A regioselective synthesis of novel pyrazolo[3,4‐d]pyrimidines, imidazo[1,2‐b]pyrazoles, pyrazolo[3,4‐d][1,2,3]triazine, pyrazolo[1,5‐c][1,3,5]triazine and pyrazolo[1,5‐c][1,3,5]thiadiazine incorporating a thiazole moiety was described via the reactions of the versatile, readily accessible 5‐amino‐3‐(phenylamino)‐N‐(4‐phenylthiazol‐2‐yl)‐1H‐pyrazole‐4‐carboxamide ( 1 ) with each of DMF‐DMA, phenylisothiocyanate, chloroacetyl chloride, phenacyl bromide, benzoylisothiocyanate and formalin, respectively. All structures of the newly synthesized compounds were elucidated by elemental analysis and spectral data. 相似文献

New series of (thieno[2,3‐c]pyrazolo‐5‐yl)‐[1,2,4]triazolo[3,4‐b][1,3,4]thiadiazoles 10a , 10b , 10c and (thieno[2,3‐c]pyrazol‐5‐yl)‐1,3,4‐oxadiazol‐3(2H)‐yl)ethanones 6a , 6b , 6c has been synthesized from thieno[2,3‐c]pyrazole‐5‐carbohydrazide 3 by multistep reaction sequence. (5‐Aryl‐1,3,4‐oxadiazol‐2‐yl)‐1H‐thieno[2,3‐c]pyrazoles 4a , 4b , 4c were also synthesized from thieno[2,3‐c]pyrazole‐5‐carbohydrazide 3 by cyclization with various aromatic carboxylic acids. The hydrazide 3 was obtained by reaction of thieno[2,3‐c]pyrazole‐5‐carboxylate 2 with hydrazine hydrate in good yield, and compound 2 was obtained by the reaction of 5‐chloro‐3‐methyl‐1‐phenyl‐1H‐pyrazole‐4‐carbaldehyde 1 and 2‐ethyl thioglycolate in presence of sodium alcoholate in good yield. 相似文献

3(5)‐Aminopyrazole derivative ( 6 ) has been synthesized by the reactions of the versatile unreported 2‐cyano‐N ′‐(1‐(3‐methyl‐6‐phenylimidazo[2,1‐b ]thiazol‐2‐yl)ethylidene)acetohydrazide ( 3 ) with phenyl isothiocyanate in KOH/DMF solution followed by reaction with methyl iodide and hydrazine hydrate. Reaction of compound 6 with some 1,3‐dicarbonyl compounds yielded pyrazolo[1,5‐a ]pyrimidine derivatives ( 14 – 17 ). Alkylation of compound 6 with various halo reagents, followed by intramolecular cyclization, yielded the corresponding imidazo[1,2‐b ]pyrazole derivatives 27 , 29 , 31 , and 33 . All newly synthesized compounds were elucidated by considering the data of both elemental analysis and spectral data. 相似文献

5‐Amino‐4‐formyl pyrazole carboxylate gave facile reactions with malononitrile, hydrazine, and ketones in the presence of piperidine furnished substituted pyrazolo[3,4‐b]pyridines and pyrazolo[3,4‐b]quinolones. Several new pyrazolo[3,4-d]pyrimidine, pyrazolo[3,4-e][1,4]diazepine, pyrazolo[3,4-d][1,2,3]triazine and pyrolo[4,3-e][1,2,4]triazolo[1,5-c]pyrimidine derivatives were prepared by the reaction of the corresponding 5-amino-pyrazole-4-carbonitrile derivative with different organic reagents under different reaction conditions. The versatile hitherto reported 3‐(1H‐indol‐3‐yl)‐1H‐pyrazol‐5‐amine ( 4 ) was synthesized by the reaction of 3‐cyanoacetylindole ( 3 ) with hydrazine hydrate in refluxing ethanol and used as a key intermediate for the synthesis of novel pyrazolo[1,5‐a ]pyrimidines via its reactions with appropriate 1,3‐biselectrophilic reagents or through three‐component condensations with triethyl orthoformate and compounds possessing an activated methylene group. Besides, the applicability and synthetic potency of ( 4 ) to attain polyfunctionally substituted imidazo[1,2‐b ]pyrazole, pyrazolo[1,5‐a ][1,3]diazepine and pyrazolo[1,5‐c ][1,3,5]thiadiazine derivatives of an expected pharmaceutical interest have been investigated. The mechanistic aspects for the formation of the newly synthesized compounds are discussed. 相似文献

E‐3‐(N,N‐Dimethylamino)‐1‐(3‐methylthiazolo[3,2‐a]benzimidazol‐2‐yl)prop‐2‐en‐1‐one ( 2 ) was synthesized by the reaction of 1‐(3‐methylthiazolo[3,2‐a]benzimidazol‐2‐yl)ethanone ( 1 ) with dimethylformamide‐dimethylacetal. The reaction of 2 with 5‐amino‐3‐phenyl‐1H‐pyrazole ( 4a ) or 3‐amino‐1,2,4‐(1H)‐triazole ( 4b ) furnished pyrazolo[1,5‐a]pyrimidine and 1,2,4‐triazolo[1,5‐a]pyrimidine derivatives 6a and 6b , while the reaction of enaminone 2 with 6‐aminopyrimidine derivatives 7a,b afforded pyrido[2,3‐d]pyrimidine derivatives 9a,b , respectively. The diazonium salts 11a or 11b coupled with compound 2 to yield the pyrazolo[5,1‐c]‐1,2,4‐triazine and 1,2,4‐triazolo[5,1‐c]‐1,2,4‐triazine derivatives 13a and 13b . Some of the newly synthesized compounds exhibited a moderate effect against some bacterial and fungal species. 相似文献

Selective Synthesis of New Tetracyclic Coumarin‐fused Pyrazolo[3,4‐b]pyridines and Pyrazolo[3,4‐b]pyridin‐6(7H)‐ones 下载免费PDF全文
A three‐component reaction for the synthesis of new coumarin‐fused tetracyclic system from 4‐hydroxycoumarin, aldehydes, and 5‐aminopyrazoles/5‐aminoisoxazole is described. In the presence of acetic acid, 4,7‐dihydro‐1H‐pyrazolo[3,4‐b]pyridines ( 4 ) and pyrazolo[3,4‐b]pyridines ( 5 ) were obtained in acetonitrile and dimethylsulfoxide medium, respectively. The reaction gave rise to 4,5‐dihydro‐1H‐pyrazolo[3,4‐b]pyridin‐6(7H)‐ones ( 6 ) in acetic acid–ethanol combination system, which involved the C–O bond cleavage. 4‐Hydroxy‐6‐methyl‐2H‐pyran‐2‐one and acenaphthylene‐1,2‐dione were also examined, affording the corresponding C–O bond cleavage products. Mechanism indicates that the reaction is reversible in acetic acid–ethanol combination system. 相似文献

A series of pyrano‐fused pyrazolo[3,4‐b]pyridine derivatives with an aryl group presenting the 2‐position of the pyridine nucleus have been synthesized by microwave‐assisted three‐component reactions of aldehydes, tetrahydropyran‐4‐one, and 3‐methyl‐1‐phenyl‐1H‐pyrazol‐5‐amine in HOAc. This method is very efficient because of short reaction times and easy work‐up, and it provides an efficient and promising synthetic strategy for the construction of the tricyclic pyrano‐fused pyrazolo[3,4‐b]pyridine skeleton. 相似文献

This research work describes the synthesis and biological properties of some novel isolated or fused heterocyclic ring systems with pyrazole, for example; enaminones containing pyrazolone ring photochromic functional unit, 4‐[(4‐chlorophenylamino)methylene]‐3‐methyl‐1‐phenyl‐1H‐pyrazol‐5(4H)‐one (3) and some analogous derivatives 4, 9, and 10, also as pyrazolo[3,4‐b]pyridine, pyrazolo[3,4‐b]quinoline, pyrazolo[3′,4′:4,5]thieno[2,3‐c]pyrazoline and pyrazolo[3,4‐c]pyrazole were synthesized and characterized. Newly synthesized compounds were characterized by IR, 1H NMR, 13C NMR, mass spectral data and quantum mechanical calculations. 2‐Benzyl‐ and 2‐aryloxymethyl‐3‐amino‐1‐phenyl‐pyrazolo[3,4‐d]pyrimidine‐4‐ones 5a–f have been synthesized by reacting the corresponding arylacetylamino derivatives 3a–f with hydrazine hydrate. Thionation of compounds 5d–f by action of P2S5 in pyridine yielded 2‐aryloxy‐methyl‐3‐amino‐1‐pheny‐lpyrazolo[3,4‐d]pyrimidin‐4‐thions 6a–c . 2,5‐Diphenyl‐2,3‐dihydro‐1H‐pyrazolo[5′,1′:4:5]pyrazolo[3,4‐d]pyrimidine‐8‐one ( 8 ) was also obtained via reaction of ethyl‐2‐cinnamoylamino‐1‐phenyl‐pyrazole‐4‐car‐boxylate ( 7 ) with hydrazine hydrate. The prepared compounds were screened in vitro for their antimicrobial activity. Some of the tested compounds were found to be active at 100 μg/ml compared with reference compounds (Ampicillin and Trivid) as antibacterial agents and claforan as antifungal agent. © 2003 Wiley Periodicals, Inc. A new convenient synthon for heterocyclic chemistry, namely 1H‐pyrazolo[3,4‐b]pyridin‐3‐ylguanidine was successfully prepared by selective guanylation of 1H‐pyrazolo[3,4‐b]pyridin‐3‐amine. A series of 3,4‐dihydropyrido[2′,3′:3,4]pyrazolo[1,5‐a][1,3,5]triazin‐2‐amines was synthesized from 1H‐pyrazolo[3,4‐b]pyridin‐3‐ylguanidine using aldehydes or ketones as one‐carbon inserting reagents. The tautomeric preferences of the products were determined using spectroscopic (e.g., 2D NOESY NMR) and single crystal X‐ray diffraction data. 相似文献
