A new procedure for determination of biogenic amines (BA): histamine, phenethylamine, tyramine and tryptamine, based on the derivatization reaction with 2-chloro-1,3-dinitro-5-(trifluoromethyl)-benzene (CNBF), is proposed. The amines derivatives with CNBF were isolated and characterized by X-ray crystallography and 1H, 13C, 19F NMR spectroscopy in solution. The novelty of the procedure is based on the pure and well-characterized products of the amines derivatization reaction. The method was applied for the simultaneous analysis of the above mentioned biogenic amines in wine samples by the reversed phase-high performance liquid chromatography. The procedure revealed correlation coefficients (R2) between 0.9997 and 0.9999, and linear range: 0.10–9.00 mg L−1 (histamine); 0.10–9.36 mg L-1 (tyramine); 0.09–8.64 mg L−1 (tryptamine) and 0.10–8.64 mg L−1 (phenethylamine), whereas accuracy was 97%–102% (recovery test). Detection limit of biogenic amines in wine samples was 0.02–0.03 mg L−1, whereas quantification limit ranged 0.05–0.10 mg L−1. The variation coefficients for the analyzed amines ranged between 0.49% and 3.92%. Obtained BA derivatives enhanced separation the analytes on chromatograms due to the inhibition of hydrolysis reaction and the reduction of by-products formation.  相似文献

The usefulness of the secondary line at 252.744 nm and the approach of side pixel registration were evaluated for the development of a method for sequential multi-element determination of Cu, Fe, Mn and Zn in soil extracts by high-resolution continuum source flame atomic absorption spectrometry (HR-CS FAAS). The influence of side pixel registration on the sensitivity and linearity was investigated by measuring at wings (248.325, 248.323, 248.321, 248.329, and 248.332 nm) of the main line for Fe at 248.327 nm. For the secondary line at 252.744 nm or side pixel registration at 248.325 nm, main lines for Cu (324.754 nm), Mn (279.482 nm) and Zn (213.875 nm), sample flow-rate of 5.0 mL min−1 and calibration by matrix matching, analytical curves in the 0.2-1.0 mg L−1 Cu, 1.0-20.0 mg L−1 Fe, 0.2-2.0 mg L−1 Mn, 0.1-1.0 mg L−1 Zn ranges were obtained with linear correlations better than 0.998. The proposed method was applied to seven soil samples and two soil reference materials (IAC 277; IAC 280). Results were in agreement at a 95% confidence level (paired t-test) with reference values. Recoveries of analytes added to soil extracts containing 0.15 and 0.30 mg L−1 Cu, 7.0 and 14 mg L−1 Fe, 0.60 and 1.20 mg L−1 Mn, 0.07 and 0.15 mg L−1 Zn, varied within the 94-99, 92-98, 93-101, and 93-103% intervals, respectively. The relative standard deviations (n = 12) were 2.7% (Cu), 1.4% (Fe - 252.744 nm), 5.7% (Fe - 248.325 nm), 3.2% (Mn) and 2.8% (Zn) for an extract containing 0.35 mg L−1 Cu, 14 mg L−1 Fe, 1.1 mg L−1 Mn and 0.12 mg L−1 Zn. Detection limits were 5.4 μg L−1 Cu, 55 μg L−1 Fe (252.744 nm), 147 μg L−1 Fe (248.325 nm), 3.0 μg L−1 Mn and 4.2 μg L−1 Zn.  相似文献

Potassium formate was extracted from airport storm water runoff by headspace solid-phase microextraction (HS-SPME) and analyzed by GC–MS. Formate was transformed to formic acid by adding phosphoric acid. Subsequently, formic acid was derivatized to methyl formate by adding methanol. Using sodium [2H]formate (formate-d) as an internal standard, the relative standard deviation of the peak area ratio of formate (m/z 60) and formate-d (m/z 61) was 0.6% at a concentration of 208.5 mg L−1. Calibration was linear in the range of 0.5–208.5 mg L−1. The detection limit calculated considering the blank value was 0.176 mg L−1. The mean concentration of potassium formate in airport storm water runoff collected after surface de-icing operations was 86.9 mg L−1 (n = 11) with concentrations ranging from 15.1 mg L−1 to 228.6 mg L−1.  相似文献

Nitrate determination in waters is generally carried out with cadmium filings and carcinogenic reagents or by reaction with phenolic compounds in highly concentrated sulfuric acid medium. In this work, it was developed a green analytical procedure for nitrate determination in natural waters based on direct spectrophotometric measurements in ultraviolet, using a flow-injection system with an anion-exchange column for separation of nitrate from interfering species. The proposed method employs only one reagent (HClO4) in a minimum amount (equivalent to 18 μL concentrated acid per determination), and allowed nitrate determination within 0.50-25.0 mg L−1, without interference of up to 200.0 mg L−1 humic acid; 1.0 mg L−1 NO2; 200.0 mg L−1 PO43−; 75.0 mg L−1 Cl; 50.0 mg L−1 SO42− and 15.0 mg L−1 Fe3+. The detection limit (99.7% confidence level) and the coefficient of variation (n = 20) were estimated as 0.1 mg L−1 and 0.7%, respectively. The results obtained for natural water samples were in agreement with those achieved by the reference method based on nitrate reduction with copperized cadmium at the 95% confidence level.  相似文献

Solid support assisted derivatization coupled to diffuse reflectance spectroscopy (DRS) was proposed and proved useful for the detection and quantification of aliphatic amines in water as an example. Dabsyl chloride (DBS), ninhydrin and sodium 1,2-naphtoquinone 4-sulphonate (NQS) were assayed as derivatization reagents. C18 and SDB-XC disks and C18 cartridges were tested for amine retention and after that derivatization. The decrease of the orange colour of dabsyl chloride on SBD-XC disks produced by the formation of its derivative with methylamine in the support (10 min at 100 °C) allowed the selective determination of the amine at concentration level equal or higher than 0.5 mg L−1. Ninhydrin can be used for methylamine, ethylamine, propylamine, butylamine and pentylamine (between 5 and 15 mg L−1) by measuring the diffuse reflectance produced by the brown derivative formed in C18 extraction disks after 15 min at 100 °C. NQS and C18 SPE columns can be also employed to estimate amines, but the detection limits were higher than those provided by DBS and Ninhydrin, around 10 mg L−1. As an example, found concentration of methylamine or total amines (expressed as -NH2-N mg L−1) in a wastewater sample is given employing dabsyl chloride or ninhydrin reagents, respectively with satisfactory results.  相似文献

This work reports a new sensitive multi-residue liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for detection, confirmation and quantification of forty-six pesticides and transformation products belonging to different chemical classes in wines. The proposed method makes use of a solid-phase extraction (SPE) procedure with Oasis HLB cartridges that combines isolation of the pesticides and sample clean-up in a single step. Analysis is performed by liquid chromatography-electrospray ionisation-tandem mass spectrometry (LC-MS/MS) operated in the selected reaction monitoring (SRM) mode, acquiring two specific precursor-product ion transitions per target compound. An investigation of matrix effects has been performed during method validation showing medium to low effects for the majority of the compounds. Limits of detection (LODs) were in the range 0.0003–0.003 mg L−1 and limits of quantification (LOQs) were in the range 0.001–0.01 mg L−1. The average recoveries, measured at two concentration levels (0.010 and 0.050 mg L−1), were in the range 70–110% for most of the compounds tested with % relative standard deviations below 20%, while a value of 0.010 mg L−1 has been established as the method limit of quantification (MLOQ) for all target species. Expanded uncertainty values were in the range 10–40% while the Horrat ratios were below 1. The method has been successfully applied to the analysis of 60 wine samples in the course of an annual monitoring study with carbendazim-benomyl, thiophanate-methyl and carbaryl being the most frequently determined pesticides.  相似文献

A sequential injection analysis (SIA) using lab-on-valve with air segmentation and spectrophotometric detection was designed for copper(II) determination. It is based on the reaction of copper(II) and 2-carboxy-2′-hydroxy-5′-sulfoformazyl benzene (Zincon) in a weak alkaline solution between the air zones. Beer's Law was obeyed over the range of 0.1-2.0 mg L−1 copper(II) with a correlation coefficient 0.9985 and a slope of 0.2893 absorbance unit/mg L−1. The relative standard deviation was 2.0% for a series of 10 measurements of 0.5 mg L−1 copper(II) solution. The detection limit (3 S/N) and the limit of quantification (LOQ) were 0.05 and 0.17 mg L−1 respectively. This method has been successfully applied to determination of copper(II) in wastewater with a sample throughput of 120 h−1. The method is superior to the batchwise method in that it provides fully automation, rapidity, less reagents and sample consumption with little waste generation.  相似文献

The present paper describes a direct procedure for the determination of catechin and epicatechin concentrations in red wines employing reverse-phase high performance liquid chromatography (RP HPLC) and detection by fluorescence. The method was performed using a sample volume of 10 µL without dilution. The separation process employed a Chromolith performance RP-18e (100 mm × 4.6 mm) column, and the mobile phase was composed of solvent A: methanol-acetic acid-water (90:8:2) and solvent B: water-acetic acid-methanol (10:2:88) at a flow rate of 1.0 mL min− 1. Linearity was observed in the range of 1 to 30 mg L− 1, with limits of detection and quantification of 0.27 and 0.89 mg L− 1, respectively, for catechin and 0.33 and 1.01 mg L− 1, respectively, for epicatechin. The precisions estimated by the relative standard deviation were 3.34 and 1.09% for catechin concentrations of 0.5 and 20 mg L− 1 respectively and 2.82 and 0.49% for epicatechin concentrations of 0.5 and 20 mg L− 1, respectively. The evaluation of the accuracy was done using an addition/recovery assay. Four wine samples were used, and the recoveries varied from 105 to 108% for catechin and from 97 to 119% for epicatechin. The method was applied to the analysis of red wine samples collected from the São Francisco region, Bahia State, Brazil. Nine samples were analyzed, and the catechin and epicatechin concentrations varied from 7.51 to 73.20 and from 5.08 to 43.32 mg L− 1, respectively. The concentrations found agree with data reported in the literature.  相似文献

Mycophenolic acid (MPA) is an immunosuppressant drug which powerfully inhibits lymphocyte proliferation. Since the early 1990s it has been used to prevent rejection in organ transplantation. The requirement of therapeutic drug monitoring shown in previous studies raises the necessity of acquiring accurate and sensitive methods to measure MPA and its major metabolite mycophenolic acid glucuronide (MPAG).The authors developed a sample cleanup-free, rapid, and highly specific method for simultaneous measurement of MPA and MPAG in human plasma and serum using the novel technology of ultra-performance liquid chromatography-electrospray ionization tandem mass spectrometry. MPA- and MPAG-determinations were performed during a 2.0-min run time. Multiple calibration curves for the analysis of MPA and MPAG exhibited consistent linearity and reproducibility in the range of 0.05-100 (r > 0.999) mg L−1 and 4-4000 mg L−1 (r > 0.999), respectively. Limits of Detection were 0.014 mg L−1 for MPA and 1.85 mg L−1 for MPAG. Lower Limits of Quantification were 0.05 mg L−1 for MPA and 2.30 mg L−1 for MPAG. Interassay imprecision was <10% for both substances. Mean recovery was 103.6% (range 78.1-129.7%) for MPA and 111.1% (range 73.0-139.6%) for MPAG. Agreement was good for MPA and MPAG between the presented method and a validated HPLC-MS/MS method. The Passing-Bablok regression line for MPA and MPAG was HPLC-MS/MS = 1.14 UPLC-MS/MS—0.14 [mg L−1], r = 0.96, and HPLC-MS/MS = 0.77 UPLC-MS/MS + 0.50 [mg L−1], r = 0.97, respectively. This sample cleanup-free and robust LC-MS/MS assay facilitates the rapid, accurate and simultaneous determination of MPA and MPAG in human body fluids.  相似文献

A spectrophotometric method was developed to quantify low polysorbate (PS) levels in biopharmaceutical formulations containing high protein concentrations. In the method, Oasis HLB solid phase extraction (SPE) cartridge was used to extract PS from high protein concentration formulations. After loading a sample, the cartridge was washed with 4 M guanidine HCl and 10% (v/v) methanol, and the retained PS was eluted by acetonitrile. Following the evaporation of acetonitrile, aqueous cobalt-thiocyanate reagent was added to react with the polyoxyethylene oxide chain of polysorbates to form a blue colored PS–cobaltothiocyante complex. This colored complex was then extracted into methylene chloride and measured spectrophotometrically at 620 nm. The method performance was evaluated on three products containing 30–40 mg L−1 PS-20 and PS-80 in ≤70 g L−1 protein formulations. The method was specific (no matrix interference identified in three types of protein formulations), sensitive (quantitation limit of 10 mg L−1 PS) and robust with good precision (relative standard deviation ≤6.4%) and accuracy (spike recoveries from 95% to 101%). The linear range of the method for both PS-20 and PS-80 was 10 to 80 mg L−1 PS. By diluting samples with 6 M guanidine HCl and/or using different methylene chloride volumes to extract the colored complexes of standards and samples, the method could accurately and precisely quantify 40 mg L−1 PS in up to 300 g L−1 protein formulations.  相似文献

A simple, automatic and practical system for successive determination of albumin and creatinine has been developed by combining sequential injection analysis (SIA) and highly sensitive dye-binding assays. Albumin detection was based on the increase in the absorbance due to complex formation between albumin and eosin Y in acidic media. The absorbance of the complex was monitored at 547 nm. For the creatinine assay, the concentration of creatinine was measured by reaction with alkaline picrate to form a colored product which absorbs at 500 nm. The influences of experimental variables such as effects of pH, reagent concentration, standard/sample volume and interferences were investigated. Under optimal conditions, the automated method showed linearity up to 20 mg L−1 for albumin and 100 mg L−1 for creatinine. The 3σ detection limits were 0.6 and 3.5 mg L−1 for albumin and creatinine, respectively, and the relative standard deviations (n = 10) were 2.49% for 20 mg L−1 albumin, and 3.14% for 20 mg L−1 creatinine. Application of the proposed method to the direct analysis of urinary samples yielded results which agreed with those obtained from the Bradford protein assay and a creatinine enzymatic assay according to a paired t-test. The results obtained should be a step towards developing a fully automated and reliable analytical system for clinical research, which requires direct determination of albumin and creatinine and/or its ratios.  相似文献

A molecularly imprinted polymer (MIP) designed to enable the selective extraction of carbamazepine (CBZ) from effluent wastewater and urine samples has been synthesised using a non-covalent molecular imprinting approach. The MIP was evaluated chromatographically in the first instance and its affinity for CBZ also confirmed by solid-phase extraction (SPE). The optimal conditions for SPE consisted of conditioning of the cartridge using acidified water purified from a Milli-Q system, loading of the sample under basic aqueous conditions, clean-up using acetonitrile and elution with methanol. The attractive molecular recognition properties of the MIP gave rise to good CBZ recoveries (80%) when 100 mL of effluent water spiked with 1 μg L−1 was percolated through the polymer. For urine samples, 2 mL samples spiked with 2.5 μg L−1 CBZ were extracted with a recovery of 65%. For urine, the linear range was 0.05-24 mg L−1, the limit of detection was 25 μg L−1 and precision, expressed as relative standard deviation at 0.5 mg L−1 (n = 3), was 3.1% and 12.6% for repeatability and reproducibility between days, respectively.  相似文献

In this work, a fully automated flow system exploiting the advantages of the association of multi-pumping, multicommutation, binary sampling and merging zones, to accomplish the sequential determination of copper in serum and urine by flame atomic absorption spectrometry, is described. The developed flow system allowed multiple tasks, such as serum samples preparation (samples and standard solutions viscosity adjustment), serum copper (SCu) measurement, urine copper (UCu) pre-concentration and its subsequent elution and measurement, to be carried out sequentially. The implemented flow manifold presented a modular configuration consisting on two quasi-independent modules, each one accountable for a specific sample manipulation and whose combined operation under computer control enabled the determination of copper in a wide concentrations range.Once optimised and with a sample consumption of about 0.250 mL of serum and 7 mL of urine, the developed flow system allowed linear calibration plots up to 5 mg L−1 with a detection limit of 0.035 mg L−1 for SCu and linear calibration plots up to 300 μg L−1 with a detection limit of 0.67 μg L−1 for UCu. The sampling rate varied according to the module employed and was about 360 determinations h−1 (SCu module), 12 determinations h−1 (UCu module) or 24 determinations h−1 (12 urine and 12 serum samples; UCu and SCu modules simultaneously). Repeatability studies (R.S.D.%, n = 10) showed good precision for UCu at concentrations of 25 μg L−1 (2.54%), 50 μg L−1 (0.90%) and 100 μg L−1 (1.62%) as well as for SCu at concentrations of 0.25 mg L−1 (8.11%), 1 mg L−1 (3.11%) and 5 mg L−1 (0.90%). A comparative evaluation showed a good agreement between the results obtained in the analysis of UCu and SCu (n = 18) by both the developed methodology and the reference procedures. Accuracy was further evaluated by means of the analysis of reference samples (Seronorm™ Trace Elements Urine and Seronorm™ Trace Elements Serum) and the obtained results complied with the certified values.  相似文献

A simple chiral high-performance liquid chromatography (HPLC) method with ultraviolet (UV) and circular dichroism (CD) detection was developed and validated for measuring benalaxyl enantiomers using (R,R) Whelk-O 1 column. The effects of mobile phase composition and column temperature on the entioseparation were investigated. A CD detector was used to determine the elution order of the enantiomers. Excellent resolution was easily obtained using n-hexane-polar organic alcohols mobile phase. The chiral recognition mechanism was also discussed. Based on the developed chiral HPLC method, enantioselective analysis methods for this fungicide in environment matrix (soil and water) were developed and validated. Good linearities were obtained over the concentration range of 0.25-25 mg L−1 for both enantiomers. Liquid-liquid extraction and solid phase extraction (SPE) were used for the enrichment and cleanup of soil and water samples. Recoveries for the two enantiomers were 79-91% at 0.02, 0.04 and 0.2 mg kg−1 levels from soil, and 89-101% at 0.0025, 0.01 and 0.05 mg L−1 levels from water. Run-to-run and day-to-day assay precisions were below 10% for both enantiomers at concentrations of 0.5, 1 and 5 mg L−1. Individual detection limits of the two enantiomers were both 2 ng. Limits of detection (LOD) were 0.004 mg kg−1 in soil and 0.001 mg L−1 in water.  相似文献

A novel, inexpensive and fast method based on the electrooxidation of glycerol on platinum electrodes by the potential cycling technique has been designed for the determination of free glycerol in biodiesel. A wide range of linearity was achieved between 15 and 150 mg L−1 (0.16 and 1.6 mmol L−1), which corresponds to concentrations ranging between 56 and 560 mg kg−1 (glycerol:biodiesel) for an extraction using 2 g biodiesel. A method for the fast extraction of glycerol from biodiesel with water followed by elimination of organic interferents has also been developed, so that the novel determination method can be applied to various biodiesel samples. The excellent repeatability allows determination of glycerol in numerous samples, with no need for recalibration.  相似文献

A simple, specific and sensitive sequential injection analysis (SIA) system based on non-immunoassay fluorescent detection has been developed for the determination of urinary albumin. The specific binding of the dye Albumin Blue 580 (AB 580) to albumin in urine generated high emission fluorescent signals. The excitation and emission wavelengths were set at 590 and 610 nm, respectively. The analytical range was obtained from 1 to 100 mg L−1, with a detection limit of 0.3 mg L−1 (S/N = 3). The SIA system gave high precision with relative standard deviations (R.S.D.s) of 0.9% and 1.4% when evaluated with 15 and 100 mg L−1 albumin (n = 15), respectively. The method exhibited good reproducibility, as assessed by performing four analytical curves on different days, and intra-run CVs (2.3-3.3%) and inter-run CVs (3.8%) were obtained. Rapid operation was achieved with a sample throughput of 37 h−1. This method was successfully applied to the determination of urinary albumin, and the method was highly correlated with the immunoturbidimetric method (r2 = 0.965; n = 72).  相似文献

A sequential injection method (SIA) for carbon speciation in inland bathing waters was developed comprising, in a single manifold, the determination of dissolved inorganic carbon (DIC), free dissolved carbon dioxide (CO2), total carbon (TC), dissolved organic carbon and alkalinity. The determination of DIC, CO2 and TC was based on colour change of bromothymol blue (660 nm) after CO2 diffusion through a hydrophobic membrane placed in a gas diffusion unit (GDU). For the DIC determination, an in-line acidification prior to the GDU was performed and, for the TC determination, an in-line UV photo-oxidation of the sample prior to GDU ensured the conversion of all carbon forms into CO2. Dissolved organic carbon (DOC) was determined by subtracting the obtained DIC value from the TC obtained value. The determination of alkalinity was based on the spectrophotometric measurement of bromocresol green colour change (611 nm) after reaction with acetic acid. The developed SIA method enabled the determination of DIC (0.24–3.5 mg C L−1), CO2 (1.0–10 mg C L−1), TC (0.50–4.0 mg C L−1) and alkalinity (1.2–4.7 mg C L−1 and 4.7–19 mg C L−1) with limits of detection of: 9.5 μg C L−1, 20 μg C L−1, 0.21 mg C L−1, 0.32 mg C L−1, respectively. The SIA system was effectively applied to inland bathing waters and the results showed good agreement with reference procedures.  相似文献

A novel method was developed for the analysis of four β-blockers, namely sotalol, carteolol, bisoprolol, and propranolol, in human urine by coupling carrier-mediated liquid phase microextraction (CM-LPME) to high performance liquid chromatography (HPLC). By adding an appropriate carrier in organic phase, simultaneous extraction and enrichment of hydrophilic (sotalol, carteolol, and bisoprolol) and hydrophobic (propranolol) drugs were achieved. High enrichment factors were obtained by optimizing the compositions of the organic phase, the acceptor solution, the donor solution, the stirring rate, and the extraction time. The linear ranges were from 0.05 to 10.0 mg L−1 for sotalol and carteolol, and from 0.05 to 8.0 mg L−1 for bisoprolol and propranolol. The limits of detection (S/N = 3) were 0.01 mg L−1 for sotalol, carteolol, and bisoprolol, and 0.005 mg L−1 for propranolol. The relative standard deviations were lower than 6%. The developed method exhibited high analyte preconcentration and excellent sample clean-up effects with little solvent consumption and was found to be sensitive and suitable for simultaneous determination of the above four drugs spiked in human urine. Furthermore, the successful analysis of propranolol in real urine specimens revealed that the determination of β-blockers in human urine is feasible using the present method.  相似文献

A flow injection (FI) method with flame atomic absorption spectrometry (FAAS) detection was developed for the determination and speciation of nitrite and nitrate in foodstuffs and wastewaters. The method is based on the oxidation of nitrite to nitrate using a manganese(IV) dioxide oxidant microcolumn where the flow of the sample through the microcolumn reduces the MnO2 solid phase reagent to Mn(II), which is measured by FAAS. The absorbance of Mn(II) are proportional to the concentration of nitrite in the samples. The injected sample volume was 400 μL with a sampling rate of analyses was 90 h−1 with a relative standard deviation better than 1.0% in a repeatability study. Nitrate is reduced to nitrite in proposed FI-FAAS system using a copperized cadmium microcolumn and analyzed as nitrite. The calibration curves were linear up to 20 mg L−1 and 30 mg L−1 with a detection limit of 0.07 mg L−1 and 0.14 mg L−1 for nitrite and nitrate, respectively. The results exhibit no interference from the presence of large amounts of ions. The method was successfully applied to the speciation of nitrite and nitrate in spiked natural water, wastewater and foodstuff samples. The precision and accuracy of the proposed method were comparable to those of the reference spectrophotometric method.  相似文献
