Novel [1,2,4]triazole derivatives were synthesized via various synthetic pathways. Among which were different substituted [1,2,4]triazole analogues that were synthesized, in addition to various fused [1,2,4]triazolo[1,5‐a]pyrimidine derivatives, [1,2,4]triazolo[1,5‐a][1,3,5]triazines, and [1,2,4]triazolo[5,1‐c][1,2,4]triazines. Besides, benzo[h][1,2,4]triazolo[5,1‐b]quinazolines, [1,2,4]triazolo‐[5,1‐b]quinazoline, [1,2,4]triazolo[1,5‐a]quinazoline and [1,2,4]triazolo[5,1‐d][1,2,3,5]tetrazine derivatives were also synthesized. The newly synthesized compounds were evaluated for their in vitro anticancer activity against liver cancer HepG2 and breast cancer MCF7 cell lines compared with the reference drug doxorubicin. Compounds 4 , 7 , 15 , 17 , 28 , 34 , and 47 were found to exert promising anticancer activity against HepG2 cell line showing IC50 values ranging from 17.69 to 25.4 μM/L, while compounds 7 , 14a , 17 , 28 , and 34 showed significant activity against MCF7 cell line with IC50 values ranging from 17.69 to 27.09 μM/L.  相似文献

Various new substituted and fused coumarin analogues have been synthesized via different synthetic pathways. Among which are variable substituted coumarin derivatives bearing either biologically active side chains or rings at 5, 6, and 3 positions of the coumarin nucleus as indicated in compounds 10 , 12 , 13 , 16–19 , 21 , 23–32 , 38 , and 42–45 . In addition, different pyranocoumarin derivatives either substituted as in compounds 2 , 3 , and 6 or fused as compounds 33–36 , pyranoxanthene analogues such as compounds 4 and 46 , coumarinotriazolothiadiazine derivative 8 , coumarinonaphthodiazocin analogue 39 and coumarinopyrazolone derivative 40 were synthesized. Thirty‐eight of the synthesized compounds were subjected to in vitro anticancer screening against mammalian liver carcinoma HepG2 and breast carcinoma MCF7 cell lines using Cisplatin as a standard reference. The anticancer activity screening results revealed that, among the tested compounds, compounds 16 , 40 , and 43 bearing 4‐chlorophenyl‐2‐aminopyridine‐3‐carbonitrile attached to C6 position, fused pyrazolone ring or attached to 4‐chlorophenyl‐2‐oxo‐dihydropyridine‐3‐carbonitrile at C3 position of the coumarin nucleus, respectively, exhibited moderate to strong activity against both cell lines.  相似文献

The novel title ruthenium(II) complex, [RuCl(C10H14)(C10H10N3OS)], was synthesized from the reaction of 1,2,4‐triazepine, a new class of bidentate ligands, with [Ru(p‐cymene)Cl2]2. The 1,2,4‐triazepine ligand is coordinated to the metal centre through the N‐4 and S atoms, forming a four‐membered chelate ring. This is the first structural example of a transition metal complex containing a 1,2,4‐triazepine ligand.  相似文献

In carrying on our interest in heteropolycyclic structures with biological activities, we projected the preparation of compounds containing the pyrido[3,2-e][1,2,4]triazine or pyrido[2,3-b][1,4]triazepine systems. The established synthetic approach for the preparation of latter system led to the triazine derivatives 5a-f while a new bicyclic triazepine structure 6 is accomplished with difficulty. In expanding the pyridotriazine structure, we obtained derivatives of a new tricyclic structure, 5-substituted-6a-ethyloxycarbonyl-5,6,6a,7-tetrahydropyrido[2,3-e]pyrrolo[2,1-c][1,2,4]triazin-9(8H)-ones 8 in which the triazine ring is fused with a pyrrole nucleus. Compounds 5a-f and 8a,b will be tested as potential CNS depressant, antiinflammatory, analgesic and antibacterial agents.  相似文献

Salts of meta‐xylyl‐linked N‐ethyl/n‐butyl/benzyl‐substituted bis‐benzimidazolium having hexafluorophosphate counterions have been synthesized. The corresponding binuclear Ag(I)‐N‐heterocyclic carbene complexes were prepared by the reaction of Ag2O. The N‐heterocyclic carbene (NHC) ligand precursor 7 and Ag(I)–NHC complexes 10 and 11 have been structurally characterized by single‐crystal X‐ray diffraction technique. All of the reported compounds have been tested for their anticancer activity using human colorectal (HCT 116) cancer cell lines. Sterically varied benzimidazolium salts displayed significant activity against HCT 116 cell line, yielding IC50 values in the range 0.1–19.4 µ m , while Ag(I)–carbene complexes showed exceptionally good activity (0.2–1.3 µ m ) against tested cancer cell lines. Six bromomethyl derivatives of the new 2,3‐dihydrooxazolo[3,2‐b]thieno[3,4‐e][1,2,4]thiadiazine 5,5‐dioxides, 2,3‐dihydrooxazolo[3,2‐b]thieno[2,3‐e][1,2,4]thiadiazine 5,5‐dioxides and 6,7‐dihydrooxazolo‐[3,2‐b]pyrazolo[4,3‐e][1,2,4]thiadiazine 9,9‐dioxides heterocyclic ring systems were synthesized. These compounds are good intermediates for the preparation and development of promising antiviral and psy‐chotropic drugs. The structures of the products are supported by different nmr spectroscopic methods and mass spectrometry.  相似文献

Chemical transformations of chromone‐3‐carbonitrile ( 1 ) with some substituted hydrazines, namely, thiosemicarbazide, S‐methyl/benzyldithiocarbazate, 7‐chloro‐4‐hydrazinoquinoline, and 3‐hydrazino‐5,6‐diphenyl‐1,2,4‐triazine, led to substituted pyrazoles 2 , 5 – 8 . Ring opening of carbonitrile 1 followed by recyclization with 3‐amino‐1,2,4‐triazole and 2‐aminobenzimidazole gave triazolo[1,5‐a]pyrimidine 9 and pyrimido[1,2‐a]benzimidazole 10 , respectively. Treatment of carbonitrile 1 with some heterocyclic amines produced 2‐amino‐3‐substituted‐chromones 11 and 12 . The novel 3‐hydroxychromeno[4,3‐b]pyrazolo[4,3‐e]pyridin‐5(1H)‐one ( 13 ) was efficiently synthesized from the ring conversion of carbonitrile 1 with cyanoacetohydrazide. A mixture of chromeno[2,3‐b]naphthyridine 14 and chromeno[4,3‐b]pyridine 15 was obtained from base catalyzed transformation of carbonitrile 1 with malononitrile dimer. A diversity of novel annulated chromeno[2,3‐b]pyridines 16 – 22 was also synthesized. Chromeno[2,3‐b]pyrrole‐2‐carboxylate 23 was obtained from the reaction of carbonitrile 1 with ethyl chloroacetate. Structures of the new synthesized products were deduced on the basis of their analytical and spectral data.  相似文献

A series of 2‐substituted 2H‐thieno[3,4‐e][1,2,4]thiadiazin‐3(4H)‐one 1,1‐dioxides ( 2 ), 2‐substituted 2H‐thieno[2,3‐e][1,2,4]thiadiazin‐3(4H)‐one 1,1‐dioxides ( 3 ), 2‐substituted 4,6‐dihydropyrazolo[4,3‐e]‐[1,2,4]thiadiazin‐3(2H)‐one 1,1‐dioxides ( 4 ), 2‐substituted 2,3‐dihydrooxazolo[3,2‐b]thieno[3,4‐e]‐[1,2,4]thiadiazine 5,5‐dioxides, ( 5 ), 6‐substituted 6,7‐dihydro‐2H‐oxazolo[3,2‐b]pyrazolo[4,3‐e][1,2,4]thia‐diazine 9,9‐dioxides ( 6 ) and 7‐substituted 6,7‐dihydro‐2H‐oxazolo[3,2‐b]pyrazolo[4,3‐e][1,2,4]thiadiazine 9,9‐dioxides ( 7 ) were synthesized as potential psychotropic agents.  相似文献

A series of symmetrically n ‐alkyl‐substituted mono benzimidazolium salts with steady increase in n ‐alkyl chain length have been prepared by stepwise N ‐alkylation resulting in salts ( 1 – 8 ). The mono N‐heterocyclic carbene (NHC)–Ag(I) complexes ( 9 – 16 ) derived from the respective salts were readily accessible by in situ deprotonation using Ag2O. All the salts and the complexes were characterized using Fourier transform infrared, 1H NMR, 13C NMR and elemental analyses. Furthermore, the structures of salts 3 and 7 and complex 16 were elucidated using X‐ray crystallography, which established that this mono NHC–Ag(I) complex has a linear bis‐carbene arrangement (C2–Ag). The proligands and the respective Ag(I) complexes were studied for their in vitro anticancer potential against human colon cancer cell line (HCT‐116) using 5‐fluorouracil as a standard. From the IC50 values of all the tested compounds, it can be postulated that there is an influential relationship between the increase in chain length of the wingtip n ‐alkyl groups and the anticancer potential. The proligands 4 – 8 and their respective complexes 12 – 16 with long n ‐alkyl chain lengths (n  = 6–10) showed better IC50 values (0.3–3.9 μM) than the standard drug with the complexes displaying markedly better antiproliferation activity against HCT‐116 cell line than the respective proligands and the standard drug (IC50 = 10.2 μM).  相似文献

In an attempt to establish novel candidate with promising anticancer activity, two derivatives of (benzo[d]thiazol‐2‐yl)thiophene backbone 1 and 14 were synthesized, and they further reacted with various chemical reagents to afford the corresponding substituted thiophene derivatives 6 , 8 , 10 , 15 , 17 , and 20 , thieno[3,2‐d]pyrimidine derivatives 2 – 5 , 7 , 9 , 16 , 21 , 23 , and 24 , thieno[3,2‐b]pyridine derivatives 11 – 13 , and thieno[3,2‐e][1,4]oxazepine derivative 18 . Structures of prepared compounds were affirmed via spectral and elemental data. Among the obtained compounds, seven derivatives 2 , 3 , 4 , 5 , 11 , 12 , and 13 were chosen by National Cancer Institute, USA. Such compounds were screened for their antitumor activity versus 60 cancer cell lines in one‐dose (10 μmol) screening assay. The outcomes showed that all selected compounds exhibited moderate to high anticancer activity towards many cancer cell lines among which compounds 5 and 11 exerted potent antitumor activity against numerous malignant growth cell lines particularly Ovarian Cancer IGROV1.  相似文献

In this paper, we present for the first time the evaluation of cytotoxicity and genotoxicity of de novo synthesized pyrazolo[4,3-e]tetrazolo[1,5-b][1,2,4]triazine sulfonamides MM129, MM130, and MM131 in human tumor cell lines: HeLa, HCT 116, PC-3, and BxPC-3. Cytotoxic and genotoxic properties of the tested compounds were estimated using the MTT assay, comet assay (alkaline and neutral version), and γ-H2AX immuno-staining. Examined sulfonamides exhibited strong anticancer properties towards tested cells in a very low concentration range (IC50 = 0.17–1.15 μM) after 72 h exposure time. The results of the alkaline and neutral version of the comet assay following 24 h incubation of the cells with tested compounds demonstrated the capability of heterocycles to induce significant DNA damage in exposed cells. HCT 116 cells were the most sensitive to the genotoxic activity of novel tricyclic pyrazolo[4,3-e]tetrazolo[1,5-b][1,2,4]triazine sulfonamides in the neutral version of the comet assay. Immunocytochemical detection of γ-H2AX showed an increase in DNA DSBs level in the HCT 116 cell line, after 24 h incubation with all tested compounds, confirming the results obtained in the neutral comet assay. Among all investigated compounds, MM131 showed the strongest cytotoxic and genotoxic activity toward all tested cell types. In conclusion, our results suggest that MM129, MM130, and MM131 exhibit high cytotoxic and genotoxic potential in vitro, especially towards the colorectal cancer cell line HCT 116. However, further investigations and analyses are required for their future implementation in the field of medicine.  相似文献

On the pharmaceutical account of the reported anticancer activity of benzothiazole derivatives, differently substituted benzothiazole derivatives 2a – c to 34a , b , attached at 2‐position to different heterocyclic moieties, were synthesized via different chemical reactions. Thirteen of the newly synthesized compounds were selected by the National Cancer Institute, Bethesda, Maryland, USA, and evaluated for their in vitro antitumor activity against 60 human tumor cell lines in a one‐dose screening panel among which two compounds 4 and 17 showed high activity and were selected for further evaluation in the five‐dose full panel assay, in which compound 4 exerted powerful growth inhibitory activity against all cell lines with GI50 ranging from 0.683 to 4.66 μM/L in addition to excellent lethal activity against most of the cell lines.  相似文献

Anticancer evaluation of pyrazole 2 and Schiff base 5 is reported. Synthesis of some important heterocyclic compounds via 2,3‐diaryloxirane‐2,3‐dicarbonitrile 1 , pyrazole 2 , and Schiff base 5 with different nitrogen nucleophiles afforded new routes for synthesized fused heterocyclic derivatives. These compounds can be used as a key starting materials to synthesize some important imidazolo‐[4,5‐c ]pyrazole, pyrazolo[3,4‐e ]1,2,4‐triaging, imidazolo[3,2‐b ]pyrazole, and pyrazolo‐pyrazine, as anticancer reagents showed good results at optimum conditions (400–500 ppm), particularly the bridge head nitrogen compounds, and could be used to improve them. Electromeric effect of the halogen atom in the aryl moieties can be controlled upon the rate of reaction and the yield of the product. The structure of the synthesized new compounds would be characterized by elemental and spectral data.  相似文献

A novel series of substituted [1,2,4]triazolo[4′,3′:1,2]pyrimido[4,5‐c ]benzo[f ]isoquinolin‐14(10H )‐one was synthesized from the reaction of hydrazonoyl chlorides with pyrimidine thione derivative or via oxidative cyclization of 3‐(2‐substituted‐benzylidene‐hydrazinyl)‐7,8‐dihydrobenzo[f ]pyrimido[4,5‐c ]isoquinolin‐1(2H )‐one. Also, some polyhetero‐cyclic ring systems were prepared through the reaction of 2‐dimethylaminomethylene‐3,4‐dihydro‐2H‐naphthalen‐1‐one and heterocyclic amines. The biological activity of some new products was evaluated, and the results obtained revealed that compounds 10e , 13a , and 18 showed excellent activities against the most bacteria and fungi species used.  相似文献

2‐[(4‐Thiazolylmethyl)thio]‐1H‐benzimidazole 3 was prepared and was allowed to react with ethyl chloroactate then with hydrazine hydrate to afford the hydrazide derivative 5 , which was then reacted with aromatic aldehydes to afford the corresponding arylidine derivatives 6 – 9 . Heterocyclization of the latter hydrazones with acetic anhydride afforded the substituted 1,3,4‐oxadiazoline derivatives 10 – 13 . In addition, new ((thiazolyl)imidazolyl) oxadiazole thioglycoside and acyclic‐C nucleoside analog were prepared via heterocylization of the hydrazide 5 then glycosylation with α‐acetobromoglucose or condensation with D‐xylose, respectively. All the new compounds were structurally characterized. The anticancer activity of some of the newly synthesized compounds was studied against human breast cancer cells (MCF‐7). The results of the anticancer activity showed that compounds 8 , 11 , 12 , 17 , and 18 revealed high activities superior to Doxorubicin; however, the other derivatives showed moderate to low inhibition activities against human breast cancer cells. Docking studies into CDK2 enzyme were investigated, which supported the anticancer activity results.  相似文献

New heterocyclic compounds 1‐(3‐methyl‐9H‐dibenzo[b,f][1,2,4]triazolo[4,3‐d][1,4]diazepin‐6‐yl)ethanone 8a , 1‐(3‐methyldibenzo[b,f][1,2,4]triazolo[4,3‐d][1,4]oxazepin‐6‐yl)ethanone 8b , and 1‐(3‐methyldibenzo[b,f][1,2,4]triazolo[4,3‐d][1,4]thiazepin‐6‐yl)ethanone 8c are synthesized from benzodiazepinone, benzoxazepinone, and benzothiazepinone derivatives. These heterocyclic scaffolds have wide medicinal importance. Best results were obtained in antibacterial screening against Escherichia coli, Enterobacter cloacae, and Staphylococcus aureus and antifungal screening against Candida albicans and Fusarium oxysporum. 1,1‐Diphenyl‐2‐picrylhydrazyl radical scavenging activities of compounds 6c , 7c , and 8c were tested in doses 10, 20, 30, 40, and 50 μg/mL and were expressed as IC50 values and percent of inhibition with means ± standard deviation of three different concentrations of synthesized compounds. The assignment of the structures of synthesized compounds was made by thin‐layer chromatography, elemental analysis, IR, 1H‐NMR, 13C‐NMR, and liquid chromatography–mass spectrometry.  相似文献

A series of novel 6‐{5‐aryl‐4H‐1,2,4‐triazol‐3‐ylthio}methyl‐3‐substituted‐[1,2,4]triazolo[3,4‐b][1,3,4]thiadiazoles and 2‐{(5‐aryl‐4H‐1,2,4‐triazol‐3‐ylthio)‐N‐4‐aryl}acetamides containing 1,2,4‐triazole moiety were synthesized and characterized by 1H, 13C, DEPT, and D2O. The synthesized compounds were evaluated for their antibacterial activity against three strains of bacteria along with antifungal activity against five fungal species. It has been found that tested compounds showed moderate activity.  相似文献

The structures of the novel triazolobenzothiazines 2,4‐dihydro‐1H‐benzo[b][1,2,4]triazolo[4,3‐d][1,4]thiazin‐1‐one (IDPH‐791), C9H7N3OS, (I), a potential muscle relaxant, its benzoyl derivative, 2‐(2‐oxo‐2‐phenylethyl)‐2,4‐dihydro‐1H‐benzo[b][1,2,4]triazolo[4,3‐d][1,4]thiazin‐1‐one, C20H17N3O4S, (II), and the β‐keto ester derivative, ethyl 3‐oxo‐2‐(1‐oxo‐2,4‐dihydro‐1H‐benzo[b][1,2,4]triazolo[4,3‐d][1,4]thiazin‐2‐yl)‐3‐phenylpropanoate, C17H13N3O2S, (III), are the first examples of benzothiazine‐fused triazoles in the crystallographic literature. The heterocyclic thiazine rings in all three structures adopt a distorted half‐chair conformation. Compound (III) exists in the trans‐β‐diketo form. Other than N—H...O hydrogen bonds in (I) forming dimers, no formal intermolecular hydrogen bonds are involved in the crystal packing of any of the three structures, which is dominated by C—H...O/N and π–π stacking interactions.  相似文献

The reaction of 2,3‐dihydro‐2,3‐epoxy‐1,4‐naphthoquinone ( 4 ) with substituted anilines furnished the corresponding benzo[fused]heterocyclic derivatives 5 , 6 , 6a , 6b , 7 , 8 . Furthermore, treatment of benzo[a]phenothiazine derivative 7 with halo compounds, namely, ethyl bromoacetate, phenacyl bromide, dibromoethane, or chloroacetone afforded ether derivatives 11 , 12 , 13 , 14 , respectively. Moreover, the reaction of 11 with o‐substituted aniline gave the corresponding benzo[a]phenothiazin‐5‐one derivatives 15 , 16 , 17 and benzo[d][1,3]oxazin‐4‐one 18 , respectively. Finally, the chromenone derivative 19 was synthesized via the reaction of ester derivative 11 with salicyaldhyde in refluxing pyridine. The newly synthesized compounds were characterized by spectroscopic measurements (IR, 1H NMR, 13C NMR, and mass spectra).  相似文献
