比较丙酮/TCA沉淀法和直接裂解法,优化提取与分离褐云玛瑙螺(Achatina fulica, AF)肝脏全蛋白.采用丙酮/TCA沉淀法,可获得约600个蛋白质斑点.用0.5 mg/L CdCl2溶液浸泡后的去梗小白菜喂养AF,并作为镉盐诱导AF肝脏表达应激蛋白质的实验材料.采用蛋白质组学技术筛选出由镉盐诱导AF肝脏表达的14个差异蛋白质斑点,并用肽质量指纹图谱技术(peptide mass fingerprinting, PMF)和数据库比对法初步鉴定出7种差异蛋白质,其中部分为热激蛋白(heat shock protein)、甲基转移酶(Methyltransferase)、三磷酸腺苷结合盒子转运体(ABC transporter)、钼酸盐转运子亚基(molybdate transporter subunit)和磷酸甘油酸变位酶(phosphoglycerate mutase).差异蛋白质均参与镉盐代谢,并适合作为监测土壤或食物中镉污染程度及危害性的指示蛋白质. 相似文献

采用电感耦合等离子体原子发射光谱(ICP-AES)法对云当归以及土壤中11种微量元素进行了分析.结果显示,土壤中微量元素的含量依次为Zn>Cu>Pb>Fe>Mg>Mn>Ni>Cr> Cd> As> Hg;云当归中Mg、Fe、Mn、Zn中含量较高,而As、Cd、Cu、Pb、Hg等元素中含量相对较低,均低于药典标准.土壤中As与Cr、Pb与Zn存在极显著正相关(r=0.785,0.808);Mn与Cr存在极显著负相关(r=-0.733),As与Cu存在显著负相关(r=-0.631).云当归中Fe与Ni极显著正相关(r=0.731),Cd与Cu极显著负相关(r=-0.714);Zn与Hg显著相关(r=0.655),Ni与As、Fe与Pb、Fe与Cd显著负相关(r=-0.658,-0.637,-0.673).云当归与土壤中的As存在显著相关(r=0.707). 相似文献

Summary An emission spectroscopic method was developed for the simultaneous determination of the trace elements Ag, Al, Cr, Cu, Fe, Ga, In, Mg, Mn, Ni, Pb and Ti in ZnSe samples. The sample to be analysed (about 10 mg) is vaporized directly from the anode and excited by a rotating d.c. arc using a homogeneous magnetic field. The influence of elements with lower ionization potential on the plasma was investigated, too. These additives (NaCl, KCl) were introduced into the plasma by means of a wick electrode (cathode). The calibration is carried out on the basis of mixtures of oxides of the trace elements with spectroscopically pure ZnSe powder. Analytical detection limits of the trace elements investigated were in the region of ppm and sub-ppm corresponding to absolute amounts of 10 ng and lower. 相似文献

Introduction Geological resource survey demands for determining various constituents including major, minor, micro, trace and ultra-trace levels of elements for preparing the map of resource distribution of our country. As a powerful and popularly used technique for multi-element analysis, inductively coupled plasma(ICP) atomic emission spectrometry (AES) has been applied to this field for a period of time[1-3]. However, ICP spectrometric determination of those micro, trace and ultratrace elements needs enrichment procedures for improving the detection limit, which is unacceptable in case a great mass of samples should be analyzed as that in the task of geological resource survey. On the other hand, although ICP mass spectrometry(MS) is considered the most powerful method for trace elements determination[4,5], it is difficult for ICP-MS to be used to determine the trace and major analytes simultaneously in a spectrum. 相似文献

For property-related characterization of inorganic materials, information is needed about bulk composition, distribution of elements, compounds, phases and structural features. Photons, electrons, charged ions or neutrons, often used as focused beams provide access to this information. The major trends in this field are the development or improvement of methods to obtain new information or to increase spatial resolution, detection power, precision and accuracy of analysis. The present state of the analysis of inorganic materials by using beam techniques is discussed in a selective manner. Emphasis is placed on the following problems, which are important for basic research and for development of high-technology materials: ultratrace bulk analysis in the pg g?1 to ng g?1 range; analysis for phases, trace elements and isotopes, including structural characterization; and surface analysis, with emphasis on the characterization of trace elements at surfaces, quantitative distribution of trace elements in heterogeneous structures, and surface structural analysis with atomic resolution. 相似文献

This paper reports on a novel procedure for determining trace element abundances (REE and Y, Cr, Mn, Co) in seawater by inductively coupled plasma sector field mass spectrometry (ICP-SFMS). The procedure uses a combination of pre-concentration using co-precipitation onto magnesium hydroxides and addition of thulium spike. The validity of the method was assessed onto 25 ml volumes of certified reference materials (NASS- and CASS-4) and in house seawater standard. Procedural blanks were determined by applying the same procedure to aliquots of seawater previously depleted in trace elements by successive Mg(OH)2 co-precipitations, yielding estimated contributions to the studied samples better than 1.1% for all elements, with the exception of Cr (<3.3%) and Co (up to 8%). The reproducibility of the method over the six month duration of the study was smaller than 11% RSD for all the studied elements. Results obtained for NASS-5 and CASS-4 agree well with published working values for trace elements. 相似文献
