Atopic dermatitis (AD ) is a widespread chronic skin condition that severely affects quality of life and can lead to more serious complications. Although ultraviolet (UV )A eye irradiation can exert various effects on the skin, it is unknown whether UVA can affect AD . To investigate potential associations, we used an NC /Nga mouse model of AD to study the effects of UVA eye irradiation. The eyes of mice were irradiated with a UVA dose of 100 kJ m−2 using a FL 20SBLB ‐A lamp. Our histological data demonstrated that AD symptoms could be ameliorated by UVA eye irradiation. We also observed an increase in the levels of adrenocorticotropic hormone (ACTH ), p53 and retinoid X receptor α (RXR α ) in mice with UVA ‐irradiated eyes. In contrast, the levels of thymic stromal lymphopoietin (TSLP ), period 2 (PER 2) and differentiated embryo chondrocytes 1 (DEC 1) protein were decreased in mice treated with UVA irradiation. Furthermore, UVA eye‐irradiated mice exhibited reduced DEC 1 and RXR α colocalization compared with nonirradiated mice. These results suggested that p53 and various clock gene proteins played important roles in the amelioration of AD symptoms observed after UVA eye irradiation; this technique may have therapeutic applications in AD .  相似文献

Itching dermatitis, which is very similar to human atopic dermatitis, arises spontaneously in NC/Nga mice. Changes in lesional skin and the plasma concentration of IgE during the development of atopic dermatitis-like disease up to 8 weeks after the start of picryl chloride (PiCl) in NC/Nga mice were examined. In the mice which atopic dermatitis had been induced, 28-day administration of α cyclodextrin (α CD) significantly inhibited aggravation of detmatitis without having effects on body weight. Clinical signs and symptoms seen in PiCl treated NC/Nga mice began with erythema and haemorrhage, following oedema, superficial erosion, deep excoriation, scaling and dryness of the skin. An increase was observed in the number of mast cells and eosinophil infiltration in the lesional skin. Almost human patients with nasal inflammation or bronchial asthmatics symptoms disappeared by oral administration of α CD (5 mg/day) for 2 month. These results suggest that α CD is useful for treatment of human atopic dermatitis.  相似文献

Summary: Equilibrium and local dynamic properties of ordered polymer networks with included rod‐like particles are considered using a simplified network model. Lagrange multipliers in the equations of motion of rigid rods are replaced by their averaged values. This approximation corresponds to modelling rod‐like particles by elastic Gaussian springs with mean‐square lengths independent of the orientational order. Nematic‐like interactions between network segments and rods are taken into account in terms of the Maier‐Saupe mean‐field approximation. Nematic ordering of rods induces network segments ordering and changes the relaxation spectrum of the network. The relaxation spectrum of the ordered network splits into two main branches for parallel and perpendicular components of chain segments with respect to the director. Relaxation times of a polymer network are calculated as functions of the wave number for the corresponding normal mode and of the order parameter taking into account both the dynamic factor (determined by friction effects) and the statistical factor (related to mean‐square fluctuations of segment projections). We compare the relaxation spectra of ordered unstretched polymer networks with fixed boundaries with those for polymer networks at free boundaries. A polymer network with free boundaries is stretched along the director. This produces additional fine structure of the two main branches in the relaxation spectrum.

Atopic dermatitis (eczema) is a condition that makes skin red and itchy. Though common in children, the condition can occur at any age. Atopic dermatitis is persistent (chronic) and tends to recur periodically. It may be accompanied by asthma or hay fever. No cure has been found for eczema. Therefore, it is very important to develop ingredients that aid the prevention and treatment of atopic dermatitis. Cycloheterophyllin is derived from Artocarpus heterophyllus and has antioxidant and anti-inflammatory activities. However, it still is not understood whether cycloheterophyllin is an anti-atopic dermatitis agent. Keratinocytes (HaCaT cells) and BALB/c mice for inducing AD-like cutaneous lesions were used to evaluate the potential of cycloheterophyllin as an anti-atopic dermatitis agent. The release of pro-inflammatory cytokines induced by treatment of TNF-α/IFN-γ was reduced after pretreatment with cycloheterophyllin. The inhibitory effects could be a contribution from the effect of the MAP kinases pathway. Moreover, the symptoms of atopic dermatitis (such as red skin and itching) were attenuated by pretreatment with cycloheterophyllin. Epidermal hyperplasia and mast cell infiltration were decreased in the histological section. Finally, damage to the skin barrier was also found to recover through assessment of transepidermal water loss. Taken together, prenylflavone-cycloheterophyllin from Artocarpus heterophyllus is a potential anti-atopic dermatitis ingredient that can be used in preventing or treating the condition.  相似文献

Photosynthesis (Pn) and photomorphogenesis (Pm) are affected by light quality, light intensity and photoperiod. Although blue light (BL) is necessary for normal development, it is less efficient in driving Pn than other wavelengths of photosynthetically active radiation. The effects of BL on Pm are highly species dependent. Here we report the interacting effects of BL and photosynthetic photon flux (PPF) on growth and development of lettuce, radish and pepper. We used light‐emitting diode (LED) arrays to provide BL fractions from 11% to 28% under broad‐spectrum white LEDs, and from 0.3% to 92% under monochromatic LEDs. All treatments were replicated three times at each of two PPFs (200 and 500 μmol m?2 s?1). Other than light quality, environmental conditions were uniformly maintained across chambers. Regardless of PPF, BL was necessary to prevent shade‐avoidance responses in radish and lettuce. For lettuce and radish, increasing BL reduced stem length, and for both species, there were significant interactions of BL with PPF for leaf expansion. Increasing BL reduced petiole length in radish and flower number in pepper. BL minimally affected pepper growth and other developmental parameters. Pepper seedlings were more photobiologically sensitive than older plants. Surprisingly, there were few interactions between monochromatic and broad‐spectrum light sources.  相似文献

The extracts of Schisandra chinensis (Turcz.) Baill. (Schisandraceae) have various therapeutic effects, including inflammation and allergy. In this study, gomisin M2 (GM2) was isolated from S. chinensis and its beneficial effects were assessed against atopic dermatitis (AD). We evaluated the therapeutic effects of GM2 on 2,4-dinitrochlorobenzene (DNCB) and Dermatophagoides farinae extract (DFE)-induced AD-like skin lesions with BALB/c mice ears and within the tumor necrosis factor (TNF)-α and interferon (IFN)-γ-stimulated keratinocytes. The oral administration of GM2 resulted in reduced epidermal and dermal thickness, infiltration of tissue eosinophils, mast cells, and helper T cells in AD-like lesions. GM2 suppressed the expression of IL-1β, IL-4, IL-5, IL-6, IL-12a, and TSLP in ear tissue and the expression of IFN-γ, IL-4, and IL-17A in auricular lymph nodes. GM2 also inhibited STAT1 and NF-κB phosphorylation in DNCB/DFE-induced AD-like lesions. The oral administration of GM2 reduced levels of IgE (DFE-specific and total) and IgG2a in the mice sera, as well as protein levels of IL-4, IL-6, and TSLP in ear tissues. In TNF-α/IFN-γ-stimulated keratinocytes, GM2 significantly inhibited IL-1β, IL-6, CXCL8, and CCL22 through the suppression of STAT1 phosphorylation and the nuclear translocation of NF-κB. Taken together, these results indicate that GM2 is a biologically active compound that exhibits inhibitory effects on skin inflammation and suggests that GM2 might serve as a remedy in inflammatory skin diseases, specifically on AD.  相似文献

Many natural terpenoid alkaloid conjugates show biological activity because their structures contain both sp3‐rich terpenoid scaffolds and nitrogen‐containing alkaloid scaffolds. However, their biosynthesis utilizes a limited set of compounds as sources of the terpenoid moiety. The production of terpenoid alkaloids containing various types of terpenoid moiety may provide useful, chemically diverse compound libraries for drug discovery. Herein, we report the construction of a library of terpenoid alkaloid‐like compounds based on Lewis‐acid‐catalyzed transannulation of humulene diepoxide and subsequent sequential olefin metathesis. Cheminformatic analysis quantitatively showed that the synthesized terpenoid alkaloid‐like compound library has a high level of three‐dimensional‐shape diversity. Extensive pharmacological screening of the library has led to the identification of promising compounds for the development of antihypolipidemic drugs. Therefore, the synthesis of terpenoid alkaloid‐like compound libraries based on humulene is well suited to drug discovery. Synthesis of terpenoid alkaloid‐like compounds based on several natural terpenoids is an effective strategy for producing chemically diverse libraries.  相似文献

One of the main targets of studies on water splitting photocatalysts is to develop semiconductor materials with narrower bandgaps capable of overall water splitting for efficient harvesting of solar energy. A series of transition‐metal oxynitrides, LaMgxTa1?xO1+3xN2?3x (x≥1/3), with a complex perovskite structure was reported as the first example of overall water splitting operable at up to 600 nm. The photocatalytic behavior of LaMg1/3Ta2/3O2N was investigated in detail in order to optimize photocatalyst preparation and water‐splitting activity. Various attempts exploring photocatalyst preparation steps, that is, cocatalyst selection, coating material and method, and synthesis method for the oxide precursor, revealed photocatalyst structures necessary for achieving overall water splitting. Careful examination of photocatalyst preparation procedures likely enhanced the quality of the produced photocatalyst, leading to a more homogeneous coating quality and semiconductor particles with fewer defects. Thus, the photocatalytic activity for water splitting on LaMg1/3Ta2/3O2N was largely enhanced.  相似文献

The surface modification of the fullerene‐like hydrogenated carbon (FL‐C:H) film was achieved by bombardment using Ar, H, and N ions, respectively. A systematic comparison of X‐ray photoelectron spectroscopy (XPS) and Fourier transformation infrared(FTIR) spectra was made between the FL‐C:H film and ion‐bombarded films. The results show that ion bombardment resulted in the increase of sp3 C content, specially, new C? N bonds were formed for N‐ion‐bombarded film. The contact angle (CA) and friction coefficient of those films were measured. The surface free energy evaluated from the contact angle increased for ion‐bombarded films, and the most obvious increase was obtained for N‐ion‐bombarded film. The friction coefficient decreased for H‐ion‐bombarded film whereas it increased for N‐ion‐bombarded film, and the friction coefficient of Ar‐ion‐bombarded film was close to that of the FL‐C:H film. The formation and the effects of laser irradiation of the complex formed by protoporphyrin IX (PPIX) and tubulin was investigated. We have used tubulin as a model protein to investigate whether docked photoactive ligands can affect the structure and function of polypeptides upon exposure to visible light. We observed that laser irradiation in the Soret band prompts bleaching of the PPIX, which is accompanied by a sharp decrease in the intensity and average fluorescence lifetime of the protein (dominated by the four tryptophan residues of the tubulin monomer). The kinetics indicate non‐trivial effects and suggest that the photosensitization of the PPIX bound to tubulin prompts structural alterations of the protein. These modifications were also observed through changes in the energy transfer between Trp residues and PPIX. The results suggest that laser irradiation produces localized partial unfolding of tubulin and that the changes prompt modification of the formation of microtubules in vitro. Measurements of singlet oxygen formation were inconclusive in determining whether the changes are prompted by reactive oxygen species or other excited state mechanisms.  相似文献

Resistance of bacterial endospores to treatments, including biocides, heat and radiation is a persistent problem. This study investigates the susceptibility of Bacillus and Clostridium endospores to 405 nm visible light, wavelengths which have been shown to induce inactivation of vegetative bacterial cells. Suspensions of B. cereus endospores were exposed to high‐intensity 405 nm light generated from a light‐emitting diode array and results demonstrate the induction of a sporicidal effect. Up to a 4‐log10 CFU mL?1 reduction in spore population was achieved after exposure to a dose of 1.73 kJ cm?2. Similar inactivation kinetics were demonstrated with B. subtilis, B. megaterium and C. difficile endospores. The doses required for inactivation of endospores were significantly higher than those required for inactivation of B. cereus and C. difficile vegetative cells, where ca 4‐log10 CFU mL?1 reductions were achieved after exposure to doses of 108 and 48 J cm?2, respectively. The significant increase in dose required for inactivation of endospores compared with vegetative cells is unsurprising due to the notorious resilience of these microbial structures. However, the demonstration that visible light of 405 nm can induce a bactericidal effect against endospores is significant, and could have potential for incorporation into decontamination methods for the removal of bacterial contamination including endospores.  相似文献

Ultraviolet light B (UVB) exposure induces cutaneous squamous cell carcinoma (cSCC), one of the most prevalent human cancers. Reoccurrence of cSCC in high‐risk patients is prevented by oral retinoids. But oral retinoid treatment causes significant side effects; and patients develop retinoid resistance. Exactly how retinoids prevent UVB‐induced cSCC is currently not well understood. Retinoid resistance blocks mechanistic studies in the leading mouse model of cSCC, the UVB‐exposed SKH‐1 hairless mouse. To begin to understand the role of retinoids in UVB‐induced cSCC we first examined the localization pattern of key retinoid metabolism proteins by immunohistochemistry 48 h after UVB treatment of female SKH‐1 mice. We next inhibited retinoic acid (RA) synthesis immediately after UVB exposure. Acute UVB increased RA synthesis, signaling and degradation proteins in the stratum granulosum. Some of these proteins changed their localization; while other proteins just increased in intensity. In contrast, acute UVB reduced the retinoid storage protein lectin:retinol acyltransferase (LRAT) in the epidermis. Inhibiting RA synthesis disrupted the epidermis and impaired differentiation. These data suggest that repair of the epidermis after acute UVB exposure requires endogenous RA synthesis.  相似文献

A series of brush‐like amphiphilic organopolysiloxanes with varying hydrophilic side‐chains was prepared, and the assembly behavior of these promising polymers was investigated in aqueous solution using a combination method of surface tension, steady‐state fluorescence, dynamic light scattering, and transmission electron microscopy. An increasing number of side‐chains could lead a higher surface tension of the polymer solution. The polymers formed regular “micelle‐like” spherical multipolymer assemblies in aqueous solution with the size distributed from the scale of hundreds to that of tens of nanometer, and the polymers that possessed more of the side‐chains would form comparatively loose and swollen assemblies with slightly higher micropolarities and bigger dimensions. The interesting discovery in this report was that the visible clearness of the solution could be improved by increasing the hydrophilicity of the assemblies in the solution. Light‐harvesting complexes (LHC) in photosynthetic organisms perform the major function of light absorption and energy transportation. Optical spectrum of LHC provides a detailed understanding of the molecular mechanisms involved in the excitation energy transfer (EET) processes, which has been widely studied. Here, we study how the geometric property of LHC in Rhodospirillum (Rs.) molischianum would affect its spectral characteristics and energy transfer process. By adopting the effective Hamiltonian and the dipole–dipole approximation, we calculate the exciton level structures for the LH2 ring and LH1 ring and the energy transfer time between different LHCs under various structural parameters and different rotational symmetries. Our numerical results show that the LHC's absorption peaks and the energy transfer time between different LHCs can be modified by changing the geometric configurations. Our study may be beneficial to the applications in designing highly efficient photovoltaic cell and other artificial photosynthetic systems.  相似文献
