免疫分析在临床医学检测领域具有重要的地位.本课题组提出了基于电感耦合等离子体质谱(ICP-MS)的免疫分析方法,利用元素标记技术结合ICP-MS检测实现多组分免疫分析.随后,研究人员在该领域做了大量研究工作,证明该方法可用于从小分子、蛋白质、核酸到细胞等一系列生物样品的检测.本文综述了基于ICP-MS免疫分析方法的特点,对其发展方向进行了展望,希望为该领域的研究工作提供参考.  相似文献

持久性有机污染物(POPs)可以通过各种环境介质(大气、水、生物体等)长距离迁移并长期存在于环境中,具有生物富集性、毒性、致畸、致癌性,对人类健康和环境具有严重的危害.用于此类物质的分析方法很多,主要可分为仪器联用分析方法和快速分析方法,如:色谱-质谱分析、光谱分析、生物分析、光/电分析、传感分析等.这些方法各有优缺点,互为补充.本文将按照分析方法,重点介绍2000年至今在POPs分析技术上的最新进展.  相似文献

人的大脑中存在数以亿计的神经元,而神经元之间的信息传递主要依赖于通过化学突触发挥作用的神经递质。精确研究神经递质的动态变化对于理解神经系统的功能和研究神经系统疾病的发病机理等均具有重要意义。本文从技术原理、应用以及局限性等方面总结了近年来发展的一系列检测神经递质的方法,着重介绍了基于G蛋白偶联受体(G protein coupled receptor, GPCR)激活原理构建的可遗传编码荧光探针,此系列探针在特异性、灵敏度及时空分辨率方面均有优异的表现,同时,在活体动物实验中也表现出良好的信噪比。此外,此类探针的构建原理具有普适性,可用于开发检测不同神经递质的荧光探针。  相似文献

This paper presents an overview of flow-based methods in food and environmental analysis using chemiluminescence (CL) detection covering the period from 2005 to the present. The review discussses both automated flow methods of analysis [such as flow-injection analysis (FIA), sequential-injection analysis (SIA) and their variants] and separation techniques [liquid chromatography (LC) and capillary electrophoresis (CE) coupled to CL detection]. The acyl-biotinyl exchange (ABE) is widely used for detection of S-palmitoylated proteins by replacing palmitic acid with biotin, which is a common method for detecting S-palmitoylated proteins. In this study, the effects of acetone precipitation and methanol-chloroform precipitation on the detection of S-palmitoylation proteins in acyl-biotin exchange method were compared, and the S-palmitoylated proteins in mouse cardiac tissue were analyzed. First, N-ethylmaleimide (NEM) was used to block free sulfhydryls within protein molecules. Then, biotinylation reagent (HPDP-Biotin) was used to label the newly produced cysteine thiols that were resulted from treatment by hydroxylamine (HA) in mouse heart tissue. During the ABE reaction, excess unreacted NEM, HA and HPDP-Biotin were removed by precipitation of the proteins. Then, the S-palmitoylated proteins from heart tissue were labeled with ABE reaction based on different precipitation methods, and the S-palmitoylated proteins labeled with biotin were enriched by streptavidin agarose beads. The enriched proteins were analyzed by mass spectrometry, and 50 S-palmitoylated proteins were identified. Specifically, 23 S-palmitoylated proteins were identified in acetone precipitation assay group, and 37 S-palmitoylated proteins in the methanol-chloroform precipitation assay group were identified. 10 palmitoylated proteins were identified in both groups. The results showed that the combination of different precipitation methods could be helpful for the identification of palmitoylated proteins.  相似文献

This report presents the single-laboratory validation of a method for the determination of melamine in dairy-based products using on-line turbulent flow extraction-tandem mass spectrometry. Liquid or powder test portions were dissolved in water, enriched with (13)C(3)(15)N(3)-Melamine internal standard, followed by protein precipitation and withdrawal of an aliquot for analysis. The turbulent flow method was validated by analyses of liquid and powdered proficiency test portions containing up to 10mg/kg melamine. Accuracy of results ranged from 96 to 106% of the assigned values for the 6 proficiency test portions tested with relative standard deviations of 4-8%. Apparent recoveries based on addition of amino-(15)N(3)-Melamine to prepared test portions were between 98 and 114%. Based on the repeat analysis of a known blank sample the limit of detection and limit of quantification were determined to be 27 and 87 μg/kg, respectively. Additionally, this report demonstrates that turbulent flow chromatography is significantly faster than traditional LC-MS, with sample analysis times of less than 2 min.  相似文献

The current trend of pesticide immunoassay is developing multi-analyte immunoassays, that is, more than one target can be detected per test. In this mini-review, the strategies to achieve multi-analyte immunoassays, which include multi-antibody strategy, broad-specificity antibody strategy, and others, are briefly introduced. In addition, the recent developments of multi-analyte immunoassays for pesticides were summarized. At last, we give a future outlook of this area, which includes rational design generic hapten with the assist of computer-assisted molecular modeling (CAMM), further improving the properties of broad-specificity antibody by recombinant antibody (rAb) technology and developing a noncompetitive immunoassay format.  相似文献

Abstract

A mass spectrometric method in the negative ion chemical ionization mode was used to analyze α- and β-endosulfan in biological samples. The selected ion recording technique was employed, monitoring the ions at m/z 35 and 37. Biological samples of animals treated with endosulfan were analyzed. The method is selective and sensitive enough to analyze crude extracts of mouse brain and plasma directly without purification, since the chromatograms were clean  相似文献

建立了基于液相色谱-电喷雾串联质谱的分析方法,对生物样本中全基因组DNA甲基化水平进行定量测定.首先将DNA从生物样本中提取出来,将DNA片段酶解为单核苷,利用液相色谱-串联质谱测定胞嘧啶核苷和5-甲基胞嘧啶核苷的含量,从而计算出其全基因组DNA甲基化率.利用该法研究了暴露于全氟辛烷磺酸的L-02细胞、10例原发性肝癌病例血浆样本和10例对照血浆样本的全基因组DNA甲基化水平,得出了它们的总甲基化率变化的初步结果.本方法操作简单,具有很高的灵敏度和稳定性,为研究生物样本,尤其是临床上易得但DNA含量极低的血浆样本的总甲基化水平提供了思路.  相似文献
