A general method was developed for the synthesis of 1,6-disubstituted 4-methyl-5-cyano-7-azalndolines from the readily available ammonium salt of 2,6-dihydroxy-3-(-hydroxyethyl)-4-methyl-5-cyanopyridine through the corresponding N-substituted ammonium salts and N-substituted 2-amino-3-(-hydroxyethyl)-4-methyl-5-cyano-6-hydroxypyridines with treatment of the latter by POCl3. This method gives a 40% yield of 4-methyl-5-cyano-7-azaindoline compounds containing various aralkyl or alkyl substituents at N-1 and a hydroxy group or halogen atom at C-6 in three steps.For Communication 65, see [1].Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 1, pp. 84–88, January, 1985.  相似文献

The reaction of esters of amino acids with 4-amino-6-chloro-5-nitropyrimidine has yielded esters of (4-amino-5-nitro-6-pyrimidyl)amino acids. The reduction of the esters of -(4-amino-5-nitro-6-pyrimidylamino) acids has yielded 4-amino-6-hydroxy-7,8-dihydropteridines. -(4-Amino-5-nitro-6-pyrimidylamino) acids and the ester of N-(4-amino-5-nitro-6-pyrimidyl)--alanine do not cyclize and on reduction give the corresponding diaminopyrimidine derivatives.  相似文献

10-Methyl-2-chloro- and 2-chloro-3-methyl-3,4-diazaphenoxazines as well as 2-oxo-3,10-dimethyl-2,3-dihydro-3, 4-diazaphenoxazine were obtained by methylation of 2-chloro-3,4-diazaphenoxazines. Depending on the conditions, alkylation of 2-chloro-3,4-diazaphenoxazine with -diethylaminoethyl chloride gave 1-ethylimidazolino[1,21,2]pyridazo[3,4-b]-1,4-benzoxazine or 2-ethoxy-10-(-diethylaminoethyl)-3,4-diazaphenoxazine. The reaction of 5-amino-4-chloro- and -4-methoxy-6~mercaptopyrimidines with 4-methoxy-, 4-amino-, and 2,4-diethoxyphenacyl chlorides has yielded 5-amino-4-chloro- and -4-methoxy-6-phenacylthiopyrimidines. The reaction of 2, 5-diamino-4-methyl-6-mercaptopyrimidine with 4-methoxyphenacyl chloride has yielded 2-amino-6-hydroxy-6-(4-methoxyphenyl)-4-methyl-5, 6-dihydropyrimido[4, 5-b]-1,4-thiazine, and that with 4-aminophenacyl chloride has yielded the corresponding 7H-pyrimido[4, 5-b]-1,4-thiazine.For part IV, see [1].  相似文献

The reaction of the acetylacetonyl radical (generated from acetylacetone by the action of manganese acetate) with vinylacetylene, 2-vinylfuran, and butadiene Was studied. The reaction proceeds nonregioselectively with vinylacetylene: 3-acetyl-2-methyl-5-ethynyl- and 3-acetyl-2-methyl-5-(4-acetyl-5-methyl-2-furyl)-4,5-dihydrofurans in a ratio of 12.2 were isolated. The reaction of 2,4-dichloro-5-(-chloroethyl)pyridine with ammonia and N-ethylaniline, leading to the formation of 5-azaindoline derivatives, has been studied. Two major compounds are formed in the reaction of 3,5-diamino-4-nitropyrazole with acetoacetic ester: 2-amino-5-methyl-3-nitro-4,7-dihydropyrazolo[1,5-a]pyrimidin-7-one and 3-amino-5-(-hydroxycrotonyl)amino-4-nitropyrazole. On the basis of a comparative study of the influence of various factors (ratio of the reactants, temperature, reaction time, polarity of the medium, and the catalytic action of metals) on the reaction of 2,6-dichloro-3-(-chloroethyl)-4-methylpyridine (trichlorocollidine) with 2,6-dimethylamine, it has been shown that, in addition to 6-chloro-1,4-dimethyl-7-azaindoline, the process gives rise to various amounts, depending on the conditions of its performance, of other products: 3-(-dimethylaminoethyl)-2,6-dichloro-4-methylpyridine, 2-chloro-3-(-chloroethyl)-6-dimethylamino-4-methylpyridine, 2-chloro-6-dimethylamino-3-(-dimethylaminoethyl)-4-methylpyridine, and 6-dimethylamino-1,4-dimethyl-7-azaindoline. A study is made of new synthetic routes, based on accessible 3-formyl-7-azaindoles, to 3-substituted 7-azaindole. 2-Phenyl-4-(1-phenyl-4-methyl-7-azaindolyl-3-rnethylene)-1, 3-oxazol-5-one is synthesized, and it is converted to 1-phenyl-4-methyl-7-azatryptophane, 1-phenyl-4-methyl-7-azaindolyl-3-acetic acid,1-phenyl-3-(,-dihydroxypropyl)-4-methyl-7-azaindole, and 1-phenyl-4-methyl-7-azaindolyl-3-pyrotartaric acid.For Part XVIII see [1].  相似文献

A number of 7H-pyrimido[4, 5-b]thiazines are obtained by reacting 5-amino-6-mercaptopyrimidines with -halogeno ketones. It is established that the reaction takes place in three stages: formation of 5-amino-6-[-ketoalkyl(aralkyl)-mercapto]pyrimidines, cyclization of these to 6-hydroxy-5, 6-dihydropyrimido[4, 5-b][1, 4]thiazines, and dehydration of the latter to 7H-pyrimido[4, 5-b][1, 4]thiazines.For Part III see [5].  相似文献

Treatment of the ammonium (I) or benzylammonium salt of 2,6-dihydroxy-3-(-hydroxyethyl)-4-methyl-5-cyanopyridine (II) with a mixture of benzylamine and phosphorus pentoxide yielded 2-benzylamino-3-(-hydroxyethyl)-4-methyl-5-cyano-6-hydroxypyri-dine (III), which, when heated with phosphorus oxychloride, is converted to 1-benzyl-4-methyl-5-cyano-6-hydroxy-7-azaindoline (IV). The products of thermal fragmentation of II with benzylamine were studied by the method of chromato-mass spectrometry. In addition to compound III, N,N-dibenzylurea (V) and the dibenzylamide of malonic acid (VI) were preparatively isolated from the reaction products. The cyclization of I and II to 4-methyl-6-hydroxy-2,3-dihydro-7-azabenzofuran (VII) and 4-methyl-5-cyano-6-hydroxy-2,3-dihydro-7-azabenzofuran (VIII) was carried out. A new type of isatin analog-2,3-dioxo-2,3-dihydro-4-methyl-6-chloro-1H-pyrrolo[2,3-b]pyridine — was obtained by oxidation of 4-methyl-6-chloro-1H-pyrrolo(2,3-b]pyridine. Condensation of L-arabinose with 4-methyl-6-chloro-2,3-dihydropyrrolo[2,3-b]pyridine and subsequent acetylation and dehydrogenation gave 1-(2,3,4-tri-O-acetyl--L-arabinopyranosyl)-4-methyl-6-chloropyrrolo[2,3-b]pyridine, which served as the starting compound for the synthesis of 1--L-arabinopyranosyl-4-methyl-6-chloropyrrolo[2,3-b]pyridine. The kinetics of the reaction of 1-methyl-3-nitro-5-chloro-1,2,4-triazoles and 1-methyl-3-nitro-5-bromo-1,2,4-triazoles with hydroxide ions were studied. The reaction leads to replacement of the halogen or nitro group, and the ratio of the rates of substitution is 301. A new method for synthesizing 5- and 7-azaindoles is given, -Chlorobutyronitrile and malonyl chloride give 2, 4, 6-trichloro-3-(ß-chloroethyl) pyridine, which is cyclized with ammonia to 4, 6-dichloro-7-azaindoline and 4, 6-dichloro-5-azaindoline. 6-Chloro derivatives of 7-azaindolines are not dehydrogenated by chlorainil, but 2, 3-dichloro-S, 6-dicyanobenzoquinone converts them to 6-chloro-7-azaindoles. It is shown that sodium in liquid ammonia is an effective means of dehydrogenating the 5-azaindoline to 5-azaindole. In this case, dehydrogenation of 4, 6-dichloro-7-azaindoline is followed by dehalogenation.For Part IX see [1].  相似文献

(5) was converted into l-(-pyridyl)-5-[5-methyl-1,2,4-oxdiazolyl(3)]-6-azauracil (6).

2-Methyl-2,4,12,13H-pyrazolo[3,4-5,6]pyrimido[1,2-b]benzo[d,e]isoquinoline-4,12-dione and its 8-nitro- and 8-amino-substituted derivatives were obtained starting from 4-amino-3-carbamoyl-1-methylpyrazole and naphthalic and 4-nitronaphthalic anhydrides.
