DFT investigations are carried out to explore the effective catalyst forms of DBU and H2O and the mechanism for the formation of 2,3‐dihydropyrido[2,3‐d]‐pyrimidin‐4(1H)‐ones. Three main pathways are disclosed under unassisted, water‐catalyzed, DBU and water cocatalyzed conditions, which involves concerted nucleophilic addition and H‐transfer, concerted intramolecular cyclization and H‐transfer, and Dimroth rearrangement to form the product. The results indicated that the DBU and water cocatalyzed pathway is the most favored one as compared to the rest two pathways. The water donates one H to DBU and accepts H from 2‐amino‐nicotinonitrile ( 1 ), forming [DBU‐H]+‐H2O as effective catalyst form in the proton migration transition state rather than [DBU‐H]+‐OH?. The hydrogen bond between [DBU‐H]+···H2O··· 1 ? decreases the activation barrier of the rate‐determining step. 1,3‐Di(thiophen‐2‐yl)prop‐2‐en‐1‐one ( 1 ) was utilized in the synthesis of 4,6‐di(thiophen‐2‐yl)‐3,4‐dihydropyrimidine‐2(1H)‐thione ( 2 ) and 5,7‐di(thiophen‐2‐yl)‐2‐thioxo‐2,3‐dihydropyrido[2,3‐d]pyrimidin‐4(1H)‐one ( 4 ). The latter thiones were used in the synthesis of two new series of [1,2,4]triazolo[4,3‐a]pyrimidines 10a – i and pyrido[2,3‐d][1,2,4]triazolo[4,3‐a]pyrimidinones 5a – i via reaction with the appropriate hydrazonoyl halides using triethylamine as a basic catalyst in dioxane. The mechanism of formation of the synthesized compounds was discussed, and the assigned structure was established via microanalysis, spectral data (infrared, 1H NMR, and Mass), and density functional calculations. Moreover, the newly synthesized products were evaluated for their antimicrobial activities, and the results show that some derivatives have been well with mild activities. Finally, quantum chemistry calculations confirmed the mechanism and structure of the products.  相似文献

The carbodiimides 4 , obtained from reactions of iminophosphorane 3 with aromatic isocyanates, were reacted with secondary amines to give 2‐dialkylamino‐5‐ethyl‐6‐methyl‐thieno[2,3‐d]pyrimidin‐4(3H)‐ones 6 in the presence of catalytic amount of EtONa. Reactions of 4 with phenols or ROH in the presence of the catalytic amount of K2CO3 or RONa gave 2‐aryloxy‐ or 2‐alkoxy‐5‐ethyl‐6‐methyl‐thieno[2,3‐d]pyrimidin‐4(3H)‐ones 6 in satisfactory yields. Their structures were clearly verified by IR spectroscopy, 1H‐NMR spectroscopy, EI‐MS, and elemental analysis, and in the case of compound 6i , further analyzed by single‐crystal X‐ray diffraction. The title compounds substituted 2‐lactosylthiothieno[2,3‐d]pyrimidin‐4‐ones 6 were synthesized by the glycosyl reaction and alcoholysis reaction of substituted 2‐thioxo‐thieno[2,3‐d]pyrimidin‐4‐ones 4 ,which is formed by the base catalytic and acetic acidify reaction of amino esters 2 with alkyl or arylisothiocyanates and hepta‐O‐acetyl‐lactosyl bromide in good yields. A synthesis of novel bis(s‐triazolo[3,4‐b][1,3,4]thiadiazines) 4 , 5 , 6 in which the triazolothiadiazine is linked to the benzene core through the thiadiazine ring via phenoxymethyl spacers was reported. First attempt to synthesize 4 , 5 , 6 by the reaction of the appropriate bis(acetophenones) with 4‐amino‐3‐mercapto‐1,2,4‐triazole derivatives using an acidified acetic acid method were unsuccessful. On the other hand, reaction of the corresponding bis(α‐bromoketones) with 4‐amino‐3‐mercapto‐1,2,4‐triazole derivatives afforded 4 , 5 , 6 in good yields. The reaction pathway is assumed to involve S‐alkylation to give bis(aminotriazole) intermediates, followed by intramolecular cyclocondensation to give 4 , 5 , 6 . The successful isolation of the corresponding bis(aminotriazole) intermediates provides strong evidence for the proposed mechanism. The novel bis(thiazoles) 23 , 24 , 25 , 26 , 27 , 28 , 29 , 30 , 31 , 32 , 33 , 34 , 35 , 36 , linked to alkyl or aryl spacers can also be synthesized by reaction of the appropriate bis(bromoacetyl) compounds 12a , 12b , 12c and 14 , 15 , 16 , 17 , 18 , 19 with the corresponding thioamide derivatives 20 , 21 , 22 .  相似文献

The reaction of 6‐aminopyrimidin‐4‐ones 1 with benzaldehydes 2 and β‐aminocrotononitrile 3 or benzoylacetonitrile 4 under microwave irradiation in dry media yields the 6‐cyano‐5,8‐dihydropyrido[2,3‐d]‐pyrimidinones 5a‐t . The structure of the synthesized compounds was determined on the basis of nmr measurements, especially by 1H,1H?, 1H,13C COSY, DEPT and NOESY experiments. In contrast with other pyrido‐[2,3‐d]pyrimidine derivatives, these compounds did not show any antifungal in vitro activity up to 250 μg/mL.  相似文献

A novel series of 2,4‐disubstituted‐1,2,4‐triazolo[1,5‐a]quinazolin‐5(4H)‐ones were prepared by Dimroth rearrangement of their respective isomers namely 1,4‐disubstituted‐[1,2,4]triazolo[4,3‐a]‐quinazolin‐5(4H)‐ones. The latter were prepared via new synthetic strategy based on 1,5‐elecrocyclization of the respective N‐(4‐oxo‐3‐phenylquinazolin‐2‐yl)nitrilimines.  相似文献

The aza‐Wittig reactions of benzaldehyde‐, acetophenone‐ and benzophenone 1‐[(triphenylphosphor‐anylidene)amino]ethylidenehydrazones ( 1 ) with 2,3‐furandiones 6 provide a new route to 4H,8H‐1,2,4‐triazolo[1,5‐c][1,3]oxazepin‐4‐ones 14 or 5,6‐dihydro‐7H,12H‐naphtho[2,1‐f|[1,2,4]triazolo[1,5‐c]‐[1,3]oxazepin‐7‐ones 17 via the thermal reaction of the expected azinoimine vinylogous lactones.  相似文献

A synthesis of novel bis(triazolothiadiazines) 11 , 12 , 13 , 14 , bis(quinoxalines) 16 and 17 , bis(thiadiazoles) 24 and 25 , and bis(oxadiazole) 31 , which are linked to the thieno[2,3‐b]thiophene core via phenoxymethyl group, was reported. Thus, reaction of the bis(α‐bromoketones) 6 and 7 with the corresponding 4‐amino‐3‐mercapto‐1,2,4‐triazole derivatives 8 , 9 , 10 in ethanol–DMF mixture in the presence of a few drops of triethylamine as a catalyst under reflux afforded the novel bis(5,6‐dihydro‐s‐triazolo[3,4‐b]thiadiazines) 11 , 12 , 13 , 14 in 60–72% yields. The bis(quinoxalines) 16 and 17 were also synthesized as a sole product in high yields by the reaction of 6 and 7 with o‐phenylenediamine 15 in refluxing acetonitrile in the presence of piperidine as a catalyst. Cyclization of the bis(aldehyde thiosemicarbazones) 20 and 21 with acetic anhydride afforded the corresponding bis(4,5‐dihydro‐1,3,4‐thiadiazolyl) derivatives 24 and 25 in good yield. Bis(5‐phenyl‐2,3‐dihydro‐1,3,4‐oxadiazole) derivative 31 could be obtained in 67% yield by cyclization of the appropriate bis(N‐phenylhydrazone) 29 in refluxing acetic anhydride for 3 h.  相似文献

A synthesis of bis(α‐bromo ketones) 5a‐c and 6b,c was accomplished by the reaction of bis(acetophenones) 3a‐c and 4b,c with N‐bromosuccinimide in the presence of p‐toluenesulfonic acid (p‐TsOH). Treatment of 5a‐c and 6b,c with each of 4‐amino‐3‐mercapto‐1,2,4‐triazoles 9a,b and 4‐amino‐6‐phenyl‐3‐mercapto‐1,2,4‐triazin‐5(4H)‐ones 13 in refluxing ethanol afforded the novel bis(s‐triazolo[3,4‐b][1,3,4]thiadiazines) 10a‐d and 11a‐c as well as bis(as‐triazino[3,4‐b][1,3,4]thiadiazines) 14a‐c and 15 , respectively, in good yields. Compounds 11b and 11c underwent NaBH4 reduction in methanol to give the target 1,ω‐bis{4‐(6,7‐dihydro‐3‐substituted‐5H‐1,2,4‐triazolo[3,4‐b][1,3,4]thiadiazin‐6‐yl)phenoxy}butanes 12a and 12b in 42 and 46% yields, respectively.  相似文献

Methane sulfonamide derivatives of 3‐amino‐2‐thioxo‐2,3‐dihydrothieno[2,3‐d]pyimidin‐4(1H)‐one, potential selective COX‐2 inhibitors, were synthesized and their structural elucidation is here reported. Some derivatives, at 10 μM concentration, showed a significant percentage of inhibition in some in vitro experiments.  相似文献

Treatment of 6‐hydroxy‐5‐phenylazo‐2‐thioxo‐4(1H)‐pyrimidinone 1 with a series of hydrazonoyl halides 2 and N,2‐diaryl‐diazinecarbohydrazonoyl halides 9 in dioxane in the presence of triethylamine under reflux furnishes 6‐phenylazo and 3,6‐bis(arylazo)‐7‐hydroxy‐1H‐[1,2,4]triazolo[4,3‐a]pyrimidin‐5(4H)‐one derivatives 7 and 10 , respectively. Sodium salt of 4‐hydroxy‐6‐methyl‐2‐phenylpyrimidine‐5‐carbonitrile ( 3 ) was subjected to alkylation with different a‐halo compounds, where the corresponding O‐alkylated products 4a‐g were obtained. Ring closure of the O‐alkylated product 4a‐c performed using sodium ethoxide in refluxing ethanol afforded furo[2,3‐d]pyrimidines 5a‐c The latter compounds on reaction with a variety of reagents gave other new furopyrimidines as well as a number of furodipyrimidines.  相似文献
