Neutrophils can responsively release reactive oxygen species (ROS) to actively combat infections by exogenous stimulus and cascade enzyme catalyzed bio-oxidation. A supramolecular nanogel is now used as an artificial neutrophil by enzymatic interfacial self-assembly of peptides (Fmoc-Tyr(H2PO3)-OH) with magnetic nanoparticles (MNPs) and electrostatic loading of chloroperoxidase (CPO). The MNPs within the nanogel can elevate H2O2 levels in cancer cells under programmed alternating magnetic field (AMF) similar to the neutrophil activator, and the loaded CPO within protective peptides nanolayer converts the H2O2 into singlet oxygen (1O2) in a sustained manner for neutrophil-inspired tumor therapy. As a proof of concept study, both the H2O2 and 1O2 in cancer cells increase stepwise under a programmed alternating magnetic field. An active enzyme dynamic therapy by magnetically stimulated oxygen stress and sustained enzyme bio-oxidation is thus shown with studies on both cells and animals.  相似文献

A simple and efficient procedure has been developed for the synthesis of biologically relevant 2‐substituted benzimidazoles through a one‐pot condensation of o‐phenylenediamines with aryl aldehydes catalysed by iron oxide magnetic nanoparticles (Fe3O4 MNPs) in short reaction times with excellent yields. In the present study, Fe3O4 MNPs synthesized in a green manner using aqueous extract of white tea (Camelia sinensis) (Wt‐Fe3O4 MNPs) were applied as a magnetically separable heterogeneous nanocatalyst to synthesize 2‐(4‐chlorophenyl)‐1H–benzo[d]imidazole which has potential application in pharmacology and biological systems. Fourier transform infrared and NMR spectroscopies were used to characterize the 2‐(4‐chlorophenyl)‐1H–benzo[d]imidazole. In vitro cytotoxicity studies on MOLT‐4 cells showed a dose‐dependent toxicity with non‐toxic effect of 2‐(4‐chlorophenyl)‐1H–benzo[d]imidazole, up to a concentration of 0.147 µM. The green synthesized Wt‐Fe3O4 MNPs as recyclable nanocatalyst could be used for further research on the synthesis of therapeutic materials, particularly in nanomedicine, to assist in the treatment of cancer.  相似文献

The development of magnetic nanoparticles (MNPs) with functional groups has been intensively pursued in recent years. Herein, a simple, versatile, and cost‐effective strategy to synthesize water‐soluble and amino‐functionalized MNPs, based on the thermal decomposition of phthalimide‐protected metal–organic precursors followed by deprotection, was developed. The resulting amino‐functionalized Fe3O4, MnFe2O4, and Mn3O4 MNPs with particle sizes of about 14.3, 7.5, and 6.6 nm, respectively, had narrow size distributions and good dispersibility in water. These MNPs also exhibited high magnetism and relaxivities of r2=107.25 mM?1 s?1 for Fe3O4, r2=245.75 mM?1 s?1 for MnFe2O4, and r1=2.74 mM?1 s?1 for Mn3O4. The amino‐functionalized MNPs were further conjugated with a fluorescent dye (rhodamine B) and a targeting ligand (folic acid: FA) and used as multifunctional probes. Magnetic resonance imaging and flow‐cytometric studies showed that these probes could specifically target cancer cells overexpressing FA receptors. This new protocol opens a new way for the synthesis and design of water‐soluble and amino‐functionalized MNPs by an easy and versatile route.  相似文献

Exploration of new property/function of nanomaterials is always a strong impetus in the nanoscience field. Here, a new method of electrochemical conversion (ECC) of magnetic nanoparticles (MNPs) is proposed to endow MNPs with signal generation ability for sensing. Briefly, high potential was applied to split H2O to generate acid, while Fe3O4 MNPs reacted with H+ and produce ferric/ferrous ions, which further reacted with K4Fe(CN)6 to yield Prussian blue (PB) through potential cycling. The ECC method worked well on both home‐made and commercial MNPs with different sizes. The generated PB possessed strong electrochemical activity for further applications. Interestingly, an uneven deposition of PB on working electrode and undesired contamination of the reference and counter electrodes were found when using commercial integrated three‐electrode chip. A 3D‐printed electrochemical cell was designed to facilitate the ECC and avoid drawbacks of commercial integrated electrode. The 3D‐printed electrochemical cell was proven to solve the problem above through spatial separation of electrodes and thus facilitated the ECC process. An electrochemical sensor for H2O2 detection based on the catalysis ability of ECC‐based PB exhibited a linear response from 5 μM to 1 mM, a high sensitivity of 269 μA mM?1 cm?2 and a low detection limit of 0.16 μM (S/N=3), which suggests its promising application prospect in electrochemistry‐related analysis.  相似文献

The exposure of peptides and proteins to reactive hydroxyl radicals results in covalent modifications of amino acid side‐chains and protein backbone. In this study we have investigated the oxidation the isomeric peptides tyrosine–leucine (YL) and leucine–tyrosine (LY), by the hydroxyl radical formed under Fenton reaction (Fe2+/H2O2). Through mass spectrometry (MS), high‐performance liquid chromatography (HPLC‐MS) and electrospray tandem mass spectrometry (HPLC‐MSn) measurements, we have identified and characterized the oxidation products of these two dipeptides. This approach allowed observing and identifying a wide variety of oxidation products, including isomeric forms of the oxidized dipeptides. We detected oxidation products with 1, 2, 3 and 4 oxygen atoms for both peptides; however, oxidation products with 5 oxygen atoms were only present in LY. LY dipeptide oxidation leads to more isomers with 1 and 2 oxygen atoms than YL (3 vs 5 and 4 vs 5, respectively). Formation of the peroxy group occurred preferentially in the C‐terminal residue. We have also detected oxidation products with double bonds or keto groups, dimers (YL–YL and LY–LY) and other products as a result of cross‐linking. Both amino acids in the dipeptides were oxidized although the peptides showed different oxidation products. Also, amino acid residues have shown different oxidation products depending on the relative position on the dipeptide. Results suggest that amino acids in the C‐terminal position are more prone to oxidation. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

In this research, green procedure was employed for biosynthesis of magnetic nanoparticles of iron oxide (Fe3O4‐MNPs) by reduction of ferric chloride solution with Orange peel water extract. Also, dihydro‐2H‐cyclopenta[d][1,3]oxazole was generated through multicomponent reaction of 1,3‐oxazole‐2(3H)‐thione, dialkyl acetylenedicarboxylates, α‐haloketones, and Fe3O4‐MNPs as catalyst at ambient temperature in good yield. Initially, 1,3‐oxazole‐2(3H)‐thione derivatives as one of the precursors are produced through the reaction of alkyl bromides, isothiocyanate, sodium hydride, and Fe3O4‐MNPs as catalyst water at ambient temperature in 83–95% yields. Also, diphenyl‐picrylhydrazine radical trapping and ferric reduction activity potential assays are used for evaluation of antioxidant activity of some synthesized compounds. Among investigated compounds, 4b has good power for radical trapping activity and 4d has good reduction power to butylated hydroxytoluene and 2‐tert‐butylhydroquinone.  相似文献

Fe3O4 magnetic nanoparticles (MNPs) were functionalized by aminopropylsilane and reacted with aromatic aldehyde, and Fe3O4‐Si‐[CH2]3‐N=CH‐Aryl and Fe3O4‐Si‐(CH2)3‐NH‐CH2‐Aryl MNPs were prepared as novel magnetic nanocatalysts. Fourier transform infrared (FT‐IR), X‐ray diffraction (XRD), and scanning and transmission electron microscopy (SEM and TEM) were used to identify the MNPs. The catalytic activity of the MNPs was evaluated in the one‐pot synthesis of some novel poly‐substituted pyridine derivatives.  相似文献

The use of nanobiocatalysts, with the combination of nanotechnology and biotechnology, is considered as an exciting and rapidly emerging area. The use of iron oxide magnetic nanoparticles, as enzyme immobilization carriers, has drawn great attention because of their unique properties, such as controllable particle size, large surface area, modifiable surface, and easy recovery. In this study, various γ‐Fe2O3/Fe3O4 magnetic nanoparticles with immobilized proteases were successfully prepared by three different immobilization strategies including A) direct binding, B) with thiophene as a linker, and C) with triazole as a linker. The oligopeptides syntheses catalyzed by these magnetic nanoparticles (MNPs) with immobilized proteases were systematically studied. Our results show that i) for magnetic nanoparticles immobilized α‐chymotrypsin, both immobilization strategies A and B furnished good reusability for the Z‐Tyr‐Gly‐Gly‐OEt synthesis, the MNPs enzymes can be readily used at least five times without significant loss of its catalytic performance: ii) In the case of Z‐Asp‐Phe‐OMe synthesis catalyzed by magnetic nanoparticles immobilized thermolysin, immobilization Strategy B provided the best recyclability: iii) For the immobilized papain, although Strategy A or B afforded an immobilized enzyme for the first cycle of Z‐Ala‐Leu‐NHNHPh synthesis in good yield, their subsequent catalytic activity decreased rapidly. In general, the γ‐Fe2O3 MNPs were better for use as an immobilization matrix, rather than the Fe3O4 MNPs, owing to their smaller particle size and higher surface area.  相似文献

A bio‐photoelectrochemical cell (BPEC) based on a fuel‐free self‐circulation water–oxygen–water system was fabricated. It consists of Ni:FeOOH modified n‐type bismuth vanadate (BiVO4) photoanode and laccase catalyzed biocathode. In this BPEC, irradiation of the photoanode generates photocurrent for photo‐oxidation of water to oxygen, which is reduced to water again at the laccase biocathode. Of note, the by‐products of two electrode reactions could continue to be reacted, which means the H2O and O2 molecules are retained in an infinite loop of water–oxygen–water without any sacrificial chemical components. As a result, the assembled fuel‐free BPEC exhibits good performance with an open‐circuit potential of 0.97 V and a maximum power density of 205 μW cm?2 at 0.44 V. This BPEC based on a self‐circulation system offers a fuel‐free model to enhance multiple energy conversion and application in reality.  相似文献

We report a simple, mild, and synthetically clean approach to accelerate the rate of enzymatic oxidation reactions by a factor of up to 100 when compared to conventional batch gas/liquid systems. Biocatalytic decomposition of H2O2 is used to produce a soluble source of O2 directly in reaction media, thereby enabling the concentration of aqueous O2 to be increased beyond equilibrium solubility under safe and practical conditions. To best exploit this method, a novel flow reactor was developed to maximize productivity (g product L?1 h?1). This scalable benchtop method provides a distinct advantage over conventional bio‐oxidation in that no pressurized gas or specialist equipment is employed. The method is general across different oxidase enzymes and compatible with a variety of functional groups. These results culminate in record space‐time yields for bio‐oxidation.  相似文献

The incorporation of highly active but also highly sensitive catalysts (e.g. the [FeFe] hydrogenase from Desulfovibrio desulfuricans) in biofuel cells is still one of the major challenges in sustainable energy conversion. We report the fabrication of a dual‐gas diffusion electrode H2/O2 biofuel cell equipped with a [FeFe] hydrogenase/redox polymer‐based high‐current‐density H2‐oxidation bioanode. The bioanodes show benchmark current densities of around 14 mA cm?2 and the corresponding fuel cell tests exhibit a benchmark for a hydrogenase/redox polymer‐based biofuel cell with outstanding power densities of 5.4 mW cm?2 at 0.7 V cell voltage. Furthermore, the highly sensitive [FeFe] hydrogenase is protected against oxygen damage by the redox polymer and can function under 5 % O2.  相似文献

A novel chiral magnetic nanocatalyst was prepared by the surface modification of Fe3O4 magnetic nanoparticles (MNPs) with a chloropropylsilane and further by arginine to form Fe3O4@propylsilan‐arginine (Fe3O4@PS‐Arg). After the structural confirmation of Fe3O4@PS‐Arg synthesized MNPs by Fourier transform‐infrared, X‐ray diffraction, field emission‐scanning electron microscopy, transmission electron microscopy, vibrating‐sample magnetometry and thermogravimetric analyses, their catalytic activity was evaluated for one‐pot enantioselective synthesis of 3‐amino‐1‐aryl‐1H‐benzo[f]chromene‐2‐carbonitrile derivatives. The results showed that in the presence of 0.07 g Fe3O4@PS‐Arg nanocatalyst and ethanol as solvent, the best reaction yield (96%) was obtained in the least time (5 min). Easy operation, reusability and stability, short reaction time, high reaction yields and good enantioselectivity are the major advantages of the newly synthesized nanocatalyst. Also, this study provides a novel strategy for further research and investigation on the synthesis of new reusable enantioselective catalysts and chiral compounds.  相似文献

We developed a novel iron‐tetrasulfophthalocyanine‐graphene‐Nafion (FeTSPc‐GR‐Nafion) modified screen‐printed electrode to determine hydrogen peroxide (H2O2) with high sensitivity and selectivity. The nanocomposite film (FeTSPc‐GR‐Nafion) exhibits an excellent electrocatalytic activity towards oxidation of H2O2 at a potential of +0.35 V in the absence of enzyme. A comparative study reveals that the FeTSPc‐GR complexes play a dual amplification role. Amperometric experiment indicates that the sensors possess good sensitivity and selectivity, with a linear range from 2.0×10?7 M to 5.0×10?3 M and a detection limit of 8.0×10?8 M. This sensor has been successfully used to develop the glucose biosensor and has also been applied to determine H2O2 in sterile water.  相似文献

Non‐aqueous Li–O2 batteries are promising for next‐generation energy storage. New battery chemistries based on LiOH, rather than Li2O2, have been recently reported in systems with added water, one using a soluble additive LiI and the other using solid Ru catalysts. Here, the focus is on the mechanism of Ru‐catalyzed LiOH chemistry. Using nuclear magnetic resonance, operando electrochemical pressure measurements, and mass spectrometry, it is shown that on discharging LiOH forms via a 4 e oxygen reduction reaction, the H in LiOH coming solely from added H2O and the O from both O2 and H2O. On charging, quantitative LiOH oxidation occurs at 3.1 V, with O being trapped in a form of dimethyl sulfone in the electrolyte. Compared to Li2O2, LiOH formation over Ru incurs few side reactions, a critical advantage for developing a long‐lived battery. An optimized metal‐catalyst–electrolyte couple needs to be sought that aids LiOH oxidation and is stable towards attack by hydroxyl radicals.  相似文献

A new type of chiral magnetic nanoparticle was prepared from covalently linked magnetic nanoparticles (Fe3O4) and heptakis‐(6‐O‐triisopropylsilyl)‐β‐cyclodextrin (6‐TIPS‐β‐CD). The resulting selectors (TIPS‐β‐CD‐MNPs) combined the good magnetic properties Fe3O4 and efficient chiral recognition ability of 6‐TIPS‐β‐CD. The enantioselectivity of TIPS‐β‐CD‐MNPs towards 1‐(1‐naphthyl)ethylamine was six times higher than that of the parent β‐CD modified Fe3O4 particles.  相似文献

Biofuel cells based on electrocatalytic oxidation of NADH and reduction of H2O2 have been prepared using carbon fiber electrodes functionalized with graphene nano‐flakes. The electrochemical oxidation of NADH was catalyzed by Meldola's blue (MB), while the reduction of H2O2 was catalyzed by hemin, both catalysts were adsorbed on the graphene flakes due to their π‐π staking. In the next set of experiments, the MB‐ and hemin‐electrodes were additionally modified with glucose dehydrogenase (GDH) and glucose oxidase (GOx), respectively. The enzyme catalyzed reactions in the presence of glucose, NAD+ and O2 resulted in the production of NADH and H2O2 in situ. The produced NADH and H2O2 were oxidized and reduced, respectively, at the bioelectrocatalytic electrodes, thus producing voltage and current generated by the biofuel cell. The enzyme‐based biofuel cells operated in a human serum solution modelling an implantable device powered from the natural biofluid. Finally, two enzyme‐based biofuel cell connected in series and operating in the serum solution produced electrical power sufficient for activation of an electronic watch used as an example device.  相似文献

A simple electrochemical approach to evaluate oxygen reduction catalysts using an inexpensive screen‐printed ring disk carbon electrode system, consisting of a ring electrode deposited with MnO2 and a disk electrode modified with the catalysts for study, is developed in this study. The as‐prepared MnO2 is selective and sensitive for H2O2 oxidation in the presence of O2 and is crucial to the proposed approach. By coupling with a wall‐jet electrochemical cell, the product generated from the reduction reaction at the disk electrode can effectively be monitored at the MnO2‐deposited ring electrode. Model catalysts of nano‐Au and nano‐Pd representing 2e? reduction of O2 to H2O2 and 4e? reduction to H2O, respectively, were evaluated as electrode materials in oxygen reduction reaction to demonstrate the applicability of the proposed method.  相似文献

A biomimetic nickel bis‐diphosphine complex incorporating the amino acid arginine in the outer coordination sphere was immobilized on modified carbon nanotubes (CNTs) through electrostatic interactions. The functionalized redox nanomaterial exhibits reversible electrocatalytic activity for the H2/2 H+ interconversion from pH 0 to 9, with catalytic preference for H2 oxidation at all pH values. The high activity of the complex over a wide pH range allows us to integrate this bio‐inspired nanomaterial either in an enzymatic fuel cell together with a multicopper oxidase at the cathode, or in a proton exchange membrane fuel cell (PEMFC) using Pt/C at the cathode. The Ni‐based PEMFC reaches 14 mW cm−2, only six‐times‐less as compared to full‐Pt conventional PEMFC. The Pt‐free enzyme‐based fuel cell delivers ≈2 mW cm−2, a new efficiency record for a hydrogen biofuel cell with base metal catalysts.  相似文献

Nanocellulose (NC) materials have some unique properties, which make them attractive as organic or inorganic supports for catalytic applications. Nanocatalysts with diameters of less than 100 nm are difficult to separate from the reaction mixture, therefore, magnetic nanoparticles (MNPs) were used as catalysts to overcome this problem. Fe3O4@NCs/BF0.2 as a green, bio‐based, eco‐friendly, and recyclable catalyst was synthesized and characterized using fourier‐transform infrared spectroscopy (FT‐IR), vibrating sample magnetometer (VSM), X‐ray diffraction (XRD), X‐ray fluorescence (XRF), Brunauer–Emmett–Teller (BET), field emission scanning electron microscopy (FESEM), transmission electron microscopy (TEM), and thermal gravimetric analysis (TGA) techniques. Fe3O4@NCs/BF0.2 was employed for the synthesis of 2,3‐dihydro‐1H‐perimidine derivatives via a reaction of 1,8‐diaminonaphthalene with various aldehydes at room temperature under solvent‐free conditions. The present procedure offers several advantages including a short reaction time, excellent yields, easy separation of catalyst, and environmental friendliness.  相似文献

Li‐O2 batteries are promising energy storage systems due to their ultra‐high theoretical capacity. However, most Li‐O2 batteries are based on the reduction/oxidation of Li2O2 and involve highly reactive superoxide and peroxide species that would cause serious degradation of cathodes, especially carbon‐based materials. It is important to explore lithium‐oxygen reactions and find new Li‐O2 chemistry which can restrict or even avoid the negative influence of superoxide/peroxide species. Here, inspired by enzyme‐catalyzed oxygen reduction/oxidation reactions, we introduce a copper(I) complex 3 N‐CuI (3 N=1,4,7‐trimethyl‐1,4,7‐triazacyclononane) to Li‐O2 batteries and successfully modulate the reaction pathway to a moderate one on reversible cleavage/formation of O?O bonds. This work demonstrates that the reaction pathways of Li‐O2 batteries could be modulated by introducing an appropriate soluble catalyst, which is another powerful choice to construct better Li‐O2 batteries.  相似文献
