benzo[d]imidazol-2-yl)ethylidene] N-(substituted) hydrazine carbothioamide (7a7t) and (E)-2-[1-(1H-benzo[d]imidazol-2-yl)ethylidene] N-(substituted) hydrazine carbothioamide (8a8t) were prepared via the synthesis of 1-(substituted-1H-benzimidazol-2-yl) ethanol (3a3b) which was synthesized by the condensation of substituted o-phenylenediamine (2a2b) with dl-lactic acid (1) followed by oxidation with sodium hypochlorite in mild acidic condition to form the corresponding ketones 4a4b. Final compounds were formed by condensation of 4a4b with different thiosemicarbazides 6a6t. A total of 40 compounds were synthesized and characterized by FT-IR, 1H NMR, 13C NMR, Mass spectral technique and elemental analysis, in addition they were evaluated for anti-malarial properties. Among the compounds tested 7o, 7p, 7q, 7r, 7s, 8e and 8h exhibited good antimalarial activity in vitro.  相似文献

A series of novel 1-(β-coumarinyl)-1-(β-indolyl)-1-(α-thiophenyl)trifluoroethane derivatives 5aaa-5hdb were prepared by one-pot reaction from 3-(trifluoroacetyl)coumarin with indole and α-substituted thiophene. Their structures were confirmed by 1H NMR, 13C NMR, 19F NMR, HRMS and X-ray single crystal diffraction, and their antifungal activities against F. moniliforme, F. graminearum, F. oxysporum, R. solani and P. nicotianae were evaluated. The title compounds displayed significant to moderate in vitro antifungal activity when compared to the standard drug triadimefon. Among the synthesized compounds, compound 5bfa showed the highest inhibitor rate of 83.5 % at 0.500 mg/mL against R. solani, while compound 5ada displayed the highest inhibitor rate of 73.3 % at 0.500 mg/mL against F. graminearum.  相似文献

Since benzo [ b ] thiophene scaffold is one of the privileged structures in drug discovery as this core exhibitsactivities for different biological problems, in this study bis (benzo[ b ]thiophene-2-yl) alkyl methanimine derivatives (1-9) were synthesized by reacting benzo[ b ]thiophene-2-carbaldehyde with diamines. All newly compounds were characterized by IR, 1H NMR and 13C NMR spectroscopic methods. Synthesized compounds were investigated using binary QSARbased models on therapeutic activity prediction of synthesized compounds and they showed high predicted activities in following diseases: bacterial, angina, allergy, depression and obesity. Thus, they were then tested for their antimicrobial and antileishmanial activities as a result of this theoretical study. Compound 1(N, N’- (propane-1,3-diyl) bis (1-(benzo [ b ] thiophene-2-yl)) methanimine) was found the most active compound in both diseases. Thus, its molecular docking studies were also carried out.  相似文献

In present work, a series of novel structural hybrids of 1,3,4-oxadiazole and carbamothioate was designed by chemical modification of 2-(4-isobutylphenyl)propanoic acid. Target compounds (7a-f) were synthesized in significant yields (84–88 %) by coupling compound (4) with different electrophiles under different reaction conditions. The structures of oxadiazole based carbamothionate derivatives were confirmed by spectroscopic (FTIR, 1H NMR, 13C NMR) and physiochemical methods. During in-vivo experimentation, all synthesized compounds were tested through 6 Hz (32 mA) and PTZ (80 mg/kg) mouse seizure models. The 7b and 7c showed significant outcomes (P < 0.05) in terms of seizure severity, protection and mortality. The behavioural outcomes of PTZ tests were further strengthened with video-electroencephalogram (vEEG) findings in which EEGs were analyzed for epileptic spikes to understand the impact of 7b and 7c treatment on these ictal activities. The 7b was found most efficient in reducing the seizure spiking activity in brains of PTZ-treated mice while both 7b and 7c significantly reduced overall PTZ-induced seizure severity. The molecular docking studies also predicted the BBB permeability, reduced binding energies and good compound interaction with GABAA receptors and SV2A protein. Therefore, the observed pharmacological outcomes might be attributed to the GABAA agonistic and SV2A modulating potential of these oxadiazole-carbamothioate hybrid compounds.  相似文献

Novel 3-(4,6-dimethoxybenzofuran-5-yl)-1-phenyl-1H-pyrazole-4-carboxaldehyde (3) and 3-chloro-3-(4,6-dimethoxybenzofuran-5-yl)propenal (4) were prepared via Vilsmeier–Haack reaction of 1-(4,6-dimethoxybenzofuran-5-yl)ethanone (1) and its hydrazone derivative 2. Reaction of compound 4 with some hydrazine derivatives, namely hydrazine hydrate, phenylhydrazine and benzylhydrazine hydrochloride led to the formation of pyrazole derivatives 58, respectively. On the other hand, reaction of compound 4 with thiourea, urea or guanidine gave the pyrimidine derivatives 911, respectively. Reaction of amino compound 11 with acetic anhydride, benzoyl chloride and benzenesulphonyl chloride yielded N-substituted pyrimidine derivatives 12–14, respectively. Reaction of diazonium salt of compound 11 with sodium azide afforded azidopyrimidine derivative 15, which upon reaction with ethyl acetoacetate gave 1,2,3-triazole derivative 16. Acid catalyzed reaction of 11 with p-nitrobenzaldehyde gave Schiff base 17, which cyclized upon reaction with thioglycolic acid or chloroacetyl chloride to give thiazolidin-4-one 18 and azetidin-2-one 19, respectively. The newly synthesized compounds were tested for their anti-inflammatory, analgesic and anticonvulsant activities. Depending on the obtained results, the newly synthesized compounds possess significant anti-inflammatory, analgesic and anticonvulsant activities.  相似文献
