Determining whether a protein regulates its net electrostatic charge during electron transfer (ET) will deepen our mechanistic understanding of how polypeptides tune rates and free energies of ET (e.g., by affecting reorganization energy, and/or redox potential). Charge regulation during ET has never been measured for proteins because few tools exist to measure the net charge of a folded protein in solution at different oxidation states. Herein, we used a niche analytical tool (protein charge ladders analyzed with capillary electrophoresis) to determine that the net charges of myoglobin, cytochrome c, and azurin change by 0.62±0.06, 1.19±0.02, and 0.51±0.04 units upon single ET. Computational analysis predicts that these fluctuations in charge arise from changes in the pKa values of multiple non‐coordinating residues (predominantly histidine) that involve between 0.42–0.90 eV. These results suggest that ionizable residues can tune the reactivity of redox centers by regulating the net charge of the entire protein–cofactor–solvent complex.  相似文献

The dinuclear complex [Cu2(HL)2(H2O)2](ClO4)2 ( 1 ) [H2L = 5′‐(pyridin‐2‐yl)‐1‐H,2′‐H‐3, 3′‐bis(1, 2,4‐triazole)] was obtained and fully characterized. It exhibits a centrosymmetry configuration, in which each copper(II) ion is pentacoordinate with four nitrogen atoms of two triazole ligands and one oxygen atom from a water molecule. The net atomic charges distribution and atomic orbital contribution to frontier molecular orbitals were obtained using the Gaussian 98 program with Hartree‐Fock method at LANL2DZ level, indicating that the copper(II) ion has the potential to accept the electron of O2 · –. The complex showed quasi‐reversible one‐electron CuII/CuI redox waves with redox potentials of –0.034 V. The SOD‐like activity (IC50) of 1 was measured to be 0.18 ± 0.01 μM by xanthine/xanthine oxidase‐NBT assay at pH 7.8. The relatively high SOD activity suggests that the positive charge of protonated triazole can effectively steer O2 · – to and from the active copper ion.  相似文献

铜锌超氧化物歧化酶(CuZnSOD)作为一种抗氧化酶, 最重要的功能是催化超氧阴离子歧化为过氧化氢和氧气。然而最近研究发现CuZnSOD具有过氧化物酶活性,能导致核酸、蛋白质和细胞膜的损伤。本工作采用光谱学和酶学方法研究外源Cu(Ⅱ)与CuZnSOD之间的相互作用,以及H2O2存在下外源Cu(Ⅱ)对 CuZnSOD断裂DNA活性的增强效应。比较CuZnSOD + nCu(Ⅱ) (n=0, 1, 2, 4, 6, 8)和单独Cu(Ⅱ)分别断裂DNA的活性,表明外源Cu(Ⅱ)的加入可显著增强CuZnSOD断裂DNA的活性。相对酶活力和稳态动力学的测定证实了这种增强效应。pH依赖性实验表明断裂DNA的最适pH范围为pH3.6-5.6和pH9.0-10,在不同的pH区域CuZnSOD + nCu(Ⅱ)断裂DNA途径可能不同。  相似文献

Direct electron transfer of immobilized copper, zinc‐superoxide dismutase (SOD) onto electrodeposited nickel‐oxide (NiOx) nanoparticle modified glassy carbon (GC) electrode displays a well defined redox process with formal potential of ?0.03 V in pH 7.4. Cyclic voltammetry was used for deposition of (NiOx) nanoparticles and immobilization of SOD onto GC electrode. The surface coverage (Γ) and heterogeneous electron transfer rate constant (ks) of immobilized SOD are 1.75×10?11 mol cm?2 and 7.5±0.5 s?1, respectively. The biosensor shows a fast amperometric response (3 s) toward superoxide at a wide concentration range from 10 µM to 0.25 mM with sensitivity of 13.40 nA µM?1 cm?2 and 12.40 nA µM?1 cm?2, detection limit of 2.66 and 3.1 µM based on anodically and cathodically detection. This biosensor exhibits excellent stability, reproducibility and long life time.  相似文献

The net electrostatic charge (Z) of a folded protein in solution represents a bird's eye view of its surface potentials—including contributions from tightly bound metal, solvent, buffer, and cosolvent ions—and remains one of its most enigmatic properties. Few tools are available to the average biochemist to rapidly and accurately measure Z at pH≠pI. Tools that have been developed more recently seem to go unnoticed. Most scientists are content with this void and estimate the net charge of a protein from its amino acid sequence, using textbook values of pKa. Thus, Z remains unmeasured for nearly all folded proteins at pH≠pI. When marveling at all that has been learned from accurately measuring the other fundamental property of a protein—its mass—one wonders: what are we missing by not measuring the net charge of folded, solvated proteins? A few big questions immediately emerge in bioinorganic chemistry. When a single electron is transferred to a metalloprotein, does the net charge of the protein change by approximately one elementary unit of charge or does charge regulation dominate, that is, do the pKa values of most ionizable residues (or just a few residues) adjust in response to (or in concert with) electron transfer? Would the free energy of charge regulation (ΔΔGz) account for most of the outer sphere reorganization energy associated with electron transfer? Or would ΔΔGz contribute more to the redox potential? And what about metal binding itself? When an apo-metalloprotein, bearing minimal net negative charge (e.g., Z=−2.0) binds one or more metal cations, is the net charge abolished or inverted to positive? Or do metalloproteins regulate net charge when coordinating metal ions? The author's group has recently dusted off a relatively obscure tool—the “protein charge ladder”—and used it to begin to answer these basic questions.  相似文献

Direct electron transfer of immobilized superoxide dismutase (Cu, Zn‐SOD) onto silicon carbide (SiC) nanoparticles displays a pair of well defined and nearly reversible redox peaks with formal potential (′) of −0.03 V in pH 7.4. The heterogeneous electron transfer rate constant (ks) and surface coverage (Γ) of immobilized SOD are 11.0±0.4 s−1 and 1.42×10−11 mol cm−2. Biosensor shows fast amperometric response (3s) with sensitivity and detection limit of 1.416 nA μM−1, 1.66 μM, and 1.375 nA μM−1, 2.1 μM for cathodically or anodically detection of superoxide, respectively. This biosensor also exhibits good stability, reproducibility and long life‐time.  相似文献

The first step in the catalytic oxidation of alcohols by molecular O2, mediated by homogeneous vanadium(V) complexes [LVV(O)(OR)], is ligand exchange. The unusual mechanism of the subsequent intramolecular oxidation of benzyl alcoholate ligands in the 8‐hydroxyquinolinato (HQ) complexes [(HQ)2VV(O)(OCH2C6H4p‐X)] involves intermolecular deprotonation. In the presence of triethylamine, complex 3 (X=H) reacts within an hour at room temperature to generate, quantitatively, [(HQ)2VIV(O)], benzaldehyde (0.5 equivalents), and benzyl alcohol (0.5 equivalents). The base plays a key role in the reaction: in its absence, less than 12 % conversion was observed after 72 hours. The reaction is first order in both 3 and NEt3, with activation parameters ΔH=(28±4) kJ mol?1 and ΔS=(?169±4) J K?1 mol?1. A large kinetic isotope effect, 10.2±0.6, was observed when the benzylic hydrogen atoms were replaced by deuterium atoms. The effect of the para substituent of the benzyl alcoholate ligand on the reaction rate was investigated using a Hammett plot, which was constructed using σp. From the slope of the Hammett plot, ρ=+(1.34±0.18), a significant buildup of negative charge on the benzylic carbon atom in the transition state is inferred. These experimental findings, in combination with computational studies, support an unusual bimolecular pathway for the intramolecular redox reaction, in which the rate‐limiting step is deprotonation at the benzylic position. This mechanism, that is, base‐assisted dehydrogenation (BAD), represents a biomimetic pathway for transition‐metal‐mediated alcohol oxidations, differing from the previously identified hydride‐transfer and radical pathways. It suggests a new way to enhance the activity and selectivity of vanadium catalysts in a wide range of redox reactions, through control of the outer coordination sphere.  相似文献

The direct electron transfer (ET) of copper–zinc superoxide dismutase (SOD) has been realized, for the first time, at Au nanoparticles electrodeposited onto GC (nano-Au/GC) electrodes. Electrodeposition of Au nanoparticles in the presence of some additive (typically cysteine) resulted in the fabrication of Au nanoparticles with a high roughness morphology (enriched in the Au(1 0 0) orientation) and thus providing a favorable adsorption orientation of SOD (key–lock interaction) suitable for a facilitated direct ET without the use of mediators or so-called ET promoters. The redox reaction of the SOD confined on the nano-Au/GC electrode was found to have a formal potential of +0.02 V vs. Ag/AgCl/KCl(sat). Whereas, round-shape spherical plumbs of Au nanoparticles were electrodeposited in the presence of iodide ions (as additive) which lack for the favorable rough surface and consequently the suitable interaction with the SOD is missing and hence the ET is not realized at this surface.  相似文献

Structural, electrochemical and spectroscopic data of a new dinuclear copper(II) complex with (±)-2-(p-methoxyphenoxy)propionic acid are reported. The complex {tetra-μ-[(±)-2-(p-methoxyphenoxy)propionato-O,O′]-bis(aqua)dicopper(II)} crystallizes in the monoclinic system, space group P21/n with a = 14.149(1) ?, b = 7.495(1) ?, c = 19.827(1) ?, β = 90.62(1) and Z = 4. X-ray diffraction data show that the two copper(II) ions are held together through four carboxylate bridges, coordinated as equatorial ligands in square pyramidal geometry. The coordination sphere around each copper ion is completed by two water molecules as axial ligands. Thermogravimetric data are consistent with such results. The ligand has an “L” type shape due to the angle formed by the β-carbon of the propionic chain and the linked p-methoxyphenoxy group. This conformation contributes to the occurrence of a peculiar structure of the complex. The complex retains its dinuclear nature when dissolved in acetonitrile, but it decomposes into the corresponding mononuclear species if dissolved in ethanol, according to the EPR measurements. Further, cyclic voltammograms of the complex in acetonitrile show that the dinuclear species maintains the same structure, in agreement with the EPR data in this solvent. The voltammogram shows two irreversible reduction waves at E pc = −0.73 and −1.04 V vs. Ag/AgCl assigned to the Cu(II)/Cu(I) and Cu(I)/Cu° redox couples, respectively, and two successive oxidation waves at E pa =− 0.01 and +1.41 V vs. Ag/AgCl, assigned to the Cu°/Cu(I) and Cu(I)/Cu(II) redox couples, respectively, in addition to the oxidation waves of the carboxylate ligand.  相似文献

The discovery of a novel superoxide dismutase (SOD) mimic which demonstrates SOD activity, chemical stability in H2O2 solution and long half‐life in circulation is reported. The SOD mimic consists of a manganese porphyrin (MnP) with SOD activity and a polymer (poly (styrene‐co‐maleic anhydride); SMA) with biological characteristics. The SOD activity of SMA‐MnP at pH = 8.1 measured using the stopped‐flow kinetic analysis technique to monitor the decay of superoxide directly was 1.1 (± 0.1) × 106 M‐1 sec‐1. It is postulated that the reduction by O2·− of the oxidized SMA‐Mn(III) is slow, while the oxidation of the reduced SMA‐Mn(II) by O2·− is very fast. The retention times of SMA‐MnP in the circulation of rabbits were determined. In vivo, SMA‐MnP that binds to the warfarin site on albumin showed an enhanced half‐life in the circulation. Additionally, in vitro SMA‐MnP indicated an excellent stability to H2O2. Copyright © 1999 John Wiley & Sons, Ltd.  相似文献

The oxidation of Na4Fe(CN)6 complex by S2O anion was found to follow an outer‐sphere electron transfer mechanism. We firstly carried out the reaction at pH=1. The specific rate constants of the reaction, kox, are (8.1±0.07)×10?2 and (4.3±0.1)×10?2 mol?1·L·s?1 at μ=1.0 mol·L?1 NaClO4, T=298 K for pH=1 (0.1 mol·L?1 HCl04) and 8, respectively. The activation parameters, obtained by measuring the rate constants of oxidation 283–303 K, were ΔH=(69.0±5.6) kJ·mol?1, ΔS=(?0.34±0.041)×102 J·mol?1·K?1 at pH=l and ΔH=(41.3±5.5) kJ·mol?1, ΔS=(?1.27±0.33)×102 J·mol?1·K?1 at pH=8, respectively. The cyclic voltammetry of Fe(CN) shows that the oxidation is a one‐electron reversible redox process with E1/2 values of 0.55 and 0.46 V vs. normal hydrogen electrode at μ=1.0 mol·L?1 LiClO4, for pH=1 and pH=8 (Tris). respectively. The kinetic results were discussed on the basis of Marcus theory.  相似文献

Subnanoparticles (SNPs) with sizes of approximately 1 nm are attractive for enhancing the catalytic performance of transition metals and their oxides. Such SNPs are of particular interest as redox-active catalysts in selective oxidation reactions. However, the electronic states and oxophilicity of copper oxide SNPs are still a subject of debate in terms of their redox properties during oxidation reactions for hydrocarbons. In this work, in situ X-ray absorption fine structure (XAFS) measurements of Cu28Ox SNPs, which were prepared by using a dendritic phenylazomethine template, during temperature-programmed reduction (TPR) with H2 achieved lowering of the temperature (T50=138 °C) reported thus far for the CuII→CuI reduction reaction because of Cu−O bond elongation in the ultrasmall copper oxide particles.  相似文献

The solution structure and the aggregation behavior of (E)-2-lithio-1-(2-lithiophenyl)-1-phenylpent-1-ene ( 1 ) and (Z)-2-lithio-1-(2-lithiophenyl)ethene ( 2 ) were investigated by one- and two-dimensional 1H-, 13C-, and 6Li-NMR spectroscopy. In Et2O, both systems form dimers which show homonuclear scalar 6Li,6Li spin-spin coupling. In the case of 2 , extensive 6Li,1H coupling is observed. In tetrahdrofuran and in the presence of 2 mol of N,N,N′,N′-tetramethylethylylenediamine (tmeda), the dimeric structure of 1 coexists with a monomer. The activation parameters for intra-aggregate exchange in the dimers of 1 and 2 ( 1 (Et2O): ΔH≠ = 62.6 ± 13.9 kJ/mol, ΔS≠ = 5.8 ± 14.0 J/mol K, ΔG≠(263) = 61.1 kJ/mol; 2 (dimethoxyethane): ΔH≠ = 36.9 ± 6.5 kJ/mol, ΔS≠ = ?61 ± 25 J/mol K, ΔG≠(263) = 54.0 kJ/mol) and the thermodynamic parameters for the dimer-monomer equilibrium for 1 (ΔH°; = 26.7 ± 5.5 kJ/mol, ΔS° = 63 ± 27 J/mol K), where the monomer is favored at low temperature, were determined by dynamic NMR studies.  相似文献

Four heterodimetallic complexes [Ru(Fcdpb)(L)](PF6) (Fcdpb=2‐deprotonated form of 1,3‐di(2‐pyridyl)‐5‐ferrocenylbenzene; L=2,6‐bis‐(N‐methylbenzimidazolyl)‐pyridine (Mebip), 2,2′:6′,2′′‐terpyridine (tpy), 4‐nitro‐2,2′:6′,2′′‐terpyridine (NO2tpy), and trimethyl‐4,4′,4′′‐tricarboxylate‐2,2′:6′,2′′‐terpyridine (Me3tctpy)) have been prepared. The electrochemical and spectroelectrochemical properties of these complexes have been examined in CH2Cl2, CH3NO2, CH3CN, and acetone. These complexes display two consecutive redox couples owing to the stepwise oxidation of the ferrocene (Fc) and ruthenium units, respectively. The potential difference, ΔE1/2 (E1/2(RuII/III)?E1/2(Fc0/+)), decreased slightly with increasing solvent donocity. The mixed‐valent states of these complexes have been generated by electrolysis and the resulting intervalence charge‐transfer (IVCT) bands have been analyzed by Hush theory. Good linear relationships exist between the energy of the IVCT band, Eop, and ΔE1/2 of four mixed‐valent complexes in a given solvent.  相似文献

The electrochemical behavior of cytochrome c (cyt‐c) that was electrostatically immobilized onto a self‐assembled monolayer (SAM) of captopril (capt) on a gold electrode has been investigated. Cyclic voltammetry, scanning electrochemical microscopy (SECM) and electrochemical impedance spectroscopy were employed to evaluate the blocking property of the capt SAM. SECM was used to measure the bimolecular electron transfer (ET) kinetics (kBI) between a solution‐based redox probe and the immobilized protein. In addition, the tunneling ET between the immobilized protein and the underlying gold electrode was calculated. A kBI value of (5.0±0.6)×108 mol?1 cm3 s?1 for the bimolecular ET and a standard tunneling rate constant (k0) of 46.4±0.2 s?1 for the tunneling ET have been obtained.  相似文献

The significance of multiple number of donor–acceptor entities on a central electron donor in a star-shaped molecular system in improving light energy harvesting ability is reported. For this, donor–acceptor–donor type conjugates comprised up to three entities ferrocenyl (Fc)-diketopyrrolopyrrole (DPP) onto a central triphenylamine (TPA), ( 4 – 6 ) by the Pd-catalyzed Sonogashira cross-coupling reactions have been newly synthesized and characterized. Donor–acceptor conjugates possessing diketopyrrolopyrrole (1 to 3 entities) onto the central triphenylamine, ( 1 – 3 ) served as reference dyads while monomeric DPP and Fc-DPP served as control compounds. Both DPP and Fc-DPP carrying conjugates exhibited red-shifted absorption compared to their respective control compounds revealing existence of ground state interactions. Furthermore, DPP fluorescence in 4 – 6 was found to be quantitatively quenched while for 1 – 3 , this property varied between 73–65 % suggesting occurrence moderate amounts of excited state events. The electrochemical investigations exhibited an additional low potential oxidation in the case of Fc-DPP-TPA based derivatives ( 4 – 6 ) owing to the presence of ferrocene unit(s). This was in addition to DPP and TPA redox peaks. Using spectral, electrochemical and computational studies, Gibbs free-energy calculations were performed to visualize excited state charge separation (ΔGCS) in these donor–acceptor conjugates as a function of different number of Fc-DPP entities. Formation of Fc+-DPP.−-TPA charge separated states (CSS) in the case of 4 – 6 was evident. Using spectroelectrochemical studies, spectrum of CSS was deduced. Finally, femtosecond transient absorption spectral studies were performed to gather information on excited state charge separation. Increasing the number of Fc-DPP entities in 4 – 6 improved charge separation rates. Surprisingly, lifetime of the charge separated state, Fc+-DPP.−-TPA was found to persist longer with an increase in the number of Fc-DPP entities in 4 – 6 as compared to Fc-DPP-control and simple DPP derived donor–acceptor conjugates in literature. This unprecedented result has been attributed to subtle changes in ΔGCS and ΔGCR and the associated electron coupling between different entities.  相似文献

Molecular organization of donor and acceptor chromophores in self‐assembled materials is of paramount interest in the field of photovoltaics or mimicry of natural light‐harvesting systems. With this in mind, a redox‐active porous interpenetrated metal–organic framework (MOF), {[Cd(bpdc)(bpNDI)] ? 4.5 H2O ? DMF}n ( 1 ) has been constructed from a mixed chromophoric system. The μ‐oxo‐bridged secondary building unit, {Cd2(μ‐OCO)2}, guides the parallel alignment of bpNDI (N,N′‐di(4‐pyridyl)‐1,4,5,8‐naphthalenediimide) acceptor linkers, which are tethered with bpdc (bpdcH2=4,4′‐biphenyldicarboxylic acid) linkers of another entangled net in the framework, resulting in photochromic behaviour through inter‐net electron transfer. Encapsulation of electron‐donating aromatic molecules in the electron‐deficient channels of 1 leads to a perfect donor–acceptor co‐facial organization, resulting in long‐lived charge‐separated states of bpNDI. Furthermore, 1 and guest encapsulated species are characterised through electrochemical studies for understanding of their redox properties.  相似文献

The study of the redox chemistry of mid-actinides (U−Pu) has historically relied on cerium as a model, due to the accessibility of trivalent and tetravalent oxidation states for these ions. Recently, dramatic shifts of lanthanide 4+/3+ non-aqueous redox couples have been established within a homoleptic imidophosphorane ligand framework. Herein we extend the chemistry of the imidophosphorane ligand (NPC=[N=PtBu(pyrr)2]; pyrr=pyrrolidinyl) to tetrahomoleptic NPC complexes of neptunium and cerium ( 1-M , 2-M , M=Np, Ce) and present comparative structural, electrochemical, and theoretical studies of these complexes. Large cathodic shifts in the M4+/3+ (M=Ce, U, Np) couples underpin the stabilization of higher metal oxidation states owing to the strongly donating nature of the NPC ligands, providing access to the U5+/4+, U6+/5+, and to an unprecedented, well-behaved Np5+/4+ redox couple. The differences in the chemical redox properties of the U vs. Ce and Np complexes are rationalized based on their redox potentials, degree of structural rearrangement upon reduction/oxidation, relative molecular orbital energies, and orbital composition analyses employing density functional theory.  相似文献

The complex formation reactions of [Cu(NTP)(OH2)]4? (NTP?=?nitrilo-tris(methyl phosphonic acid)) with some selected bio-relevant ligands containing different functional groups, are investigated. Stoichiometry and stability constants for the complexes formed are reported. The results show that the ternary complexes are formed in a stepwise mechanism whereby NTP binds to copper(II), followed by coordination of amino acid, peptide or DNA. Copper(II) is found to form Cu(NTP)H n species with n?=?0, 1, 2 or 3. The concentration distribution of the various complex species has been evaluated. The kinetics of base hydrolysis of glycine methyl ester in the presence of copper(II)-NTP complex is studied in aqueous solution at different temperatures. It is proposed that the catalysis of GlyOMe ester occurs by attack of OH? ion on the uncoordinated carbonyl carbon atom of the ester group. Activation parameters for the base hydrolysis of the complex [Cu(NTP)NH2CH2CO2Me]4? are, ΔH±?=?9.5?±?0.3?kJ?mol?1 and ΔS±?=??179.3?±?0.9?J?K?1?mol?1. These show that catalysis is due to a substantial lowering of ΔH±.  相似文献
