Soft tissues, such as fat and skin, present high flexibility and are capable of withstanding large deformation in various functions. Hydrogels that can resemble the mechanical performance of soft tissue are unique and widely demanded. In this study, micellar hydrogels based on biocompatible poly(l ‐glutamic acid) (PLGA) were designed with the enhanced capacity to bear large deformation. Amphipathic triblock copolymer poly(ethylene glycol) acrylate‐co‐poly(ε‐caprolactone)‐co‐poly (ethylene glycol) acrylate (APEG‐PCL‐APEG) with two terminal double bonds was synthesized and self‐assembled into micelles. At the same time, graft copolymers, poly(l ‐glutamic acid)‐g‐hydroxyethyl methacrylate (PLGA‐g‐HEMA) with double bonds were synthesized. APEG‐PCL‐APEG micelles and PLGA‐g‐HEMA were mixed to construct micellar hydrogel via radical polymerization. The crystalline structure and hydrophobic aggregation of copolymers (APEG‐PCL‐APEG) were found to associate with PCL molecular weight. Due to the hydrophobic stress dissipation and crystalline structure of the micelles, the softness and toughness of hydrogels were promoted, exhibiting a 25% increase in ultimate strain. Moreover, the micellar hydrogels were able to load proteins with long‐term retention. In addition, under dynamic mechanical stimulation, the release of proteins could be accelerated. Besides, the micellar hydrogels also supported rabbit adipose‐derived stem cells (rASCs) growth, thus exhibiting the potential toward soft tissue engineering. © 2019 Wiley Periodicals, Inc. J. Polym. Programmed deformations are widespread in nature, providing elegant paradigms to design self‐morphing materials with promising applications in biomedical devices, flexible electronics, soft robotics, etc. In this emerging field, hydrogels are an ideal material to investigate the deformation principle and the structure‐deformation relationship. One crucial step is to construct heterogeneous structures in a facile yet effective way. Herein, we provide a focus review on different deformation modes and corresponding structural features of hydrogels. Photolithography is a versatile approach to control the outer shape of the hydrogel and spatial distribution of the component in the hydrogel, endowing the patterned hydrogels with programmed internal stress and thus controllable deformations. Specifically, cooperative deformations take place in periodically patterned hydrogels with in‐plane gradients, and multiple morphing structures are formed in one patterned hydrogel using selective preswelling to direct the buckling of each unit. The structural control strategy and deformation principles should be applicable to other materials with broad applications in diverse areas.  相似文献

Immobilization of compartmentalized microscale objects in 3D hydrogels provides a step towards the modular assembly of soft functional materials with tunable architectures and distributed functionalities. Herein, we report the use of a combination of micro‐compartmentalization, immobilization, and modularization to fabricate and assemble hydrogel‐based microreactor assemblies comprising millions of functionalized polysaccharide–polynucleotide coacervate droplets. The heterogeneous hydrogels can be structurally fused by interfacial crosslinking and coupled as input and output modules to implement a UV‐induced photocatalytic/peroxidation nanoparticle/DNAzyme reaction cascade that generates a spatiotemporal fluorescence read‐out depending on the droplet number density, intensity of photoenergization, and chemical flux. Our approach offers a route to heterogeneous hydrogels with endogenous reactivity and reconfigurable architecture, and provides a step towards the development of soft modular materials with programmable functionality.  相似文献

Supramolecular structures with strain‐stiffening properties are ubiquitous in nature but remain rare in the lab. Herein, we report on strain‐stiffening supramolecular hydrogels that are entirely produced through the self‐assembly of synthetic molecular gelators. The involved gelators self‐assemble into semi‐flexible fibers, which thereby crosslink into hydrogels. Interestingly, these hydrogels are capable of stiffening in response to applied stress, resembling biological intermediate filaments system. Furthermore, strain‐stiffening hydrogel networks embedded with liposomes are constructed through orthogonal self‐assembly of gelators and phospholipids, mimicking biological tissues in both architecture and mechanical properties. This work furthers the development of biomimetic soft materials with mechanical responsiveness and presents potentially enticing applications in diverse fields, such as tissue engineering, artificial life, and strain sensors.  相似文献

Conductive hydrogels with ionic compounds possess great potential for the development of soft smart devices. A dielectric scarfskin is typically required for these devices to prevent short circuiting, leading to devices with lower stretchability than the hydrogel. Henceforth, commonly used dielectric materials, such as PDMS and Ecoflex, cannot be largely stretched. Hydrogel devices with ultrastretchability are required to accommodate hostile application environments. Herein, we propose a hydrogel fiber coated with a dielectric layer that can be stretched to over 2000% of its initial length. The fiber remains conductive when stretched to ~1300%. In addition, the core/sheath hydrogel fiber can be endowed with a variety of functional properties, such as electroluminescence (EL), photoluminescence (PL), and magnetic‐responsiveness, demonstrating scalability of the resultant fiber. A novel class of functional poly(N-isopropylacrylamide) (PNIPAAm) hydrogels with pendent micellar structure resulting from the pending amphiphilic polymers was designed and prepared. The influence of the pendent micellar structure on the properties of the resulted PNIPAAm hydrogels was examined in terms of morphology observed by scanning electron microscopy, thermal response through differential scanning calorimetry, and deswelling/reswelling kinetics upon external temperature changes. In comparison with the conventional ones, the novel PNIPAAm hydrogels with pendent micellar structure presented improved temperature-sensitive properties, i.e., enlarged water containing capability at room temperature, as well as improved deswelling rate upon heating.  相似文献

Local, micromechanical environment is known to influence cellular function in heterogeneous hydrogels, and knowledge gained in micromechanics will facilitate the improved design of biomaterials for tissue regeneration. In this study, a system comprising microstructured resilin‐like polypeptide (RLP)–poly(ethylene glycol) (PEG) hydrogels is utilized. The micromechanical properties of RLP‐PEG hydrogels are evaluated with oscillatory shear rheometry, compression dynamic mechanic analysis, small‐strain microindentation, and large‐strain indentation and puncture over a range of different deformation length scales. The measured elastic moduli are consistent with volume averaging models, indicating that volume fraction, not domain size, plays a dominant role in determining the low strain mechanical response. Large‐strain indentation under a confocal microscope enables the visualization of the microstructured hydrogel micromechanical deformation, emphasizing the translation, rotation, and deformation of RLP‐rich domains. The fracture initiation energy results demonstrate that failure of the composite hydrogels is controlled by the RLP‐rich phase, and their independence with domain size suggested that failure initiation is controlled by multiple domains within the strained volume. This approach and findings provide new quantitative insight into the micromechanical response of soft hydrogel composites and highlight the opportunities in employing these methods to understand the physical origins of mechanical properties of soft synthetic and biological materials.  相似文献

Thermosensitive guar‐based hydrogels are obtained in water solutions by copper‐catalyzed 1,3‐dipolar cycloaddition between alkyne‐functionalized guars and α,ω‐diazido‐poly[(ethylene glycol)‐co‐(propylene glycol)]. Characterization by TGA, HR‐MAS 1H NMR, and rheology have shown that hydrogels with tunable physico‐chemical properties, such as crosslinking density, viscoelasticity, swelling ratio, and so forth, could be obtained by varying the guar molar mass, the degree of alkyne functionality, the guar/crosslinker weight ratio, and the reaction temperature. Based on swelling measurements, it has been shown that the thermal sensitiveness of guar‐based hydrogels is fast, reversible, and intimately related to the weight fraction of the thermosensitive crosslinker in the network. Finally, the monitoring of doxorubicin hydrochloride release has demonstrated the potential of these hydrogels as temperature‐dependent drug release devices. The robust, efficient, and orthogonal approach described herein represents a general approach towards the development of well‐controlled guar‐based hydrogels using α,ω‐diazido crosslinkers. © 2010 Wiley Periodicals, Inc. Three‐dimensional porous graphene hydrogels have been prepared by a green and facile but very efficient approach using glucose as an assistant. Based on a one‐step hydrothermal reaction with optimal experimental conditions such as the reaction time and temperature, the graphene hydrogels exhibit a superior electrical conductivity (95.3 S/m) and can be used as supercapacitor electrode without any binder or conducting additives but showing a high specific capacitance of 384.6 F/g at a current density of 1 A/g. The results show that addition of glucose can not only greatly decrease the reaction temperature but also shorten the reaction time. The superior performance of the three‐dimensional porous graphene hydrogels as electrode for supercapacitor suggests its promising potentials in the field of energy storage devices.  相似文献

In this study, fracture toughness of nanocomposite hydrogels is quantified, and active mechanisms for dissipation of energy of nanocomposite hydrogels are ascertained. Poly(N,N‐dimethylacrylamide) nanocomposite hydrogels are prepared by in situ free radical polymerization with the incorporation of Laponite, a hectorite synthetic clay. Transmission electron microscopy proves exfoliation of clay platelets that serve as multifunctional crosslinkers in the created physical network. Extraordinary high fracture energies of up to 6800 J m?2 are determined by the pure shear test approach, which shows that these soft and stretchable hydrogels are insensitive to notches. In contrast to single‐ and double‐network hydrogels, dynamic mechanic analysis and stress relaxation experiments clarify that significant viscoelastic dissipation occurs during deformation of nanocomposite hydrogels. Similar to double‐network hydrogels, crack tip blunting and plastic deformation also contribute to the observed massive fracture energies. © 2015 Wiley Periodicals, Inc. J. Polym. Rational design and development of tailorable simple synthesis process remains a centerpiece of investigational efforts toward engineering advanced hydrogels. In this study, a green and scalable synthesis approach is developed to formulate a set of gelatin‐based macroporous hybrid hydrogels. This approach consists of four sequential steps starting from liquid‐phase pre‐crosslinking/grafting, unidirectional freezing, freeze‐drying, and finally post‐curing process. The chemical crosslinking mainly involves between epoxy groups of functionalized polyethylene glycol and functional groups of gelatin both in liquid and solid state. Importantly, this approach allows to accommodate different polymers, chitosan or hydroxyethyl cellulose, under identical benign condition. Structural and mechanical anisotropy can be tuned by the selection of polymer constituents. Overall, all hydrogels show suitable structural stability, good swellability, high porosity and pore interconnectivity, and maintenance of mechanical integrity during 3‐week‐long hydrolytic degradation. Under compression, hydrogels exhibit robust mechanical properties with nonlinear elasticity and stress‐relaxation behavior and show no sign of mechanical failure under repeated compression at 50% deformation. Biological experiment with human bone marrow mesenchymal stromal cells (hMSCs) reveals that hydrogels are biocompatible, and their physicomechanical properties are suitable to support cells growth, and osteogenic/chondrogenic differentiation, demonstrating their potential application for bone and cartilage regenerative medicine toward clinically relevant endpoints.  相似文献

This work aims at developing an approach to Ru(II)(Tpy)2‐functionalized hydrogels and exploring the coupling of the hydrogels with the Belousov‐Zhabotinsky (BZ) reaction. Based on free radical polymerization, two synthetic routes are developed. The first one is the direct gelation by copolymerization of acrylamide as hydrophilic component and Ru(II)(Tpy)2 as the functional group. The second one is carried out through a combined approach. A terpyridine‐containing hydrogel is first prepared and then post‐functionalized by coordination between Ru(III)(Tpy)Cl3 and terpyridine groups in the hydrogel network. Utilizing the synthetic hydrogels, the reversible redox responsiveness, the coupling with the BZ reaction, the occurrence and the self‐oscillating properties of the BZ reaction in the hydrogel networks are studied. © 2015 Wiley Periodicals, Inc. Soft conducting materials in the shape of microfibers with various functional geometries are crucial for soft electronics. To develop highly stretchable conducting microfibers, a microfluidic method is used to prepare hydrogels in a double-network structure. Based on the coagulation of chitosan in cold water and simultaneous photopolymerization and photocrosslinking of N-isopropylacrylamide and N-diethylacrylamide, long microfibers with controlled uniform diameters can be obtained at the junction of a coaxially aligned microchannel device. After further reinforcement of the chitosan chain and exchange of the medium of the hydrogel microfiber with an aqueous electrolyte of lithium bis(trifluoromethanesulfonyl)imide, the prepared ionic hydrogel exhibits high conductivity and stretchability and dry-free properties. Owing to its mechanical robustness and ionic conductivity, we envision a highly stretchable soft electrode with the prepared ionic hydrogel microfiber that can be stretched up to 900%. This fiber has potential for applications in soft electronics and wearable devices.  相似文献

Despite their remarkably mechanical features, tough hydrogels exhibit two flaws (freezing around the icing temperatures of water and drying under arid conditions). Inspired by cryoprotectants (CPAs) used in the inhibition of the icing of water in biological samples, a versatile and straightforward method is reported to fabricate extreme anti‐freezing, non‐drying CPA‐based organohydrogels with long‐term stability by partially displacing water molecules within the pre‐fabricated hydrogels. CPA‐based Ca‐alginate/polyacrylamide (PAAm) tough hydrogels were successfully fabricated with glycerol, glycol, and sorbitol. The CPA‐based organohydrogels remain unfrozen and mechanically flexible even up to −70 °C and are stable under ambient conditions or even vacuum.  相似文献

This Review presents polysaccharides, oligosaccharides, nucleic acids, peptides, and proteins as functional stimuli‐responsive polymer scaffolds that yield hydrogels with controlled stiffness. Different physical or chemical triggers can be used to structurally reconfigure the crosslinking units and control the stiffness of the hydrogels. The integration of stimuli‐responsive supramolecular complexes and stimuli‐responsive biomolecular units as crosslinkers leads to hybrid hydrogels undergoing reversible triggered transitions across different stiffness states. Different applications of stimuli‐responsive biomolecule‐based hydrogels are discussed. The assembly of stimuli‐responsive biomolecule‐based hydrogel films on surfaces and their applications are discussed. The coating of drug‐loaded nanoparticles with stimuli‐responsive hydrogels for controlled drug release is also presented.  相似文献

Hydrogels are hydrophilic, porous polymer networks that can absorb up to thousands of times their own weight in water. They have many potential applications, one of which is the encapsulation of freestanding black lipid membranes (BLMs) for novel separation technologies or biosensor applications. We investigated gels for in situ encapsulation of multiple BLMs formed across apertures in a hydrophobic ethylene tetrafluoroethylene (ETFE) support. The encapsulation gels consisted of networks of poly(ethylene glycol)‐dimethacrylate or poly(ethylene glycol)‐diacrylate polymerized using either a chemical initiator or a photoinitiator. The hydrogels were studied with regards to volumetric stability, porosity, and water permeability. All hydrogels had pore sizes around 7 nm with volumetric changes >2% upon crosslinking. Photoinitiated hydrogels had a lower hydraulic water permeability compared to chemically initiated hydrogels; however, for all hydrogels the permeability was several‐fold higher than the water permeability of conventional reverse osmosis (RO) membranes. Lifetimes of freestanding BLM arrays in gel precursor solutions were short compared to arrays formed in buffer. However, polymerizing (crosslinking) the gel stabilized the membranes and resulted in BLM arrays that remained intact for days. This is a substantial improvement over lifetimes for freestanding BLM arrays. Optical images of the membranes and single channel activity of incorporated gramicidin ion channels showed that the lipid membranes retained their integrity and functionality after encapsulation with hydrogel. Our results show that hydrogel encapsulation is a potential means to provide stability for biomimetic devices based on functional proteins reconstituted in biomimetic membrane arrays. A generally applicable strategy to obtain mechanically robust hydrogels for the incorporation and containment of functional reporter bacteria for the microarray and microparticle‐based detection and signaling of N‐acyl homoserine lactone autoinducers (3OC12HSL) at relevant concentrations is reported. For reinforcing hydrogels of 1,4‐bi(phenylalanine‐diglycol)‐benzene (PDB), a hybrid hydrogel is formed by the combination of PDB self‐assembly with Ca2+ mediated alginate crosslinking. The different assembly mechanisms are shown not to interfere with each other and despite the more than four‐fold increased moduli of the hydrogels, diffusion of autoinducers into the gels remains efficient and Escherichia coli pLuxR‐green fluorescent protein (GFP) reporter bacteria are proliferating. Templating affords reporter bacteria‐loaded hydrogels with controllable shape and size. Upon exposure to 3OC12HSL, the embedded bacteria exhibit an up to 12 ± 3 times increase in fluorescence intensity due to autoinducer‐triggered GFP expression. This approach can serve as a potentially generally applicable strategy to sensitively detect bacteria via their secreted autoinducers.  相似文献
