The asymmetric synthesis of new tetrahydropyrrolo[2,3-b]indole 19 and tetrahydropyrano[2,3-b]indole 20 rings, substituted in position C-3a and C-4a with a hydroxy- and an amino functionalized chain, respectively, was performed starting from the racemic spiro[cyclohexane-1,3′-indoline]-2′,4-diones 7. The enantiopure spiro oxo-azepinoindolinone (+)-10, obtained from (±)-7 by the way of an asymmetric ring enlargement, and the amino acid (+)-14, obtained by the hydrolysis of 10, were prepared as key intermediates for the synthesis of enantiopure compounds (−)-19 and (−)-20. Since the amino acid 14 is the common intermediate for the chemoselective preparation of derivatives 19 and 20, experimental and computational studies were performed in order to selectively obtain these compounds and to provide a mechanistic rationalization for their formation.  相似文献

Sb-chirogenic organoantimony compounds (±)-5a-c bearing heteroatom moieties such as 4,4-dimethyl-2-oxazolinyl, methoxymethyl, and diphenylphosphanyl substituents on the o-position of an aryl group have been prepared by nucleophilic displacement of the ethynyl moiety on (1-naphthyl)(phenylethynyl)(p-tolyl)stibane (3) with aryllithium reagents (2a-c). The optical resolution of the racemic (±)-5a,b was attained via separation of a diastereomeric mixture of their palladium complexes (S)-7 formed from the reactions of (±)-5a,b with di-μ-chlorobis[(S)-dimethyl(1-ethyl-α-naphthyl)aminato-C2,N]dipalladium(II) (6). The enantiomerically pure Sb-chirogenic stibanes isolated here were optically stable, and no racemization on the chiral antimony center was observed even when they were allowed to stand at room temperature for over 72 h in chloroform. The structure of 5a,b including the absolute configuration was determined by single crystal X-ray analyses of (+)-5aB and antimony-palladium complex (7bB), respectively. The analyses also revealed the presence of intramolecular interaction between the antimony and sp2-nitrogen atoms in the molecule Sb(S)-(+)-5aB.  相似文献

A convenient enantioselective synthesis of 3-asymmetrically substituted oxindoles is reported. Compound (2) prepared by radical cyclisation of (1) was used for the synthesis of racemic and enantiomerically pure 3-asymmetrically substituted oxindoles. Desulfurisation of (2) using Raney Ni yielded the racemate (5). Addition of (S)-1-phenylethanol to compound (2) yielded the diastereoisomer (21) the structure of which was determined using X-ray crystallography. Using a sequence of steps (21) was converted to the enantiomer (8). The enantiomer (9) was similarly prepared from (2) using (R)-1-phenylethanol.  相似文献

The synthesis of (+)-nangustine [(+)-2] has been achieved, for the first time, using the enantiomerically pure cis-1,2-dihydrocatechol 3 as starting material. The latter compound is available, in multi-gram quantities, through a whole-cell-mediated biotransformation of chlorobenzene using genetically engineered organisms that over-express the responsible enzyme, namely toluene dioxygenase. Since the enantiomer of compound 3 is available by related means, the present work also represents a formal total synthesis of the montanine alkaloid (−)-nangustine [(−)-2]. Single-crystal X-ray analyses of compounds 13 and (+)-2 are reported.  相似文献

A new route to racemic diethyl 2-methyl-bicyclo[3.1.1]hept-2-ene-6,6-dicarboxylate (±)-1a by means of a palladium-catalyzed intramolecular allylic alkylation exercised on malonate-ester derivatives has been developed from R-(−)-carvone. The stereocontrolled synthesis of the enantiomerically pure (−)-1b by application of a six-step reactions sequence with a 28% overall yield from acetal 8, has been accomplished.  相似文献

Both racemic ethyl 5-iodo-2-methylcyclohexanecarboxylate (1), known as Mediterranean fruit fly attractant ceralure B1, and its (−)-(1R,2R,5R) enantiomer 1a were conveniently synthesized from commercially available racemic trans-6-methyl-3-cyclohexenecarboxylic acid 2 or its (1R,6R) enantiomer 2a. Key steps included an asymmetric Diels-Alder reaction using a sultam auxiliary and cyclization of the unwanted trans-5-iodo-trans-2-methylcyclohexanecarboxylic acid (8) to the intermediate lactone 7 (or 8a to 7a). The new method may circumvent chromatographic separations and seems amenable to scale-up.  相似文献

The γ-trifluoromethyl γ-sulfone hydroxamate 1 was synthesized both in racemic and enantiomerically pure forms by means of a thia-Michael reaction of p-methoxythiophenol on achiral and chiral 3,3,3-trifluorocrotonoyl Michael acceptors. The (R)-1 enantiomer was the most potent inhibitor of MMP-3 (stromelysin-1), showing an IC50 = 3.2 nM, as well as the most selective with respect to MMP-9 (65-fold).  相似文献

The novel optically active derivatives of 2,2′-disubstituted-1-aminocyclopropane-1-carboxylic acid (−)-2 and (+)-3 were synthesised from the spiro-azlactone (+)-1. Oxidation of the diol moiety of (+)-3 gave by ring enlargement the racemic mixture of 2,3-dihydrofuran derivative (±)-6. This conversion is explained by stepwise rearrangement of the initially formed tetrasubstituted cyclopropanecarbaldehyde 4 through zwitterionic's reactive intermediate 5. The formation of (±)-6 is preferred energetically as established by ab initio calculations of the ground states and possible intermediates for that rearrangement. The crystal structure and absolute configuration of the compounds (+)-1, (−)-2, (+)-3 and (−)-7 were determined by single-crystal X-ray diffraction method. All four compounds possess Z-configuration of the cyclopropane ring. The dioxolane ring in the structures (+)-1 and (−)-2 adopts half-chair conformation, while the cyclopropane ring and geminally substituted groups in the structures (−)-2, (+)-3 and (−)-7 possess the anticlinal conformation. The molecules of the compound (+)-1 are connected by very weak intermolecular hydrogen bond of C-H?O type. In the compounds (−)-2, (+)-3 and (−)-7inter- and intramolecular hydrogen bonds of N-H?O type were observed. The spiro-compound (+)-1 exhibited a more pronounced inhibitory activity against the proliferation of murine leukemia and human T-lymphocytes cells than other type of tumor cell lines and normal human fibroblast cells.  相似文献

We describe an intriguing new example of a parallel kinetic resolution; an asymmetric cyclization-carbonylation of propargyl ketols catalyzed by palladium(II) with chiral bisoxazoline (box) ligands. The 2S,3S enantiomer of (±)-6 was preferentially converted to 13 (45-49% yields, 37-46% ee), and the 2R,3R enantiomer of (±)-6 was preferentially converted to 14 (21-23% yields, 92-97% ee). As an application of this reaction, formal synthesis of (+)-bakkenolide A was achieved.  相似文献

A new atropisomeric dihydrobenzofuran-based bisphosphine ligand 1 was easily prepared from 1,4-dibromo-2-fluorobenzene. This racemic bisphosphine (±)-1 was used as a ligand for the palladium-catalyzed Suzuki-Miyaura reaction of aryl chloride with arylboronic acids and Hartwig-Buchwald amination of aryl bromides with aniline derivatives. The optical resolution of (±)-1 was also carried out by HPLC with a chiral stationary phase column.  相似文献

The hydrochlorothiazide, 6-chloro-3,4-dihydro-2H-1,2,4-benzothiadiazine-7-sulfonamide-1,1-dioxide, (HCTZ), widely used as a diuretic and anti-hypertensive drug, was transformed into its N-nitroso-derivative, 6-chloro-4-nitroso-3,4-dihydro-2H-1,2,4-benzothiadiazine-7-sulfonamide-1,1-dioxide (ON-HCTZ) by sodium nitrite in an acidic medium. The crystalline complexes of ON-HCTZ with 18-crown-6 (18C6) and cis-anti-cis-dicyclohexyl-18C6 (DCH6B) demonstrated different H-bonding modes from those present in the co-crystals of HCTZ with the same crown ethers. The influence of the nitroso-group on the binding mode and crystal packing is discussed.  相似文献

A new non-C2-symmetrical antimony-phosphorous ligand, (±)-2-diphenyl-phosphano-2′-di(p-tolyl)stibano-1,1′-binaphthyl (BINAPSb) 3, has been prepared from 2-bromo-2′-diphenylphosphano-1,1′-naphthyl 4 via its borane complex 6, and could be resolved by the separation of a mixture of the diastereomeric palladium complexes 8A and 8B derived from the reaction of (±)-3 with optically active palladium reagent (S)-7. The enantiomerically pure BINAPSb 3 has proved to be highly effective in the palladium-catalyzed asymmetric hydrosilylation of styrene as a chiral auxiliary.  相似文献

Flash vacuum pyrolysis (fvp) of some substituted [1,2,4]triazolo[3,4-c][1,2,4]benzotriazine derivatives (1a-d) has been studied between 450 and 600 °C. The only transformation observed up to 525 °C was the unexpected valence bond isomerization of the angularly fused starting compounds to the isomeric linearly fused [1,2,4]triazolo[4,3-b][1,2,4]benzotriazine derivatives (9a-d), whereas at higher temperatures fragmentation products such as aromatic nitriles were also formed. Kinetic measurements revealed negative entropies of activation in the isomerization process, which suggest a concerted ring closure reaction to an intermediate antiaromatic diazirine. Reversibiblity of the title isomerization reaction was also proved by FVP experiments.  相似文献

In this study, a novel and convenient route for the construction of 5-((1H-1,2,4-triazol-1-yl)methyl)-1H-indoles (8) is presented starting from (1H-1,2,4-triazol-1-yl)methanol (5) and indolines (6) under 98% H2SO4 at room temperature for 4–24 h, followed by deacetylation and dehydrogenation. Based on this finding, a novel route to synthesize Rizatriptan starting from tryptamine was designed and accomplished with 48.5% overall yield in 6 steps. Compared with operational art, the new route afforded higher yield and more pure products requiring no chromatographic purification, which may further be applied in industrialization.  相似文献

The synthesis of the title compound (4), starting from commercial 3-bromo-4-methylthiophene, is described. The single crystal packing mode of 4, as well as the absorption and photoluminescence properties in solution and in the solid state are reported and analyzed in relation to those of the isomeric 3,5-dimethyl-dithieno[3,2-b:2′,3′-d]thiophene-4,4-dioxide (7). The different reactivities of 4 and 7 towards bromination are analyzed in the light of the results of ab initio B3LYP/6-31G calculations on both compounds.  相似文献

Lipase-catalyzed hydrolysis of (E)-2-[α-(acetoxyimino)benzyl]-1,1′-binaphthyl [(±)-1a] and (Z)-2-[α-(acetoxyimino)benzyl]-1,1′-binaphthyl [(±)-1b] yielded optically active (E)-2-[α-(hydroxyimino)benzyl]-1,1′-binaphthyl [(S)-2a] and (Z)-2-[α-(hydroxyimino)benzyl]-1,1′-binaphthyl [(R)-2b], respectively, with high enantiomeric excess. Selectivity for the opposite enantiomer of the axial binaphthyl skeleton was shown by (Z)-isomer 1b against (E)-isomer 1a.  相似文献

A synthesis of functionalized phenolic α-amino-alcohol (±)-13 as synthetic precursor of the catechol tetrahydroisoquinoline structure of phthalascidin 650 is disclosed. Starting from 3-methylcatechol 5, eight steps of synthesis give rise to the synthesis of phenolic α-amino-alcohol (±)-13 in 27% overall yield. This synthetic strategy involves the elaboration of fully functionalized aromatic aldehyde 8 and its transformation into a phenolic α-amino-alcohol (±)-13, through a Knoevenagel condensation, simultaneous reduction of nitroketene and ester functions and hydrogenolysis of the benzyl protecting group. The pentacycle (±)-18 was obtained after four additional steps. The Pictet-Spengler cyclisation between the phenolic α-amino-alcohol (±)-13 and N-protected α-amino-aldehyde 4 allowed to obtain (1,3′)-bis-tetrahydroisoquinoline 14 with N-methylated and N-Fmoc removed. The last step was a Swern oxidation for allowing an intramolecular condensation.  相似文献

The hemisphere-shaped calixarene 1a and its split-hemisphere-shaped isomer 1b were synthesized from [2.1.2.1]metacyclophane (MCP) 3 by pinacol rearrangement and subsequent intramolecular acetalization. Their structures were revealed by X-ray crystallography and 1H NMR spectroscopy. The temperature-dependence of the intramolecular acetalization to provide 1a and 1b was examined. The results indicated that 1a is the dominant product at high temperatures, and the values of ΔH° and ΔS° were estimated to be −18.3±0.37 kJ mol−1, −59.1±1.12 kJ mol−1 K−1, respectively. The dinitro derivative 7 and tetranitro derivatives8 were obtained by ipso-nitration at the upper rims of 1a. The optical resolution and chiral recognition ability of racemic mixture 7 were investigated by HPLC systems.  相似文献

A series of racemic 4-aryl-5-(tert-butoxycarbonyl)-6-methyl-3,4-dihydro-2(1H)-pyridones have been prepared by means of a modified Hantzsch reaction using commercially available starting materials. An easy removal of the tert-butyl group of these pyridones and subsequent reaction with cesium carbonate and chloromethyl 2-methylpropanoate provided us suitable substrates (±)-5 to be used in lipase-catalyzed hydrolysis reactions. Lipase B from Candida antarctica (CAL-B) was the most adequate lipase in the hydrolysis of (±)-5. Despite the low enantioselectivity values obtained (E≤12), several optically active pyridone derivatives were finally isolated with high enantiomeric excesses (ee≥91%) and moderate yields.  相似文献

A simple and convergent approach to enantiomerically pure 5-[[2-[1-[3,5-bis(trifluoromethyl)phenyl]ethoxy-3-(4-fluorophenyl)morpholin-4-yl]methyl]-1,2-dihydro-1,2,4-triazol-3-one 1, a potent orally active antagonist of the human neurokinin-1 (NK-1) receptor, is described. The synthetic procedure starts from p-fluorobenzaldehyde to access the racemic morpholinone 2 via a modified Strecker synthesis and utilizes a diastereomeric salt resolution technique to accomplish the synthesis of 1 in enantiomerically pure form and good yield.  相似文献
