A sensitive liquid chromatography-electrospray tandem mass spectrometry method combined with solid-phase extraction and silica cartridge cleanup was established for 16 sulfonamides and trimethoprim in various water matrices. Signal suppression of all target analytes in sewage treatment plant influent, effluent and river water was improved by this method developed in this study. The method detection limits for 17 analytes were 20-200 pg/L for influent, 16-120 pg/L for effluent and 8.0-60 pg/L for river water with overall mean recoveries of 62-102% in all studied matrices. This method was used to analyze residual sulfonamides and trimethoprim in wastewater and river samples from Japan, and 8 analytes (0.08 (sulfadimethoxine)-161 ng/L (sulfapyridine) in wastewater and 10 (0.03 (sulfamethizol)-8.9 ng/L (sulfaquinoxaline) in river samples were detected.  相似文献

A sensitive and reliable method using capillary zone electrophoresis with UV-diode array detection (CZE-DAD) has been developed and validated for trace determination of beta-lactam antibiotics in waste, well and river water matrices. Due to the lack of sensitivity of the UV-vis detection, a solvent extraction/solid-phase extraction (SPE) method applied for off-line preconcentration and cleanup of water samples, in combination with an on-line preconcentration methodology named large volume sample stacking (LVSS) have been applied. The analytes included nafcillin, dicloxacillin, cloxacillin, oxacillin, ampicillin, penicillin G and amoxicillin. Average recoveries for water samples fortified with the studied beta-lactams at different concentration levels (1.0, 2.0 and 4.0 microg/L) were ranging between 94 and 99%, with relative standard deviations (RSDs) lower than 10%. The precision, calculated as intra-day and inter-day standard deviations fell within acceptable ranges (3.3-7.2%). The limits of detection were estimated to range between 0.08 and 0.80 microgL(-1) for the studied compounds. All the samples analyzed were negative for all the analytes at these levels of concentration and the method showed its usefulness for the detection of these widely applied beta-lactam antibiotics in different kinds of waters.  相似文献

In this study, a highly sensitive and robust method using an ultra-high-performance liquid chromatography-tandem mass spectrometry combined with solid-phase extraction and ultrasonic extraction for pretreatment and silica gel purification steps has been developed for determination of 21 natural and synthetic progestagens in river surface water and sediments, and influents, effluents, and sludge from municipal wastewater treatment plants, and flush water and feces from swine farms. For the various matrices considered, the optimized method showed satisfactory performance with recoveries of 70–129 % (except AD, 5α-DHP, DPT, HPC), the limits of quantification below 2.30 ng/L for liquid samples and 2.59 ng/g for solid samples (except AD), and good linearity and reproducibility. This developed method was successfully applied in the analysis of progestagens in environmental samples from Liuxi Reservoir, Xintang municipal wastewater treatment plant, and Shunfeng swine farm in South China. Six analytes were detected at trace levels in surface water, effluent, and sediment samples. Seven analytes (0.7 (HPA)–35.1 ng/L (DGT)) were found in the influent samples and three analytes (5.6 (DGT)–11.8 ng/g (5α-DHP)) in the dewatered sludge samples. Moreover, 13 analytes were detected in swine farm, with high concentrations ranging from 23.8 ng/L (ET) to 5,024 ng/L (P) in flush water, and from 20.0 ng/g (MPA) to 1952 ng/g (P) in feces.  相似文献

A sensitive ultra-performance liquid chromatography-electrospray tandem mass spectrometry method, combined with solid-phase extraction and silica cartridge cleanup, was established for nine androgens (androstenedione, 19-nor-4-androstene-3,17-diol, androsterone, epiandrosterone, testosterone, methyltestosterone, trenbolone, nandrolone, stanozolol) and nine progestogens (progesterone, 17alpha-hydroxyprogesterone, 21alpha-hydroxyprogesterone, 6alpha-methyl-hydroxyprogesterone, 17alpha,20beta-dihydroxy-4-pregnene-3-one, megestrol acetate, norethindrone, norgestrel, medroxyprogesterone acetate) in environmental waters. For the various water matrices considered, the overall method recoveries were from 78 to 100%, and no apparent signal suppression was found. The method detection limits for the eighteen analytes in the influent, effluent and surface water samples were 0.20-50, 0.04-20 and 0.01-12 ng/L, respectively. This method was used to analyze the residual androgens and progestogens in the wastewater and surface water samples from Japan, and ten analytes (0.03 (medroxyprogesterone acetate)-1441 ng/L (androsterone)) were detected in the wastewater samples, and four analytes (0.06 (progesterone)-0.46 ng/L (androstenedione)) were detected in the surface water samples.  相似文献

A method was developed to determine 11 pharmaceutical compounds in water samples. The method uses SPE and HPLC coupled to MS (LC/MS) using ESI in both positive and negative modes. Three different sorbents were compared for the extraction of analytes from river and sewage treatment plant (STP) waters and OASIS HLB provided the best results. For the solid-phase extraction of 500 mL of river water samples, the recoveries were between 41 and 101% with the exception of acetaminophen, salicylic acid and naproxen. The LODs were between 3 and 5 ng/L for all the compounds, except naproxen which had an LOD of 15 ng/L. Acetaminophen, caffeine, carbamazepine, bezafibrate and ibuprofen were found in three of the tested river samples at ng/L levels and among them, the highest values were for caffeine and bezafibrate with 305 and 363 ng/L, respectively. For the influent and effluent water samples of the STP, volumes of 100 and 250 mL were used, respectively, to obtain acceptable recoveries. All the compounds showed recoveries between 33 and 91% for effluent samples and 33-72% for influent samples, with the exception of acetaminophen, salicylic acid and bezafibrate, which had lower recoveries. The method developed enabled pharmaceuticals in the influent and effluent sewage waters to be determined in five campaigns carried out between February 2004 and June 2005. Several pharmaceuticals were found in the influent samples: for instance, maximum concentrations of ibuprofen and caffeine were 6 and 40 microg/L, respectively.  相似文献

Pharmaceuticals constitute one of the most important emerging classes of environmental pollutants. A three‐phase solvent system of water, water containing 0.1% of formic acid and acetonitrile was successfully used to separate, by liquid chromatography with mass spectrometry (LC‐MS), polarity‐matched pharmaceuticals, that is, carbamazepine, clarithromycin, and erythromycin, as well as amoxicillin and metformin. Despite of polarity similarities, these pharmaceuticals were completely resolved in the analytical run time of 15 min. The optimized three‐phase solvent system based‐method was validated for the simultaneous analysis of six matched‐polarity pharmaceuticals in wastewater samples. Good linearity (coefficient of determination more than 0.993) and precision (relative standard deviation less than 15.66%) were achieved. Recovery of analytes from the wastewater was between 0.70 and 1.18. Limits of detections ranged from 0.0001 to 0.5114 µg/L. No significant matrix effect, evaluated by post extraction addition, was observed in the electrospray ionization (ESI) source. Then, this methodology has been successfully applied to environmental study of pharmaceutical residues occurring in influent and effluent wastewater samples, from the main wastewater treatment plant in Potenza (Basilicata, Southern Italy).  相似文献

建立了一种同时测定制药废水中3类10种抗生素的超高效液相色谱-串联质谱分析方法。水样用固相萃取柱富集净化,通过比较在不同的固相萃取柱和洗脱液等条件下水样中目标物的回收率,优化了前处理方法。采用Agilent C18色谱柱(75 mm×2.1 mm, 2.7 μm),以0.2%(v/v)甲酸水溶液和乙腈为梯度洗脱的流动相,在电喷雾-多反应监测模式下进行定性定量分析。实验结果表明:在0.1~1000 μg/L范围内,6种氨基糖苷类抗生素、螺旋霉素及3种氟喹诺酮类抗生素的峰面积与质量浓度的线性关系良好(r2 > 0.995),方法检出限为0.07~4.37 ng/L,定量限为0.22~14.55 ng/L;目标抗生素的加标水平为0.002~40 μg/L时,平均回收率为50.4%~114.1%,相对标准偏差均不高于9.89%(n=3)。基于上述方法,对江苏省某制药厂废水中相关物质进行检测,在各废水处理单元中检出3种目标抗生素,质量浓度范围为0.46~1033.60 μg/L。该方法准确可靠、灵敏度高,适用于制药厂废水中氨基糖苷类抗生素、螺旋霉素和氟喹诺酮类抗生素的检测。  相似文献

A multi-class method for the simultaneous quantification and confirmation of 47 pharmaceuticals in environmental and wastewater samples has been developed. The target list of analytes included analgesic and anti-inflammatories, cholesterol lowering statin drugs and lipid regulators, antidepressants, anti-ulcer agents, psychiatric drugs, ansiolitics, cardiovasculars and a high number (26) of antibiotics from different chemical groups. A common pre-concentration step based on solid-phase extraction with Oasis HLB cartridges was applied, followed by ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) measurement. All compounds were satisfactorily determined in just one single injection, with a chromatographic run time of only 10 min. The process efficiency (combination of the matrix effect and the extraction process recovery) for the 47 selected compounds was evaluated in nine effluent wastewater (EWW) samples, and the use of different isotope-labelled internal standards (ILIS) was investigated to correct unsatisfactory values. Up to 12 ILIS were evaluated in EWW and surface water (SW). As expected, the ILIS provided satisfactory correction for their own analytes. However, the use of these ILIS for the rest of pharmaceuticals was problematic in some cases. Despite this fact, the correction with analogues ILIS was found useful for most of analytes in EWW, while was not strictly required in the SW tested. The method was successfully validated in SW and EWW at low concentration levels, as expected for pharmaceuticals in these matrices (0.025, 0.1 and 0.5 μg/L in SW; 0.1 and 0.5 μg/L in EWW). With only a few exceptions, the instrumental limits of detection varied between 0.1 and 8 pg. The limits of quantification were estimated from sample chromatograms at the lowest spiked levels tested and normally were below 20 ng/L for SW and below 50 ng/L for EWW. The developed method was applied to the analysis of around forty water samples (river waters and effluent wastewaters) from the Spanish Mediterranean region. Almost all the pharmaceuticals selected in this work were detected, mainly in effluent wastewater. In both matrices, analgesics and anti-inflammatories, lipid regulators and quinolone antibiotics were the most detected groups.  相似文献

Phytoestrogens activate a biological response in vertebrata where they can mime or modulate the action of endogenous estrogens. For this reason they have been subjected to several studies about their physiological effects on humans and many analytical methodologies for their determination in food matrices and physiological fluids have been developed. On the contrary, little information can be found in literature about the presence of isoflavones and coumestrol in the environment, even if it is known that this may have significance. In the present study we investigated the presence of nine selected free and conjugated phytoestrogens in environmental water. A liquid chromatography-mass spectrometry (LC-MS/MS) based analytical methodology was developed and employed for detection of target compounds in surface water and wastewater.The methodology uses solid-phase extraction, followed by high performance liquid chromatography coupled to tandem mass spectrometry using an electrospray (ESI) interface operating in positive ion mode (LC-ESI-MS/MS). The extraction was made with 200 mg, 6 mL OASIS HLB® cartridges. Recoveries for the selected compounds were in the 67-97% range for all the considered analytes. The method was employed for environmental monitoring. Samples of river water and wastewater collected over a 4-month period were analyzed with the developed procedure. Results showed the presence of isoflavones in most of the samples analyzed. Average concentration of target analytes found in wastewater sewage treatment plant influent ranged from 454 to 12 ng/L. In effluent water and river water the analytes were present at lower concentration.  相似文献

This study dealt with the potential of MEKC with LIF detection involving derivatization with sulfoindocyanine succinimidyl ester (Cy5) for the separation and determination of beta-lactam antibiotics (ampicillin, amoxicillin, cephradine, and cephalexin) in environmental water samples. Water samples of 50 mL were enriched by SPE by passage through a weak base-cation Amberlite(R) IRA-93 exchange column. SDS micelles play important roles in the whole analytical process by improving the yield (sensitivity) and the kinetics of the labeling reaction, the elution of the retained antibiotics from the SPE preconcentration system and the electrophoretic resolution of their Cy5-derivatives. The optimum procedure includes a derivatization step of the antibiotics at 25 degrees C for 10 min and direct injection for MEKC analysis, which is conducted within about 15 min using 15 mM SDS in the running buffer (35 mM sodium borate at pH 9.3). LODs from 30 to 45 ng/L and RSDs (within-day precision) from 3.5 to 5.9% were obtained for the antibiotics in water samples with average recoveries ranging from 96.4 to 99.4%. These results indicate that the method proposed is a straightforward and sensitive tool for the determination of these antibiotics in environmental water samples providing similar quantitative results to those using more expensive equipment like LC-electrospray MS/MS.  相似文献

A sensitive method was developed for the trace determination of six tetracyclines and ten of their degradation products in influent, effluent, and river waters using liquid chromatography–electrospray tandem mass spectrometry detection, combined with Oasis hydrophilic–lipophilic balance (HLB) cartridge extraction and Oasis mixed-mode strong anion exchange (MAX) cartridge cleanup. Tetracyclines and their products were separated by liquid chromatography in 9.5 min, and the instrument detection limits were generally between 0.03 and 0.1 μg/L except for minocycline (0.5 μg/L). The chromatograms were improved through the MAX cleanup and no apparent matrix effect was found. The recoveries of all the target compounds except for 4-epianhydrochlortetracycline and anhydrochlortetracycline (34–52%) were 75–120% for influent, 61–103% for effluent, and 64–113% for river waters. The method detection limits (MDLs) of the analytes varied in the range of 0.8–17.5 ng/L in all studied matrices. The method was applied for the determination of tetracyclines and their products in a sewage treatment plant (STP) and surface waters in Beijing, China. Oxytetracycline (3.8–72.5 ng/L), tetracycline (1.9–16.5 ng/L), and five products including 4-epitetracycline, 4-epioxytetracycline, isochlortetracycline, anhydrotetracycline, and 4-epianhydrochlortetracycline (5.7–25.3 ng/L) were detected in wastewater, while only oxytetracycline and tetracycline (2.2 and 2.1 ng/L) were detected in surface water samples.  相似文献

A new, simple and sensitive method for isolating nine compounds from the bisphenol group (analogues: A, B, C, E, F, G, Cl2, Z, AP) based on one-step liquid–liquid microextraction with in situ acylation followed by gas chromatography-mass spectrometry was developed and validated using influent and effluent wastewaters. The chemometric approach based on the Taguchi method was used to optimize the main conditions of simultaneous extraction and derivatization. The recoveries of the proposed procedure ranged from 85 to 122%, and the repeatability expressed by the coefficient of variation did not exceed 8%. The method’s limits of detection were in the range of 0.4–64 ng/L, and the method’s limits of quantification ranged from 1.3 to 194 ng/L. The developed method was used to determine the presence of the tested compounds in wastewater from a municipal wastewater treatment plant located in northeastern Poland. From this sample, eight analytes were detected. Concentrations of bisphenol A of 400 ng/L in influent and 100 ng/L in effluent were recorded, whereas other bisphenols reached 67 and 50 ng/L for influent and effluent, respectively. The removal efficiency of bisphenol analogues in the tested wastewater treatment plant ranged from 7 to approximately 88%.  相似文献

A high-performance liquid chromatography/electrospray ionization ion trap mass spectrometry (HPLC/ESI-MSn) method has been developed for the trace determination of phytoestrogens in aquatic environmental samples. The method includes solid-phase extraction (SPE) and analysis using liquid chromatography/electrospray ionization ion trap mass spectrometry. The aquatic environmental samples, influent of a wastewater treatment plant (WWTP) and creek water, were adjusted to pH approximately 5 before extraction. The analyzed phytoestrogens were identified by an MSn method and quantified against a deuterated internal standard (genistein-3',5',6,8-D4). In negative ion mode, 0.1% formic acid was employed in acetonitrile/water mobile phase. The method detection limits ranged from 0.5 to 10 ng/L in WWTP influent and from 0.1 to 5 ng/L in creek water. Average SPE recoveries for the analyzed phytoestrogens ranged from 85 to 95%, with a relative standard deviation (RSD) (%) ranging from 3.9 to 6.5. The concentrations of the six analyzed phytoestrogens varied from 0.2 to 600 ng/L with high levels of enterolignans (enterolactone and enterodiol) found in the collected wastewater. The method is shown to be suitable for the determination of phytoestrogens in aquatic environmental samples at nano- and sub-nanogram per liter levels.  相似文献

A simple and reliable method was developed to detect two basic synthetic dyes, rhodamine B (RB) and rhodamine 6G (R6G), in wastewater and surface water samples by high performance liquid chromatography with fluorescence detection (HPLC‐FLD). These dyes have been reported to be both mutagenic and carcinogenic in various organisms. The contents of these two dyes in water samples were extracted by Oasis HLB solid‐phase extraction (HLB‐SPE), and were then determined by an isocratic HPLC using an Atlantis® T3‐C18 column. Water samples at various pH conditions and the compositions of eluents for SPE were evaluated. The results indicate that the proposed method is precise and sensitive in analyzing these two basic synthetic dyes, and the limits of quantitation were 1.5 ng/L for RB and 0.3 ng/L for R6G in 100 mL of water samples. The recovery of analytes in spiked surface water and municipal wastewater treatment plant (WWTP) effluent samples ranged from 61 to 90% with the precision (RSD) ranging from 2 to 12%. The concentrations of analytes were detected in various water samples ranging from 0.7 to 81 ng/L.  相似文献

A sensitive rapid resolution liquid chromatography-tandem mass spectrometry (RRLC-MS/MS) method, combined with solid-phase extraction, ultrasonic extraction and silica gel cartridge cleanup, was developed for 28 steroids including 4 estrogens (estrone (E1), 17β-estradiol (E2), 17α-ethynyl estradiol (EE2), diethylstilbestrol (DES)), 14 androgens (androsta-1,4-diene-3,17-dione (ADD), 17α-trenbolone, 17β-trenbolone, 4-androstene-3,17-dione, 19-nortestoserone, 17β-boldenone, 17α-boldenone, testosterone (T), epi-androsterone (EADR), methyltestosterone (MT), 4-hydroxy-androst-4-ene-17-dione (4-OHA), 5α-dihydrotestosterone (5α-DHT), androsterone (ADR), stanozolol (S)), 5 progestagens (progesterone (P), ethynyl testosterone (ET), 19-norethindrone, norgestrel, medroxyprogesterone (MP)), and 5 glucocorticoids (cortisol, cortisone, prednisone, prednisolone, dexamethasone) in surface water, wastewater and sludge samples. The recoveries of surface water, influents, effluents and sludge samples were 90.6-119.0% (except 5α-DHT was 143%), 44.0-200%, 60.7-123% and 62.6-138%, respectively. The method detection limits for the 28 analytes in surface water, influents, effluents and freeze-dried sludge samples were 0.01-0.24 ng/L, 0.02-1.44 ng/L, 0.01-0.49 ng/L and 0.08-2.06 ng/g, respectively. This method was applied in the determination of the residual steroidal hormones in two surface water of Danshui River, 12 wastewater and 8 sludge samples from two wastewater treatment plants (Meihu and Huiyang WWTPs) in Guangdong (China). Ten analytes were detected in surface water samples with concentrations ranging between 0.4 ng/L (17β-boldenone) and 55.3 ng/L (5α-DHT); twenty analytes in the wastewater samples with concentrations ranging between 0.3 ng/L (P) and 621 ng/L (5α-DHT); and 12 analytes in the sludge samples with concentrations ranging between 1.6 ng/g (E1) and 372 ng/g (EADR).  相似文献

A chiral liquid chromatography-tandem mass spectrometry (HPLC-MS-MS) method was developed and validated for measuring individual enantiomers of three beta-blocker drugs (atenolol, metoprolol, and propranolol) in wastewater treatment plant (WWTP) influents and effluents. Mean recoveries of the pharmaceuticals ranged from 67 to 106%, and the limits of detection of the analytes were 2-17 ng/L in wastewater effluents. The method was demonstrated by measuring, for the first time, the stereoisomer composition of target analytes in raw and treated wastewaters of two Canadian WWTPs. In these trials, racemic amounts of the three drugs were observed in influent of one wastewater treatment plant, but nonracemic amounts were observed in another. Effluents of the two plants contained nonracemic amounts of the drugs. These results indicate that biologically-mediated stereoselective processes that differ among WWTPs had occurred to eliminate individual enantiomers of the target analytes.  相似文献

A fully automated method for determining nine Environmental Protection Agency N-nitrosamines in several types of environmental waters at ng/L levels is presented. The method is based on a headspace solid-phase microextraction followed by GC-MS-MS using chemical ionization. Three different fibers (carboxen/PDMS, divinylbenzene/carboxen/PDMS, and PEG) were tested. Solid-phase microextraction conditions were best when a divinylbenzene/carboxen/PDMS fiber was exposed for 60?min in the headspace of 10?mL water samples at pH 7 containing 360?g/L of NaCl, at 45°C. All compounds were analyzed by GC-MS-MS within 18?min. The method was validated using effluent from an urban wastewater treatment plant and the LODs ranged from 1 to 5?ng/L. The method was then applied to determine the N-nitrosamines in samples of different complexities, such as tap water and several influent and effluent wastewater samples from urban and industrial wastewater treatment plants and a potable water treatment plant. Although the analysis of influent industrial wastewater revealed high concentrations of some compounds (N-nitrosomorpholine and N-nitrosodimethylamine at μg/L levels), in industrial effluents and other samples, the concentrations were substantially lower (ng/L levels). The new method is suitable for the simple and reliable determination of N-nitrosamines in highly complex water samples in a completely automated procedure.  相似文献

This paper presents a fully automated method for determining ten primary amines in wastewater at ng/L levels. The method is based on simultaneous derivatization with pentafluorobenzaldehyde (PFBAY) and headspace solid-phase microextraction (HS-SPME) followed by gas chromatography coupled to ion trap tandem mass spectrometry (GC–IT-MS–MS). The influence of main factors on the efficiency of derivatization and of HS-SPME is described in detail and optimized by a central composite design. For all species, the highest enrichment factors were achieved using a 85 μm polyacrylate (PA) fiber exposed in the headspace of stirred water samples (750 rpm) at pH 12, containing 360 g/L of NaCl, at 40 °C for 15 min. Under optimized conditions, the proposed method achieved detection limits ranging from 10 to 100 ng/L (except for cyclohexylamine). The optimized method was then used to determine the presence of primary amines in various types of wastewater samples, such as influent and effluent wastewater from municipal and industrial wastewater treatment plants (WWTPs) and a potable water treatment plant. Although the analysis of these samples revealed the presence of up to 1500 μg/L of certain primary amines in influent industrial wastewater, the concentration of these compounds in the effluent and in municipal and potable water was substantially lower, at low μg/L levels. The new derivatization–HS-SPME–GC–IT-MS–MS method is suitable for the fast, reliable and inexpensive determination of primary amines in wastewater in an automated procedure.  相似文献

A novel and reliable analytical method based on a graphene adsorbent for solid-phase extraction (SPE) derivatized with N-tert-butyldimethylsilyl-N- methyltrifluoroacetamide and analyzed by gas chromatography–mass spectrometry was developed for determination of nine pharmaceuticals and personal care products (PPCPs) in wastewater samples. Different ratios of graphene/silica gel were tested, with 20 % graphene/silica gel giving the best performance as an SPE adsorbent. The mean recoveries of the target analytes obtained by 20 % graphene/silica gel SPE ranged from 58.1 to 87.6 %. The limit of quantification ranged from 30 to 259 ng/L and from 13 to 115 ng/L for the influent and effluent, respectively. By comparing the accuracy and precision of 20 % graphene/silica gel and Oasis HLB SPE cartridges, we demonstrated that the method can be satisfactorily used for the analysis of PPCPs in wastewater samples. We applied the method to wastewater samples from a sewage treatment plant near Riverside, California, to track the concentration change of PPCPs in the treatment processes.  相似文献
