Herein we report the development of a photocatalytic strategy for the divergent preparation of functionalized bicyclo[1.1.1]pentylamines. This approach exploits, for the first time, the ability of nitrogen-radicals to undergo strain-release reaction with [1.1.1]propellane. This reactivity is facilitated by the electrophilic nature of these open-shell intermediates and the presence of strong polar effects in the transition-state for C−N bond formation/ring-opening. With the aid of a simple reductive quenching photoredox cycle, we have successfully harnessed this novel radical strain-release amination as part of a multicomponent cascade compatible with several external trapping agents. Overall, this radical strategy enables the rapid construction of novel amino-functionalized building blocks with potential application in medicinal chemistry programs as p-substituted aniline bioisosteres.  相似文献

A new generation of saturated benzene mimetics, 2‐oxabicyclo[2.1.1]hexanes, was developed. These compounds were designed as analogues of bicyclo[1.1.1]pentane with an improved water solubility. Crystallographic analysis of 2‐oxabicyclo[2.1.1]hexanes revealed that they occupy a novel chemical space, but, at the same time, resemble the motif of meta‐disubstituted benzenes.  相似文献

1,3‐Disubstituted bicyclo[1.1.1]pentanes (BCPs) are important motifs in drug design as surrogates for p‐substituted arenes and alkynes. Access to all‐carbon disubstituted BCPs via cross‐coupling has to date been limited to use of the BCP as the organometallic component, which restricts scope due to the harsh conditions typically required for the synthesis of metallated BCPs. Here we report a general method to access 1,3‐C‐disubstituted BCPs from 1‐iodo‐bicyclo[1.1.1]pentanes (iodo‐BCPs) by direct iron‐catalyzed cross‐coupling with aryl and heteroaryl Grignard reagents. This chemistry represents the first general use of iodo‐BCPs as electrophiles in cross‐coupling, and the first Kumada coupling of tertiary iodides. Benefiting from short reaction times, mild conditions, and broad scope of the coupling partners, it enables the synthesis of a wide range of 1,3‐C‐disubstituted BCPs including various drug analogues.  相似文献

1,3‐Disubstituted bicyclo[1.1.1]pentanes (BCPs) are valuable bioisosteres of para‐substituted aromatic rings. The most direct route to these structures is via multicomponent ring‐opening reactions of [1.1.1]propellane. However, challenges associated with these transformations mean that difunctionalized BCPs are more commonly prepared by multistep reaction sequences with BCP‐halide intermediates. Herein, we report three‐ and four‐component 1,3‐difunctionalizations of [1.1.1]propellane with organometallic reagents, organoboronic esters, and a variety of electrophiles. This process is achieved by trapping intermediate BCP‐metal species with boronic esters to form boronate complexes, which are versatile intermediates whose electrophile‐induced 1,2‐metallate rearrangement chemistry enables a broad range of C?C bond‐forming reactions.  相似文献

The realization of controllable multicomponent self‐assembly through reversible supramolecular interactions is a challenging goal, and is an important strategy for the fabrication of switchable nanomaterials. Herein we show that the self‐assembly of TiO2 nanoparticles (NP) functionalized with methyl viologen can be controlled both by light irradiation and chemical reduction through cucurbit[8]uril‐enhanced radical cation dimerization interactions. Moreover, the controlled assembly and disassembly of this system are accompanied by switchable photocatalytic activity of the TiO2 NPs, which shows potential application as a novel smart and recyclable photocatalyst.  相似文献

Bicyclo[1.1.1]pentanes (BCPs) have sparked the interest of medicinal chemists due to their recent discovery as bioisosteres of aromatic rings. To study the biological activity of this relatively new class of bioisosteres, reliable methods to incorporate BCPs into target molecules are in high demand, as reflected by a flurry of methods for BCP synthesis in recent years. In this work, we disclose a general method for the synthesis of BCP‐containing dithianes which, upon deprotection, provide access to BCP analogues of medicinally abundant diarylketones. A broad scope of 2‐aryl‐1,3‐dithianes, including several heterocyclic derivatives, react with [1.1.1]propellane to afford 26 new derivatives in good to excellent yields. Further transformation of the dithiane portion into a variety of functional groups demonstrates the robustness of the products. A computational study indicates that the reaction of 2‐aryl‐1,3‐dithianes and [1.1.1]propellane proceeds via a two‐electron pathway.  相似文献

An efficient and green strategy for the improved synthesis of a biologically and pharmaceutically interesting multi‐functionalized diverse novel spiro‐benzo[a ]phenazine annulated heterocycles was developed with the assistance of microwave irradiation. A sequential one‐pot, two‐step domino reaction starting from 2‐hydroxynaphthalene‐1,4‐dione, benzene‐1,2‐diamines, a cyclic carbonyl compound, and 1,3‐indandione in the presence of a basic ionic liquid (1‐butyl‐3‐methylimidazolium hydroxide) as an expedient, ecofriendly and reusable catalyst afforded the corresponding novel spiro[benzo[a ]indeno[2′,1′:5,6]pyrano[2,3‐c ]phenazine] derivatives with high yield under solvent‐free conditions. This domino Knoevenagel–Michael annulation reaction provided five new bonds (two C–C, two CN, and one C–O) and two new rings through multiple operations in a single flask.  相似文献

In the room‐temperature X‐ray structure of the N‐Boc‐protected derivative of the novel 3‐amino­bi­cyclo[1.1.1]­pentanecarboxyl­ic acid, C11H17NO4, the interbridgehead distance in the bi­cyclo­[1.1.1]­pentane cage is 1.852 (2) Å. The carboxyl and parts of the blocked amino group are almost in plane with one of the cage triangles. N—H?O and O—H?O hydrogen bonds generate infinite corrugated molecular chains in the crystal lattice.  相似文献

1-Aryl-substituted bicyclo[1.1.1]pentanes (BCPs) are an important class of BCP derivatives with widespread application in drug development. Most syntheses of these materials require multiple chemical steps via BCP electrophiles or nucleophiles derived from [1.1.1]propellane. Although one-step, multicomponent radical cross-coupling reactions could provide a more sustainable and rapid route to access diverse heteroarylated BCPs, current approaches are limited to tertiary alkyl radicals, leading to a decrease in their practical value. In this study, a conceptually different approach enabled by a radical multicomponent heteroarylation of [1.1.1]propellane to access functionalized heteroarylated BCPs is described. Importantly, this protocol is compatible with primary-, secondary-, and tertiary aliphatic radicals, as well as various fluoroalkyl radical sources, thus enabling rapid library generation of sought-after BCP derivatives for drug development.  相似文献

A novel strategy to generate functionalized 1‐azatriene intermediates for 6π electrocyclizations was developed by using readily accessible dienyne‐imides and various terminal olefins under PdII catalysis. Taking advantage of the sequential cooperation between preloaded and incorporated functional handles at 1,3‐dien‐5‐yne skeletons, this method not only enables the selective generation of putative 1‐azatrienes but significantly accelerates their thermal 6π‐electrocyclic ring‐closure processes to a series of highly substituted furo[2,3‐b]dihydropyridine derivatives in good yields.  相似文献

These reactions proceeded with a broad range of substrates at room temperature in high yield. A visible‐light‐driven radical‐mediated strategy for the in situ generation of aza‐ortho ‐quinone methides from 2‐vinyl‐substituted anilines and alkyl radical precursors is described. This process enables an efficient multicomponent reaction of 2‐vinylanilines, halides, and sulfur ylides, and has a wide substrate scope and good functional group tolerance. Treatment of the cycloaddition products with a base leads to densely functionalized indoles in a single‐flask operation.  相似文献

We report a general preparation of arylated bicyclo[1.1.1]pentanes through the opening of [1.1.1]propellane with various arylmagnesium halides. After transmetalation with ZnCl2 and Negishi cross‐coupling with aryl and heteroaryl halides, bis‐arylated bicyclo[1.1.1]pentanes are obtained. These bis‐arylated bicyclo[1.1.1]pentanes may be considered as bioisosteres of internal alkynes. Bioisosteres of tazarotene and the metabotropic glutamate receptor 5 (mGluR5) antagonist 2‐methyl‐6‐(phenylethynyl)pyridine were prepared and their physicochemical properties were evaluated.  相似文献

A new synthetic route to functionalized neutral and anionic azadiphospholes from easily accessible starting materials is described. Equimolar reaction of Na(OCP) and N‐(2,6‐dimethylphenyl)pivalimidoyl chloride 2 a cleanly affords the imidoxy‐functionalized 1,2,4‐azadiphosphole 3 a . Using Na(OCP) and imidoyl chloride in a 2:1 ratio leads to an anionic four‐membered ring Na[ 4 a ], which has been structurally characterized. During 16 h at room temperature, Na[ 4 a ] rearranges to the anionic 1,3,4‐azadiphospholide Na[ 5 a ] with release of carbon monoxide. Applying the more sterically demanding N‐(2,6‐diisopropylphenyl)pivalimidoyl chloride allows isolation of the 1,3,4‐azadiphospholide Na[ 5 b ] in good yield (>70 %). Possible mechanisms leading to the new isomeric azadiphospholides have been investigated with the aid of high‐level composite calculations.  相似文献

RhII‐catalyzed intermolecular [3+2] cycloaddition of 2 H‐azirines with N‐sulfonyl‐1,2,3‐triazoles is disclosed, in which a series of fully functionalized pyrroles is produced via rhodium azavinyl carbene intermediates. A distinct feature of this reaction is that the azavinyl carbene serves as a [2 C] equivalent, instead of as [1 C] or aza‐[3 C] synthons, which have been reported previously in cyclopropanations and [3+n] cycloadditions. Moreover, this methodology has also been successfully applied in the total synthesis of URB447 as well as the formal synthesis of Atorvastatin (Lipitor).  相似文献

The synthesis of novel triazolo[1,5‐a]triazin‐7‐ones is presented. Starting from 3‐amino‐5‐sulfanyl‐1,2,4‐triazole, the synthetic sequence involved alkylation with benzyl bromide, reaction with p‐nitrophenyl chloroformate followed by treatment with a primary amine, and condensation with diethoxymethyl acetate. Final oxidation of the thioether moiety with 3‐chloroperbenzoic acid provided 2‐(benzylsulfonyl)[1,2,4]triazolo[1,5‐a][1,3,5]triazin‐7‐ones 5a and 5b in good overall yields. Treatment of 5a and 5b with secondary amines provided highly functionalized [1,2,4]triazoles through an unexpected triazinone ring opening. A mechanism for this transformation is proposed.  相似文献

Fe3O4 magnetic nanoparticles functionalized with 5,10‐dihydropyrido[2,3‐b]quinoxaline‐7,8‐diol were synthesized as was their complex with copper as a novel nanomagnetic iron oxide catalyst via a simple and green method, and characterized using various techniques. The capability of the catalyst was evaluated in the one‐pot three‐component synthesis of different tetrazoles, which showed very good results. Mild reaction conditions, good reusability and simple magnetic work‐up make this methodology interesting for the efficient synthesis of tetrazoles.  相似文献

A highly fluorescent (ΦF=0.60) and water‐soluble two‐dimensional (2D) honeycomb‐shaped supramolecular organic framework (SOF) was successfully synthesized in pure aqueous solution via self‐assembly of novel cyanostilbene‐functionalized trilateral guest molecules and cucurbit[8]uril hosts. The size of this fluorescent 2D SOF was >500 nm in diameter, 1.7 nm in thickness, and 3.9 nm in the honeycomb pore diameter. This 2D SOF holds potential as a new all‐organic photosensitizer template for photocatalytic H2 evolution from pure water.  相似文献
