Microbes synthesize cell-associated nanoparticles (NPs) and utilize their physicochemical properties to produce energy under unfavorable metabolic conditions. Iron sulfide (FeS) NPs are ubiquitous and are predominantly biosynthesized by sulfate-reducing bacteria (SRB). However, the biological role of FeS NPs in SRB remains understudied. Now, conductive FeS NPs function is demonstrated as an electron conduit enabling Desulfovibrio vulgaris Hildenborough, an SRB strain, to utilize solid-state electron donors via direct electron uptake. After forming FeS NPs on the cell surface, D. vulgaris initiated current generation coupled with sulfate reduction on electrodes poised at −0.4 V versus standard hydrogen electrode. Single-cell activity analysis showed that the electron uptake and metabolic rate via FeS NPs in D. vulgaris were about sevenfold higher than those via native cell-surface proteins in other SRB.  相似文献

研究了以再生水作为循环冷却系统补水的北京某热电厂冷却塔底粘泥中分离纯化培养出来的硫酸盐还原菌(SRB)生长特性.采用原子力显微镜(AFM)、扫描电镜(SEM)、能谱分析仪(EDS)和电化学交流阻抗(EIS)方法研究了304不锈钢(SS304)表面生物膜特征及其主要成分和不锈钢/生物膜界面电化学行为.结果显示,再生水环境下304不锈钢表面形成的生物膜是由吸附的SRB菌体及以含碳有机物为主的胞外聚合物和FeS腐蚀产物构成.浸泡前期(前7 d)SS304电极表面阻抗值主要由SS304表面钝化膜的贡献;浸泡后期(14 d后),电极体系阻抗值由不锈钢表面钝化膜和生物膜共同贡献.  相似文献

We had previously developed surface-modified poly (D,L-lactide-co-glycolide) (PLGA) nanoparticles (NPs) for use as a cellular drug delivery system. The cellular uptake of PLGA-NPs was mediated predominantly by endocytosis, and this uptake was increased by surface modifications with polymers, such as chitosan (CS) and polysorbate 80 (P80). In the present study, we prepared a cell-sized giant unilamellar vesicle (GUV) that mimics a cell membrane to investigate the interaction between cell membranes and NPs. Endocytosis-like uptake of NPs into a GUV was observed when the NPs were modified with nonionic surfactant P80 probably due to change in viscoelasticity and enhanced fusion activity of the membrane induced by P80. In contrast, unmodified NPs and those modified with CS were not internalized into a GUV. These results suggest that surface properties of PLGA-NPs are an important formulation parameter for their interaction with lipid membranes.  相似文献

Voltammetric response of FeS nanoparticles (NPs) dispersion based on the oxidation exchange voltammetric peak between Hg electrode and FeS NPs at around ?0.45 V was studied in different electrolytes (chloride, acetate, perchlorate). Several experiments were designed to monitor in parallel to voltammetric measurements, physicochemical and surface characteristics of the formed FeS NPs (ζ-potential and size) under same experimental conditions. It was shown that recorded voltammetric peak produced by FeS NPs from bulk solution is changing with electrolyte concentration and composition, as well as observed size and ζ-potential of the studied FeS NPs. Our measurements indicate relationship between measured ζ-potential of FeS NPs dispersions and recorded voltametric peak charge and potential, pointing to a promising potential of voltammetry in characterization of physicochemical and surface chemistry features of metal sulphide NPs in water environment. The best voltammetric response is obtained in presence of small NPs, <100 nm.  相似文献

Samples of FeS were oxidized by oxygen‐bearing acidic solutions at 25 °C and different initial pH values (2.75 ≤ pH ≤ 3.45). The reacted FeS samples were investigated by scanning electron microscopy (SEM), nitrogen adsorption/desorption isotherms measurements, and FT‐Raman spectroscopy. A sulfur rich phase (layer) is inferred to be form on reacting FeS surface based on sequential extraction with acidic chromium(II) chloride solutions. The sulfur‐rich layer (SRL) formed on oxidized FeS samples is traversed by various mesopores with average pore size < 450Å. It is reasonable to assume that the mesopores facilitate the migration of iron and sulfur from FeS structure into solution across SRL. The FT‐Raman spectra of oxidized FeS samples show an intense peak at 162 cm?1, which can be attributed to SRL developed on FeS surface. A model for SRL development on FeS during its oxidation by dissolved oxygen has been proposed from the present results and previous studies. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

Biodegradable self‐assembled polymeric nanoparticles (NPs) composed of poly(6‐O‐methacryloyl‐D‐galactopyranose)‐b‐poly(L‐lactide)‐b‐poly(6‐O‐methacryloyl‐D‐galactopyranose) (PMAGP‐b‐PLA‐b‐PMAGP) are prepared as carriers for the hydrophobic anticancer drug paclitaxel (PTX), to achieve target delivery to hepatoma cells. PTX can be encapsulated by the NPs with various molar ratios of L‐lactide (LA) and 6‐O‐methacryloyl‐D‐galactopyranose (MAGP) during the process of self‐assembly, and the resulting NPs exhibit high drug loading efficacy and substantial stability in aqueous solution. The size, size distribution, and morphology of the NPs are characterized using a Zetasizer Nano ZS and transmission electron microscopy. The hemolysis assay and cell cytotoxicity assay indicate that the polymeric NPs are biocompatible and non‐toxic. The cellular uptake assay demonstrates that the galactose‐containing NPs can be selectively recognized and subsequently accumulate in HepG2 cells. All of these results demonstrate that galactose‐containing polymeric NPs are potential carriers for hepatoma‐targeted drug delivery and liver cancer therapy in clinical medicine.

Abstract

The present study reports ecofriendly synthesis of CuO nanoparticles (NPs) using an extract of Rhus punjabensis as a reducing agent. NPs structural and composition analysis are evaluated by X-rays diffraction (XRD), Fourier transform infrared, Energy dispersive spectroscopy, Scanning electron microscopy, Transmission electron microscopy, and Thermal analysis. The NPs have pure single phase monoclinic geometry with spherical structure and high stability toward heat and with average particle size of about 36.6 and 31.27?nm calculated by XRD and SEM, respectively. NPs are tested for antibacterial, protein kinase (PK) inhibition, SRB cytotoxic, and NF-κB activities. Antibacterial activity is observed against B. subtilis and E. coli. Significant PK and SRB cytotoxic activity is observed with some NF-κB inhibition. NPs IC50 values against HL-60 and PC-3 prostate cancer cells are 1.82?±?1.22 and 19.25?±?1.55?μg/mL. The results encourage further studies for antibacterial and anticancer drug development of NPs using animal models.  相似文献

This work was aimed to synthesize and characterize poly(2‐hydroxyethyl methacrylate) [poly (HEMA)]‐based molecularly imprinted polymer nanoparticles (MIP NPs) containing timolol maleate (TM) via precipitation polymerization. The molecular structures of the MIP and non‐imprinted polymer (NIP) NPs were compared by means of Fourier transform infrared spectroscopy. The morphological observations by using scanning electron microscopy and transmission electron microscopy confirmed the formation of MIP NPs as small as 128 nm in average diameter with appropriate synthesis conditions. Thermal behaviors of the samples were also studied by the use of thermogravimetric analysis and differential scanning calorimetry. By considering a series of key factors such as monomer : template ratio, cross‐linker type, pH, and temperature, the sample with promising characteristics was found to be that of HEMA : TM ratio of 10:1, 40 mmol of ethylene glycol dimethacrylate as cross‐linker, and polymerization temperature of 60°C in acetonitrile as porogenic solvent. Furthermore, the ultraviolet‐visible (UV‐vis) spectrophotometry results proved a controlled release of TM from the MIP NP samples compared with NIP ones at extended periods. Moreover, the cytotoxicity of the MIP and NIP NPs samples was evaluated on mesenchymal stem cells, and the obtained observations showed that they had no adverse side effect on the living cells; especially the surface of the MIP NPs sample depicted highly cell's biocompatibility. Finally, the outcomes from designed different experiments conducted us that the HEMA‐based MIP NPs have great potential as an ocular nanocarrier for TM delivery. Electrocatalytic reduction of CO2 to formate on carbon based electrodes is known to suffer from low electrochemical reaction activity and product selectivity. Pd/three‐dimensional graphene (Pd/3D‐RGO), In/3D‐RGO and Pd‐In/3D‐RGO for the electrochemical reduction of CO2 were prepared by a mild method that combines chemical and hydrothermal. The metal/3D‐graphenes (metal/3D‐RGO) were characterized by scanning electron microscopy, X‐ray diffraction, transmission electron microscopy and X‐ray photoelectron spectroscopy (XPS). Cyclic voltammetry and the ion chromatography were performed to investigate the electrochemical performance of the metal/3D‐RGO. The morphology and dispersion of metal/3D‐RGO are 3D structure with amount of interconnected pores with metal NPs loading on the fold. And the Pd0.5‐In0.5/3D‐RGO show excellent surface performance with well dispersion and smallest particle size (12.8 nm). XPS reveal that binding energy of Pd (In) NPs is shifted to negative energy, for the metal lose electrons in metal and combine with C, which is demonstrated in the HNO3 experiment. The peak potential of Pd0.5‐In0.5/3D‐RGO is −0.70 V (vs. Ag/AgCl), which is more positive than In1.0/3D‐RGO (−0.73 V) and Pd1.0/3D‐RGO (−1.2 V). The highest faradaic efficiency (85.3 %) happens in Pd0.5‐In0.5/3D‐RGO at −1.6 V vs. Ag/AgCl. In these experiments, the special structure that metal NPs combine with C and the bimetal NPs give a direction to convert CO2 to formate.  相似文献

In the present study, modification of nanoparticles (NPs) was investigated to mitigate aggregation of SiO2 nanoparticles and improve the polymeric membrane's performance. For this purpose, the surface of SiO2 nanoparticles was activated with amine groups, and polymethacrylic acid (PMAA) was grafted on the surface of NPs by atom transfer radical polymerization. Modified NPs were characterized by Fourier transform infrared spectroscopy (FTIR) and thermogravimetric analysis (TGA) tests. Polyethersulfone (PES) membranes were fabricated with both SiO2 and SiO2‐g‐PMAA NPs via nonsolvent‐induced phase separation method. The fabricated membranes were characterized regarding their permeability, hydrophilicity, and porosity properties, and their separation efficiency was tested using the synthetic oil‐in‐water emulsion. The surface and cross‐sectional morphologies of membranes were observed by field emission scanning electron microscopy (FESEM). The experimental trials showed that modified NPs dispersed more uniformly in the structure of membranes and hydroxyl groups on the surface of NPs acted more effectively. Modification of NPs enhance the membrane performance in terms of permeate flux, hydrophilicity, and porosity. NPs modification improved the permeate flux about 46%. Oil rejection for all tested membranes was more than 98%, and modification of NPs did not reduce the rejection of membranes. The optimum concentration was obtained as 1 wt.% and 1.5 wt.% for SiO2 and SiO2‐g‐PMAA, respectively. Aggregation effect dominated at concentrations beyond the optimum values that decreased the permeate flux, consequently.  相似文献

By a combination of theoretical and experimental design, we probed the effect of a quasi‐single electron on the surface plasmon resonance (SPR)‐mediated catalytic activities of Ag nanoparticles. Specifically, we started by theoretically investigating how the E‐field distribution around the surface of a Ag nanosphere was influenced by static electric field induced by one, two, or three extra fixed electrons embedded in graphene oxide (GO) next to the Ag nanosphere. We found that the presence of the extra electron(s) changed the E‐field distributions and led to higher electric field intensities. Then, we experimentally observed that a quasi‐single electron trapped at the interface between GO and Ag NPs in Ag NPs supported on graphene oxide (GO‐Ag NPs) led to higher catalytic activities as compared to Ag and GO‐Ag NPs without electrons trapped at the interface, representing the first observation of catalytic enhancement promoted by a quasi‐single electron.  相似文献

When nanoparticles (NPs) are introduced to a biological fluid, different proteins (and other biomolecules) rapidly get adsorbed onto their surface, forming a protein corona capable of giving to the NPs a new “identity” and determine their biological fate. Protein–nanoparticle conjugation can be used in order to promote specific interactions between living systems and nanocarriers. Non‐covalent conjugates are less stable and more susceptible to desorption in biological media, which makes the development of engineered nanoparticle surfaces by covalent attachment an interesting topic. In this work, the surface of poly(globalide‐co‐ε‐caprolactone) (PGlCL) nanoparticles containing double bonds in the main polymer chain is covalently functionalized with bovine serum albumin (BSA) by thiol‐ene chemistry, producing conjugates which are resistant to dissociation. The successful formation of the covalent conjugates is confirmed by flow cytometry (FC) and fluorescence correlation spectroscopy (FCS). Transmission electron microscopy (TEM) allows the visualization of the conjugate formation, and the presence of a protein layer surrounding the NPs can be observed. After conjugation with BSA, NPs present reduced cell uptake by HeLa and macrophage RAW264.7 cells, in comparison to uncoated NP. These results demonstrate that it is possible to produce stable conjugates by covalently binding BSA to PGlCL NP through thiol‐ene reaction.  相似文献

The use of 4.2 nm gold nanoparticles wrapped in an adsorbates shell and embedded in a TiO2 metal oxide matrix gives the opportunity to investigate ultrafast electron-electron scattering dynamics in combination with electronic surface phenomena via the surface plasmon lifetimes. These gold nanoparticles (NPs) exhibit a large nonclassical broadening of the surface plasmon band, which is attributed to a chemical interface damping. The acceleration of the loss of surface plasmon phase coherence indicates that the energy and the momentum of the collective electrons can be dissipated into electronic affinity levels of adsorbates. As a result of the preparation process, gold NPs are wrapped in a shell of sulfate compounds that gives rise to a large density of interfacial molecules confined between Au and TiO2, as revealed by Fourier-transform-infrared spectroscopy. A detailed analysis of the transient absorption spectra obtained by broadband femtosecond transient absorption spectroscopy allows separating electron-electron and electron-phonon interaction. Internal thermalization times (electron-electron scattering) are determined by probing the decay of nascent nonthermal electrons (NNEs) and the build-up of the Fermi-Dirac electron distribution, giving time constants of 540 to 760 fs at 0.42 and 0.34 eV from the Fermi level, respectively. Comparison with literature data reveals that lifetimes of NNEs measured for these small gold NPs are more than four times longer than for silver NPs with similar sizes. The surprisingly long internal thermalization time is attributed to an additional decay mechanism (besides the classical e-e scattering) for the energy loss of NNEs, identified as the ultrafast chemical interface scattering process. NNEs experience an inelastic resonant scattering process into unoccupied electronic states of adsorbates, that directly act as an efficient heat bath, via the excitation of molecular vibrational modes. The two-temperature model is no longer valid for this system because of (i) the temporal overlap between the internal and external thermalization process is very important; (ii) a part of the photonic energy is directly transferred toward the adsorbates (not among "cold" conduction band electrons). These findings have important consequence for femtochemistry on metal surfaces since they show that reactions can be initiated by nascent nonthermal electrons (as photoexcited, out of a Fermi-Dirac distribution) besides of the hot electron gas.  相似文献

We investigate the chronoamperometric noise characteristics of electron‐transfer reactions occurring on single nanoparticles (NPs) and assemblies of well‐separated NPs on a supporting surface. To this end, we combine a formerly described expression for the steady‐state current of a single particle with the shot‐noise model and derive an expression for the signal‐to‐noise ratio as a function of bulk concentration and particle radius. Our findings are supported by random‐walk simulations, which closely match the analytical results.  相似文献

We report that human mesenchymal stem cells (hMSCs) were successfully labeled with poly(lactide‐co‐glycolide) nanoparticles (PLGA NPs) surface‐conjugated quantum dots (QDs) (PLGA‐QD NPs) via endocytosis pathway. These NPs were not toxicity even treated with PLGA‐QD NPs at high concentrations for at least four weeks. Besides, PLGA‐QD NPs‐labeled hMSCs did not change their proliferation and differentiation capability toward the cell fates of adipocytes, osteocytes, and chrondrocytes. It's known that PLGA has been widely employed to act as delivery carrier which encapsulates drugs and releases them under a controlled way. Currently, we have also demonstrated that FITC‐loaded PLGA‐QD NPs degraded in hMSCs to achieve intracellular release of FITC. The aim of this research is to investigate viability, proliferation and differentiation capability and the potential for gene delivery of MSCs labeled with PLGA‐QD NPs. In addition to PLGA‐QD NPs, QDs alone were used to serve as a control set for comparison.  相似文献

A large volume of bacterially produced precipitates are generated during the bio-treatment of heavy metal wastewater. The composition of the bacterially produced precipitates and its effects on sulfate reducing bacteria (SRB) in copper-containing waste stream were evaluated in this study. The elemental composition of the microbial precipitate was studied using electrodispersive X-ray spectroscopy (EDX), and it was found that the ratio of S:Cu was 1.12. Combining with the results of copper distribution in the SRB metabolism culture, which was analyzed by the sequential extraction procedure, copper in the precipitates was determined as covellite (CuS). The bacterially produced precipitates caused a decrease of the sulfate reduction rate, and the more precipitates were generated, the lower the sulfate reduction rate was. The particle sizes of bacterially generated covellite were ranging from 0.03 to 2 μm by particles size distribution (PSD) analysis, which was smaller than that of the SRB cells. Transmission electron microscopy (TEM) analysis showed that the microbial covellite was deposited on the surface of the cell. The effects of the microbial precipitate on SRB metabolism were found to be weakened by increasing the precipitation time and adding microbial polymeric substances in later experiments. These results provided direct evidence that the SRB activity was inhibited by the bacterially produced covellite, which enveloped the bacterium and thus affected the metabolism of SRB on mass transfer.  相似文献

Recent work regarding the Layer by Layer (LbL) engineering of poly(lactide-co-glycolide) nanoparticles (PLGA NPs) is reviewed here.The LbL engineering of PLGA NPs is applied as a means of generating advanced drug delivery devices with tailored recognition,protection,cargo and release properties.LbL in combination with covalent chemistry is used to attach PEG and folic acid to control cell uptake and direct it towards cancer cells.LbL coatings composed of chitosan and alginate show low protein interactions and can be used as an alternative to Pegylation.The assembly on top of LbL coatings of lipid layers composed of variable percentages of 1,2-dioleoyl-sn-glycero-3-choline (DOPC) and 1,2-dioleoyl-sn-glycero-3-phosphoL-serine (DOPS) increases NP uptake and directs the NPs towards the endoplasmic reticulum.The antibody anti-TNF-α is encapsulated forming a complex with alginate that is assembled LbL on top of PLGA NPs.The antibody is released in cell culture following first order kinetics.The release kinetics of encapsulated molecules inside PLGA NPs are studied when the PLGA NPs are coated via LbL with different polyelectrolytes.The intracellular release of encapsulated Doxorubicin is studied in the HepG2 cell line by means of Fluorescence Lifetime Imaging.  相似文献

We report here a facile method to obtain folic acid (FA)‐protected gold nanoparticles (Au NPs) by heating an aqueous solution of HAuCl4/FA in which FA acts as both the reducing and stabilizing agent. The successful formation of FA‐protected Au NPs is demonstrated by UV/Vis spectroscopy, transmission electron microscopy (TEM), selected‐area electron diffraction (SAED), X‐ray photoelectron spectroscopy (XPS), and Fourier transform infrared spectroscopy (FTIR). The intracellular uptake of these nanoparticles is facilitated by HeLa cells overexpressing the folate reporter, which itself is significantly inhibited by free FA in a competitive assay as quantified by inductively coupled plasma mass spectroscopy (ICP‐MS). This simple one‐step approach affords a new perspective for creating functional nanomaterials, and the resulting biocompatible, functional Au NPs may find some prospective applications in various biomedical fields.  相似文献

Au/Pt core shell nanoparticles (NPs) have been prepared via a layer‐by‐layer growth of Pt layers on Au NPs using underpotential deposition (UPD) redox replacement technique. A single UPD Cu monolayer replacement with Pt(II) yielded a uniform Pt film on Au NPs, and the shell thickness can be tuned by controlling the number of UPD redox replacement cycles. Oxygen reduction reaction (ORR) in air‐saturated 0.1 M H2SO4 was used to investigate the electrocatalytic behavior of the as‐prepared core shell NPs. Cyclic voltammograms of ORR show that the peak potentials shift positively from 0.32 V to 0.48 V with the number of Pt layers increasing from one to five, suggesting the electrocatalytic activity increases with increasing the thickness of Pt shell. The increase in electrocatalytic activity may originate mostly from the large decrease of electronic influence of Au cores on surface Pt atoms. Rotating ring‐disk electrode voltammetry and rotating disk electrode voltammetry demonstrate that ORR is mainly a four‐electron reduction on the as‐prepared modified electrode with 5 Pt layers and first charge transfer is the rate‐determining step.  相似文献

This study analyzes the varying electrophoretic mobility and zeta potential of bone marrow stromal cells (BMSCs) during their differentiation towards neurons. Electrophoresis of primary BMSCs and neuron growth factor (NGF)‐induced neuron‐like cells with the uptake of heparin‐functionalized poly(lactide‐co‐glycolide) (PLGA) nanoparticles (NPs) are also investigated. Immunofluorescent images revealed that a high concentration of NGF accelerated the differentiation of BMSCs into neurons. When the concentration of NGF increased, the absolute values of electrophoretic mobility and zeta potential of the differentiating BMSCs increased. In addition, a longer inductive period yielded higher charge of the differentiating BMSCs and a smaller uptake amount of heparin‐functionalized PLGA NPs. However, an increase in the loading efficiency of heparin on PLGA NPs enhanced the uptake and reduced the electrical characteristics of the primary and differentiating BMSCs. Hence, a general rule is drawn that an increase in the uptake of heparin‐functionalized PLGA NPs decreased the electrophoretic mobility and zeta potential of BMSCs during differentiation towards neurons.  相似文献
