Supramolecular structures with strain‐stiffening properties are ubiquitous in nature but remain rare in the lab. Herein, we report on strain‐stiffening supramolecular hydrogels that are entirely produced through the self‐assembly of synthetic molecular gelators. The involved gelators self‐assemble into semi‐flexible fibers, which thereby crosslink into hydrogels. Interestingly, these hydrogels are capable of stiffening in response to applied stress, resembling biological intermediate filaments system. Furthermore, strain‐stiffening hydrogel networks embedded with liposomes are constructed through orthogonal self‐assembly of gelators and phospholipids, mimicking biological tissues in both architecture and mechanical properties. This work furthers the development of biomimetic soft materials with mechanical responsiveness and presents potentially enticing applications in diverse fields, such as tissue engineering, artificial life, and strain sensors.  相似文献

Self‐healing hydrogels as wound dressings still face challenges in infection prevention, especially in the dressing of mass wounds, due to their inflexibility and the slow formation of the protective film on the wound. Therefore, designing a spray‐filming (rapid‐forming) hydrogel that can serve as a bacterial barrier is of particular significance in the development of wound dressings. Here, a self‐healing hydrogel based on adipic acid dihydrazide‐modified gelatin (Gel‐ADH) and monoaldehyde‐modified sodium alginate(SA‐mCHO) is prepared. Using dynamic, Schiff base bonds, the hydrogels exhibit excellent self‐healing properties. Moreover, the gelation time of SA‐mCHO/Gel‐ADH (SG) hydrogels is shortened to 2–21 s, resulting in rapid filming by spraying the two precursor solutions. In addition, the rapid spray‐filming ability might offer sufficient flexibility and rapidity for dealing with mass and irregular wounds. Notably, the bacterial barrier experiments show that the SG hydrogel films could form an effective barrier to Staphylococcus aureus and Candida albicans for 12 h. Therefore, SG hydrogels could be used in wound dressings and they show great promise in applications associated with mass and irregular traumas.  相似文献

A new family of isomeric tetrapeptides containing aromatic and polar amino acid residues that are able to form molecular hydrogels following a smooth change in pH is described. The hydrogels have been studied by spectroscopic and microscopic techniques showing that the peptide primary sequence has an enormous influence on the self‐assembly process. In particular, the formation of extended hydrophobic regions and the appearance of π‐stacking interactions have been revealed as the driving forces for aggregation. Moreover, the interaction of these compounds with amyloid peptidic fragment Aβ1‐40 has been studied and some of them have been shown to act as templates for the aggregation of this peptide into non‐β‐sheet fibrillar structures. These compounds could potentially be used for the capture of toxic, soluble amyloid oligomers.  相似文献

Ionizable amino acids in protein‐based hydrogels can confer pH‐responsive behavior. Because elastin‐like polypeptides (ELPs) have an established sequence and can crosslink to form hydrogels, they are an ideal system for creating pH‐sensitive materials. This study examines different parameters that might affect pH‐sensitive behavior and characterizes the mechanical and physical properties between pH 3 and 11 of three ELP‐based crosslinked hydrogels. The first finding is that varying the amount of crosslinker affects the overall stiffness and resilience of the hydrogels but does not strongly affect water content, swelling ratio, or pH sensitivity. Second, the choice of two popular tag sequences, which vary in histidine and aspartic acid content, does not have a strong effect on pH‐sensitive properties. Last, selectively blocking lysine and tyrosine residues through acetylation significantly decreases the pH‐sensitive zeta potential. Acetylated hydrogels also demonstrate different behavior at low pH values with reduced swelling, reduced water content, and higher stiffness. Overall, this work demonstrates that ELP hydrogels with ionizable groups are promising materials for environmentally‐responsive applications such as drug delivery, tissue engineering, and microfluidics.  相似文献

A synthetic toolbox for the introduction of aldehydo and hydrazido groups into the polysaccharides hyaluronic acid, alginate, dextran, pullulan, glycogen, and carboxymethyl cellulose and their use for hydrogel formation is reported. Upon mixing differently functionalized polysaccharides derived from the same natural precursor, hydrazone cross‐linking takes place, which results in formation of a hydrogel composed of one type of polysaccharide backbone. Likewise, hydrogels based on two different polysaccharide strands can be formed after mixing the corresponding aldehydo‐ and hydrazido‐modified polysaccharides. A second line of these studies paves the way to introduce a biomedically relevant ligand, namely, the adhesion factor cyclic RGD pentapeptide, by using an orthogonal click reaction. This set of modified polysaccharides served to create a library of hydrogels that differ in the combination of polysaccharide strands and the degree of cross‐linking. The different hydrogels were evaluated with respect to their rheological properties, their ability to absorb water, and their cytotoxicity towards human fibroblast cell cultures. None of the hydrogels studied were cytotoxic, and, hence, they are in principal biocompatible for applications in tissue engineering.  相似文献

However, water‐based conductive hydrogels inevitably lose elasticity and conductivity at subzero temperatures, which severely limits their applications at low temperatures. Herein we report anti‐freezing conductive organohydrogels by using an H2O/ethylene glycol binary solvent as dispersion medium. Owing to the freezing tolerance of the binary solvent, our organohydrogels exhibit stable flexibility and strain‐sensitivity in the temperature range from −55.0 to 44.6 °C. Meanwhile, the solvent molecules could form hydrogen bonds with polyvinyl alcohol (PVA) chains and induce the crystallization of PVA, greatly improving the mechanical strength of the organohydrogels. Furthermore, the non‐covalent crosslinks endow the conductive organohydrogels with intriguing remoldability and self‐healing capability, which are important for practical applications.  相似文献

Two types of chitosan hydrogel systems have been prepared that have a laser damage threshold (LDT) up to 35 times higher than commercial PMMA bulk materials. For these samples, the LDT increases with increasing water content. The mechanism of laser damage and the contribution of water to their high laser damage resistance have been examined. DSC measurements indicate water within the hydrogels exist in various states, each with different laser damage resistance properties. These various states play a key role in determining the LDT by controlling the dissipation of laser energy and providing a mechanism for self‐healing. This preliminary research shows that polymer hydrogels have potential for high power laser applications because they combine good mechanical integrity due to the polymer frame and good energy dissipation and healing characteristics due to the molecular mobility of water. These two traits allow for bulk shape‐retaining films with high laser damage thresholds and potential reversibility in damage processes. © 1999 John Wiley & Sons, Inc. Self‐sorting, simultaneous, and orthogonal operations during the self‐assembly of complex mixtures are commonly observed for biological species but rare in artificial systems. In this study, we designed two gelators (LPF and LPFEG) containing the same chiral phenylalanine core but different achiral peripheral substituents to give hydrogels with opposite supramolecular handedness. When the two hydrogels were mixed, double‐network nanofibers with opposite handedness were formed by spontaneous high‐order organization and self‐sorting of the two gelators. The chiroptical activity of the double‐network hydrogels could be tuned by varying the molar ratio of LPF and LPFEG in the mixture, thus showing that the two gelators were highly independent of each other. Enhanced mechanical properties were observed for the interpenetrating networks when the LPF/LPFEG molar ratio was 3:7, with a more than fourfold increase in both the storage (G′) and loss modulus (G′′) relative to those of the individual hydrogels.  相似文献

DNA hydrogels hold great potential for biological and biomedical applications owing to their programmable nature and macroscopic sizes. However, most previous studies involve spontaneous and homogenous gelation procedures in solution, which often lack precise control. A clamped hybridization chain reaction (C‐HCR)‐based strategy has been developed to guide DNA self‐assembly to form macroscopic hydrogels. Analogous to catalysts in chemical synthesis or seeds in crystal growth, we introduced DNA initiators to induce the gelation process, including crosslinked self‐assembly and clamped hybridization in three dimensions with spatial and temporal control. The formed hydrogels show superior mechanical properties. The use of printed, surface‐confined DNA initiators was also demonstrated for fabricating 2D hydrogel patterns without relying on external confinements. This simple method can be used to construct DNA hydrogels with defined geometry, composition, and order for various bioapplications.  相似文献

Supramolecular hydrogels are a class of self‐assembled network structures formed via non‐covalent interactions of the hydrogelators. These hydrogels capable of responding to external stimuli are considered to be smart materials due to their ability to undergo sol–gel and/or gel–sol transition upon subtle changes in their surroundings. Such stimuli‐responsive hydrogels are intriguing biomaterials with applications in tissue engineering, delivery of cells and drugs, modulating tissue environment to promote innate tissue repair, and imaging for medical diagnostics among others. This review summarizes the recent developments in stimuli‐responsive supramolecular hydrogels and their potential applications in regenerative medicine. Specifically, various structural aspects of supramolecular hydrogelators involved in self‐assembly, the role of external stimuli in tuning/controlling their phase transitions, and how these functions could be harnessed to advance applications in regenerative medicine are focused on. Finally, the key challenges and future prospects for these versatile materials are briefly described.  相似文献

Injectable hydrogels have been commonly used as drug‐delivery vehicles and tried in tissue engineering. Injectable self‐healing hydrogels have great advantage over traditional injectable hydrogels because they can be injected as a liquid and then rapidly form bulk gels in situ at the target site under physiological conditions. This study develops an injectable thermosensitive self‐healing hydrogel based on chain‐extended F127 (PEO90‐PPO65‐PEO90) multi‐block copolymer (m‐F127). The rapid sol–gel transition ability under body temperature allows it to be used as injectable hydrogel and the self‐healing property allows it to withstand repeated deformation and quickly recover its mechanical properties and structure through the dynamic covalent bonds. It is hoped that the novel strategy and the fascinating properties of the hydrogel as presented here will provide new opportunities with regard to the design and practical application of injectable self‐healing hydrogels.

Recently, supramolecular hydrogels have attracted increasing interest owing to their tunable stability and inherent biocompatibility. However, only few studies have been reported in the literature on self‐healing supramolecular nucleoside hydrogels, compared to self‐healing polymer hydrogels. In this work, we successfully developed a self‐healing supramolecular nucleoside hydrogel obtained by simply mixing equimolar amounts of guanosine (G) and isoguanosine (isoG) in the presence of K+. The gelation properties have been studied systematically by comparing different alkali metal ions as well as mixtures with different ratios of G and isoG. To this end, rheological and phase diagram experiments demonstrated that the co‐gel not only possessed good self‐healing properties and short recovery time (only 20 seconds) but also could be formed at very low concentrations of K+. Furthermore, nuclear magnetic resonance (NMR), powder X‐ray diffraction (PXRD), and circular dichroism (CD) spectroscopy suggested that possible G2isoG2‐quartet structures occurred in this self‐healing supramolecular nucleoside hydrogel. This co‐gel, to some extent, addressed the problem of isoguanosine gels for the applications in vivo, which showed the potential to be a new type of drug delivery system for biomedical applications in the future.  相似文献

Natural and synthetic gel‐like materials have featured heavily in the development of biomaterials for wound healing and other tissue‐engineering purposes. More recently, molecular gels have been designed and tailored for the same purpose. When mixed with, or conjugated to therapeutic drugs or bioactive molecules, these materials hold great promise for treating/curing life‐threatening and degenerative diseases, such as cancer, osteoarthritis, and neural injuries. This focus review explores the latest advances in this field and concentrates on self‐assembled gels formed under aqueous conditions (i.e., self‐assembled hydrogels), and critically compares their performance within different biomedical applications, including three‐dimensional cell‐culture studies, drug delivery, and tissue engineering. Although stability and toxicity issues still need to be addressed in more detail, it is clear from the work reviewed here that self‐assembled gels have a bright future as novel biomaterials.  相似文献

In this study, biodegradable and antibacterial poly(azomethine‐urethane) (PAMU)‐ and chitosan (CS)‐based hydrogels have been prepared for controlled drug delivery applications. Structural and morphological characterizations of the hydrogels were performed via Fourier transform‐infrared and scanning electron microscopy analyses. Thermal stability, hydrophilicity, swelling, mechanical, biodegradation, protein absorption properties, and drug delivery application of PAMU‐ and CS‐based hydrogels were also investigated. The swelling performance of the hydrogels was studied in acidic, neutral, and alkaline media. Swelling results showed that the hydrogels have higher swelling capacity in acidic and alkaline media than neutral medium. Biodegradation experiments of the hydrogels were also studied via hydrolytic and enzymatic experiments. The drug release property of the hydrogel was carried out using 5‐fluoro uracil (5‐FU), and 5‐FU release capacity of the hydrogels was found in the range from 40.10% to 58.40% after 3 days.  相似文献

Hyaluronic acid (HA), a naturally occurring linear polysaccharide, has been widely used as a key biomaterial in a range of cosmetic and therapeutic applications. Its excellent biocompatibility and bio‐functions related to tissue regeneration encourage the development of HA‐based hydrogels to expand its applications. This study details an in situ forming surgical glue based on photocrosslinkable HA, providing tunable mechanical properties and firm tissue adhesion under wet and dynamic conditions. Depending on the degree of photocrosslinkable methacrylate groups in HA polymer chains, the mechanical properties of hyaluronate methacrylate (HAMA) hydrogels prepared by UV photocrosslinking was improved. Ex vivo adhesion tests revealed that HAMA hydrogels exhibited 3‐fold higher shear adhesive strength compared to gelatin methacryloyl hydrogels and achieved firm adherence to the porcine skin tissue for several weeks. The high adhesive strength of HAMA hydrogels, under dry and wet conditions, suggests that it may have great promise as a tissue adhesive. © 2018 Wiley Periodicals, Inc. J. Polym. Sci., Part A: Polym. An electrochromic system based on a self‐assembled dipeptide‐appended redox‐active quinquethiophene π‐gel is reported. The designed peptide‐quinquethiophene consists of a symmetric bolaamphiphile that has two segments: a redox‐active π‐conjugated quinquethiophene core for electrochromism, and peptide motif for the involvement of molecular self‐assembly. Investigations reveal that self‐assembly and electrochromic properties of the π‐gel are strongly dependent on the relative orientation of peptidic and quinquethiophene scaffolds in the self‐assembly system. The colors of the π‐gel film are very stable with fast and controlled switching speed at room temperature.  相似文献

Polymer hydrogels and small‐molecule‐based (SMB) supramolecular hydrogels have been widely explored. But oligomeric hydrogels have remained a challenge because synthetic difficulties of the oligomers and control of their amphiphilicities. Reported herein is the rational design of two precursors Cys(SEt)‐Lys‐CBT ( 1 ) and (Cys‐Lys‐CBT)2 ( 2 ) (CBT=2‐cyano‐6‐aminobenzothiazole) and the use of a biocompatible condensation to prepare oligomeric hydrogels. Glutathione reduction of 1 or 2 yields the same gelator Cys‐Lys‐CBT ( 3 ) which condenses with each other to yield amphiphilic cyclic oligomers. The oligomers instantly self‐assemble into nanofibers and form oligomeric hydrogels with similar mechanic properties. Chemical analyses indicated that the major condensation product in both two hydrogels is a cyclic dimer. Considering its biocompatibility, optimal mechanical strength, and biodegradability, we believe that our oligomeric hydrogel might be useful for long‐term drug delivery in the future.  相似文献

Tough hydrogels have great potentials in soft robotics, artificial muscles, tissue replacement, and so on. Here we introduce novel tough hydrogels crosslinked by triblock copolymer (F127DA) micelles and metal coordination. The gels showed outstanding tensile strength (∼1–11 MPa), toughness (∼4–32 MJ m−3), and excellent self‐recovery properties (∼56.8–87.2% toughness recovery in 9 min at room temperature). The mechanical and self‐recovery properties could be manipulated by varying contents of micelles and/or COO groups. Dynamic mechanical analysis of the hydrogels revealed apparent activation energy and relaxations for both physical interactions. In situ small‐angle X‐ray scattering measurements on hydrogels upon stretching revealed micelle deformations. XPS measurements on hydrogels before and after stretching revealed significant changes in the binding energy of Fe3+ ions in the gels, suggesting the rupture of coordination bonds. The experimental results strongly suggest a synergistic effect from the micelle‐crosslinking and Fe3+–COO coordination on the strength, toughness, and self‐recovery of the hydrogels. © 2018 Wiley Periodicals, Inc. Keratin is widely recognized as a high‐quality renewable protein resource for biomedical applications. Despite their extensive existence, keratin resources such as feathers, wool, and hair exhibit high stability and mechanical properties because of their high disulfide bond content. Consequently, keratin extraction is challenging and its application is greatly hindered. In this work, a biological extraction strategy is proposed for the preparation of bioactive keratin and the fabrication of self‐assembled keratin hydrogels (KHs). Based on moderate and controlled hydrolysis by keratinase, keratin with a high molecular weight of approximately 45 and 28 kDa that retain its intrinsic bioactivities is obtained. The keratin products show excellent ability to promote cell growth and migration and are conferred with significant antioxidant ability because of their intrinsically high cysteine content. In addition, without the presence of any cross‐linking agent, the extracted keratin can self‐assemble into injectable hydrogels. The KHs exhibit a porous network structure and 3D culture ability, showing potential in promoting wound healing. This enzyme‐driven keratin extraction strategy opens up a new approach for the preparation of keratin that can self‐assemble into injectable hydrogels for biomedical engineering.  相似文献
