Well‐defined star‐shaped hydrophobic poly(ε‐caprolactone) (PCL) and hydrophilic poly(ethylene glycol) (PEG) amphiphilic conetworks (APCNs) have been synthesized via the combination of ring opening polymerization (ROP) and click chemistry. Alkyne‐terminated six arm star‐shaped PCL (6‐s‐PCLx‐C?CH) and azido‐terminated PEG (N3‐PEG‐N3) are characterized by 1H NMR and FT‐IR. The swelling degree of the APCNs is determined both in water and organic solvent. This unique property of the conetworks is dependent on the nanophase separation of hydrophilic and hydrophobic phases. The morphology and thermal behaviors of the APCNs are investigated by SEM and DSC respectively. The biocompatibility is determined by water soluble tetrazolium salt reagents (WST‐1) assay, which shows the new polymer networks had good biocompatibility. Through in vitro release of paclitaxel (PTX) and doxorubicin (DOX), the APCNs is confirmed to be promising drug depot materials for sustained hydrophobic and hydrophilic drugs. © 2015 Wiley Periodicals, Inc. Degradable polyester‐based star polymers with a high level of functionality in the arms were synthesized via the “arms first” approach using an acetylene‐functional block copolymer macroinitiator. This was achieved by using 2‐hydroxyethyl 2′‐methyl‐2′‐bromopropionate to initiate the ring‐opening polymerization (ROP) of caprolactone monomer followed by an atom transfer radical polymerization (ATRP) of a protected acetylene monomer, (trimethylsilyl)propargyl methacrylate. The hydroxyl end‐group of the resulting block copolymer macroinitiator was subsequently crosslinked under ROP conditions using a bislactone monomer, 4,4′‐bioxepanyl‐7,7′‐dione, to generate a degradable core crosslinked star (CCS) polymer with protected acetylene groups in the corona. The trimethylsilyl‐protecting groups were removed to generate a CCS polymer with an average of 1850 pendent acetylene groups located in the outer block segment of the arms. The increased functionality of this CCS polymer was demonstrated by attaching azide‐functionalized linear polystyrene via a copper (I)‐catalyzed cycloaddition reaction between the azide and acetylene groups. This resulted in a CCS polymer with “brush‐like” arm structures, the grafted segment of which could be liberated via hydrolysis of the polyester star structure to generate molecular brushes. © 2009 Wiley Periodicals, Inc. New hyperbranched hydrophobic cross‐linkers with peripheral azide groups were synthesized as follows: First, star‐shaped polycaprolactones (sPCL) were synthesized by ring‐opening polymerization of caprolactone in the presence of pentaerythritol and tin (II) octoate. In the next step, sequential acrylation, Micheal addition, tosylation, and azidation by acryloyl chloride, diethanol amine, tosyl chloride, and sodium azide were respectively exploited to synthesize azide‐functionalized hyperbranched star‐shaped polycaprolactones which were named sPCL‐acrylate‐diethanolamine‐azide (sPCL‐AC‐DEA‐N3) and sPCL‐acrylate‐diethanolamine‐acrylate‐diethanolamine‐azide (sPCL‐AC‐DEA‐AC‐N3). All steps were thoroughly characterized by FT‐IR and 1H NMR spectroscopy. The GPC analysis showed that the molecular weight of sPCL increased after two azide functionalizations. Amphiphilic hydrogels based on sPCL‐AC‐DEA‐N3 (Mn = 8130 g/mol) and sPCL‐AC‐DEA‐AC‐N3 (Mn = 10112 g/mol) with linear alkyne‐terminated polyethylene glycols (PEG) (Mn = 2000, 4000, and 6000 g/mol) were synthesized through click coupling between azide and alkyne groups. In both hydrogels, the swelling ratio increased by increasing the molecular weight of PEG. The obtained results showed that the branching of the cross‐linker, significantly affected the swelling ratio of hydrogels. For instance, the swelling ratio of sPCL‐AC‐DEA‐AC‐N3 and PEG‐6000 (Q = 900) was higher than sPCL‐AC‐DEA‐N3 and PEG‐6000 (Q = 600). Despite the high cross‐linking density of sPCL‐AC‐DEA‐AC‐DEA‐N3–based hydrogels, the amount of released theophylline was higher than sPCL‐AC‐DEA‐N3–based hydrogels, due to the high content of PEG in these hydrogels.  相似文献

Well‐defined AB3‐type miktoarm star‐shaped polymers with cholic acid (CA) core were fabricated with a combination of “click” chemistry and ring opening polymerization (ROP) methods. Firstly, azide end‐functional poly(ethylene glycol) (mPEG), poly(methyl methacrylate) (PMMA), polystyrene (PS), and poly(ε‐caprolactone) (PCL) polymers were prepared via controlled polymerization and chemical modification methods. Then, CA moieties containing three OH groups were introduced to these polymers as the end groups via Cu(I)‐catalyzed click reaction between azide end‐functional groups of the polymers ( mPEG‐N3 , PMMA‐N3 , PS‐N3 , and PCL‐N3 ) and ethynyl‐functional CA under ambient conditions, yielding CA end‐functional polymers ( mPEG‐Cholic , PMMA‐Cholic , PS‐Cholic , and PCL‐Cholic ). Finally, the obtained CA end‐capped polymers were employed as the macroinitiators in the ROP of ε‐caprolactone (ε‐CL) yielding AB3‐type miktoarm star polymers ( mPEG‐Cholic‐PCL3 , PMMA‐Cholic‐PCL3 , and PS‐Cholic‐PCL3 ) and asymmetric star polymer [ Cholic‐(PCL)4 ]. The chemical structures of the obtained intermediates and polymers were confirmed via Fourier transform infrared and 1H nuclear magnetic resonance spectroscopic techniques. Base‐catalyzed reaction between a thiol and an epoxide group is a simple fusion process that leads to the formation of a β‐hydroxythio‐ether linkage. This reaction is efficient, regio‐selective, and fast. In addition, it produces a reactive hydroxyl group upon completion. Therefore, it is of considerable potential in synthesis of reactive and functional soft materials. Here, we discuss the fundamental aspects of this process, the so‐called thiol‐epoxy “click” reaction, and its utility in the preparation and post‐polymerization functionalization of polymers and crosslinked networks. Furthermore, its application in surface modification of solid substrates is also considered. Aliphatic polyesters bearing pendant alkyne groups were successfully prepared by step‐growth polymerization of different building blocks such as adipic acid and succinic acid in combination with an acetylene‐based diol, 2‐methyl‐2‐propargyl‐1,3‐propanediol, besides 1,4‐butanediol and ethylene glycol. It was demonstrated that the alkyne groups survive the high reaction temperatures (200 °C) in the presence of a radical inhibitor. The alkyne loading has been tuned by the ratio of the different monomers used, up to 25 mol % of alkyne groups. Subsequently, the alkyne groups have been reacted with azides by the copper‐catalyzed Huisgen 1,3‐dipolar cycloaddition reaction, a popular type of “click” chemistry. “Click” reactions have been performed quantitatively in the presence of benzyl azide and azide‐terminated poly(ethylene glycol), yielding brush copolymers in the latter case. Kinetic investigations about this click reaction have been performed by means of on‐line Fourier transform mid‐infrared spectroscopy, which was reported for the first time in the field of the click chemistry research. A whole range of functionalized polyesters, based on poly(ethylene succinate) and poly(butylene adipate), is available, the properties of which can be tailored by choosing the appropriate azide compound. © 2008 Wiley Periodicals, Inc. Hetero‐arm star ABC‐type terpolymers, poly(methyl methacrylate)‐polystyrene‐poly(tert‐butyl acrylate) (PMMA‐PS‐PtBA) and PMMA‐PS‐poly(ethylene glycol) (PEG), were prepared by using “Click” chemistry strategy. For this, first, PMMA‐b‐PS with alkyne functional group at the junction point was obtained from successive atom transfer radical polymerization (ATRP) and nitroxide‐mediated radical polymerization (NMP) routes. Furthermore, PtBA obtained from ATRP of tBA and commercially available monohydroxyl PEG were efficiently converted to the azide end‐functionalized polymers. As a second step, the alkyne and azide functional polymers were reacted to give the hetero‐arm star polymers in the presence of CuBr/N,N,N′,N″,N″‐pentamethyldiethylenetriamine ( PMDETA) in DMF at room temperature for 24 h. We describe a facile, one‐pot, two‐step polymerization towards synthesizing block co‐polymers bearing reactive isocyanate functional groups. Reversible addition fragmentation chain transfer (RAFT) polymerization is used to mediate the co‐polymerization of isocyanate‐bearing monomers dimethyl meta‐isopropenyl benzyl isocyanate (TMI) and 2‐isocyanatoethyl methacrylate (ICEMA) with styrene and methyl methacrylate (MMA), respectively. ICEMA was incorporated into the polymer at a faster rate than TMI and its unhindered isocyanate group was found to be more reactive than the hindered isocyanate group of TMI. Both the TMI/styrene and the MMA/ICEMA systems maintain the reactivity of the isocyanate functionality, which was exploited by attaching representative hydroxyl‐bearing small and large molecules as well as solid substrates to the block co‐polymers. Thus, we demonstrate the versatility of the block co‐polymer system as a basis for forming branched polymers or as grafts for a solid substrate. © 2011 Wiley Periodicals, Inc. Well‐defined ABCD 4‐Miktoarm star‐shaped quarterpolymers of [poly(styrene)‐poly(tert‐butyl acrylate)‐poly(ethylene oxide)‐poly(isoprene)] [star(PS‐PtBA‐PEO‐PI)] were successfully synthesized by the combination of the “click” chemistry and multiple polymerization mechanism. First, the poly(styryl)lithium (PS?Li+) and the poly(isoprene)lithium (PI?Li+) were capped by ethoxyethyl glycidyl ether (EEGE) to form the PS and PI with both an active ω‐hydroxyl group and an ω′‐ethoxyethyl‐protected hydroxyl group, respectively. After these two hydroxyl groups were selectively modified to propargyl and 2‐bromoisobutyryl group for PS, the resulted PS was used as macroinitiator for ATRP of tBA monomer and the diblock copolymer PS‐b‐PtBA with a propargyl group at the junction point was achieved. Then, using the functionalized PI as macroinitiator for ROP of EO monomer and bromoethane as blocking agent, the diblock copolymer PI‐b‐PEO with a protected hydroxyl group at the conjunction point was synthesized. After the hydrolysis, the recovered hydroxyl group of PI‐b‐PEO was modified to bromoacetyl and then azide group successively. Finally, the “click” chemistry between them was proceeded smoothly. The obtained star‐shaped quarterpolymers and intermediates were characterized by 1H NMR, FT‐IR, and SEC in detail. © 2008 Wiley Periodicals, Inc. The star graft copolymers composed of hyperbranched polyglycerol (HPG) as core and well defined asymmetric mixed “V‐shaped” identical polystyrene (PS) and poly(tert‐butyl acrylate) as side chains were synthesized via the “click” chemistry. The V‐shaped side chain bearing a “clickable” alkyne group at the conjunction point of two blocks was first prepared through the combination of anionic polymerization of styrene (St) and atom transfer radical polymerization of tert‐butyl acrylate (tBA) monomer, and then “click” chemistry was conducted between the alkyne groups on the side chains and azide groups on HPG core. This contribution describes our recent efforts geared toward the use of a general, thio‐bromo “click” reaction as a post‐polymerization method for the preparation of flame‐resistant polymeric materials. α‐bromo ester‐containing polymers could easily be prepared using ROMP and a subsequent, facile thio‐bromo click reaction was used for the installation of a phosphorus‐moiety that was shown to impart flame‐resistant/self‐extinguishing properties to these polymers. The extent of their flame resistance was then ascertained by treating paper (previously coated with polymer) to standard burn tests as well as measuring the limiting oxygen index (LOI). © 2017 Wiley Periodicals, Inc. Poly(ethylene glycol) (PEG)‐containing quasi‐model amphiphilic polymer conetworks (APCNs) were prepared by reversible addition fragmentation chain transfer (RAFT) polymerization using α,ω‐bis(2‐cyanoprop‐2‐yl dithiobenzoate)‐PEG as a bifunctional RAFT macrochain transfer agent (macro‐CTA) and stepwise additions of a hydrophobic monomer and a crosslinker (crosslinker: macro‐CTA = 10:1, reaction time 24 h). Three different types of monomers, methyl methacrylate (MMA), n‐butyl acrylate and styrene, were employed as the hydrophobic monomers, whereas ethylene glycol dimethacrylate, ethylene glycol diacrylate and 1,4‐divinylbenzene served as the respective crosslinkers. PEG homopolymer hydrophilic quasi‐model networks were also prepared by RAFT‐polymerizing the three crosslinkers directly onto the two active ends of the PEG‐based macro‐CTA. From the three ABA triblock copolymers prepared, the MMA‐containing one was obtained at the highest polymerization yields. The crosslinking yields of the three ABA triblock copolymers with the corresponding crosslinkers were higher than those of the PEG‐based macro‐CTA with the same crosslinkers. The degrees of swelling (DSs) of all conetworks were measured in water and in tetrahydrofuran (THF). The DSs of the APCNs in THF were higher than those in water, whereas the reverse was true for the DSs of the hydrophilic homopolymer networks. Finally, the aqueous DSs of the APCNs were lower than those of the corresponding hydrophilic homopolymer networks. © 2008 Wiley Periodicals, Inc. Conetworks based on end‐linked homopolymers and amphiphilic gradient copolymers were synthesized by the atom transfer radical polymerization (ATRP) of 2‐(dimethylamino)ethyl methacrylate (DMAEMA, hydrophilic monomer), methyl methacrylate (MMA, hydrophobic monomer), and ethylene glycol dimethacrylate (EGDMA, hydrophobic cross‐linker). Sequential, rather than step‐wise polymerizations, were performed to enhance the livingness of the polymerization, particularly for the end‐linking step, and to ultimately obtain conetworks based on gradient rather than pure block copolymers. Amphiphilic conetworks based on end‐linked MMA‐DMAEMA‐MMA gradient copolymers of different compositions were successfully synthesized as confirmed by the narrow molecular weight distributions of the linear precursors, the rigidity of the amphiphilic conetwork products and the low sol‐fraction extracted from the conetworks. Similarly successful was the ATRP synthesis of an end‐linked conetwork based on a DMAEMA‐MMA statistical copolymer and of a randomly cross‐linked conetwork that resulted from the simultaneous terpolymerization of DMAEMA, MMA and EGDMA. An amphiphilic conetwork based on an end‐linked DMAEMA‐MMA‐DMAEMA gradient copolymer presented a less rigid, mucous‐like, texture. The degrees of swelling (DS) in tetrahydrofuran of all the conetworks were higher than those measured in pure water, whereas the aqueous DS values increased by lowering the pH and increasing the DMAEMA content of the conetworks. © 2010 Wiley Periodicals, Inc. Nitroxide‐mediated radical polymerization has been used for the preparation of pentafluorostyrene (PFS) homopolymers and random copolymers of PFS and oligo(ethyleneglycol) methacrylate (OEGMA8.5). The poly(pentafluorostyrene) homopolymers were reacted with thiophenol at different ratios at room temperature in the presence of triethylamine. The “clicked” polymers were characterized by 1H and 19F NMR spectroscopy and size exclusion chromatography. Moreover, the copolymerization kinetics of the PFS and OEGMA8.5 copolymers was followed, and the phase transition behavior of random copolymers with different compositions was discussed. Furthermore, copolymers of PFS and 2‐(dimethylamino) ethyl methacrylate (DMAEMA) were prepared at various mole ratios, and the copolymer with a 10:90 ratio, respectively, was soluble in water at room temperature. Turbidimetry measurements were performed for PFS and OEGMA8.5 or DMAEMA copolymers to determine their cloud points. Finally, the PFS and OEGMA8.5 copolymer with a mole ratio of 60:40 was reacted further with thiophenol to increase the hydrophobic part in the copolymer. The cloud points of the obtained copolymers could be tuned from 87 to 33 °C by using not only the controlled radical polymerization but also the “click” reaction in a controlled fashion. © 2010 Wiley Periodicals, Inc. The use of a thio‐bromo click strategy as an efficient postpolymerization tool is described. Norbornene derivatives bearing an α‐bromo ester could be polymerized using Grubbs 2nd generation initiator to provide α‐bromo ester‐containing homo‐and block copolymers that could be efficiently functionalized through reactions with various thiols. A one‐pot strategy was also used, in which up to four different thiols were reacted simultaneously. This chemistry could also be used as an efficient cross‐linking strategy to form ROMP‐based gels as well as a tool for terminal functionalization of polypropylene‐based oligomers. © 2015 Wiley Periodicals, Inc. Amphiphilic, biodegradable block glycopolymers based on poly(ε‐caprolactone) (PCL) with various pendent saccharides were synthesized by combination of ring‐opening polymerization (ROP) and “click” chemistry. PCL macroinitiators obtained by ROP of ε‐caprolactone were used to initiate the ROP of 2‐bromo‐ε‐caprolactone (BrCL) to get diblock copolymers, PCL‐b‐PBrCL. Reaction of the block copolymers with sodium azide converted the bromine groups in the PBrCL block to azide groups. In the final step, click chemistry of alkynyl saccharides with the pendent azide groups of PCL‐b‐PBrCL led to the formation of the amphiphilic block glycopolymers. These copolymers were characterized by 1H NMR spectroscopy and gel permeation chromatography. The self‐assembly behavior of the amphiphilic block copolymers was investigated using transmission electron microscopy and atomic force microscope, spherical aggregates with saccharide groups on the surface were observed, and the aggregates could bind reversibly with Concanavalin A. © 2009 Wiley Periodicals, Inc. In this article, the synthesis and the functionalization of well‐defined, narrow polydispersity (polydispersity index < 1.2) star polymers via reversible addition‐fragmentation chain transfer polymerization is detailed. In this arm first approach, the initial synthesis of a poly(pentafluorophenyl acrylate) polymer, and subsequent, cross‐linking using bis‐acrylamide to prepare star polymers, has been achieved by reversible addition fragmentation chain transfer polymerization. These star polymers were functionalized using a variety of amino functional groups via nucleophilic substitution of pentafluorophenyl activated ester to yield star polymers with predesigned chemical functionality. This approach has allowed the synthesis of star glycopolymer using a very simple approach. Finally, the core of the stars was modified via thiol‐ene click chemistry reaction using fluorescein‐o‐acrylate and DyLigh 633 Maleimide. © 2011 Wiley Periodicals, Inc. Novel hexa‐armed and pyrene (Pyr) end‐capped phosphazene dendrimer [N3P3‐(Pyr)6] and star polymer with poly(ε‐caprolactone) (PCL) arms [N3P3‐(PCL‐Pyr)6] were prepared via two series of reactions. In these series, core‐first approach was used starting from a hexa‐hydroxy functional phosphazene derivative (N3P3‐(OH)6). It was used as an initiator in the ring‐opening polymerization of ε‐caprolactone to prepare a hexa‐armed PCL star polymer (N3P3‐(PCL‐OH)6). Hydroxyl functionalities of N3P3‐(OH)6 and N3P3‐(PCL‐OH)6 were then successfully converted into bromide and azide, in turn. Further end‐group modifications of azide functional dendrimer precursor (N3P3‐(N3)6) and star polymer (N3P3‐(PCL‐N3)6) were achieved quantitatively via the Cu(I) catalyzed click reaction between azide functional groups and 1‐ethynyl pyrene in the final step. pH‐ and temperature‐responsive poly(N‐isopropylacrylamide‐block?4‐vinylbenzoic acid) (poly(NIPAAm‐b‐VBA)) diblock copolymer brushes on silicon wafers have been successfully prepared by combining click reaction, single‐electron transfer‐living radical polymerization (SET‐LRP), and reversible addition‐fragmentation chain‐transfer (RAFT) polymerization. Azide‐terminated poly(NIPAAm) brushes were obtained by SET‐LRP followed by reaction with sodium azide. A click reaction was utilized to exchange the azide end group of a poly(NIPAAm) brushes to form a surface‐immobilized macro‐RAFT agent, which was successfully chain extended via RAFT polymerization to produce poly(NIPAAm‐b‐VBA) brushes. The addition of sacrificial initiator and/or chain‐transfer agent permitted the formation of well‐defined diblock copolymer brushes and free polymer chains in solution. The free polymer chains were isolated and used to estimate the molecular weights and polydispersity index of chains attached to the surface. Ellipsometry, contact angle measurements, grazing angle‐Fourier transform infrared spectroscopy, and X‐ray photoelectron spectroscopy were used to characterize the immobilization of initiator on the silicon wafer, poly(NIPAAm) brush formation via SET‐LRP, click reaction, and poly(NIPAAm‐b‐VBA) brush formation via RAFT polymerization. The poly(NIPAAm‐b‐VBA) brushes demonstrate stimuli‐responsive behavior with respect to pH and temperature. The swollen brush thickness of poly(NIPAAm‐b‐VBA) brush increases with increasing pH, and decreases with increasing temperature.
