Selective extraction of low molecular weight (LMW) proteins and peptides from complex biological samples plays an important role in the discovery of useful biomarkers and signaling molecules. Various methods, such as solid-phase extraction (SPE), ultrafiltration, and size-exclusion chromatography have been developed for such extraction purpose. In this study, we present, to our knowledge, the first demonstration of alkyl-diol@SiO2 mesoporous material MCM-41 (alkyl-diol group on the external surface of mesoporous material) for selective extraction of LMW proteins and peptides from complex biological samples. The adsorption kinetics of LMW proteins, the influence of pH on adsorption and the desorption recovery by different elution solvents were investigated by using standard proteins as model samples. It was demonstrated that the modification of alkyl-diol group on the external surface could efficiently decrease the adsorption of HMW protein and increase the desorption recovery of LMW protein. Furthermore, the mesoporous materials were applied to selectively extract LMW proteins and peptides (<10 kDa) from crude human plasma. And the modified MCM-41 material had much better extraction selectivity and efficiency for LMW proteins and peptides than unmodified one.  相似文献

This study aimed to evaluate the heavy metal adsorptivity of calcium-alginate-modified diethylenetriamine-silica gel (CaAD) and incorporate this biosorbent into a flow analytical system for heavy metal ions using flame atomic absorption spectrometry (FAAS). The biosorbent was synthesized by electrostatically coating calcium alginate onto diethylenetriamine (dien)-silica gel. Copper ion adsorption tests by a batch method showed that CaAD exhibited a higher adsorption rate compared with other biosorbents despite its low maximum adsorption capacity. Next, CaAD was packed into a 1 mL microcolumn, which was connected to a flow analytical system equipped with an FAAS instrument. The flow system quantitatively adsorbed heavy metals and enriched their concentrations. This quantitative adsorption was achieved for pH 3–4 solutions containing 1.0 × 10−6 M of heavy metal ions at a flow rate of 5.0 mL min−1. Furthermore, the metal ions were successfully desorbed from CaAD at low nitric acid concentrations (0.05–0.15 M) than from the polyaminecarboxylic acid chelating resin (Chelex 100). Therefore, CaAD may be considered as a biosorbent that quickly adsorbs and easily desorbs analyte metal ions. In addition, the flow system enhanced the concentrations of heavy metals such as Cu2+, Zn2+, and Pb2+ by 50-fold. This new enrichment system successfully performed the separation and determination of Cu2+ (5.0 × 10−8 M) and Zn2+ (5.7 × 10−8 M) in a river water sample and Pb2+ (3.8 × 10−9 M) in a ground water sample.  相似文献

A novel dual-function material was synthesized by anchoring a molecularly imprinted polymer (MIP) layer on CdTe/ZnS quantum dots (QDs) using a sol–gel with surface imprinting. The material exhibited highly selective and sensitive determination of ractopamine (RAC) through spectrofluorometry and solid-phase extraction (SPE) coupled with high performance liquid chromatography (HPLC). A series of adsorption experiments revealed that the material showed high selectivity, good adsorption capacity and a fast mass transfer rate. Fluorescence from the MIP-coated QDs was more strongly quenched by RAC than that of the non-imprinted polymer, which indicated that the MIP-coated QDs acted as a fluorescence sensing material could recognize RAC. In addition, the MIP-coated QDs as a sorbent was also shown to be promising for SPE coupled with HPLC for the determination of trace RAC in feeding stuffs and pork samples. Under optimal conditions, the spectrofluorometry and SPE-HPLC methods using the MIP-coated QDs had linear ranges of 5.00 × 10−10–3.55 × 10−7 and 1.50 × 10−10–8.90 × 10−8 mol L−1, respectively, with limits of detection of 1.47 × 10−10 and 8.30 × 10−11 mol L−1, the relative standard deviations for six repeat experiments of RAC (2.90 × 10−9 mol L−1) were below 2.83% and 7.11%.  相似文献

Experimental determination and modeling of IgG binding on a new protein A adsorbent based on a macroporous resin were performed. The new adsorbent consists of polymeric beads based on hydrophilic acrylamido and vinyl monomers with a pore structure optimized to allow favorable interactions of IgG with recombinant protein A coupled to the resin. The particles have average diameter of 57 μm and a narrow particle size distribution. The IgG adsorption equilibrium capacity is 46 mg/cm3 and the effective pore diffusivity determined from pulse response experiments for non-binding conditions is 8.0 × 10−8 cm2/s. The IgG adsorption kinetics can be described with the same effective diffusivity by taking into account a heterogeneous binding mechanism with fast binding sites, for which adsorption is completely diffusion controlled, and slow binding sites for which adsorption is controlled by the binding kinetics. As a result of this mechanism, the breakthrough curve exhibits a tailing behavior, which appears to be associated with the slow binding sites. A detailed rate model taking into account intraparticle diffusion and binding kinetics is developed and is found capable of predicting both batch adsorption and breakthrough behavior over an ample range of experimental conditions. The corresponding effective diffusivity is independent of protein concentration in solution over the range 0.2–2 mg/cm3 and of protein binding as a result of the large pore size of the support matrix. Overall, the small particle size and low diffusional hindrance allow capture of IgG with short residence times while attaining substantial dynamic binding capacities.  相似文献

Cross-linked chitosan was chemically modified with di-2-propanolamine via an arm of chloromethyloxirane (CCTS-DPA resin). The adsorption behavior of the resin towards 62 elements was examined using a mini-column pretreatment method, and the collected elements were eluted with 1 mol L− 1 nitric acid before measurement by inductively coupled plasma-mass spectrometry (ICP-MS).The CCTS-DPA resin can adsorb several metal cations and several oxoanionic elements at appropriate pH. However, di-2-propanolamine (DPA) attached to cross-linked chitosan (CCTS) showed excellent ability and selectivity for the adsorption of germanium at pH 6 to 9. The adsorption capacity of the resin for germanium (IV) was found to be 106 mg g− 1 resin, whereas the adsorption rate constant was 9.82 × 10− 2 min− 1. Through the column treatment, alkali and alkaline earth matrices in river water and seawater matrices could be completely removed. The resin can also successfully remove chloride and selenium that can interfere with the direct determination of germanium by ICP-MS. The applicability of the CCTS-DPA resin was further demonstrated for the collection/preconcentration of germanium in environmental water samples and its determination by ICP-MS. The concentrations of germanium in tap water, river water and seawater samples were found in the range of 0.011 to 0.022 μg L− 1.  相似文献

A novel chromatographic medium for high-capacity protein adsorption was fabricated by grafting dextran (40 kDa) onto the pore surfaces of superporous agarose (SA) beads. The bead was denoted as D-SA. D-SA, SA and homogeneous agarose (HA) beads were modified with sulfopropyl (SP) group to prepare cation exchangers, and the adsorption and uptake of lysozyme on all three cation-exchange chromatographic beads (SP-HA, SP-SA and SP-D-SA) were investigated at salt concentrations of 6–50 mmol/L. Static adsorption experiments showed that the adsorption capacity of SP-D-SA (2.24 mmol/g) was 78% higher than that of SP-SA (1.26 mmol/g) and 54% higher than that of SP-HA (1.45 mmol/g) at a salt concentration of 6 mmol/L. Moreover, salt concentration had less influence on the adsorption capacity and dissociation constant of SP-D-SA than it did on SP-HA, suggesting that dextran-grafted superporous bead is a more potent architecture for chromatographic beads. In the dynamic uptake of lysozyme to the three cation-exchange beads, the De/D0 (the ratio of effective pore diffusivity to free solution diffusivity) values of 1.6–2.0 were obtained in SA-D-SA, indicating that effective pore diffusivities of SP-D-SA were about two times higher than free solution diffusivity for lysozyme. At 6 mmol/L NaCl, the De value in SA-D-SA (22.0 × 10−11 m2/s) was 14.4-fold greater than that in SP-HA. Due to the superior uptake kinetics in SA-D-SA, the highest dynamic binding capacity (DBC) and adsorption efficiency (the ratio of DBC to static adsorption capacity) was likewise found in SP-D-SA. It is thus confirmed that SP-D-SA has combined the advantages of superporous matrix structure and drafted ligand chemistry in mass transport and offers a new opportunity for the development of high-performance protein chromatography.  相似文献

The use of asymmetrical flow field-flow fractionation (AsFlFFF) in the study of heat-induced aggregation of proteins is demonstrated with bovine serum albumin (BSA) as a model analyte. The hydrodynamic diameter (dh), the molar mass of heat-induced aggregates, and the radius of gyration (Rg) were calculated in order to get more detailed understanding of the conformational changes of BSA upon heating. The hydrodynamic diameter of native BSA at ambient temperature was ∼7 nm. The particle size was relatively stable up to 60 °C; above 63 °C, however, BSA underwent aggregation (growth of hydrodynamic diameter). The hydrodynamic diameters of the aggregated particles, heated to 80 °C, ranged from 15 to 149 nm depending on the BSA concentration, duration of incubation, and the ionic strength of the solvent. Heating of BSA in the presence of sodium dodecyl sulfate (1.7 or 17 mM) did not lead to aggregation. The heat-induced aggregates were characterized in terms of their molar mass and particle size together with their respective distributions with a hyphenated technique consisting of an asymmetrical field-flow fractionation device and a multi-angle light scattering detector and a UV-detector. The carrier solution comprised 8.5 mM phosphate and 150 mM sodium chloride at pH 7.4. The weight-average molar mass (Mw) of native BSA at ambient temperature is 6.6 × 104 g mol−1. Incubation of solutions with BSA concentrations of 1.0 and 2.5 mg mL−1 at 80 °C for 1 h resulted in aggregates with Mw 1.2 × 106 and 1.9 × 106 g mol−1, respectively. The average radius of gyration and the average hydrodynamic radius of the heat-induced aggregate samples were calculated and compared to the values obtained from the size distributions measured by AsFlFFF. For comparison static light scattering measurements were carried out and the corresponding average molar mass distributions of solutions with BSA concentrations of 1.0 and 2.5 mg mL−1 at 80 °C for 1 h gave aggregates with Mw 1.7 × 106 and 3.5 × 106 g mol−1, respectively.  相似文献

We determine the association constants for ligand–protein complex formation using the flow injection method. We carry out the measurements at high flow rates (F = 1 mL min−1) of a carrier phase. Therefore, determination of the association constant takes only a few minutes. Injection of 1 nM of the ligand (10 μL of 1 μM concentration of the ligand solution) is sufficient for a single measurement. This method is tested and verified for a number of complexes of selected drugs (cefaclor, etodolac, sulindac) with albumin (BSA). We obtain K = 4.45 × 103 M−1 for cefaclor, K = 1.00 × 105 M−1 for etodolac and K = 1.03 × 105 M−1 for sulindac in agreement with the literature data. We also determine the association constants of 20 newly synthesized 3β- and 3α-aminotropane derivatives with potential antipsychotic activity – ligands of 5-HT1A, 5-HT2A and D2 receptors with the albumin. Results of the studies reported here indicate that potential antipsychotic drugs bind weakly to the transporter protein (BSA) with ≈ 102–103 M−1. Our method allows measuring K in a wide range of values (102–109 M−1). This range depends only on the solubility of the ligand and sensitivity of the detector.  相似文献

A fluorometric method for quantity analysis of biothiols was developed using a graphene oxide (GO)-based “molecular beacon”-like probe, which consisted of FITC labeled thymine (T)-rich single-stranded DNA (ssDNA), GO and Hg2+ ions. The labeled ssDNA containing T–T mismatches would self-hybridize to duplex in the presence of Hg2+, which can avoid its adsorption on GO and the fluorescence of this GO-based probe was recovered. The fluorescence of the probe quenched after the addition of biothiols such as glutathione (GSH) and cysteine (Cys) owing to thiol groups can selectively competitive ligation of Hg2+ ions with T–T mismatches. In the present work, the GO-based probe was used for the determination of GSH and Cys. Under the optimal conditions, a linear correlation was established between fluorescence intensity ratio I0/I and the concentration of GSH in the range of 2.0 × 10−9–5.0 × 10−7 mol L−1 with a detection limit of 1.0 × 10−9 mol L−1. The linear range for Cys is from 5.0 × 10−9 to 4.5 × 10−7 mol L−1 with a detection limit of 2.0 × 10−9 mol L−1. The proposed method was applied to the determination of GSH in human serum and cell extract samples with satisfactory results.  相似文献

Serum low-molecular weight (LMW) proteins potentially contain useful biological information and their identification can be used to discover novel potential biomarkers. Given the high complexity of serum samples, in the last years several different prefractionation and enrichment strategies have been developed. In this study three different methods, i.e. hydrogel nanoparticles, Proteominer® peptide ligand affinity beads and Sartorius Vivaspin® centrifugal ultrafiltration device, were compared and evaluated in order to select the best strategy for the enrichment and prefractionation of LMW proteins. A shotgun proteomics approach was adopted, with in-solution proteolytic digestion of the whole protein mixture and determination of the resulting peptides by nanoHPLC coupled with a high-resolution Orbitrap LTQ-XL mass spectrometer. Data analysis, focusing on the LMW proteome (MW ≤ 40 kDa), has shown that the hydrogel nanoparticles performed better in enriching the LMW protein profiles, with 115 proteins identified against 93 and 95 for Proteominer® beads and Sartorius Vivaspin® device, respectively.  相似文献

A novel flow injection chemiluminescence (FI-CL) sensor for determination of sulfadiazine (SDZ) using core–shell magnetic molecularly imprinted polymers (MMIPs) as recognition element is developed. Briefly, a hydrophilic MMIPs layer was produced at the surface of Fe3O4@SiO2 magnetic nanoparticles (MNPs) via combination of molecular imprinting and reversible stimuli responsive hydrogel. And it provided the MMIPs with excellent adsorption capacity and rapid adsorption rate due to the imprinted sites mostly situated on the surface of MMIPs. Then the prepared SDZ-MMIPs were packed into flow cell to establish a novel FI-CL sensor. The sensor provided a wide linear range for SDZ of 4.0 × 10−7 to 1.0 × 10−4 mol L−1 with a detection limit of 1.54 × 10−7 mol L−1. And the relative standard deviation (RSD) for the determination of 1.0 × 10−6 mol L−1 SDZ was 2.56% (n = 11). The proposed method was applied to determine SDZ in urine samples and satisfactory results were obtained.  相似文献

Metallothioneins (MTs) belong to cysteine-rich proteins with unique higher structure. One of the most known MT's functions is metals detoxification and maintaining their homeostasis in a cell. Structure of MT with naturally occurred zinc(II) ions can be affected by concentration of metal ions as well as redox milieu inside a cell, however the exact explanation and biochemical effects of the structural changes are still missing. In this study we used capillary electrophoresis on chip coupled with fluorescence detection to determine structural changes of MT with increasing concentration of zinc(II) ions and under various redox conditions. To investigate the structural-dependent effects, reduced and/or oxidized apo-MT (MT without natural occurred metal ion) was prepared. Zinc binding into reduced and/or oxidized apo-MT was compared. MT was incubated with 0, 5, 15, 25, 50 and 100 μM ZnCl2 for 1 h in 37 °C. Formation of MT aggregates with increasing zinc concentration was observed by spectrophotometry, chip capillary electrophoresis, and SDS-PAGE. We found out that reduced MT forms aggregates more readily compared to oxidized MT. Using the chip capillary electrophoresis allowed us relative quantification of MT aggregation as a decrease in the area of the signal corresponding to the monomer form of MT (Mw 15 kDa, migration time 26.5 s) and its ratio to total signal (sum of all signals measured by the electrophoresis). The dependences had an exponential character with equation y = 2.4 × e−0.01x, R2 = 0.945 for 15 kDa peak area and y = 0.11 × e−0.01x, R2 = 0.938 for decrease of 15 kDa peak area ratio to the total signal. Zn–MT interaction was 30% faster during the first 15 min and 50% faster during the whole experiment for reduced MT. It can be concluded that formation of MT aggregates is dependent on redox state and Zn(II) concentration.  相似文献

Total metal concentrations were determined in the serum of 12 continuous ambulatory peritoneal dialysis (CAPD) patients and in fresh and spent CAPD fluids by electrothermal and flame atomic absorption spectrometry (ETAAS, FAAS). Concentrations of Cu in serum of CAPD patients ranged from 720 to 1780 ng cm−3, Rb from 128 to 346 ng cm−3, Al from 10 to 72 ng cm−3, Fe from 800 to 2300 ng cm−3 and Zn from 659 to 1310 ng cm−3. The accuracy of the analytical procedure was checked by the analysis of the reference material Seronom™, Trace Elements in Serum. Good agreement between the certified and determined values was obtained for Al, Cu, Fe and Zn. The data on the total metal concentrations in CAPD fluids indicated that during CAPD fluid exchange the losses of Cu from 5.0 to 35 ng cm−3, of Rb from 50 to 110 ng cm−3 and of Al from 3.0 to 14.0 ng cm−3 occurred through the peritoneal membrane. Although fresh CAPD fluids contained traces of Fe (3.0-5.0 ng cm−3), the transfer of this element took place through the peritoneal membrane into spent CAPD fluid (13.0-38.0 ng cm−3). Zn concentrations were in general lower in spent (20.0-80 ng cm−3) than in fresh CAPD fluids (∼100 ng cm−3). To follow the mechanisms of the transfer of trace elements through the peritoneal membrane of CAPD patients, fractionation of metals was carried out in spent CAPD fluids by size exclusion chromatography with UV and AAS detection, applying Superdex HR 10/30 column. The chromatographic run was followed at 278 nm and separated metal species also determined ‘off line’ in 1 cm3 fractions by ETAAS or FAAS. From the UV chromatograms and AAS analysis of trace elements in the separated fractions it was demonstrated that Cu, Al, Fe and Zn were bound to proteins and only partially to low molecular weight (LMW) species, while Rb was associated exclusively with LMW species. For characterisation of the high molecular weight (HMW) binding proteins, fractions containing trace elements were subjected to SDS-PAGE electrophoresis. Al and Fe were presumably bound to transferrin, but due to its low concentration in spent CAPD fluids, it was not possible to confirm its presence in the separated fractions. About 10% of Al and 15% of Fe corresponded to LMW species. A fraction of HMW proteins of Cu in spent CAPD fluids was most probably bound to albumin and Zn to albumin and globulins. About 50% of Cu and Zn existed in LMW proteins, while Zn was also found partially in ionic form.  相似文献

The microphase adsorption-spectral correction (MPASC) technique was described and applied to the study of the interactions of fluorinated surfactants such as potassium perfluorooctanesulfonate (PFOS) and potassium perfluorobutanesulfonate (PFBS) with human serum albumin (HSA). Sodium octanesulfonate (SOS) was also studied as non-fluorinated surfactant. The aggregation of PFOS, PFBS and SOS obeys the Langmuir monolayer adsorption. The results show that the adsorption ratios of surfactants to HSA are PFOS:HSA = 120:1, PFBS:HSA = 205:1 and SOS:HSA = 18:1. The adsorption constants are KPFOS-HSA = 5.01 × 103, KPFBS-HSA = 9.79 × 102 and KSOS-HSA = 4.03 × 103. The detection limits are 2.7 mg/L for BSA using PFOS, 3.1 mg/L using PFBS and 3.1 mg/L using SOS. It was found that fluorinated surfactant exhibited stronger interaction with protein than hydrogenated one, and fluorinated surfactant with long hydrophobic chain exhibited stronger interaction with protein than that with short hydrophobic chain.  相似文献

A novel sulfopropyl gigaporous polystyrene (SP-GP) microsphere enhancing the separation of poly(ethylene glycol)–protein (PEGylated protein) was first presented. The SP-GP microspheres were successfully prepared by introducing sulfopropyl groups into agarose-coated gigaporous polystyrene microspheres and used as chromatography media. Compared with a commercial medium, SP-GP microspheres exhibited improved column efficiency and reduced backpressure with increasing flow velocity, which could ensure its use in high-speed chromatography. Furthermore, a higher protein recovery and purity of the PEGylated protein could be obtained, even when SP-GP was applied at a flow velocity of 1224 cm h−1. Additionally, the dynamic binding capacity (DBC) of SP-GP was significantly improved, which was higher than 10 mg mL−1 medium even at a flow velocity of 306 cm h−1. Further investigation using a laser scanning confocal microscope (LSCM) demonstrated that the static adsorption equilibrium of the PEGylated protein on SP-GP could be completed in 5 min, whereas a much longer period (ca. 60 min) was required for the commercial medium, indicating that the mass transfer of SP-GP was much faster with the gigaporous structure. All of these results strongly support that our developed SP-GP could serve as a promising cation exchange chromatography resin for high-speed separation, especially for biomolecules of high molecular weight.  相似文献

A flow injection system with anion exchange resin minicolumns was coupled with dynamic reaction cell (DRC™) ICP-MS for the determination and speciation of selenite and selenate at sub μg L−1 levels. The charged selenate and uncharged selenite were separated on the first resin column in which only selenate was retained. The unretained selenite was then deprotonated with alkaline solution, and the resulting anionic selenite species was collected on the second column serially connected downstream. By setting a sample loop, total selenium can be determined together with selenite and selenate. The selenium species was eluted by nitric acid and carried to DRC™ ICP-MS for their detection. Using ammonia as reaction gas, the detection of 78Se was improved. The enrichment factor was 20 for 10 mL of sample. The standard deviations (n = 5) of peak heights were 4.9%, 4.1%, and 7.0% for a 5.0 × 10−2 μg L−1 selenite and selenate, and total Se, respectively. The calibration graphs were linear from 2.0 × 10−2 to 1.0 μg L−1 selenite and selenate. And, the linearity for total selenium was good in the range of 10.0 × 10−2 to 1.0 μg L−1. The proposed method has been demonstrated for the application to natural and bottled drinking water samples.  相似文献

The dependence of Th recovery on hydrofluoric acid (HF) concentration in nitric acid (HNO3) solutions (1–5 mol/dm3) containing 1 × 10−6 mol/dm3 of Th and various concentrations of HF and the elution behavior were studied using a commercially available UTEVA (for uranium and tetravalent actinide) resin column. Thorium recovery decreased with an increase in HF concentration in the sample solutions. The concentration of HF at which Th recovery started to decrease was ∼1 × 10−4 mol/dm3 in 1 mol/dm3 HNO3 solution, ∼1 × 10−3 mol/dm3 in 3 mol/dm3 HNO3 solution, and ∼1 × 10−2 mol/dm3 in 5 mol/dm3 HNO3 solution. When Al(NO3)3 (0.2 mol/dm3) or Fe(NO3)3 (0.6 mol/dm3) was added as a masking agent for F to the Th solution containing 1 × 10−1 mol/dm3 HF and 1 mol/dm3 HNO3, Th recovery improved from 1.4 ± 0.3% to 95 ± 5% or 93 ± 3%. Effective extraction of Th using UTEVA resin was achieved by selecting the concentration of HNO3 and/or adding masking agents such as Al(NO3)3 according to the concentration of HF in the sample solution.  相似文献

A method for determination of nine brominated phenols as environmental risk compounds was developed by on-line coupled capillary isotachophoresis and capillary zone electrophoresis (ITP–CZE). For ITP step, 1 × 10−2 mol L−1 hydrochloric acid with 3 × 10−2 mol L−1 ammediol pH 9.1 was used as the leading electrolyte, and 3 × 10−2 mol L−1 β-alanine with 2 × 10−2 mol L−1 sodium hydroxide pH 10.05 was used as the terminating electrolyte. As the background electrolyte for CZE separation, 2.5 × 10−2 mol L−1 β-alanine with 2.5 × 10−2 mol L−1 lysine pH 9.6 was used. All electrolytes contained 0.05% or 0.1% (m/v) hydroxyethylcellulose to suppress the electroosmotic flow. UV detection at wavelength 220 nm was used. Detection limits in order of tens of nmol L−1 were achieved. Good repeatability of migration times (less than 0.33% RSD) and good repeatability of peak areas (less than 7.19% RSD) at concentration level 5 × 10−8 mol L−1 were observed. Developed ITP–CZE method was applied to determination of brominated phenols in spiked tap and river water samples.  相似文献

The suitability of the use of macroporous adsorbent Amberlite XAD7HP in expanded bed adsorption processes for the isolation of flavonoids from crude extracts of Ginkgo biloba L. has been assessed. The expansion and hydrodynamic properties of expanded beds were investigated and analyzed. The bed expansion as a function of operational fluid velocity was measured and correlated with the Richardson–Zaki equation. Theoretical predictions of the correlation parameters (the terminal settling velocity ut and exponent n) were improved by modifying equations in the literature. Residence time distributions (RTDs) were studied using acetone as a tracer. Three measures of liquid phase dispersion (the height equivalent of theoretical plate, Bodenstein number and axial distribution coefficient) were investigated and compared to values previously obtained with commercial EBA adsorbents developed for protein purification. A suitable bed expansion ratio was found to be 1.25 times the settled bed height, which occurred at a corresponding flow velocity of 183 cm/h. For an initial settled bed height of 42 cm, the mean residence time of liquid in the expanded bed was around 28 min. Under these flow conditions, the axial mixing coefficient Dax was 7.54 × 10−6 m2/s and the Bodenstein number was 28; the number of theoretical plates (N) was 19 and the height equivalent of a theoretical plate (HETP) was 2.77 cm. Rutin trihydrate was used as a model flavonoid for the characterization of the adsorption properties of Amberlite XAD7HP. Adsorption was observed to reach equilibrium within 3 h with 70% of the adsorption capacity being achieved within 30 min. The estimated maximum equilibrium adsorption capacity for rutin was estimated to be 43.0 mg/(g resin) when the results were fitted to Langmuir isotherms. The adsorption performance was not seriously impaired by the physical presence of G. biloba leaf powders. Assessment of the kinetics of the adsorption of rutin revealed that the rate constant for adsorption was only reduced by 15% in the presence of leaf powders at a concentration of 50 mg/mL. The results demonstrated that Amberlite XAD7HP should be suitable for expanded bed adsorption of flavonoids from crude extracts of G. biloba L.  相似文献
