The hydrolysis of the sulfonylurea herbicide ethametsulfuron-methyl [methyl 2-[[[[(4-ethoxy-6-methylamino-1,3,5-triazin-2-yl)amino]carbonyl]amino]sulfonyl]benzoate] was studied in aqueous buffers of different pH values. The reaction was first-order and pH-dependent. Ethametsulfuron-methyl was more persistent in neutral or weakly basic than in acidic solution. Eleven degradation products were detected and tentatively identified by LC/MS/MS analysis. At all pH values studied, the primary pathway of degradation was the cleavage of the sulfonylurea bridge. However, minor degradation pathways have also been observed, such as O-de-ethylation, N-demethylation, and opening of the triazine ring.  相似文献

The alterations produced by microbiological attack on terpenoid resin-based varnishes from panel and canvas paintings have been evaluated using pyrolysis–gas chromatography–mass spectrometry (Py–GC–MS) and gas chromatography–mass spectrometry (GC–MS). The proposed methods include the on-line derivatisation of drying oils and diterpenoid resins using hexamethyldisilazane during pyrolysis and the application of methyl chloroformate as a derivatisation reagent for triterpenoid resins in GC–MS. Two types of specimens, consisting of model oil medium prepared from linseed oil and model spirit varnishes prepared from colophony and mastic resins dissolved in turpentine, have been used as reference materials. For a series of specimens upon which different genera of bacteria and fungi were inoculated and encouraged to grow, analyses indicated that no mechanisms that commonly occur during the attack of enzymes on drying oils and terpenoid biodegraders were observed to occur in the oil medium and varnishes studied. Thus, the degradation pathways observed in the performed trials usually occur as consequence of natural ageing. Specific trials consisting of the application of biocides to uninoculated colophony varnish resulted in the identification of processes that produce undesirable degradation of the varnish due to interactions between the biocide and the varnish components. Finally, the studied biocides—Biotin, New-Des and Nipagine—generally exhibited good inhibiting effects on the microorganisms studied, although some interesting differences were found between them regarding the application method and type of biocide.  相似文献

A multiresidue method using gas chromatography coupled to ion trap tandem mass spectrometry (GC–ITD–MS/MS) associated with solid phase microextraction (SPME) was developed for the analysis of 20 pesticides commonly used in the Alsace region in rainwater samples. Since the pesticides were expected to be present at very low concentrations and in complex matrices, the analytical method used was both highly selective and sensitive. Therefore, fibers coated with polyacrylate (PA), polydimethylsiloxane (PDMS) and polydimethylsiloxane-divinylbenzene (PDMS-DVB) were tested, and the parameters affecting the precision and accuracy of the SPME method were investigated and optimized. These parameters include the type of fiber, the adsorption time, the effect of salt, and the extraction temperature. The PDMS fiber was the most polyvalent for the extractions of the different pesticides studied. Detection limits of between 5 and 500 ng L−1, depending on the compounds under study (except for those which could not be analyzed: captan and mevinphos), were obtained with this analytical procedure. This method was applied to the analysis of rainwater samples collected simultaneously on a weekly basis at one rural and one urban site between March 2002 and July 2003. While some of the 20 pesticides analyzed were constantly detected (such as lindane and atrazine), a strong temporal variability was observed for some of the others (including alachlor, metolachlor, atrazine).  相似文献

Penindolone (PND) is a novel influenza A virus dual inhibitor that blocks hemagglutinin-mediated adsorption and membrane fusion. A sensitive and specific ultra-performance liquid chromatography–tandem mass spectrometry method was developed and validated to determine PND in rat plasma. Plasma sample preparation was a simple deproteinization with acetonitrile followed by centrifugation. Chromatographic separation was performed on a C18 column with a gradient mobile phase of acetonitrile–water containing 0.1% formic acid. Detection was carried out by electrospray ionization in negative ion multiple reaction monitoring mode. Linear detection responses were obtained for PND ranging from 1 to 1,000 ng/ml. The intra- and inter-day precision (relative standard deviations, RSD) were within 6.5%, and accuracy (relative error, RE) was within ±11.0%. The extraction recovery data for PND and internal standard (IS) were >96.0%. PND was proved to be stable during the sample storage, preparation and analytic procedures. The validated method was successfully applied to pharmacokinetic and bioavailability studies for PND in rats. The results showed the existence of twin peaks, gender difference and nonlinear pharmacokinetics for PND. In addition, two oxidation metabolites and three glucuronidation metabolites of PND were detected by ultra-high-performance liquid chromatography–high resolution mass spectrometry.  相似文献

Diosmin is a flavonoid often administered in the treatment of chronic venous insufficiency, hemorrhoids, and related affections. Diosmin is rapidly hydrolized in the intestine to its aglicone, diosmetin, which is further metabolized to conjugates. In this study, the development and validations of three new methods for the determination of diosmetin, free and after enzymatic deconjugation, and of its potential glucuronide metabolites, diosmetin-3-O-glucuronide, diosmetin-7-O-glucuronide, and diosmetin-3,7-O-glucuronide from human plasma and urine are presented. First, the quantification of diosmetin, free and after deconjugation, was carried out by high-performance liquid chromatography coupled with tandem mass spectrometry, on an Ascentis RP-Amide column (150?×?2.1 mm, 5 μm), in reversed-phase conditions, after enzymatic digestion. Then glucuronide metabolites from plasma were separated by micro-liquid chromatography coupled with tandem mass spectrometry on a HALO C18 (50?×?0.3 mm, 2.7 μm, 90 Å) column, after solid-phase extraction. Finally, glucuronides from urine were measured using a Discovery HSF5 (100?×?2.1 mm, 5 μm) column, after simple dilution with mobile phase. The methods were validated by assessing linearity, accuracy, precision, low limit of quantification, selectivity, extraction recovery, stability, and matrix effects; results in agreement with regulatory (Food and Drug Administration and European Medicines Agency) guidelines acceptance criteria were obtained in all cases. The methods were applied to a pharmacokinetic study with diosmin (450 mg orally administered tablets). The mean C max of diosmetin in plasma was 6,049.3?±?5,548.6 pg/mL. A very good correlation between measured diosmetin and glucuronide metabolites concentrations was obtained. Diosmetin-3-O-glucuronide was identified as a major circulating metabolite of diosmetin in plasma and in urine, and this finding was confirmed by supplementary experiments with differential ion-mobility mass spectrometry.  相似文献

A rapid and sensitive method for the speciation and quantification of glucosinolates in rapeseed is described. The method combines liquid chromatography (LC) with ion trap mass spectrometry (ITMS) detection. Electrospray ionization (ESI) has been chosen as the ionization technique for the on-line coupling of LC with ITMS. Glucosinolates are extracted from different rapeseeds with MeOH and the extracts are cleaned-up by solid phase extraction with Florisil cartridges. Aqueous extracts are injected into LC system coupled to an ITMS, leading to accurately quantify eight of the most important glucosinolates in rapeseed, by MS2 mode and confirming their structure by MS3 acquisition. All the glucosinolates found in rapeseeds provide good signals corresponding to the deprotonated precursor ion [M-H]. The method is reliable and reproducible, and detection limits range from 0.5 nmol g−1 to 3.7 nmol g−1 when 200 mg of dried seeds of certified reference material are analyzed. Within-day and between-day RSD percentages range between 2.4–14.1% and 3.9–16.9%, respectively. The LC-ESI-ITMS-MS method described here allows for a rapid assessment of these metabolites in rapeseed without a desulfatation step. The overall process has been successfully applied to identify and quantify glucosinolates in rapeseed samples.  相似文献

This study used reversed-phase liquid chromatography–tandem mass spectrometry and supercritical fluid chromatography–tandem mass spectrometry for determination of the stereoisomers of chlorfenvinphos and dimethylvinphos in tobacco. Tobacco samples were extracted and purified with a modified quick, easy, cheap, effective, rugged, and safe technique using spherical carbon. The performance of both methodologies was comprehensively compared in terms of methods validation parameters (separation efficiency, linearity, selectivity, recovery, repeatability, sensitivity, matrix effect, etc.). Under optimized conditions, the calibration curves of the stereoisomers of chlorfenvinphos and dimethylvinphos in the range of 10–500 ng/mL showed excellent linearity with R2 ≥ 0.997 in both methods. The adequate recoveries of analytes from three different spiked tobaccos were obtained using reversed-phase liquid chromatography–tandem mass spectrometry (86.1–95.7%) as well as supercritical fluid chromatography–tandem mass spectrometry (86.5–94.0%). The relative standard deviations for spiked samples were all below 7.0%. Compared with supercritical fluid chromatography–tandem mass spectrometry, lower matrix effects and LODs can be obtained in reversed-phase liquid chromatography–tandem mass spectrometry.  相似文献

Several sport nutrition products and doping drugs sold in the period from 2014 to 2016 were studied using gas and liquid chromatography coupled with mass-spectrometry. In the study, WADA-banned substances were detected in the composition of pre-workout supplements, fat burners, and prohormones. A series of selective androgen receptor modulators and peptide doping drugs were also studied. It was shown that, in some cases, preparations can be adulterated.  相似文献

Over the past years, ion mobility spectrometry (IMS) as a well established method within the fields of military and security has gained more and more interest for biological and medical applications. This highly sensitive and rapid separation technique was crucially enhanced by a multi-capillary column (MCC), pre-separation for complex samples. In order to unambiguously identify compounds in a complex sample, like breath, by IMS, a reference database is mandatory. To obtain a first set of reference data, 16 selected volatile organic substances were examined by MCC-IMS and comparatively analyzed by the standard technique for breath research, thermal desorption–gas chromatography–mass spectrometry. Experimentally determined MCC and GC retention times of these 16 compounds were aligned and their relation was expressed in a mathematical function. Using this function, a prognosis of the GC retention time can be given very precisely according to a recorded MCC retention time and vice versa. Thus, unknown MCC-IMS peaks from biological samples can be assigned—after alignment via the estimated GC retention time—to analytes identified by GC/MS from equivalent accomplished data. One example of applying the peak assignment strategy to a real breath sample is shown in detail.  相似文献

Single-drop microextraction (SDME) followed by gas chromatography–mass spectrometry detection was used for the determination of some carbamate pesticides in water samples. The studied pesticides were thiofanox, carbofuran, pirimicarb, methiocarb, carbaryl, propoxur, desmedipham and phenmedipham. Two alternative sample introduction methods have been examined and compared; SDME followed by cool on-column injection (without derivatization) and SDME followed by in-microvial derivatization and splitless injection. Acetic anhydride was used as derivatization reagent. Parameters that affect the derivatization reaction yield and the extraction efficiency of the SDME method were studied and optimized. The analytical performances and possible applications of both approaches were investigated. Relative standard deviations for the studied compounds ranged from 3.2 to 8.3%. The detection limits obtained by the derivatization method were found to be in the range 3–35 ng/L. Using cool on-column injection (without derivatization), the detection limits were between 30 and 80 ng/L.  相似文献

In recent years, a new class of designer drugs has appeared on the drugs of abuse market in many countries, namely, the so-called beta-keto (bk) designer drugs such as mephedrone (bk-4-methylmethamphetamine), butylone (bk-MBDB), and methylone (bk-MDMA). The aim of the present study was to identify the metabolites of mephedrone in rat and human urine using GC-MS techniques and to include mephedrone, butylone, and methylone within the authors’ systematic toxicological analysis (STA) procedure. Six phase I metabolites of mephedrone were detected in rat urine and seven in human urine suggesting the following metabolic steps: N-demethylation to the primary amine, reduction of the keto moiety to the respective alcohol, and oxidation of the tolyl moiety to the corresponding alcohols and carboxylic acid. The STA procedure allowed the detection of mephedrone, butylone, methylone, and their metabolites in urine of rats treated with doses corresponding to those reported for abuse of amphetamines. Besides macro-based data evaluation, an automated evaluation using the automated mass spectral deconvolution and identification system was performed. Mephedrone and butylone could be detected also in human urine samples submitted for drug testing. Assuming similar kinetics in humans, the described STA procedure should be suitable for proof of an intake of the bk-designer drugs in human urine.  相似文献

The present study describes a new environmentally friendly sample pretreatment system based on solid-phase microextraction (SPME) for the sensitive determination of polyphenols. A derivatization process was necessary to convert the polar non-volatile compounds into volatile derivatives. Direct immersion (DI) SPME was used for the adsorption of polyphenols, and then the fiber was placed in the headspace of the derivatizing reagent, bis(trimethylsilyl)trifluoroacetamide (BSTFA). The separation was carried out by coupling gas chromatography with mass spectrometry in the selected ion monitoring mode, after silylation. Optimal extraction conditions were 25 °C for 10 min under continuous stirring using DI and a polyacrylate fiber. After extraction, the fiber was inserted into the headspace of BSTFA (10 μL) and the polyphenols were derivatized for 15 min at 50 °C. Desorption was carried out at 280 °C for 5 min. The method allowed the determination of both isomers cis- and trans-resveratrol, piceatannol, catechin and epicatechin in wine and grapes, and it was validated for linearity, detection and quantitation limits, selectivity, accuracy and precision. Detection limits ranged from 0.05 to 0.9 ng mL−1 at a signal-to-noise ratio of 3, depending on the compound. Recoveries obtained for spiked samples were satisfactory for all compounds.  相似文献

Over the last two decades, coupled capillary electrophoresis (CE)–mass spectrometry (MS) has developed into a generally accepted technique with a wide applicability. A growing number of CE-MS applications make use of capillaries where the internal wall is modified with surface coating agents. In CE-MS, capillary coatings are used to prevent analyte adsorption and to provide appropriate conditions for CE-MS interfacing. This paper gives an overview of the various capillary coating strategies used in CE-MS. The main attention is devoted to the way coatings can contribute to a proper CE-MS operation. The foremost capillary coating methods are discussed with emphasis on their compatibility with MS detection. The role of capillary coatings in the control of the electroosmotic flow and the consequences for CE-MS coupling are treated. Subsequently, an overview of reported applications of CE-MS employing different coating principles is presented. Selected examples are given to illustrate the usefulness of the coatings and the overall applicability of the CE-MS systems. It is concluded that capillary coatings can enhance the performance and stability of CE-MS systems, yielding a highly valuable and reproducible analytical tool.  相似文献

A confirmatory method for the simultaneous determination of nandrolone (α and β) and trenbolone (α and β) in urine samples by liquid chromatography electrospray mass spectrometry (LC–MS-MS) was developed. After an enzymatic deconjugation, the urine was subjected to a one-step cleanup on a commercially available immunoaffinity chromatography cartridge. The analytes were detected by liquid chromatography–positive ion electrospray tandem mass spectrometry using deuterium labelled internal standards. The analytical procedure was applicable to bovine and swine urine samples. The procedure was validated as a quantitative confirmatory method according to the Commission Decision 2002/657/EC criteria. The results obtained showed that the method was suitable for statutory residues testing regarding the following performance characteristics: instrumental linearity, specificity, precision (repeatability and intra-laboratory reproducibility), recovery, decision limit (CCα), detection capability (CCβ) and ruggedness. The decision limits (CCα) obtained, were between 0.54 and 0.60 μg L−1; the recovery was above 64% for all the analytes. Repeatability was between 1.6% and 5.7% and within-laboratory reproducibility between 1.6% and 6.0% for all the steroids.  相似文献

Fuzhuan brick tea, a distinctive dark tea fermented by microorganisms, is a traditional beverage in China throughout history. Recently, it has attracted considerable attention owing to its unique quality characteristics and potential health benefits. The aim of this study was to establish a method for the quality control of Fuzhuan brick tea for stable production. Ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry was used to identify Fuzhuan brick tea, and the major components were chosen for further quantitative analysis. Subsequently, a quantification method was developed using ultra-high-performance liquid chromatography coupled with triple-quadrupole mass spectrometry, and its reliability was verified through methodological validation. Finally, a total of 30 compounds were identified, including catechins, flavonoids, alkaloids, and fatty acids. The established method was reliable for methodological validation and was applied to the quantitative analysis of Fuzhuan brick tea. This study provides a fundamental basis for the quality control and further studies on the component analysis of Fuzhuan brick tea.  相似文献

Data precision in the analysis by purge-and-trap coupled on-line to gas chromatography–mass spectrometry (P&T-GC–MS) of honey volatiles has been studied by statistical analysis. The contribution of non-random factors to dispersion of quantitative results was proven by comparing several statistical parameters (correlation coefficients, principal component analysis (PCA) eigenvalues and loadings) from both experimental and simulated data. PCA was also useful for grouping volatiles with similar dispersion behaviour; these groups being generally related to compounds with common properties or structural features. The use of area ratios improves data precision for compounds within the same group. Results from this study could be used for a better selection of internal standards in quantitative analysis of volatiles by P&T-GC–MS.  相似文献

This paper describes the characterisation of liposome-type nanoparticles (NPs) dispersed in a beverage matrix. Characterisation is based on a two-step procedure: first, liposomes are separated on the basis of size in the nanometre range by use of hydrodynamic chromatography (HDC); second, chemical characterisation is performed by use of MALDI–TOF mass spectrometry (MS). Characterisation of three types of Coatsome liposome, a commercially available type of empty liposome, was investigated. All three liposome types, Coatsome A?=?anionic, N?=?neutral, and C?=?cationic, gave single peaks in HDC, reflecting diameters of 153, 187, and 205 nm, respectively. Subsequent MALDI–TOF MS in positive mode furnished major signals at m/z?=?734.5 ([M?+?H]+ adduct) and m/z?=?756.6 ([M?+?Na]+ adduct) of l-(α)-dipalmitoylphosphatidylcholine (DPPC) monomer and dimeric adducts at m/z?=?1468.1 and m/z?=?1490.1, respectively. MALDI–TOF MS in negative mode gave a signal at m/z?=?721.3 ([M???H]? adduct) of l-(α)-dipalmitoylphosphatidylglycerol (DPPG), except for Coatsome C which lacks this phospholipid. After HDC separation of Coatsome A NPs the major DPPC and DPPG signals can be detected in the expected fractions by use of MALDI–TOF MS in positive and negative modes, respectively. Validation of the analytical strategy revealed linearity (R 2?>?0.99), repeatability (relative standard deviation <10 %), and reproducibility (relative standard deviation between days <10 %) were good, recovery was 61?±?5 %, and the limit of quantification was 1 mg?mL?1 in this matrix. With 4 mg Coatsome A mL?1 20 out of 20 samples furnished the 734.5 and 756.6 signals typical of DPPC in MALDI–TOF MS characterisation.  相似文献

A novel method based on the coupling of membrane-supported headspace single-drop microextraction with gas chromatography?mass spectrometry (GC–MS) is developed for the determination of chlorobenzenes in water samples. For the determination of five chlorobenzenes, a 15 μL toluene microdrop was placed inside the plastic membrane and exposed for 10 min for headspace extraction while stirring at 1000 rpm. The microdrop was then picked up by a microsyringe and directly injected into the injector block of the GC–MS instrument. Under the optimized operation conditions, the calculated calibration curves gave a high level of linearity for all targets with correlation coefficients range from 0.9945 to 0.9987. The limits of detection range from 0.01 to 0.05 μg/L and the RSDs for most of chlorobenzenes were below 7%. The method is simple, sensitive, and stable for single drop microextraction. Its applicability is demonstrated by the determination of chlorobenzenes in tap water samples.  相似文献
