To determine the structures of two isomeric products, 2-phenacylidene-1,2-dihydro-4H-pyrido[2,3-b]pyrazin-3-one (2) and 3-phenacylidene-3,4-dihydro-1H-pyrido[2,3-b]pyrazin-2-one (3) obtained by condensation of 2,3-diaminopyridine (1) with ethyl benzoylpyruvate [1–3], these compounds were hydrolyzed to give 2-methyl-4H-pyrido[2,3-b]pyrazin-3-one (4) and 3-methyl-1H-pyrido[2,3-b]pyrazin-2-one (5) , respectively [4,5]. Both hydrolysates 4 and 5 were hydrogenated to afford 2-methyl-1,2-dihydro-4H-pyrido[2,3-b]pyrazin-3-one (6) and 3-methyl-3,4-dihydro-1H-pyrido[2,3-b]pyrazin-2-one (7) . The latter compound was identical with an unequivocally synthesized compound providing proof for the structures of all these compounds.  相似文献

Twelve phenacyl derivatives of 1H-pyrido[3,4-b]pyrazin-2-ones and 4H-pyrido[3,4-b]pyrazin-3-ones have been synthesized. In these compounds, C = N double bonds at the 3 and 4 positions in the former compounds and those at the 1 and 2 positions in the latter compounds migrate onto the side chains to form phenacyli-dene structures. These migrations are facilitated by chelation between side chain carbonyl and the proton attached to the ring nitrogen atom which was generated by the migration. All the hydrogen-bonded structures appear to be stable as shown by their ir spectra in the crystalline state, and by their 1H nmr spectra in solution.  相似文献

A general and versatile method for the preparation of 2H-thiopyrano[2,3-b]pyridin-2-ones is described. The starting materials, the β, β-disubstituted vinyl-1-t-butyl-2-(1H)pyridinethiones were prepared from the synthon 3-formyl-1-t-butyl-2-(1H)pyridinethione by condensation. 13H nmr spectra showed the vinyl double bond of the condensation products to have the trans configuration with the smallest group close to the sulfur atom. Some reactions of these new azaanalogues of thiocoumarins are reported.  相似文献

5,11-Dihydro-6H-pyrido[2,3-6][1,4]benzodiazepin-6-one (1), a basic intermediate in the preparation of 11-α-aminoacetyl derivatives with important biological activities, has been obtained by a three-step synthesis starting from easily available isatoic anhydride and anhydro ornithine. Some model cyclisation reactions leading to 5-member ring derivatives 10 and 12 instead of 7-member ring analogues of 1 , are reported. Easy transformations of the tetrahydro congener of 1 , i.e., compound 4 into 19 , which actually represents a tetrahedral intermediate in the transformation of 5 into 4 , is noticed. Further rearrangement of 19 into spiro compound 20 , and return of the latter into 5 is described.  相似文献

The 2-C, methylene group of 1-aroyl-1,2-dihydro-3H-naphtho[2,1-b]pyran-3-ones (2) have been showed to possess a superior reactivity towards many of organic reagents. The naphthopyrones (2) condensed with aromatic aldehydes to give the 2-arylidine derivatives (3). Bromination of 2 gave 1,2-dibromo (4) or 5-bromo (5) derivatives depending on the reaction conditions. The base catalyzed addition or cycloaddition of naphthopyrones (2) to 4-methoxy benzalacetophenone gave the expected Michael adduct (6) or the cyclic adducts (7–9) depending on the type of catalytic base and reaction conditions. Structures of the products have been established by elemental analyses, IR and 1H NMR spectroscopic methods.  相似文献

4H,6H-[1,3,4]Thiadiazino[2,3-b]quinazolin-6-one with a methyl group in position 3 (6a) has been synthesised by the condensation of 3-amino-2-mercapto-3H-quinazolin-4-one (1) with allyl bromide (2) followed by treatment with bromine and subsequent dehydrohalogenation of the brominated product (4) with ethanolic sodium hydroxide. Its isomeric 3-methyl-2H,6H-[1,3,4]thiadiazino[2,3-b]quinazolin-6-one (6b) has also been obtained by condensation of1 and bromoacetone (7) followed by cyclisation of the intermediates (8 or9) with hydrobromic acid or with concentrated sulphuric acid. The structures have been established on the basis of IR and PMR data.

Zusammenfassung Zur Synthese von 4H,6H-[1,3,4]thiadiazino[2,3-b]chinazolin-6-on (6a) wurde die Kondensation von 3-Amino-2-mercapto-3H-chinazolin-4-on (1) mit Allylbromid mit nachfolgender Behandlung mit Brom und Dehydrohalogenierung des bromierten Produktes4 mit ethanolischer Natronlauge herangezogen. Das zu6a isomere 2H,6H-Produkt6b wurde ebenfalls durch Kondensation von1 mit Bromaceton und nachfolgender Cyclisierung der Zwischenprodukte8 bzw.9 mit HBr oder H2SO4 erhalten. Die Strukturen wurden mittels IR und NMR abgesichert.

New synthesis of 11-acyl-5,11-dihydro-6H-pyrido[2,3-b][1,4]benzodiazepin-6-ones ( 42-44 ) is reported. The crucial steps (Scheme VI) represented N-oxydation of 1 ( 1A ) to 35 ( 35A ), facilitated ring-closure of 36 into 37 , its subsequent N-α-chloroacetylation to 38 , aminolysis to 39-41 (involving N-O anchimeric assistance as depicted in 38A ) and deoxygenation to 42-44 (Scheme VII). The central intermediate 37 is also obtained on oxygenation of 2 , a new synthesis of which was reported in the previous paper of this series [3]. Other attempts of cyclisation “from the top” or “from the bottom” (Scheme I) are described. Thus, interaction of 1 with acetamide afforded 3 and 4 instead of the expected 2A . Compound 5 cyclised into 3-pyridoquinazolone 6 while its 2-(4′-methylpiperazin-1′-yl analogue 9 was observed to be unstable for the attempted ring-opening and reclosure to 42 . “From the bottom” cyclisations of 10A-10C , via intermediary amines 11A-11C failed and pyridoquinazolinone 13 was isolated (Scheme V). The attempted oxidative cyclisation of the compounds 15 and 18 into 2 and 42 , respectively, 13 afforded imidazolo[5,4-b]pyridine derivative (18–19), while 15 remained unchanged. 3-Acylamino-2-arylaminopyridines ( 21-24 ), cyclised into imidazolopyridines 29-30 . Model compounds 45-50 were prepared to study selective aminolysis of the chlorine atoms in 2-chloro-3-(2′-chlorobenzoyl)aminopyridine 1 , and its N-oxide 35 .  相似文献

Diastereoisomers of 4-hydroxy-2-methyl-3,4-dihydro-2H-pyrano[2,3-b]quinolines 8 and 9 were synthesized starting from the appropriate 2-chloroquinoline-3-carboxaldehydes 1. The relative configuration of the 1,3-diol intermediates 4 and 5 was determined on the basis of the 13C-nmr spectra of their acetonides. The relative stereochemistry of title compounds was confirmed by using homonuclear NOE and selective decoupling experiments, as well as by analysis of the coupling patterns observed in their 1H-nmr spectra.  相似文献

In solution the 9-phenylaminotetrahydro-4H-pyrido[1,2-a]pyrimidin-4-ones 1-5 were oxidized into the 9-aminodihydro compounds 15-19 by atmospheric oxygen at ambient temperature. Autoxidation is most probably a free-radical chain process, which takes place with ground-state triplet oxygen via the radical cation of the enamine form. The 9-aminodihydro derivatives were also prepared from 9,9-dibromo compounds 10 and 11 and from 9-hydroxydihydro compounds 12-14 . The 9-hydroxydihydro derivatives, obtained from the 9-amino compounds 16, 19 and 21 by acidic hydrolysis, showed a solvent-dependent and R1 substituent-dependent oxo-enol tautomerism. The enol form was stabilized by electron-withdrawing R1 groups and a polar solvent. However, for the 9-aminodihydropyrido[1,2-a]pyrimidines 15-26 only the enamine tautomer (E) could be identified independently of the substituent and the solvent. The chemical structures of the synthesized products were studied by uv, ir, 1H- and 13C-nmr spectroscopy.  相似文献

The unexpected formation of 11H-pyrido[2,1-b]quinazolin-11-one derivative 6 from 5,11-dihydro-6H-pyrido[2,3-b]-1,4-benzodiazepin-6-one (2) has been observed. Its structure 6 was determined by X-ray crystallography. Detailed nmr study provided a complete set of proton and carbon-13 nmr parameters of compound 6 in solution.  相似文献

Treatment of 2-(4,9-dihydro-3H-pyrido[3,4-b]indol-1-yl)-1-methylcyclohexanol ( 2a ) with acetic anhydride or methyl isocyanate gave 2-acetyl-2,3,4,9-tetrahydro-1-(6-oxoheptylidene)-1H-pyrido[3,4-b]indole ( 3 ) or 1,3,4,9-tetrahydro-N-methyl-1-(6-oxoheptylidene)-2H-pyrido[3,4-b]indole-2-carboxamide ( 4 ), respectively. Simpler analogues, 1-alkyl-4,9-dihydro-3H-pyrido[3,4-b]indoles, 7 , subjected to identical reaction conditions, gave 2-acetyl-1-alkylidene-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indoles 8 and 1,3,4,9-tetrahydro-N-methyl-1-alkyli-dene-2H-pyrido[3,4-b]indole-2-carboxamides 9 , respectively. A limited lanthanide shift reagent study to determine stereochemical assignments was also performed.  相似文献

The acid catalyzed condensation of 4,5-diaminopyrazoles and chalcones gave the hitherto unknown 1-benzyl-2,4,6-triphenyl-2,3-dihydropyrazolo[3,4-b][1,4]diazepines derivatives. The structure of all products was supported by ir, 1H and 13C-nmr and mass spectra.  相似文献

This paper reports the synthesis of new pyrido[2,3-d]pyrimidin-4-one derivatives as diuretic agents. Starting with 1,2-dihydro-5-nitro-2-oxo-3-pyridinecarboxylic acid 1 , ethyl 2-ethoxy-5-nitro-3-pyridincarboxylate 4 was obtained. Compound 4 reacts with ammonia, methylamine or S-methylpseudothiourea to give the respective 2-amino-5-nitro-3-pyridinecarboxamide derivatives 5 and 6 or 2-methylthio-6-nitro-3H-pyrido[2,3-d]pyrimidin-4-one 8. Treating carboxamide 5 with arylaldehydes and zinc dichloride, new 2-aryl-1,2-dihydro-6-nitro-3H-pyrido[2,3-d]pyrimidin-4-ones 9 were synthetised. These compounds reduced with iron(II) hydroxide gave 6-amino-2-aryl-1,2-dihydro-3H-pyrido[2,3-d]pyrimidin-4-ones 10 as expected.  相似文献

Iodide-catalyzed ring expansion of 2-[(1-aziridinylcarbonyl)amino]benzoic acid methyl ester (2) gave 2,3-dihydro-5H-oxazolo[2,3-b]quinazolin-5-one (3) in quantitative yield. Treatment of the dimethyl analog of 2 (9) with sodium iodide in acetone gave a mixture of the 2,3-dihydro-2,2-dimethyl- (10) and 2,3-dihydro-3,3-dimethyl-5H-oxazolo[2,3-b]quinazolin-5-ones (11). However, rearrangement of 9 with sulfuric acid produced only 10. Synthesis of 11 by another route for comparison is described, and the known syntheses of 2,3-dihydro-5H-oxazolo[2,3-b]quinazolin-5-ones are reviewed.  相似文献

2,3-Dihydro-5H-oxazolo[2,3-b]quinazolin-5-one 5a was synthesized from anthranilamide 1 and 2-chloro-ethyl isocyanate either by a direct reflux in methanol, or by stirring at room temperature in acetonitrile leading to the intermediate, 2-(2-chloroethyl ureido)benzamide 6a which was subsequently cyclized on heating with an organic base. However, when compound 6a was refluxed with concentrated hydrochloric acid, it furnished 3-(2-chloroethyl)-2,4-dioxo-1H,3H-quinazoline 2a in good yields. 3,4-Dihydro-2H,6.H-[1,3]-oxazino[2,3-b]quinazolin-6-one 5b , 3-(3-chloropropyl)-2,4-dioxo-1H,3.H-quinazoline 2b and 2-(3-chloropropyl ureido)benzamide 6b were obtained similarly from 1 and 3-chloropropyl isocyanate.  相似文献

A convenient general synthesis of 2,2-dimethyl-3,4-dihydro-2H-pyrano[2,3-b]quinolines using the Wittig reaction is described. The o-nitrobenzaldehydes (1a–d) on reaction with phosphorane 2 provided ( E )-ethyl-α-(2,2-dimethylprop-2-ene)-2-nitrocinnamates (3a–d) in excellent yields, which on cyclization with polyphosphoric acid followed by reductive cyclization using Fe/HCl afforded dihydropyranoquinolines (5a–d). Alternatively, the pyranoquinolines 5a–d were also synthesised from esters 3a–d by employing domino reductive cyclization in a single step.  相似文献

The 13C nmr spectra of 2- or 3-monosubstituted furo[2,3-b]- 1a-1j , furo[3,2-b]- 2a-2j , furo[2,3-c]- 3a-3j and furo[3,2-c]pyridine derivatives 4a-4j are reported. Effects by change in annelation and substituent effects on 13C chemical shifts and carbon-proton coupling constants are discussed. The spectra of benzo[b]furan derivatives 5a-5j having the corresponding substituent are also reported for comparison.  相似文献

Summary A new series of thiophenic isosters of thioxanthones, namely: 2-substituted-4H-thieno[2,3-b][1]benzothiopyran-4-ones and 5-substituted-2-nitro-8-methyl-4H-thieno[2,3-b][1]benzothiopyran-4-ones were synthesized as potential schistosomicidal agents. The synthesized compounds were characterized by their1H-NMR data.

Zusammenfassung Es wurde eine neue Serie von thiophenischen Isosteren des Thioxanthons, nämlich 2-substituierte 4H-Thieno[2,3-b][1]benzothiopyran-4-one und 5-substituierte 2-Nitro-8-methyl-4H-thieno[2,3-b][1]benzothiopyran-4-one als potentielle schistosomicide Wirkstoffe synthetisiert. Die synthetisierten Verbindungen wurden mittels ihrer1H-NMR Daten charakterisiert.
