Burn injuries represent a major life‐threatening event that impacts the quality of life of patients, and places enormous demands on the global healthcare systems. This study introduces the fabrication and characterization of a novel wound dressing made of core–shell hyaluronic acid–silk fibroin/zinc oxide (ZO) nanofibers for treatment of burn injuries. The core–shell configuration enables loading ZO—an antibacterial agent—in the core of nanofibers, which in return improves the sustained release of the drug and maintains its bioactivity. Successful formation of core–shell nanofibers and loading of zinc oxide are confirmed by transmission electron microscopy, Fourier‐transform infrared spectroscopy, and energy dispersive X‐ray. The antibacterial activity of the dressings are examined against Escherichia coli and Staphylococcus aureus and it is shown that addition of ZO improves the antibacterial property of the dressing in a dose‐dependent fashion. However, in vitro cytotoxicity studies show that high concentration of ZO (>3 wt%) is toxic to the cells. In vivo studies indicate that the wound dressings loaded with ZO (3 wt%) substantially improves the wound healing procedure and significantly reduces the inflammatory response at the wound site. Overall, the dressing introduced herein holds great promise for the management of burn injuries.  相似文献

Wound healing, one of the most complex processes of the body involving the cooperation of several important biomolecules and pathways, is one of the major therapeutic and economic issues in regenerative medicine. The present study aimed to introduce a novel electrospun curcumin (Cur)‐incorporated chitosan/polyvinyl alcohol/carbopol/polycaprolactone nanofibrous composite for concurrent delivery of the buccal fat pad‐derived mesenchymal stem cells (BFP‐MSCs) and Cur to a full‐thickness wound on the mouse model. Scaffolds were characterized structurally using scanning electron microscopy (SEM), fluorescence microscopy imaging and Fourier‐transform infrared spectroscopy, and toxicity of the scaffolds was also evaluated after BFP‐MSC seeding by SEM imaging and 3‐(4,5 dimethyiazol‐2‐1)‐2‐5‐diphenyl tetrazolium bromide (MTT) assay. Then, its influence on the wound‐healing process was investigated as a wound dressing for a full‐thickness skin defect in mouse model. Results demonstrated that the designed composite scaffolds have the capability for cell seeding and support their growth and proliferation. Macroscopic and histopathological characteristics were evaluated at the end of the 7 and 14 days after surgery, and their results showed that our designed scaffold groups accelerated the wound‐healing process compared with the control group. Among those, scaffold/Cur, scaffold/Cur/BFP‐MSC and scaffold/BFP‐MSC groups demonstrated more wound repair efficacy. These results indicated that the combined grafts can be used to improve the wound‐healing process, and therefore, the electrospun nanofibers presented in this study, Cur and BFP‐MSC together, were demonstrated to have promising potential for wound‐dressing applications.  相似文献

In this study, wound dressings consisting of dexpanthenol (Dex)-loaded electrospun nanofibers were fabricated using polyvinyl alcohol (PVA)/sodium alginate (SA), and chitosan as the core and the shell, respectively. Considering the remarkable properties of chitosan, it was used as a shell against drug release and to improve the thermal stability, and tensile strength of the scaffold. By comparing the thermogravimetric, and tensile strength results of nanofibers with and without shell, it was revealed that the presence of chitosan in the shell side could improve the thermal stability and increased the tensile strength by about three times. The isotherm models of dexpanthenol release from the PVA/SA/Dex-CS scaffold was best described by the Langmuir model. Besides, Fourier transform infrared, scanning electron microscopy, and X-ray diffraction techniques were performed to characterize nanofibers. Furthermore, an in vivo investigation of a wound dressing with dexpanthenol showed better healing compared to the wound dressings without dexpanthenol.  相似文献

A facile fabrication of a cross-linked hyaluronic acid (HA) hydrogel nanofibers by a reactive electrospinning method is described. A thiolated HA derivative, 3,3'-dithiobis(propanoic dihydrazide)-modified HA (HA-DTPH), and poly(ethylene glycol) diacrylate (PEGDA) are selected as the cross-linking system. The cross-linking reaction occurs simultaneously during the electrospinning process using a dual-syringe mixing technique. Poly(ethylene oxide) (PEO) is added into the spinning solution as a viscosity modifier to facilitate the fiber formation and is selectively removed with water after the electrospinning process. The nanofibrous structure of the electrospun HA scaffold is well preserved after hydration with an average fiber diameter of 110 nm. A cell morphology study on fibronectin (FN)-adsorbed HA nanofibrous scaffolds shows that the NIH 3T3 fibroblasts migrate into the scaffold through the nanofibrous network, and demonstrate an elaborate three-dimensional dendritic morphology within the scaffold, which reflects the dimensions of the electrospun HA nanofibers. These results suggest the application of electrospun HA nanofibrous scaffolds as a potential material for wound healing and tissue regeneration. Poly(lactic acid) (PLA) is a versatile, bioabsorbable, and biodegradable polymer with excellent biocompatibility and ability to incorporate a great variety of active agents. Silver sulfadiazine (SDZ) is an antibiotic used to control bacterial infection in external wounds. Aiming to combine the properties of PLA and SDZ, hydrotalcite ([Mg–Al]‐LDH) was used as a host matrix to obtain an antimicrobial system efficient in delivering SDZ from electrospun PLA scaffolds intended for wound skin healing. The structural reconstruction method was successfully applied to intercalate silver sulfadiazine in the [Mg–Al]‐LDH, as evidenced by X‐ray diffraction and thermogravimetric analyses. Observations by scanning electron microscopy revealed a good distribution of SDZ‐[Mg–Al]‐LDH within the PLA scaffold. Kinetics studies revealed a slow release of SDZ from the PLA scaffold due to the intercalation in the [Mg–Al]‐LDH. In vitro antimicrobial tests indicated a significant inhibitory effect of SDZ‐[Mg–Al]‐LDH against Escherichia coli and Staphylococcus aureus. This antibacterial activity was sustained in the 2.5‐wt% SDZ‐[Mg–Al]‐LDH–loaded PLA nanofibers, which also displayed excellent biocompatibility towards human cells. The multifunctionality of the PLA/SDZ‐[Mg–Al]‐LDH scaffold reported here is of great significance for various transdermal applications.  相似文献

Effective management of full-thickness wounds faces significant challenges due to poor angiogenesis and impaired healing. Biomimetic tissue-engineered scaffolds with angiogenic properties can, however, enhance the regeneration capacity of the damaged skin. Here, we developed a hybrid double-layer nanofibrous scaffold, comprised of egg white (EW) and polyvinyl alcohol (PVA), loaded with niosomal Deferoxamine (NDFO) for enhanced angiogenesis and wound healing features. The hybrid scaffold showed enhanced mechanical properties with comparable modulus and shape-recovery behavior of the human skin. Thanks to the porous morphology and uniform distribution of NDFO within the nanofibers, in vitro drug release studies indicated controlled and sustained release of DFO for up to 9 days. The constructs also promoted a significant increase in vascular sprouting area in vitro and enhanced vascular branches ex vivo. In vivo, implantation of the hybrid scaffold in full-thickness wounds in rats revealed early angiogenic response, a higher number of neo-formed vessels, a faster healing rate and complete epithelialization as early as day 10, compared to the control groups. Thus, the presented biomimetic hybrid scaffold with DFO control release features holds great promise in accelerated full-thickness wound healing and soft tissue regeneration.  相似文献

Controlling the self‐assembly morphology of π‐conjugated block copolymer is of great interesting. Herein, amphiphilic poly(3‐hexylthiophene)‐block‐poly(phenyl isocyanide)s (P3HT‐b‐PPI) copolymers composed of π‐conjugated P3HT and optically active helical PPI segments were readily prepared. Taking advantage of the crystallizable nature of P3HT and the chirality of the helical PPI segment, crystallization‐driven asymmetric self‐assembly (CDASA) of the block copolymers lead to the formation of single‐handed helical nanofibers with controlled length, narrow dispersity, and well‐defined helicity. During the self‐assembly process, the chirality of helical PPI was transferred to the supramolecular assemblies, giving the helical assemblies large optical activity. The single‐handed helical assemblies of the block copolymers exhibited interesting white‐light emission and circularly polarized luminescence (CPL). The handedness and dissymmetric factor of the induced CPL can be finely tuned through the variation on the helicity and length of the helical nanofibers.  相似文献

Unique features of nanofibers provide enormous potential in the field of biomedical and healthcare applications. Many studies have proven the extreme potential of nanofibers in front of current challenges in the medical and healthcare field. This review highlights the nanofiber technologies, unique properties, fabrication techniques (i.e., physical, chemical, and biological methods), and emerging applications in biomedical and healthcare fields. It summarizes the recent researches on nanofibers for drug delivery systems and controlled drug release, tissue‐engineered scaffolds, dressings for wound healing, biosensors, biomedical devices, medical implants, skin care, as well as air, water, and blood purification systems. Attention is given to different types of fibers (e.g., mesoporous, hollow, core‐shell nanofibers) fabricated from various materials and their potential biomedical applications.  相似文献

The novel use of nanofibers as a physical barrier between blood and medical devices has allowed for modifiable, innovative surface coatings on devices ordinarily plagued by thrombosis, delayed healing, and chronic infection. In this study, the nitric oxide (NO) donor S‐nitrosoglutathione (GSNO) is blended with the biodegradable polymers polyhydroxybutyrate (PHB) and polylactic acid (PLA) for the fabrication of hemocompatible, antibacterial nanofibers tailored for blood‐contacting applications. Stress/strain behavior of different concentrations of PHB and PLA is recorded to optimize the mechanical properties of the nanofibers. Nanofibers incorporated with different concentrations of GSNO (10, 15, 20 wt%) are evaluated based on their NO‐releasing kinetics. PLA/PHB + 20 wt% GSNO nanofibers display the greatest NO release over 72 h (0.4–1.5 × 10?10 mol mg?1 min?1). NO‐releasing fibers successfully reduce viable adhered bacterial counts by ≈80% after 24 h of exposure to Staphylococcus aureus. NO‐releasing nanofibers exposed to porcine plasma reduce platelet adhesion by 64.6% compared to control nanofibers. The nanofibers are found noncytotoxic (>95% viability) toward NIH/3T3 mouse fibroblasts, and 4′,6‐diamidino‐2‐phenylindole and phalloidin staining shows that fibroblasts cultured on NO‐releasing fibers have improved cellular adhesion and functionality. Therefore, these novel NO‐releasing nanofibers provide a safe antimicrobial and hemocompatible coating for blood‐contacting medical devices.  相似文献

A new positive working photosensitive poly(benzoxazole) (PBO) precursor based on poly(o‐hydroxyazomethine) ( 3 ) and 1‐{1,1‐bis[4‐(2‐diazo‐1‐(2H)naphthalenone‐5‐sulfonyloxy)phenyl]ethyl}‐4‐{1‐[4‐(2‐diazo‐1(2H)naphthalenone‐5‐sulfonyloxy)phenyl]methylethyl}benzene (S‐DNQ) as a photosensitive compound was developed. 3 was prepared by the condensation of 2,2‐bis(3‐amino‐4‐hydroxyphenyl)hexafluoropropane with isophthalaldehyde in 1‐methyl‐2‐pyrrolidinone/toluene under azeotropic conditions. The photosensitive PBO precursor containing 30 wt % S‐DNQ showed a sensitivity of 120 mJ cm?2 and a contrast of 2.2 when it was exposed to 436‐nm light and developed with a 2.38 wt % aqueous tetramethylammonium hydroxide solution at room temperature. A fine positive image featuring 10‐μm line and space patterns was observed on the film of the photoresist exposed to 200 mJ cm?2 ultraviolet light at 436 nm by the contact mode. An injectable nanofibrous hydrogel scaffold integrated with growth factors (GFs) loaded polysaccharide nanoparticles was developed that specifically allows for targeted adipose‐derived stem cells (ASCs) encapsulation and soft tissue engineering. The nanofibrous hydrogel was produced via biological conjugation of biotin‐terminated star‐shaped poly(ethylene glycol) (PEG‐Biotin) and streptavidin‐functionalized hyaluronic acid (HA‐Streptavidin). The polysaccharide nanoparticles were noncovalently assembled via electrostatic interactions between low‐molecular‐weight heparin (LMWH) and N,N,N‐trimethylchitosan chloride (TMC). Vascular endothelial growth factor (VEGF) was entrapped in the LMWH/TMC nanoparticles by affinity interactions with LMWH.  相似文献

Summary: The complexation between polystyrene‐block‐poly(acrylic acid) (PS‐b‐PAA) micelles and poly(ethylene glycol)‐block‐poly(4‐vinyl pyridine) (PEG‐b‐P4VP) is studied, and a facile strategy is proposed to prepare core‐shell‐corona micellar complexes. Micellization of PS‐b‐PAA in ethanol forms spherical core‐shell micelles with PS block as core and PAA block as shell. When PEG‐b‐P4VP is added into the core‐shell micellar solution, the P4VP block is absorbed into the core‐shell micelles to form spherical core‐shell‐corona micellar complexes with the PS block as core, the combined PAA/P4VP blocks as shell and the PEG block as corona. A model is suggested to characterize the core‐shell‐corona micellar complexes.

A versatile template‐assisted strategy for the preparation of monodispersed rattle‐type hybrid nanospheres, encapsulating a movable Au nanocore in the hollow cavity of a hairy electroactive polymer shell (Au@air@PTEMA‐g‐P3HT hybrid nanorattles; PTEMA: poly(2‐(thiophen‐3‐yl)ethyl methacrylate; P3HT: poly(3‐hexylthiophene), was reported. The Au@silica core‐shell nanoparticles, prepared by the modified Stöber sol–gel process on Au nanoparticle seeds, were used as templates for the synthesis of Au@silica@PTEMA core‐double shell nanospheres. Subsequent oxidative graft polymerization of 3‐hexylthiophene from the exterior surface of the Au@silica@PTEMA core‐double shell nanospheres allowed the tailoring of surface functionality with electroactive P3HT brushes (Au@silica@PTEMA‐g‐P3HT nanospheres). The Au@air@ PTEMA‐g‐P3HT hybrid nanorattles were obtained after etching of the silica interlayer by HF. The as‐prepared nanorattles were dispersed into an electrically insulating polystyrene matrix and for the first time used to fabricate nonvolatile memory devices. As a result, unique electrical behaviors, including insulator behavior, write‐once‐read‐many‐times and rewritable memory effects, and conductor behavior as well, were observed in the Al/Au@air@PTEMA‐g‐P3HT+PS/ITO (ITO: indium‐tin oxide) sandwich thin‐film devices.  相似文献

Worm‐like aggregates with a PAA/P4VP complex core and a PEG/PNIPAM mixed shell were prepared in ethanol by the comicellization of poly(ethylene glycol)‐block‐poly(acrylic acid) (PEG‐b‐PAA) and poly(N‐isopropylacrylamide)‐block‐poly(4‐vinylpyridine) (PNIPAM‐b‐P4VP) through hydrogen‐bonding. The formed aggregates were studied by dynamic light scattering, static light scattering, 1H NMR, and transmission electron microscopy. The length of worm‐like aggregates could be adjusted by changing the weight ratio of W(PNIPAM‐b‐P4VP)/W(PEG‐b‐PAA). When the ratio changed from 20 to 150%, the length changed from about 100 nm to several microns, and the diameter stayed almost unchanged at about 15 nm.

Novel rubbery wound closures containing various proportions and molecular weights of polyisobutylene (PIB) and poly(2‐octyl cyanoacrylate) [P(OctCA)] for potential clinical use were designed, synthesized, characterized, and tested. Homo‐networks were prepared by crosslinking 3‐arm star‐shaped PIBs fitted with terminal cyanoacrylate groups, [Ø(PIB‐CA)3], and co‐networks by copolymerizing Ø(PIB‐CA)3 with OctCA using N‐dimethyl‐p‐toluidine (DMT). Neat Ø(PIB‐CA)3, and Ø(PIB‐CA)3/OctCA blends, upon contact with initiator, polymerize within seconds to optically transparent strong rubbery co‐networks, Ø(PIB‐CA)3co‐P(OctCA). Homo‐ and co‐network formation was demonstrated by sol/gel studies, and structures and properties were characterized by a battery of techniques. The Tg of P(OctCA) is 58 °C by DSC, and 75 °C by DMTA. Co‐networks comprising 25% Ø(PIB‐CA)3 (Mn = 2400 g/mol) and 75% P(OctCA) are stronger and more extensible than skin. Short and long term creep studies show co‐networks exhibit high dimensional stability and <6% creep strain at high loading. When deposited on porcine skin co‐networks yield hermetically‐adhering clear rubbery coatings. Strips of porcine skin coated with co‐networks could be stretched and twisted without compromising membrane integrity. Current therapeutic interventions in bone defects are mainly focused on finding the best bioactive materials for inducing bone regeneration via activating the related intracellular signaling pathways. Integrins are trans‐membrane receptors that facilitate cell‐extracellular matrix (ECM) interactions and activate signal transduction. To develop a suitable platform for supporting human bone marrow mesenchymal stem cells (hBM‐MSCs) differentiation into bone tissue, electrospun poly L‐lactide (PLLA) nanofiber scaffolds were coated with nano‐hydroxyapatite (PLLA/nHa group), gelatin nanoparticles (PLLA/Gel group), and nHa/Gel nanoparticles (PLLA/nHa/Gel group) and their impacts on cell proliferation, expression of osteoblastic biomarkers, and bone differentiation were examined and compared. MTT data showed that proliferation of hBM‐MSCs on PLLA/nHa/Gel scaffolds was significantly higher than other groups (P < .05). Alkaline phosphatase activity was also more increased in hBM‐MSCs cultured under osteogenic media on PLLA/nHa/Gel scaffolds compared to others. Gene expression evaluation confirmed up‐regulation of integrin α2β1 as well as the osteogenic genes BGLAP, COL1A1, and RUNX2. Following use of integrin α2β1 blocker antibody, the protein level of integrin α2β1 in cells seeded on PLLA/nHa/Gel scaffolds was decreased compared to control, which confirmed that most of the integrin receptors were bound to gelatin molecules on scaffolds and could activate the integrin α2β1/ERK axis. Collectively, PLLA/nHa/Gel scaffold is a suitable platform for hBM‐MSCs adhesion, proliferation, and osteogenic differentiation in less time via activating integrin α2β1/ERK axis, and thus it might be applicable in bone tissue engineering.  相似文献

A novel nanofibrous mat featuring an ultraviolet (UV)‐induced CO2‐responsive behavior was fabricated via electrospinning and used as a controlled drug release system. First, a random copolymer for electrospinning, poly(N,N‐diethylaminoethyl acrylamide‐coN‐benzylacrylamide‐coN,N‐dimethyl‐N‐(2‐nitrobenzyl)‐ethaneamine acrylamide‐co‐4‐acryloyloxy benzophenone) [P(DEEA‐co‐BA‐co‐DMNOBA‐co‐ABP)], was prepared based on pentafluorophenyl esters via an “active ester‐amine” chemistry reaction. Subsequently, doxorubicin hydrochloride (DOX)‐loaded P(DEEA‐co‐BA‐co‐DMNOBA‐co‐ABP) nanofibers were fabricated, yielding a new drug‐loaded nanofibrous mat as a potential wound dressing. These DOX‐loaded nanofibers can respond to UV irradiation and CO2 stimulation. Interestingly, without UV irradiation, the fabricated nanofibers cannot exhibit any responsiveness. Therefore, the majority of the DOX was steadily stored in the nanofibers, even in the presence of CO2. However, upon UV irradiation, the CO2‐responsive behavior of the nanofibers was activated and the prepared nanofibers swelled slightly, resulting in the release of around 42% DOX from the nanofibers. Upon further purging with CO2, the release amount of DOX from the nanofibers could reach up to approximately 85%, followed by the morphological transition from a nanofibrous mat to a porous hydrogel film. © 2019 Wiley Periodicals, Inc. Scaffold, an essential element of tissue engineering, should provide proper physical and chemical properties and evolve suitable cell behavior for tissue regeneration. Polycaprolactone/Gelatin (PCL/Gel)‐based nanocomposite scaffolds containing hydroxyapatite nanoparticles (nHA) and vitamin D3 (Vit D3) were fabricated using the electrospinning method. Structural and mechanical properties of the scaffold were determined by scanning electron microscopy (SEM) and tensile measurement. In this study, smooth and bead‐free morphology with a uniform fiber diameter and optimal porosity level with appropriate pore size was observed for PCL/Gel/nHA nanocomposite scaffold. The results indicated that adding nHA to PCL/Gel caused an increase of the mechanical properties of scaffold. In addition, chemical interactions between PCL, gelatin, and nHA molecules were shown with XRD and FT‐IR in the composite scaffolds. MG‐63 cell line has been cultured on the fabricated composite scaffolds; the results of viability and adhesion of cells on the scaffolds have been confirmed using MTT and SEM analysis methods. Here in this study, the culture of the osteoblast cells on the scaffolds showed that the addition of Vit D3 to PCL/Gel/nHA scaffold caused further attachment and proliferation of the cells. Moreover, DAPI staining results showed that the presence and viability of the cells were greater in PCL/Gel/nHA/Vit D3 scaffold than in PCL/Gel/nHA and PCL/Gel scaffolds. The results also approved increasing cell proliferation and alkaline phosphatase (ALP) activity for MG‐63 cells cultured on PCL/Gel/nHA/Vit D3 scaffold. The results indicated superior properties of hydroxyapatite nanoparticles and vitamin D3 incorporated in PCL/Gel scaffold for use in bone tissue engineering.  相似文献

We report the synthesis and gradient stimuli‐responsive properties of cyclodextrin‐overhanging hyperbranched core‐double‐shell miktoarm architectures. A ionic hyperbranched poly(β‐cyclodextrin) (β‐CD) core was firstly synthesized via a convenient “A2+B3” approach. Double‐layered shell architectures, composed of poly(N‐isopropyl acrylamide) (PNIPAm) and poly(N,N‐dimethylaminoethyl methacrylate) (PDMAEMA) miktoarms as the outermost shell linked to poly(N,N‐diethylaminoethyl methacrylate) (PDEAEMA) homoarms which form the inner shell, were obtained by a sequential atom transfer radical polymerization (ATRP) and parallel click chemistry from the modified hyperbranched poly(β‐CD) macroinitiator. The combined characterization by 1H NMR, 13C NMR, 1H‐29Si heteronuclear multiple‐bond correlation (HMBC), FTIR and size exclusion chromatography/multiangle laser light scattering (SEC/MALLS) confirms the remarkable hyperbranched poly(β‐CD) core and double‐shell miktoarm architectures. The gradient triple‐stimuli‐responsive properties of hyperbranched core‐double‐shell miktoarm architectures and the corresponding mechanisms were investigated by UV–vis spectrophotometer and dynamic light scattering (DLS). Results show that this polymer possesses three‐stage phase transition behaviors. The first‐stage phase transition comes from the deprotonation of PDEAEMA segments at pH 9–10 aqueous solution under room temperature. The confined coil‐globule conformation transition of PNIPAm and PDMAEMA arms gives rise to the second‐stage hysteretic cophase transition between 38 and 44 °C at pH 10. The third‐stage phase transition occurs above 44 °C at pH = 10 attributed to the confined secondary conformation transition of partial PDMAEMA segments. This cyclodextrin‐overhanging hyperbranched core‐double‐shell miktoarm architectures are expected to solve the problems of inadequate functionalities from core layer and lacking multiresponsiveness for shell layers existing in the dendritic core‐multishell architectures. © 2011 Wiley Periodicals, Inc. A novel and facile bottom‐up strategy for preparing core‐shell nanofibers with selectively localized carbon nanotubes is developed using hierarchical composite micelles of crystalline‐coil copolymer and carbon nanotubes as the building blocks. An amphiphilic di‐block copolymer of poly (p‐dioxanone) (PPDO) and PEG (polyethylene glycol) functionalized with pyrene moieties at the chain ends of PPDO blocks (Py‐PPDO‐b‐PEG) is designed for constructing composite micelles with multiwalled carbon nanotubes (MWCNTs). The self‐assembly of Py‐PPDO‐b‐PEG and MWCNTs is co‐induced by the crystallization of PPDO blocks and the π–π stacking interactions between pyrene moieties and MWCNTs, resulting in composite micelles with “shish kebab”‐like nanostructure. A mixture of composite micelles and polyvinyl alcohol (PVA) water solution is then used as the spinning solution for preparing electrospun nanofibers. The morphologies of the nanofibers with different composition are investigated by SEM and TEM. The results suggest that the MWCNTs selectively localized in the core of the nanofibers of MWCNTs/Py‐PPDO‐b‐PEG/PVA. The alignment and interfusion of composite micelles during the formation of nanofibers may confine the carbon nanotubes in the hydrophobic core region. In contrast, the copolymer without pyrene moieties cannot form composite micelles, thus these nanofibers show selective localization of MWCNTs in the PVA shell region.
