Multi-way partial least-squares (N-PLS) is combined to the residual bi-linearization procedure (RBL) for the direct analysis of metabolites of polycyclic aromatic hydrocarbons in urine samples. Metabolite analysis is carried out via a two-step experimental procedure based on solid-phase extraction and room temperature fluorescence spectroscopy. Excitation-emission matrices are recorded from octadecyl (C18) membranes that serve as solid substrates for sample extraction and spectroscopic measurements. Excellent metabolite recoveries were obtained in all cases, which varied from 96.2 ± 1.35% (9-hydroxyphenanthrene) to 99.7 ± 0.49% (3-hydroxybenzo[a]pyrene). Background correction of extraction membranes is carried out with a new alternating least-squares (ALS) procedure adapted to second order data. The performance of N-PLS/RBL is compared to the well-established multivariate curve resolution-alternating least-squares (MCR-ALS) algorithm. Both algorithms provided similar analytical figures of merit, including their ability to handle unknown interference in urine samples. With only 10 mL of sample, the limits of detection varied between 0.06–0.08 ng mL−1 (1-hydroxypyrene) and 0.016–0.018 ng mL−1 (2-hydroxyfluorene). When compared to previously reported univariate calibration data, the limits of detection via N-PLS/RBL and MCR-ALS are approximately one order of magnitude higher. This was somehow expected due to the effect of unexpected components in multivariate figures of merit, i.e. a more realistic approach to the analysis of metabolites in human urine samples.  相似文献

Urine analysis of monohydroxy metabolites is recognized as an accurate assessment of human exposure to polycyclic aromatic hydrocarbons. Despite the sophisticated arsenal of analytical tools, monitoring of monohydroxy metabolites via simple, cost effective and direct methods of analysis still remains a challenge. This article evaluates the analytical potential of solid-phase extraction room-temperature fluorescence spectroscopy for the problem at hand. Extraction membranes serve the dual purpose of sample pre-concentration and solid substrate for RTF measurements. The potential of our proposition is demonstrated with the analysis of 2-hydroxy-fluorene, 1-hydroxy-pyrene, 3-hydroxy-benzo[a]pyrene and 9-hydroxy-phenanthrene in synthetic urine samples. Signal reproducibility is improved with the aid of a sample holder specifically designed for the manual optimization of luminescence signals. Background correction of solid substrates is carried out with the aid of Asymmetric Least Squares. Recovery values for the studied metabolites varied from 99.0 ± 1.2% (3-hydroxy-benzo[a]pyrene) to 99.9 ± 0.05% (1-hydroxy-pyrene). With only 10 mL of urine sample, the limits of detection varied from 57 pg mL−1 (2-hydroxy-fluorene) to 2 pg mL−1 (1-hydroxy-pyrene). Additional figures of merit include a simple experimental procedure for routine screening of numerous samples and compatibility with portable instrumentation for field analysis. Because of the non-destructive nature of fluorescence measurements, membranes can be brought to the lab for subsequent elution and confirmation of compounds via high-resolution techniques.  相似文献

Low picograms of the hormone 17β-estradiol were detected at an electrochemical immunosensor. This immunosensor features a gold nanoparticle|Protein G-(LC-SPDP)1-scaffold, to which a monoclonal anti-estradiol capture antibody was immobilised to facilitate a competitive immunoassay between sample 17β-estradiol and a horseradish peroxidase-labelled 17β-estradiol conjugate. Upon constructing this molecular architecture on a disposable gold electrode in a flow cell, amperometry was conducted to monitor the reduction current of benzoquinone produced from a catalytic reaction of horseradish peroxidase. This current was then quantitatively related to 17β-estradiol present in a sample. Calibration of immunosensors in blood serum samples spiked with 17β-estradiol yielded a linear response up to ∼1200 pg mL−1, a sensitivity of 0.61 μA/pg mL−1 and a detection limit of 6 pg mL−1. We attribute these favourable characteristics of the immunosensors to the gold nanoparticle|Protein G-(LC-SPDP) scaffold, where the gold nanoparticles provided a large electrochemically active surface area that permits immobilisation of an enhanced quantity of all components of the molecular architecture, while the Protein G-(LC-SPDP) component aided in not only reducing steric hindrance when Protein G binds to the capture antibody, but also providing an orientation-controlled immobilisation of the capture antibody. Coupled with amperometric detection in a flow system, the immunosensor exhibited excellent reproducibility.  相似文献

An amplified electrochemical impedimetric aptasensor for ochratoxin A (OTA) was developed with picomolar sensitivity. A facile route to fabricate gold nanoparticles covalently bound reduced graphene oxide (AuNPs–rGO) resulted in a large number of well-dispersed AuNPs on graphene sheets with tremendous binding sites for DNA, since the single rGO sheet and each AuNP can be loaded with hundreds of DNA strands. An aptasensor with sandwich model was fabricated which involved thiolated capture DNA immobilized on a gold electrode to capture the aptamer, then the sensing interface was incubated with OTA at a desired concentration, followed by AuNPs–rGO functionalized reporter DNA hybridized with the residual aptamers. By exploiting the AuNPs–rGO as an excellent signal amplified platform, a single hybridization event between aptamer and reporter DNA was translated into more than 107 redox events, leading to a substantial increase in charge-transfer resistance (Rct) by 7∼ orders of magnitude compared with that of the free aptamer modified electrode. Such designed aptasensor showed a decreased response of Rct to the increase of OTA concentrations over a wide range of 1 pg mL−1–50 ng mL−1 and could detect extremely low OTA concentration, namely, 0.3 pg mL−1 or 0.74 pM, which was much lower than that of most other existed impedimetric aptasensors. The signal amplification platform presented here would provide a promising model for the aptamer-based detection with a direct impedimetric method.  相似文献

In this paper, a novel gold nanoparticles based protein immobilization method was designed. Biocomposites of gold nanoparticles and proteins were successfully coated on poly(methyl methacrylate) (PMMA) plates and polystyrene microtiter plates. The proteins could be immobilized on solid materials with high density and better bioactivity. Based on above design, chemiluminescence (CL) imaging assay for determination of H2O2 and recombinant human interleukin-6 (rHu IL-6) was developed. The linear range and the loading capability were greatly improved when compared with imaging assay performed with direct proteins immobilization. Under the selected experimental conditions, a linear relationship was obtained between the CL intensity and the concentration of H2O2 in the range of 1.0 × 10−6 to 1.0 × 10−4 mol L−1, and rHu IL-6 in the range of 2.0-312.0 pg mL−1. The detection limits were 2 × 10−7 mol L−1 (3σ) for H2O2 and 0.5 pg mL−1 for rHu IL-6 with relative standard deviation of 3.8% for 3.0 × 10−5 mol L−1 H2O2, and 4.4% for 39.0 pg mL−1 rHu IL-6. This method has been applied to the determination of rHu IL-6 in human serum with satisfactory results.  相似文献

The influence of different parameters on the sorption profiles of trace and ultra traces of gold (I) species from the aqueous cyanide media onto the solid sorbents ion exchange polyurethane foams (IEPUFs) and commercial unloaded polyurethane foams (PUFs) based polyether type has been investigated. The retention of gold (I) species onto the investigated solid sorbents followed a first-order rate equation with an overall rate constant k in the range 2.2-2.8 ± 0.2 s−1. The sorption data of gold (I) followed Freundlich and Langmuir isotherm models. Thus, the a dual-mode of sorption mechanism involving absorption related to “weak base anion exchanger” and an added component for “surface adsorption” seems the most likely proposed dual mechanism for retention profile of gold (I) by the IEPUFs and PUFs solid sorbents. The capacity of the IEPUFs and PUFs towards gold (I) sorption calculated from the sorption isotherms was found to be 11.21 ± 1.8 and 5.29 ± 0.9 mg g−1, respectively. The chromatographic separation of the spiked inorganic gold (I) from de ionized water at concentrations 5-15 μg mL−1 onto the developed IEPUFs and PUFs packed columns at 10 mL min−1 flow rate was successfully achieved. The retained gold (I) species were then recovered quantitatively from the IEPUFs (98.4 ± 2.4%, n = 5) and PUFs (95.4 ± 3.4%, n = 5) packed columns using perchloric acid (60 mL, 1.0 mol L−1) as a proper eluating agent. Thiourea (1.0 mol L−1)-H2SO4 (0.1 mol L−1) system was also used as eluating agent for the recovery of gold (I) from IEPUFS (95.4 ± 5.4%, n = 3) and also PUFs (93.4 ± 4.4%, n = 3) packed columns. The performance of the IEPUFs and PUFs packed columns in terms of the height equivalent to the theoretical plates (HETP), number of plates (N), and critical and breakthrough capacities towards gold (I) species were evaluated. The developed IEPUFs packed column was applied successfully for complete retention and recovery (98.5 ± 2.7) of gold (III) species spiked onto tap- and industrial wastewater samples at <10 μg Au mL−1 after reduction to gold (I). The IEPUFs packed column was applied satisfactorily for complete retention and recovery (98.5 ± 2.7) of total inorganic gold (I) and/or gold (III) species spiked to tap- and industrial wastewater samples at <10 μg mL−1 gold. Chromatographic separation of gold (I) from silver (I) and base metal ions (Fe, Ni, Cu and Zn) using IEPUFS packed columns was satisfactorily achieved. The proposed method was applied successfully for the pre-concentration and separation from anodic slime and subsequent FAAS determination of analyte with satisfactory results (recoveries >95%, relative standard deviations <4.0%).  相似文献

In this work, a simple, rapid and sensitive sample pretreatment technique, dispersive liquid-liquid microextraction (DLLME) coupled with high performance liquid chromatography-fluorescence detection (HPLC-FLD), has been developed to determine carbamate (carbaryl) and organophosphorus (triazophos) pesticide residues in water and fruit juice samples. Parameters, affecting the DLLME performance such as the kind and volume of extraction and dispersive solvents, extraction time and salt concentration, were studied and optimized. Under the optimum extraction conditions (extraction solvent: tetrachloroethane, 15.0 μL; dispersive solvent: acetonitrile, 1.0 mL; no addition of salt and extraction time below 5 s), the performance of the proposed method was evaluated. The enrichment factors for the carbaryl and triazophos were 87.3 and 275.6, respectively. The linearity was obtained in the concentration range of 0.1-1000 ng mL−1 with correlation coefficients from 0.9991 to 0.9999. The limits of detection (LODs), based on signal-to-noise ratio (S/N) of 3, ranged from 12.3 to 16.0 pg mL−1. The relative standard deviations (RSDs, for 10 ng mL−1 of carbaryl and 20 ng mL−1 of triazophos) varied from 1.38% to 2.74% (n = 6). The environmental water (at the fortified level of 1.0 ng mL−1) and fruit juice samples (at the fortified level of 1.0 and 5.0 ng mL−1) were successfully analyzed by the proposed method, and the relative recoveries of them were in the range of 80.4-114.2%, 89.8-117.9% and 86.3-105.3%, respectively.  相似文献

To address people's concerns of exogenous oxytocin (OT) administration to lactating bovines, a study was undertaken to (a) establish an enzyme immunoassay (EIA) for OT determination in milk, (b) quantify OT in milk of cows administered OT, and (c) study influence of pasteurization on OT stability in milk. A sensitive EIA validated according to the criteria of European Union—Decision 2002/657/EC was developed for OT in skim milk in an analytical range of 10-250 pg mL−1 with a decision limit (CCα) of 30 pg mL−1 and detection capability (CCβ) of 41.5 pg mL−1. Milk samples collected from cows (n = 38) administered either 25 or 50 IU OT prior to milking were investigated for the presence of OT. There was no significant difference among both groups with the mean concentrations of OT being 15.8 and 14.9 pg mL−1 for cows subjected to 25 and 50 IU OT administration, respectively. The OT levels in skim milk of control cows (n = 30; untreated) were basal (around 10 pg mL−1). All the analyzed milk samples were below the CCα value of 30 pg mL−1. Pasteurization of OT spiked milk samples at different temperature and sample holding conditions reduced the immunological activity of OT to 43% at 110 °C. However, no further decline occurred in the immunological activity with increased pasteurization temperature and time. It was concluded that the milk OT concentrations after OT administrations were minimal and below the assay decision limit. However, OT was quite stable to pasteurization in OT spiked milk.  相似文献

In the presented work, a disposable immunosensor for the detection of testosterone, an endogenous steroid hormone, in bovine urine has been developed using screen-printed electrodes (SPEs). Due to concerns over the use of steroid hormones as growth promoters, the EU prohibits their use in food producing animals. Consequently, rigorous screening procedures have been implemented in all member states to detect the illegal administration of such compounds. Competitive immunoassays were developed, initially by enzyme linked immunosorbent assay (ELISA), and subsequently transferred to an electrochemical immunosensor format using disposable screen-printed carbon electrodes. Horseradish peroxidase (HRP) was the enzyme label of choice and chronoamperometric detection was carried out using a tetramethylbenzidine/hydrogen peroxide (TMB/H2O2) substrate system, at +100 mV. The EC50 values obtained for the assay in buffer and urine gave relatively comparable results, 710 pg mL−1 and 960 pg mL−1, respectively. The linear range obtained for the assay in buffer extended from 0.03 ng mL−1 to 40 ng mL−1; while that in urine ranged from 0.03 ng mL−1 to 1.6 ng mL−1. The corresponding limits of detection (LOD) in buffer and urine were 26 pg mL−1 and 1.8 pg mL−1. Cross reactivity profiles of the antibody have been examined, with notable cross reactivities with 19-nortestosterone (11.6%) and boldenone (9.86%). Precision studies for the sensor demonstrated adequate reproducibility (CV < 13%, n = 3) and repeatability (CV < 9%, n = 3). Recovery data obtained showed good agreement between spiking studies and known concentrations of analyte. Sensors showed stability for 4 days at +4 °C. A sensitive, highly specific, inexpensive, disposable immunosensor, showing excellent overall performance for the detection of testosterone in bovine urine, has been developed.  相似文献

A one-step electrochemical aptasensor using the thiol- and methylene blue- (MB-) dual-labeled aptamer modified gold electrode for determination of ochratoxin A (OTA) was presented in this research. The aptamer against OTA was covalently immobilized on the surface of the electrode by the self-assembly effect and used as recognition probes for OTA detection by the binding induced folding of the aptamer. Under the optimal conditions, the developed electrochemical aptasensor demonstrated a wide linear range from 0.1 pg mL−1 to 1000 pg mL−1 with the limit of detection (LOD) of 0.095 pg mL−1, which was an extraordinary sensitivity compared with other common methods for OTA detection. Moreover, as a practical application, this proposed electrochemical aptasensor was used to monitor the OTA level in red wine samples without any special pretreatment and with satisfactory results obtained. Study results showed that this electrochemical aptasensor could be a potential useful platform for on-site OTA measurement in real complex samples.  相似文献

A simple, rapid and efficient method, dispersive liquid-liquid microextraction (DLLME), has been developed for the extraction and preconcentration of polybrominated diphenyl ethers (PBDEs) in water samples. The factors influencing microextraction efficiencies, such as the kind and volume of extraction and dispersive solvent, the extraction time and the salt effect, were optimized. Under the optimum conditions (sample volume: 5 mL; extraction solvent: tetrachloroethane, 20.0 μL; dispersive solvent: acetonitrile, 1.00 mL; extraction time: below 5 s and without salt addition), the enrichment factors and extraction recoveries were high and ranged from 268 to 305 and 87.0 to 119.1%, respectively. Linearity was observed in the range 0.05-50 ng mL−1 for BDE-28 and BDE-99, and 0.1-100 ng mL−1 for BDE-47 and BDE-209, respectively. Coefficients of correlation (r2) ranged from 0.9995 to 0.9999. The repeatability study was carried out by extracting the spiked water samples at concentration levels of 50 ng mL−1 for BDE-28 and BDE-99, and 100 ng mL−1 for BDE-47 and BDE-209, respectively. The relative standard deviations (R.S.D.s) varied between 3.8 and 6.3% (n = 5). The limits of detection (LODs), based on signal-to-noise ratio (S/N) of 3, ranged from 12.4 to 55.6 pg mL−1 (the wavelength of detector at 226 nm). The relative recoveries of PBDEs from tap, lake water and landfill leachate samples at spiking levels of 5, 10 and 50 ng mL−1 were in the range of 89.7-107.6%, 114.3-119.1% and 87.0-90.9%, respectively. As a result, this method can be successfully applied for the determination of PBDEs in landfill leachate and environmental water samples.  相似文献

A sensitive and label-free electrochemical impedance immunosensor via covalent coupling the antibody with functionalized gold nanoparticles (FAuNP) for probing apolipoprotein A-I was presented. The hybrid gold nanoparticles were prepared with a two-in-one strategy, i.e. via the stepwise employment of self-assembled monolayer (SAM) and sol-gel techniques, to improve the performance of such a label-free immunosensor, which was investigated by electrochemical impedance spectroscopy. It was found that this novel FAuNP immunosensor showed higher protein-loading capacity and better response properties (6-17 times) than that fabricated by normal SAM technique did. The remarkably improved properties of the immunosensor were ascribed to FAuNP with the larger surface-to-volume ratio, more free amino linkage groups, and the lower nonspecific protein adsorption. As a result, the thus-prepared antibody-modified immunosensor showed reproducible (R.S.D. = ±3.2%, n = 10) linear response to apolipoprotein A-I (Apo A-I) antigens in the range of 0.1-10 ng mL−1. The detection limit of this immunosensor was 50 pg mL−1 (corresponding to 1.8 pmol L−1), which was two orders of magnitude lower than that of the traditional methods. These results exhibited the novel immunosensor had a high sensitivity, stability and selectivity for the determination of Apo A-I, especially in clinic microanalysis.  相似文献

建立了固相萃取-液相色谱-串联质谱同时测定尿中2-羟基萘、1-羟基萘、2-羟基芴、3-羟基菲、1-羟基芘等9种多环芳烃代谢物的液相色谱-串联质谱测定方法。尿样中结合态的多环芳烃代谢物在β-葡萄糖苷酸酶-芳基硫酸酯酶缓冲液(pH 5.0)作用下,于37℃水浴中避光水解4 h后,以C18固相萃取小柱富集、净化,以甲醇洗脱,采用Waters Symmetry C18色谱柱,流动相为乙腈-0.2%氨水(72∶27,V/V)等度淋洗分离后进入质谱测定。在喷雾电压4 kV,毛细管温度300℃下,以3-羟基菲13C为内标,采用SRM模式负离子扫描方式测定,内标法定量。9种多环芳烃代谢物在尿中的线性范围为0.90~100μg/L;相关系数为0.9970~0.9990;回收率为79.0%~119.8%;相对标准偏差为4.3%~12.4%;检出限为0.04~0.90μg/L;结果表明,本方法可用于尿中9种多环芳烃代谢物的测定。  相似文献

Nanoporous gold (NPG) has recently received considerable attention in analytical electrochemistry because of its good conductivity and large specific surface area. A facile layer-by-layer assembly technique fabricated NPG was used to construct an electrochemical immunosensor for carcinoembryonic antigen (CEA). NPG was fabricated on glassy carbon (GC) electrode by alternatively assembling gold nanoparticles (AuNPs) and silver nanoparticles (AgNPs) using 1,4-benzenedimethanethiol as a cross-linker, and then AgNPs were dissolved with HNO3. The thionine was absorbed into the NPG and then gold nanostructure was electrodeposited on the surface through the electrochemical reduction of gold chloride tetrahydrate (HAuCl4). The anti-CEA was directly adsorbed on gold nanostructure fixed on the GC electrode. The linear range of the immunosensor was from 10 pg mL−1 to 100 ng mL−1 with a detection limit of 3 pg mL−1 (S/N = 3). The proposed immunosensor has high sensitivity, wide linear range, low detection limit, and good selectivity. The present method could be widely applied to construct other immunosensors.  相似文献

Measurements of Pb isotope ratios in ice containing sub-pg g−1 concentrations are easily compromised by contamination, particularly where limited sample is available. Improved techniques are essential if Antarctic ice cores are to be analysed with sufficient spatial resolution to reveal seasonal variations due to climate. This was achieved here by using stainless steel chisels and saws and strict protocols in an ultra-clean cold room to decontaminate and section ice cores. Artificial ice cores, prepared from high purity water were used to develop and refine the procedures and quantify blanks. Ba and In, two other important elements present at pg g−1 and fg g−1 concentrations in Polar ice, were also measured. The final blank amounted to 0.2 ± 0.2 pg of Pb with 206Pb/207Pb and 208Pb/207Pb ratios of 1.16 ± 0.12 and 2.35 ± 0.16, respectively, 1.5 ± 0.4 pg of Ba and 0.6 ± 2.0 fg of In, most of which probably originates from abrasion of the steel saws by the ice. The procedure was demonstrated on a Holocene Antarctic ice core section and was shown to contribute blanks of only ∼5%, ∼14% and ∼0.8% to monthly resolved samples with respective Pb, Ba and In concentrations of 0.12 pg g−1, 0.3 pg g−1 and 2.3 fg g−1. Uncertainties in the Pb isotopic ratio measurements were degraded by only ∼0.2%.  相似文献

We have developed a simple, rapid, and sensitive method for the determination of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) by on-line in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-mass spectrometry (LC-MS). PFOA and PFOS were separated within 10 min by high-performance liquid chromatography using an Inertsil ODS-3 column and 10 mM ammonium acetate/methanol (35/65, v/v) as a mobile phase at a flow rate of 0.25 mL min−1. Electrospray ionization conditions in the negative ion mode were optimized for MS detection of PFOA and PFOS. The optimum in-tube SPME conditions were 20 draw/eject cycles with a sample size of 40 μL using a CP-Pora PLOT amine capillary column as the extraction device. The extracted compounds could be desorbed easily from the capillary by passage of the mobile phase, and no carryover was observed. Using the in-tube SPME LC-MS method, good linearity of the calibration curve (r = 0.9990 for PFOA, r = 0.9982 for PFOS) was obtained in the range of 0.05-5 ng mL−1 each compound. The detection limits (S/N = 3) for PFOA and PFOS were 1.5 and 3.2 pg mL−1, respectively. The method described here showed about 100-fold higher sensitivity than the direct injection method. The within-day and between-day precisions (relative standard deviations) were below 3.7 and 6.0%, respectively. This method was applied successfully to the analysis of PFOA and PFOS in environmental water samples and to the elution test from a Teflon®-coated frying pan without interference peaks. The recoveries of PFOA and PFOS spiked into river samples were above 81%, and PFOA was detected at pg mL−1 levels in environmental water samples and eluate from the frying pan.  相似文献

A sensitive electrochemical approach for the detection of thrombin was designed by using densely packed hierarchical dendritic gold microstructures (HDGMs) with secondary and tertiary branches as matrices, and thionine-functionalized mesoporous silica nanospheres as signal tags. To prepare the signal tags, the positively charged thionine (as an indicator) was initially adsorbed onto the mesoporous silica nanoparticles (MSNs). Then [AuCl4] ions were in situ reduced on the thionine-modified MSNs by ascorbic acid to construct nanogold-decorated MSNs (GMSNs). The formed GMSNs were employed as label of the aminated aptamers. The assay was carried out in PBS, pH 7.4 with a sandwich-type assay mode by using the assembled thionine in the GMSNs as indicators. Compared with the pure silica nanoparticles, mesoporous silica could provide a larger surface for the immobilization of biomolecules and improve the sensitivity of the aptasensor. Under optimal conditions, the electrochemical aptasensors exhibited a wide linear range from 0.001 to 600 ng mL−1 (i.e. 0.03 pM to 0.018 μM thrombin) with a low detection limit (LOD) of 0.5 pg mL−1 (≈15 fM) thrombin at 3σ. No obvious non-specific adsorption was observed during a series of analyses to detect target analyte. The precision, selectivity and stability of the aptasensors were acceptable. Importantly, the methodology was evaluated with thrombin spiked samples in blank fetal calf serum, and the recoveries were 94.2–112%, indicating an exciting potential for thrombin detection.  相似文献

For the first time, an enzyme-linked immunosorbent assay (ELISA) has been developed and validated for the determination of fluvastatin (FLV) in plasma samples at picogram level. The assay employed a polyclonal antibody that specifically recognizes FLV with high affinity, and FLV conjugate of bovine serum albumin (FLV-BSA) immobilized onto microplate wells as a solid-phase. The assay involved a competitive binding reaction between FLV, in plasma sample, and the immobilized FLV-BSA for the binding sites on a limited amount of the anti-FLV antibody. The bound anti-FLV antibody was quantified with horseradish peroxidase-labeled second anti-rabbit IgG antibody (HRP-IgG) and 3,3′,5,5′-tetramethylbenzidine (TMB) as a substrate for the peroxidase enzyme. The concentration of FLV in the sample was quantified by its ability to inhibit the binding of the anti-FLV antibody to the immobilized FLV-BSA and subsequently the color intensity in the assay wells. The conditions for the proposed ELISA were investigated and the optimum conditions were employed in the determination of FLV in plasma samples. The assay limit of detection was 10 pg mL−1 and the effective working range at relative standard deviations (RSD) of ≤5% was 20-1000 pg mL−1. Analytical recovery of FLV from spiked plasma was 97.1-102.7 ± 2.85-6.25%. The precision of the assay was satisfactory; RSD was 2.46-5.37 and 3.19-6.64% for the intra- and inter-assay precision, respectively. The analytical procedure is convenient, and one can analyze ∼200 samples per working day, facilitating the processing of large-number batch of samples. The proposed ELISA has a great value in routine analysis of FLV for its therapeutic monitoring and pharmacokinetic studies.  相似文献

The paper reports a highly sensitive enzyme free electrochemical immunoassay (EFEIA) for the detection of herbicide chlorsulfuron. The assay is based upon oxidative gold nanoparticle (GNP) dissolution in an acidic solution. The consequent release of large amounts of gold (Au) metal ions after dissolution of gold nanoparticles tagged to antibody leads to the development of sensitive stripping voltammetry based immunoassay. The detection is made possible by the reduction of Au3 + ions at the screen printed electrode surface followed by metal analysis by using the square wave voltammetry technique. The sensitivity of chlorsulfuron detection by competitive assay procedure was 6.7 pg mL− 1 for EFEIA in marked contrast to optical detection using Standard ELISA procedure that gives a sensitivity of 4.97 ng mL− 1.  相似文献

A simple, efficient, innovative and environmentally friendly analytical technique was successfully applied for the first time for the extraction and preconcentration of polybrominated diphenyl ethers (PBDEs) from water samples. The PBDEs selected for this work were those most commonly found in the literature in natural water samples: 2,2′,4,4′-tetraBDE (BDE-47), 2,2′,4,4,5-pentaBDE (BDE-99), 2,2′,4,4,6-pentaBDE (BDE-100) and 2,2,4,4′,5,5′-hexaBDE (BDE-153). The extracted PBDEs were separated and determined by gas chromatography–mass spectrometry (GC–MS). The extraction/preconcentration technique is based on ultrasound-assisted emulsification-microextraction (USAEME) of a water-immiscible solvent in an aqueous medium. Several variables including, solvent type, extraction time, extraction temperature and matrix modifiers were studied and optimized over the relative response the target analytes. Chloroform was used as extraction solvent in the USAEME technique. Under optimum conditions, the target analytes were quantitatively extracted achieving enrichment factors (EF) higher than 319. The detection limits (LODs) of the analytes for the preconcentration of 10 mL sample volume were within the range 1–2 pg mL−1. The relative standard deviations (RSD) for five replicates at 10 pg mL−1 concentration level were <10.3%. The calibration graphs were linear within the concentration range of 5–5000 pg mL−1 for BDE-47 and BDE-100; and 5–10,000 pg mL−1 for BDE-99 and BDE-153, respectively. The coefficients of estimation were ≥0.9985. Validation of the methodology was performed by standard addition method at two concentration levels (10 and 50 pg mL−1). Recovery values were ≥96%, which showed a successful robustness of the analytical methodology for determination of picogram per milliliter of PBDEs in water samples. Significant quantities of PBDEs were not found in the analyzed samples.  相似文献
