This paper reports a multiresidue analysis procedure which permits the identification and quantification of sixty-three water-soluble pollutants. Subsequent to their solid-phase extraction (SPE) enrichment, analyses of species have been carried out from one solution, by a single injection, as their trimethylsilyl-oxime ether/ester derivatives, by gas chromatography–mass spectrometry, within 31 min. Based on our optimized extraction, derivatization and mass fragmentation studies separation have been performed in the total ion current mode, identification and quantification of compounds have been carried out on the basis of their selective fragment ions. Including various pharmaceuticals, benzoic acid, its substituted species, different aromatic carboxylic acids, cholic acids, unsaturated and saturated fatty acids, aliphatic dicarboxylic acids, as well as synthetic pollutants of various origins (2,4-di-tert-butylphenol, different phthalates). Standard compounds were added to 500 mL effluent wastewater samples, at three concentrations (1–5 μg/L, 5–10 μg/L and 10–20 μg/L). Recoveries, using the Waters Oasis cartridges performing extractions at pH 2, pH 4 and pH 7 proved to be the optimum at pH 4 (average recoveries (94.5%), except for cholesterol (10%), paracetamol (18%) and 2,5-dihydroxybenzoic acid (25%). Carbamazepine could be recovered at pH 7, only. Responses, obtained with derivatized standards proved to be linear in the range of 4–80 μg/L levels. Limit of quantitation values varied between 0.92 ng/L (4-hydroxyphenylacetic acid) and 600 ng/L (dehydrocholic acid) concentrations. One of the most important messages of this work is the confirmation of the origin of blank values. It was shown that contaminants, mainly 2,4-di-tert-butylphenol, different phthalates and fatty acids, are sourced both from the reagents and mainly from the SPE procedure, independent on the cartridge applied. Reproducibilities, characterized with the relative standard deviations (RSDs) of measurements, varied between 0.71% and 10%, with an average of 4.38% RSD. The practical utility of the method was shown by the identification and quantification of the pollutant contents of Hungarian influent and effluent wastewaters (for six consecutive months and that of the Danube River for 2 months).  相似文献

Complex mixtures of polychlorostyrenes are produced and released into the environment by reaction of chlorine with graphite at high temperatures, e.g. in electrodes. The occurrence of these compounds in the environment or human tissues is often indicative of pollution from electrolytic processes. Combined gas chromatography coupled to electron impact and negative ion chemical ionization mass spectrometry is described here for the analysis of these compounds in sediments, human venous sera and cord sera. This method has allowed a specific congener identification and quantification of the chlorostyrene mixtures present in fluvial sediments polluted by effluents from a chlorine-alkali plant. Besides octachlorostyrene, the mixture of compounds identified involved the six possible heptachlorostyrenes, fifteen hexachlorostyrenes and seven pentachlorostyrenes, having concentrations in the range of 76–16 000 ng g−1 dry weight. In human sera from the population exposed to airborne emissions from this plant these compounds ranged between 17 and 63 ng L−1 and the distributions were dominated by octachlorostyrene, three heptachlorostyrenes, including β,β-2,3,4,5,6-heptachlorostyrene, and α-2,3,4,5,6-hexachlorostyrene. Both distribution patterns showed major differences in composition despite the common pollution source that was influencing both types of samples. The method also revealed qualitative differences between maternal and cord sera from the exposed population.  相似文献

Urine samples have been the predominant matrix for doping controls for several decades. However, owing to the complementary information provided by blood (as well as serum or plasma and dried blood spots (DBS)), the benefits of its analysis have resulted in continuously increasing appreciation by anti-doping authorities. On the one hand, blood samples allow for the detection of various different methods of blood doping and the abuse of erythropoiesis-stimulating agents (ESAs) via the Athlete Biological Passport; on the other hand, targeted and non-targeted drug detection by means of chromatographic–mass spectrometric methods represents an important tool to increase doping control frequencies out-of-competition and to determine drug concentrations particularly in in-competition scenarios. Moreover, blood analysis seldom requires in-depth knowledge of drug metabolism, and the intact substance rather than potentially unknown or assumed metabolic products can be targeted. In this review, the recent developments in human sports drug testing concerning mass spectrometry-based techniques for qualitative and quantitative analyses of therapeutics and emerging drug candidates are summarized and reviewed. The analytical methods include both low and high molecular mass compounds (e.g., anabolic agents, stimulants, metabolic modulators, peptide hormones, and small interfering RNA (siRNA)) determined from serum, plasma, and DBS using state-of-the-art instrumentation such as liquid chromatography (LC)–high resolution/high accuracy (tandem) mass spectrometry (LC-HRMS), LC–low resolution tandem mass spectrometry (LC-MS/MS), and gas chromatography–mass spectrometry (GC-MS).  相似文献

A simple and rapid analytical method based on in-matrix ethyl chloroformate (ECF) derivatization has been developed for the quantitative determination of bisphenol-A (BPA) in milk and water samples. The samples containing BPA were derivatised with ECF in the presence of pyridine for 20 s at room temperature, and the non-polar derivative thus formed was extracted using polydimethylsiloxane solid-phase microextraction (SPME) fibres with thicknesses of 100 μm followed by analysis using gas chromatography–mass spectrometry. Three alkyl chloroformates (methyl, ethyl and isobutyl chloroformate) were tested for optimum derivatisation yields, and ECF has been found to be optimum for the derivatisation of BPA. Several parameters such as amount of ECF, pyridine and reaction time as well as SPME parameters were studied and optimised in the present work. The limit of detection for BPA in milk and water samples was found to be 0.1 and 0.01 μg L−1, respectively, with a signal-to-noise ratio of 3:1. The limit of quantitation for BPA in milk and water was found to be 0.38 and 0.052 μg L−1, respectively, with a signal-to-noise ratio of 10:1. In conclusion, the method developed was found to be rapid, reliable and cost-effective in comparison to silylation and highly suitable for the routine analysis of BPA by various food and environmental laboratories.  相似文献

Solid-phase microextraction (SPME) coupled with gas chromatography–mass spectrometry (GC–MS) and selected ion monitoring (SIM) was used for the analytical determination of priority pesticide residues. Fibers coated with a 65-µm film thickness of polydimethylsiloxane divinylbenzene (PDMS-DVB) were used to extract 31 pesticides of different chemical groups. The quality parameters of the method demonstrated a good precision with detection limits of 1–56?ng/L. Linearity was controlled in the range of 0.1–50?µg/L. The proposed method was applied for the trace-level determination of the target pesticides in surface water samples including three rivers and one lake at the Epirus region (north-west Greece) for a period of one year. The results demonstrate the suitability of the SPME–GC–MS approach for the analysis of multi-residue pesticides in environmental water samples.  相似文献

A simple and sensitive procedure, using p-tolylpiperazine (pTP) as internal standard (IS), has been developed and validated for the qualitative and quantitative analysis of 1-(3-trifuoromethylphenyl)piperazine (TFMPP), 1-(3-chlorophenyl)piperazine (mCPP) and 1-(4-methoxyphenyl)piperazine (MeOPP) in hair. Drug extraction was performed by incubation with 1 M sodium hydroxide at 50 °C for 40 min, and the extracts were cleaned up using mixed-mode solid-phase extraction. The analytes were derivatized with N-methyl-N-(trimethylsilyl) trifluoroacetamide with 5% trimethylchlorosilane and analysed by gas chromatography–mass spectrometry in the selected ion monitoring mode. The method was linear from 0.05 (lower limit of quantitation) to 4 ng mg−1, with correlation coefficients higher than 0.99 for all the compounds. Intra- and interday precision and accuracy were in conformity with the criteria normally accepted in bioanalytical method validation, and the sample cleanup step presented a mean efficiency higher than 90% for all the analytes. Due to its simplicity and speed, this method can be successfully applied in the screening and quantitation of these compounds in hair samples, and is suitable for application in forensic toxicology routine analysis.  相似文献

A rapid, sensitive and selective liquid chromatography–mass spectrometry (LC–MS) assay has been developed for determination of cyclosporin A (CyA) in human plasma; cyclosporin B (CyB) was used as internal standard (IS). The method utilized a combination of a column-switching valve and a reversed-phase symmetry column. The mobile phase was a 25:75 (v/v) mixture of 10% aqueous glacial acetic acid and acetonitrile. Running time per single run was less than 10 min. Sample preparation included C8 SPE of human plasma spiked with the analyte and internal standard, evaporation of the eluate to dryness at 50°C under N2 gas, and finally reconstitution in the mobile phase. Detection of cyclosporin A and the IS was performed in selected ion-monitoring mode at m/z 601.3 and 594.4 Da for CyA and IS, respectively. Quantitation was achieved by use of the regression equation of relative peak area of cyclosporin to IS against concentration of cyclosporin. The method was validated according to FDA guideline requirements. The linearity of the assay in the range 5.0–400.0 ng mL–1 was verified as characterized by the least-squares regression line Y=(0.00268±1.9×10–4)X+(0.00078±1.8×10–3), correlation coefficient, r=0.9986±1.1×10–3 (n=48). Intra and inter-day quality-control measurements in the range 5.0–350.0 ng mL–1 revealed almost 100% accuracy and 9% CV for precision. The mean absolute recovery of CyA was found to be 84.01±9.9% and the respective relative recovery was 100.3±9.19. The limit of quantitation (LOQ) achieved was 5 ng mL–1. Eventually, stability testing of the analyte and IS in plasma or stock solution revealed that both chemicals were very stable when stored for long or short periods of time at room temperature or –20°C.  相似文献

Stir-bar-sorptive extraction followed by liquid desorption and large-volume injection capillary gas chromatography with mass spectrometric detection (SBSE–LD–LVI-GC–MS), had been applied for the determination of ultra-traces of eight pyrethroid pesticides (acrinathrin, cypermethrin, deltamethrin, esfenvalerate, fenpropathrin, fenvalerate, and permethrin cis and trans isomers) in water samples. Instrumental calibration for selected-ion monitoring acquisition and conditions that could affect the SBSE–LD efficiency are fully discussed. By performing systematic assays on 30-mL water samples spiked at the 0.10 g L–1 level it was established that stir-bars coated with 47 L polydimethylsiloxane, an equilibrium time of 60 min (750 rpm), 5% methanol as organic modifier, and acetonitrile as back-extraction solvent, provided the best analytical performance to monitor pyrethroid pesticides in water matrices. Good accuracy (81.8–105.0%) and remarkable reproducibility (<11.7%) were obtained, and the experimental recovery data were in good agreement with the theoretical equilibrium described by octanol–water partition coefficients (log KO/W), with the exception of acrinathrin for which lower yields were measured. Excellent linear dynamic ranges between 25 and 400 ng L–1 (r2>0.994), low quantification (3.0–7.5 ng L–1) and detection (1.0–2.5 ng L–1) limits were also achieved for the eight pyrethroid pesticides studied. The method was successfully used for analysis of tapwater and groundwater matrices spiked at the 0.10 g L–1, revealing the suitability of the method for determination of pyrethroid pesticides in real samples. The method was shown be reliable and sensitive and a small volume of sample was required to monitor pyrethroids at ultra-trace levels, in compliance with international regulatory directives on water quality.  相似文献

This research work represents a synthetic approach for the development of hybrids derivatives of oxadiazole-based benzothiazole (117) and diversity in derivatives was achieved using variety of aryl ring of S-substituted benzothiazole to see the effect on the biological activities. All the synthesized derivatives were evaluated for their in vitro α-glucosidase and urease inhibitory potential. The α-glucosidase and urease inhibition profile of the new derivatives represents moderate to good inhibitory potential with IC50 values ranging from 4.60 ± 1.20 µM to 48.40 ± 7.70 µM (α-glucosidase) and 8.90 ± 2.80 to 57.30 ± 7.70 µM (urease) respectively. The results were compared to standard acarbose (38.60 ± 4.50 µM) and thiourea (58.70 ± 6.80 µM) drugs respectively. Among the synthesized series, the analogs 1 having IC50 values of and 4.60 ± 1.20 (α-glucosidase), 8.90 ± 2.80 (urease) and 2 with IC50 values of 5.60 ± 1.60 (α-glucosidase) and 10.90 ± 2.10(urease) were found to be significantly active against targeted α-glucosidase and urease enzymes. The structure of all the newly synthetics scaffolds were confirmed by using different types of spectroscopic techniques such as HREI-MS, 1H- and 13C- NMR spectroscopy. The molecular docking studies of the synthesized derivatives showed good correlations with the experimental findings. The binding modes of active compounds and their interactions with active site residues revealed them as possible anti-diabetics and anti-urease leads. The degree of activity and docking studies displayed by the novel innovative structural hybrids of oxadiazole-based benzothiazole moieties make these compounds new active leads and promising candidates for the development of anti-diabetics and anti-urease agents.  相似文献

An on-line screening method to analyse volatile organic compounds (VOCs) in faecal samples was developed. VOCs were isolated from a standard solution or faecal samples using a purge-and-trap system and identified and quantified by GC–MS. The experimental conditions were optimised and the performance of the system was evaluated. Linear calibration curves were obtained with correlation coefficients of at least 0.992. RSDs within and between days were less than 10%. The method was successfully applied to the analysis of faecal samples, yielding 135 different volatile organic compounds identified in 11 faecal samples. Of those, 22 VOCs were found in all volunteers, whereas 34 VOCs were person-specific.  相似文献

Dynamic single-drop microextraction (SDME) was automatized employing an Arduino-based lab-made Cartesian robot and implemented to determine parabens in wastewater samples in combination with liquid chromatography–tandem mass spectrometry. A dedicated Arduino sketch controls the auto-performance of all the stages of the SDME process, including syringe filling, drop exposition, solvent recycling, and extract collection. Univariate and multivariate experiments investigated the main variables affecting the SDME performance, including robot-dependent and additional operational parameters. Under selected conditions, limit of detections were established at 0.3 µg/L for all the analytes, and the method provided linear responses in the range between 0.6 and 10 µg/L, with adequate reproducibility, measured as intraday relative standard deviations (RSDs) between 5.54% and 17.94%, (n = 6), and inter-days RSDs between 8.97% and 16.49% (n = 9). The robot-assisted technique eased the control of dynamic SDME, making the process more feasible, robust, and reliable so that the developed setup demonstrated to be a competitive strategy for the automated extraction of organic pollutants from water samples.  相似文献

The application of combined Raman spectroscopic and GC–MS analytical techniques for the characterisation of organic varnish residues from Egyptian Dynastic funerary sarcophagal and cartonnage fragments from the Graeco-Roman period, ca. 2200 BP, is described. The nondestructive use of Raman spectroscopy was initially employed to derive information about the specific location of organic material on the specimens, which were then targeted in specific areas using minimal sampling for GC–MS analysis. In the case of the sarcophagal fragment, a degraded yellow-brown surface treatment was identified as a Pistacia spp. resin; this provides additional evidence for the use of this resin, which has previously been identified in Canaanite transport amphorae, varnishes and “incense” bowls in an Egyptian Late Bronze Age archaeological context. The cartonnage fragment also contained an organic coating for which the Raman spectrum indicated a degradation that was too severe to facilitate identification, but the GC–MS data revealed that it was composed of a complex mixture of fatty acid residues. The combined use of GC–MS and Raman spectroscopy for the characterisation of organic materials in an archaeological context is advocated for minimisation of sampling and restriction to specifically identified targets for museum archival specimens.  相似文献

Non-steroidal anti-inflammatory drugs are widely used for treatment of animals. According to Council Directive 96/23/EC, residues of these drugs must be monitored because of the potential risk they pose to the consumers' health. For this reason an LC-MS-MS method was developed for detection of wide range of NSAIDs, including both "acidic" NSAIDs (carprofen, diclofenac, flunixin, meloxicam, phenylbutazone, oxyphenbutazone, tolfenamic acid, mefenamic acid, naproxen, ketoprofen, ibuprofen, firocoxib, rofecoxib, and celecoxib) and "basic" NSAIDs (four metamizole metabolites). Analytes were extracted from milk samples with acetonitrile in the presence of ammonium acetate. One portion of the extract was directly analyzed for the presence of metamizole metabolites; a second portion was cleaned with an amino cartridge. All NSAIDs were separated on a Phenomenex Luna C8(2) column and analyzed by LC-MS-MS in negative (acidic NSAIDs) and positive (metamizole metabolites) ion modes. The method was validated in accordance with the requirements of Commission Decision 2002/657/EC. Within-laboratory reproducibility was in the range 7-28%, and accuracy was in the range 71-116%. The method enabled detection of all the analytes with the expected sensitivity, below the recommended concentrations. The method fulfills the criteria for confirmatory methods and, because of its efficiency, may also be used for screening purposes. The procedure was also successfully verified in the proficiency test organized by EU-RL in 2010. As far as the authors are aware, this is one of the first methods capable of detecting diclofenac residues below the MRL in milk (0.1 μg kg(-1)). An additional advantage is the possibility of simultaneous determination of "acidic" NSAIDs and metamizole metabolites.  相似文献

Four different isolation techniques, conventional hydrodistillation (HD), microwave-assisted hydrodistillation (MWHD), microdistillation (MD) and micro-steam distillation-solid-phase microextraction (MSD-SPME), have been used to analyze the volatile constituents from the aerial parts of Salvia rosifolia Sm. by gas chromatography and gas chromatography coupled to mass spectrometry. HD and MWHD techniques produced quantitatively (yield, 0.39% and 0.40%) and qualitatively (aromatic profile) similar essential oils. α-Pinene (15.7–34.8%), 1,8-cineole (16.6–25.1%), β-pinene (6.7–13.5%), β-caryophyllene (1.4–5.0%) and caryophyllene oxide (1.4–4.4%) were identified as major constituents of this Turkish endemic species. Besides, the hydrodistilled oil of S. rosifolia was evaluated for antibacterial, antifungal, anticancer, antioxidant and cytotoxic activities. The hydrodistilled oil of S. rosifolia showed antibacterial activity against Methicillin-resistant Staphylococcus aureus (MRSA) with a MIC value of 125 μg/mL. Other human pathogenic microorganisms (Escherichia coli, Pseudomonas aeruginosa, Enterobacter aerogenes, Salmonella typhimurium, Staphylococcus epidermidis, Candida albicans) were also inhibited within a moderate range (MIC = 125–1000 μg/mL). Antifungal activity of the oil was also observed against the strawberry anthracnose-causing fungal plant pathogens Colletotrichum acutatum, C. fragariae and C. gloeosporioides. No cytotoxicity was observed for S. rosifolia oil up to 25 mg/mL against malignant melanoma, epidermal, ductal and ovary carcinoma.  相似文献

The development of nontargeted screening strategy for veterinary drugs and their metabolites is very important for food safety. In this study, a nontargeted screening strategy was developed to find the potentially hazardous substances based on mass defect filtering (MDF) using liquid chromatography–high-resolution mass spectrometry. First, the drug metabolites of 112 veterinary drugs from seven classes of antimicrobials were predicted. Second, three MDF models were established, including the traditional rectangular MDF, the enhanced parallelogram MDF, and the polygonal MDF. Finally, the strategy was applied to nontargeted screening of veterinary drugs in 36 milk samples. The polygonal MDF model based on the distribution area of parent drugs and their metabolites showed a better filtering effect. After removing food components and performing MDF, about 10% of the substances remained, and four veterinary drugs and six drug metabolites were discovered and identified, showing the effectiveness of this strategy. The nontargeted screening strategy can rapidly remove interfering substances and find the suspected compounds. It can also be used for nontargeted screening of veterinary drugs and their metabolites in other food matrices.  相似文献

Current strategies in the LC–MS analysis of pesticides and related compounds in environmental samples, fruits and vegetables, and biological samples mostly rely on the selection of appropriate precursor/product-ion combinations (transitions) for selected reaction monitoring (SRM), often based on automated parameter optimization and selection of the transition. Such a procedure does not require any information on the type of fragmentation reaction involved in the generation of the product ion from the selected precursor ion. However, such information does become important in untargeted screening for unknown contaminants in environmental and food samples, which are generally based on a combination of high-resolution mass spectrometry and (multistage) tandem mass spectrometry. With this in mind, the group-specific fragmentation behaviour has been studied for six classes of pesticides and herbicides, i.e., triazines, organophosphorous pesticides, phenylurea herbicides, carbamates, sulfonylurea herbicides, and chlorinated phenoxy acid herbicides. When relevant, some comparison was made between fragmentation of protonated molecules in MS–MS and of molecular ions generated by electron ionization in GC–MS.  相似文献

Peptides with a molecular mass between 2 and 10 kDa that are prohibited in elite sports usually require dedicated sample preparation and mass spectrometric detection that commonly cannot be combined with other (lower molecular mass) substances. In most instances, the physicochemical differences are too significant to allow for a generic analytical procedure. A simplification of established and comparably complex analytical approaches is therefore desirable and has been accomplished in the context of this study. With urine samples representing still the most frequently collected doping control specimens, efficient extraction of peptidic analytes from this matrix was a major goal of this method, as demonstrated for the included compounds such as insulins (human, lispro, aspart, glulisine, tresiba, glargine metabolite, bovine insulin, porcine insulin), growth hormone-releasing hormones (sermorelin, CJC-1295, tesamorelin) incl. their respective metabolites, insulin-like-growth factors (long-R3-IGF-I, R3-IGF-I, des1–3-IGF-I), synacthen, gonadorelin and mechano growth factors (human MGF, MGF-Goldspink). Sample preparation and detection are controlled by five internal standards, covering all five included peptide drug categories. Nearly all requirements of the recent technical documents from the World Anti-Doping Agency (WADA) considering their minimum required performance levels (MRPL) are fulfilled, and the method was validated for its utilisation as initial testing procedure in doping controls. Finally, the approach was applied to authentic post-administration study urine samples (for insulins and gonadorelin) in order to provide proof of principle.  相似文献
