Ghost membranes prepared from human erythrocytes exhibit 2 distinct (Ca + Mg)-ATPase1 activities (Quist and Roufogalis, Arch Biochem Biophys 168:240, 1975). (Ca + Mg)-ATPase activity dependent on a water soluble protein fraction is selectively lost from ghost membranes during preparation of vesicles under low ionic strength, slightly alkaline conditions. In this study, the Ca2+ dependence of the remaining membrane bound (Ca + Mg)-ATPase activity and ATP-dependent Ca uptake in vesicles were compared. The Ca2+ activation curves for (Ca + Mg)-ATPase activity and Ca uptake into vesicles were parallel over a Ca2+ range of 0.3-330 micrometer, and both curves have 2 apparent KA values for Ca2+ of 0.45 and 100 micrometer. Addition of a concentrated soluble protein fraction containing predominantly spectrin to the vesicles increased (Ca + Mg)-ATPase activity over twofold but did not affect the rate of Ca uptake. These findings suggest that the (Ca + Mg)-ATPase activity remaining in vesicles after extraction of the water soluble proteins is associated with the Ca pump whereas (Ca + Mg)-ATPase activity dependent on the soluble protein fraction is associated with some other function.  相似文献

Ca2+, "a signal of life and death", controls numerous cellular processes through interactions with proteins. An effective approach to understanding the role of Ca2+ is the design of a Ca2+-binding protein with predicted structural and functional properties. To design de novo Ca2+-binding sites in proteins is challenging due to the high coordination numbers and the incorporation of charged ligand residues, in addition to Ca2+-induced conformational change. Here, we demonstrate the successful design of a Ca2+-binding site in the non-Ca2+-binding cell adhesion protein CD2. This designed protein, Ca.CD2, exhibits selectivity for Ca2+ versus other di- and monovalent cations. In addition, La3+ (Kd 5.0 microM) and Tb3+ (Kd 6.6 microM) bind to the designed protein somewhat more tightly than does Ca2+ (Kd 1.4 mM). More interestingly, Ca.CD2 retains the native ability to associate with the natural target molecule. The solution structure reveals that Ca.CD2 binds Ca2+ at the intended site with the designed arrangement, which validates our general strategy for designing de novo Ca2+-binding proteins. The structural information also provides a close view of structural determinants that are necessary for a functional protein to accommodate the metal-binding site. This first success in designing Ca2+-binding proteins with desired structural and functional properties opens a new avenue in unveiling key determinants to Ca2+ binding, the mechanism of Ca2+ signaling, and Ca2+-dependent cell adhesion, while avoiding the complexities of the global conformational changes and cooperativity in natural Ca2+-binding proteins. It also represents a major achievement toward designing functional proteins controlled by Ca2+ binding.  相似文献

Sodium alginate (Alg) hydrogel films were crosslinked with either calcium poly(γ‐glutamate) (Ca‐PGA) or CaCl2. The hydrophilicity of the resulting hydrogel films was evaluated through swelling tests, water retention capacity tests, and water vapor permeation tests. The swelling ratio, water retention capacity, and the water vapor transmission rate (WVTR) of Alg/Ca‐PGA were higher than those of Ca‐Alg. The swelling ratio of Alg/Ca‐PGA was 651 and 190% at pH 7.4 and pH 1.2, respectively. The tensile strength of Alg/Ca‐PGA hydrogel was lower than that of Ca‐Alg. The results of hemocompatibility test showed that Alg/Ca‐PGA caused shorter activated partial thromboplastin time (APTT) than Ca‐Alg. Both Ca‐Alg and Alg/Ca‐PGA exhibited almost no adsorption of human serum albumin (HSA), whereas the adsorption of human plasma fibrinogen (HPF) of Ca‐Alg was 10 times of that of Alg/Ca‐PGA. In addition, Alg/Ca‐PGA exhibited platelet adhesion higher than Ca‐Alg. Furthermore, both Alg/Ca‐PGA and Ca‐Alg exhibited no cytotoxicity. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

Doxorubicin (DOX) is one of the most potent anticancer drugs and induces acute cardiac arrhythmias and chronic cumulative cardiomyopathy. Though DOX-induced cardiotoxicity is known to be caused mainly by ROS generation, a disturbance of Ca2+ homeostasis is also implicated one of the cardiotoxic mechanisms. In this study, a molecular basis of DOX-induced modulation of intracellular Ca2+ concentration ([Ca2+]i) was investigated. Treatment of adult rat cardiomyocytes with DOX increased [Ca2+]i irrespectively of extracellular Ca2+, indicating DOX-mediated Ca2+ release from intracellular Ca2+ stores. The DOX-induced Ca2+ increase was slowly processed and sustained. The Ca2+ increase was inhibited by pretreatment with a sarcoplasmic reticulum (SR) Ca2+ channel blocker, ryanodine or dantrolene, and an antioxidant, alpha-lipoic acid or alpha-tocopherol. DOX-induced ROS generation was observed immediately after DOX treatment and increased in a time-dependent manner. The ROS production was significantly reduced by the pretreatment of the SR Ca2+ channel blockers and the antioxidants. Moreover, DOX-mediated activation of caspase-3 was significantly inhibited by the Ca2+ channel blockers and a-lipoic acid but not a-tocopherol. In addition, cotreatment of ryanodine with alpha-lipoic acid resulted in further inhibition of the casapse-3 activity. These results demonstrate that DOX-mediated ROS opens ryanodine receptor, resulting in an increase in [Ca2+]i and that the increased [Ca2+]i induces ROS production. These observations also suggest that DOX/ROS-induced increase of [Ca2+]i plays a critical role in damage of cardiomyocytes.  相似文献

Treatment of calcium bromide with 3,5-di-tert-butylpyrazolatopotassium (2 equiv) in tetrahydrofuran afforded Ca(tBu2pz)2(THF)2 (69%). The reaction of this compound with pyridine (3 equiv), tetramethylethylenediamine (TMEDA, 1 equiv), N,N,N',N',N"-pentamethyldiethylenetriamine (PMDETA, 1 equiv), triglyme (1 equiv), and tetraglyme (1 equiv) yielded Ca(tBu2pz)2(py)3 (51%), Ca(tBu2pz)2(TMEDA) (74%), Ca(tBu2pz)2(PMDETA) (50%), Ca(tBu2pz)2(triglyme) (73%), and Ca(tBu2pz)2(tetraglyme) (57%), respectively. Treatment of the tetrahydrofuran adduct of Ca(Me2pz)2, generated in situ, with PMDETA (1 equiv), triglyme (1 equiv), and tetraglyme (1 equiv) afforded Ca(Me2pz)2(PMDETA) (65%), Ca(Me2pz)2(triglyme) (54%), and Ca(Me2pz)2(tetraglyme) (40%), respectively. The X-ray crystal structures of Ca(tBu2pz)2(py)3, Ca(tBu2pz)2(TMEDA), Ca(tBu2pz)2(PMDETA), Ca(tBu2pz)2(triglyme), and Ca(Me2pz)2(PMDETA) revealed six-, seven-, or eight-coordinate calcium centers with eta 2-pyrazolato ligands. Ca(tBu2pz)2(triglyme) sublimes at 160 degrees C (0.1 mmHg). The potential utility of these complexes as source compounds for chemical vapor deposition processes is discussed.  相似文献

Altered intracellular Ca2+ dynamics are characteristically observed in cardiomyocytes from failing hearts. Studies of Ca2+ handling in myocytes predominantly use Fluo-3 AM, a visible light excitable Ca2+ chelating fluorescent dye in conjunction with rapid line-scanning confocal microscopy. However, Fluo-3 AM does not allow for traditional ratiometric determination of intracellular Ca2+ concentration and has required the use of mathematic correction factors with values obtained from separate procedures to convert Fluo-3 AM fluorescence to appropriate Ca2+ concentrations. This study describes methodology to directly measure intracellular Ca2+ levels using inactivated, Fluo-3-AM-loaded cardiomyocytes equilibrated with Ca2+ concentration standards. Titration of Ca2+ concentration exhibits a linear relationship to increasing Fluo-3 AM fluorescence intensity. Images obtained from individual myocyte confocal scans were recorded, average pixel intensity values were calculated, and a plot is generated relating the average pixel intensity to known Ca2+ concentrations. These standard plots can be used to convert transient Ca2+ fluorescence obtained with experimental cells to Ca2+ concentrations by linear regression analysis. Standards are determined on the same microscope used for acquisition of unknown Ca2+ concentrations, simplifying data interpretation and assuring accuracy of conversion values. This procedure eliminates additional equipment, ratiometric imaging, and mathematic correction factors and should be useful to investigators requiring a straightforward method for measuring Ca2+ concentrations in live cells using Ca2+-chelating dyes exhibiting variable fluorescence intensity.  相似文献

The effect of the calcium-binding protein regucalcin on the Ca2+ transport system in rat liver mitochondria was investigated. Ca2+ transport was assayed by the method of Millipore filtration to estimate mitochondrial 45Ca2+ accumulation. 45Ca2+ uptake was stimulated by the presence of regucalcin (1.0 and 2.0 microM). This stimulation was remarkable during 1.0 min after 45Ca2+ addition, while appreciable stimulation was no longer seen at 3 min. Regucalcin (2.0 microM)-induced stimulation of 45Ca2+ uptake was prevented by the presence of ruthenium red (1.0 microM) and lanthanum chloride (0.1 mM). Regucalcin (2.0 microM) did not increase the mitochondrial adenosine triphosphatase (ATPase) activity during 3.0 min after Ca2+ addition. Meanwhile, 45Ca2+, which accumulated in the mitochondria during 5.0 min after 45Ca2+ addition, was not released by the addition of regucalcin. Regucalcin may stimulate Ca2+ uptake in rat liver mitochondria independently of the energy.  相似文献

Na+-Ca2+ exchanger (NCX) transports Ca2+ coupled with Na+ across the plasma membrane in a bi-directional mode. Ca2+ flux via NCX mediates osteogenic processes, such as formation of extracellular matrix proteins and bone nodules. However, it is not clearly understood how the NCX regulates cellular Ca2+ movements in osteogenic processes. In this study, the role of NCX in modulating Ca2+ content of intracellular stores ([Ca2+]ER) was investigated by measuring intracellular Ca2+ activity in isolated rat osteoblasts. Removal of extracellular Na+ elicited a transient increase of intracellular Ca2+ concentration ([Ca2+]i). Pretreatment of antisense oligodeoxynucleotide (AS) against NCX depressed this transient Ca2+ rise and raised the basal level of [Ca2+]i. In AS-pretreated cells, the expression and activity of alkaline phosphatase (ALP), an osteogenic marker, were decreased. However, the cell viability was not affected by AS-pretreatment. Suppression of NCX activity by the AS-pretreatment decreased ATP-activated Ca2+ release from intracellular stores and significantly enhanced Ca2+ influx via store operated calcium influx (SOCI), compared to those of S-pretreated or control cells. These results strongly suggest that NCX has a regulatory role in cellular Ca2+ pathways in osteoblasts by modulating intracellular Ca2+ content.  相似文献

Human red blood cells (RBCs) were loaded with the Ca(2+)-sensitive fluorescent dye fura-2 to investigate the effects of media ionic strength and prostaglandin E2 (PGE2) on the intracellular free Ca2+ concentration ([Ca2+]i). [Ca2+]i of intact RBCs in a Ca(2+)-containing physiological (high) ionic strength (HIS) solution was 75.1 +/- 8.3 nM after 5 min incubation, increasing to 114.9 +/- 9.6 nM after 1 h. In Ca(2+)-containing low ionic strength (LIS) solutions, [Ca2+]i was significantly lower than in the Ca(2+)-containing HIS solution (p = 0.041 or 0.0385 for LIS solutions containing 200 or 250 mM sucrose, respectively), but, as in HIS solution, an increase of [Ca2+]i was seen after 1 h. In Ca(2+)-free (0 Ca2+ plus 15 microM EGTA) media, [Ca2+]i decreased (ranging from 15 to 21 nM), but were not significantly different in HIS or LIS, and did not change following 1 h incubation. The effect of the ionic strength and PGE2 on passive Ca2+ influx was investigated on ATP-depleted RBCs. Ca2+ influx was faster during the initial 10 min in comparison with the subsequent time period (10-45 min), both in HIS and LIS media, decreasing from 20.3 +/- 1.9 to 12.9 +/- 1.3 micromol/(lcells x h) in HIS, and from 36.7 +/- 5.3 to 8.6 +/- 1.2 micromol/(lcells x h) in LIS. Prostaglandin E2 (PGE2; 10(-7)-10(-11) M), dissolved in deionised water or in ethanol, did not affect [Ca2+]i in either normal or in ATP-depleted RBCs suspended in Ca(2+)-containing HIS medium. Finally, the addition of carbachol (100 microM) did not affect [Ca2+]i. The present findings suggest that stimulation of the Ca(2+)-activated K+ channel by PGE2, reported in [J. Biol. Chem. 271 (1996) 18651], cannot be mediated via increased [Ca2+]i.  相似文献

Changes of the intracellular Ca2+ content in human red blood cells (RBCs) in glycerol-containing solutions and after freeze-thawing the cells with glycerol and subsequent deglycerolization were investigated with the Ca2+-sensitive fluorescent dye fluo-4 using fluorescence microscopy. In the glycerol-containing solutions the Ca2+ content increased when compared with a physiological medium (Hepes buffered saline solution (HBSS)). This effect was most likely a result of an inhibition of the Ca2+ pump. After inhibiting the Ca2+ pump using o-vanadate, the Ca2+ uptake was not significantly different in the cells in glycerol-containing and physiological medium. Freeze-thawing and deglycerolization of RBCs resulted in a more pronounced increase in the Ca2+ content. Also in this case, the Ca2+ pump seemed to play a major role.  相似文献

Ca(2+) signals regulate diverse physiological processes through tightly regulated fluxes varying in location, time, frequency, and amplitude. Here, we developed LOVS1K, a genetically encoded and photoactivated synthetic protein to generate local or global Ca(2+) signals. With 300?ms blue light exposure, LOVS1K translocated to Orai1, a plasma membrane Ca(2+) channel, within seconds, generating a local Ca(2+) signal on the plasma membrane, and returning to the cytoplasm after tens of seconds. With repeated photoactivation, global Ca(2+) signals in the cytoplasm were generated to modulate engineered Ca(2+)-inducible proteins. Although Orai1 is typically associated with global store-operated Ca(2+) entry, we demonstrate that Orai1 can also generate local Ca(2+) influx on the plasma membrane. Our photoactivation system can be used to generate spatially and temporally precise Ca(2+) signals and to engineer synthetic proteins that respond to specific Ca(2+) signals.  相似文献

The effects of hydrophilic headgroups of Ca surfactants, calcium dodecylsulfate (Ca(DS)(2)), calcium dodecylsulfonate (Ca(DSA)(2)), and calcium laurate (CaL(2)) and hydrophobic chains of alkyldimethylamine oxide (C(n)DMAO, n = 12, 14, 16) on the formation of Ca(2+)-ligand coordinated vesicles was investigated in detail. On the basis of phase behavior studies, rheological properties and freeze-fracture transmission electron microscope (FF-TEM) images were measured. Quite different phase behaviors were observed in different surfactant systems. For a Ca surfactant with a highly polar group, Ca(DS)(2), vesicles were observed in all Ca(DS)(2)/C(n)DMAO (n = 12, 14, and 16) systems, whereas for Ca surfactant with lower polar group, Ca(DSA)(2), vesicles can form in Ca(DSA)(2)/C(n)DMAO systems of n = 14 and 16 but not for n = 12. For CaL(2), the surfactant with the least polar group, vesicles form only in the CaL(2)/C(16)DMAO system. The results demonstrate that in the systems formed by Ca surfactants and C(n)DMAO, the formation of vesicles is driven not only by interaction between Ca(2+) and the N → O groups of C(n)DMAO but also by electrostatic and hydrophobic interactions. Vesicles prefer to form in Ca surfactants with highly polar headgroups and C(n)DMAO with long chain length.  相似文献

The Ca, Cl, Mg, Na, and P content and Ca/P, Ca/Mg, Ca/Na, Cl/Ca, and Cl/Na ratios in samples of intact cortical bone, inflamed bone and osteogenic sarcoma tissue were investigated by neutron activation analysis with high resolution spectrometry of short-lived radionuclides. In osteogenic sarcoma tissue the mass fractions of Cl and Na are higher and the mass fraction of Ca is lower than that of both normal and inflamed bone tissues. It was shown that the differences between the Cl/Ca ratio can be used as an additional test for differential diagnosis between normal or inflamed bone and osteogenic sarcoma.  相似文献

Recently, it was shown that the double Ca−H−Ca bridged calcium hydride cation dimer complex [LCaH2CaL]2+ (macrocyclic ligand L=NNNN-tetradentate Me4TACD) exhibited remarkable activity in catalyzing the hydrogenation of unactivated 1-alkenes as well as the H2 isotope exchange under mild conditions, tentatively via the terminal Ca−H bond of cation monomer LCaH+. In this DFT mechanistic work, a novel substrate-dependent catalytic mechanism is disclosed involving cooperative Ca−H−Ca bridges for H2 isotope exchange, competitive Ca−H−Ca bridges and terminal Ca−H bonds for anti-Markovnikov addition of unactivated 1-alkenes, and terminal Ca−H bonds for Markovnikov addition of conjugation-activated styrene. THF-coordination plays a key role in favoring the anti-Markovnikov addition while strong cation-π interactions direct the Markovnikov addition to terminal Ca−H bonds.  相似文献

We have carried out an extensive ab initio quantum chemical (QC)43Ca NMR study on a series of Ca-O organic compounds and three different Ca-bound proteins and found that the HF/6-31G* level of function can reliably predict 43Ca NMR interaction parameters (delta(iso) and chi(q)), especially for organic solids. This QC study finds correlations between Ca-O bond environment (mean distance and coordination number) and delta(iso)(43Ca). Although relatively small values of chi(q)(43Ca) are found for Ca-O organic compounds with a coordination number between 6 and 10, the QC shows that chi(q)(43Ca) is sensitive to the Ca-O coordination geometry of the Ca2+ sites in metalloproteins--a potentially important observation. An application of such ab initio QC 43Ca NMR studies is in characterizing the Ca-O bonding environment around target Ca2+ sites. As an example, we propose a new potential analytical approach using the absolute (43)Ca chemical shielding constant to investigate the hydration shell of Ca2+ in a dilute CaCl2 aqueous solution. Furthermore, by adopting a NMR methodology similar to that reported in Wong et al. Chem. Phys. Lett. 2006, 427, 201, natural abundance 43Ca MAS NMR spectra of Ca(L-glutamate)(2) x 4H2O were recorded, and delta(iso)(43Ca) and the quadrupolar parameter (Pq) were estimated to be 6.6 ppm and 0.8 MHz, respectively.  相似文献

Ca2+ as a signaling molecule carries information pivotal to cell life and death via its reversible interaction with a specific site in a protein. Although numerous Ca2+-dependent activities are known, the proteins responsible for some of these activities remain unidentified. We synthesized and characterized a photoreactive reagent, azido ruthenium (AzRu), which interacts specifically with Ca2+ binding proteins and strongly inhibits their Ca2+-dependent activities, regardless of their catalytic mechanisms or functional state as purified proteins, embedded in the membrane or in intact cells. As expected from a Ca2+ binding protein-specific reagent, AzRu had no effect on Ca2+-independent and Mg2+-dependent activities. Az103Ru covalently bound, and specifically labeled, known Ca2+ binding proteins. AzRu is a photoreactive reagent that provides an approach for identification of Ca2+ binding proteins, characterization of their binding sites, and exploration of new Ca2+-dependent processes.  相似文献

Many studies on intracellular calcium ([Ca2+](i)) and intracellular pH (pH(i)) have been carried out due to their importance in regulation of different cellular functions. However, most of the previous studies are focused on human or mammalian cells. The purpose of the present study was to characterize the effect of Rhodojaponin-III (R-III) on [Ca2+](i) and pH(i) and the proliferation of Sf9 cells. R-III strongly inhibited Sf9 cells proliferation with a time- and dose-dependent manner. Flow cytometry established that R-III interfered with Sf9 cells division and arrested them in G2/M. By using confocal scanning technique, effects of R-III on intracellular free calcium ([Ca2+](i)) and intracellular pH (pH(i)) in Sf9 cells were determined. R-III induced a significant dose-dependent (1, 10, 100, 200 μg/mL) increase in [Ca2+](i) and pH(i) of Sf9 cells in presence of Ca2+-containing solution (Hanks) and an irreversible decrease in the absence of extra cellular Ca2+. We also found that both extra cellular Ca2+ and intracellular Ca2+ stores contributed to the increase of [Ca2+](i), because completely treating Sf9 cells with CdCl(2) (5 mM), a Ca2+ channels blocker, R-III (100 μg/mL) induced a transient elevation of [Ca2+](i) in case of cells either in presence of Ca2+ containing or Ca2+ free solution. In these conditions, pH(i) showed similar changes with that of [Ca2+](i) on the whole. Accordingly, we supposed that there was a certain linkage for change of [Ca2+](i), cell cycle arrest, proliferation inhibition in Sf9 cells induced by R-III.  相似文献
